Rapamycin Pretreatment Attenuates High Glucose-induced Alteration of Synaptic Transmission in Hippocampal Dentate Gyrus Neurons.

Diabetic neuropathy is one of the most common complications in patients with diabetes and leads to cognitive impairment. It is suggested that protracted hyperglycemia is the main trigger of cognitive deficits in diabetes and causes hippocampal abnormalities. Rapamycin, an inhibitor of mammalian target of rapamycin complex 1 (mTORC1), can significantly ameliorate cognitive deficits in neurodegenerative diseases, such as Alzheimer's disease, Parkinson's disease and Huntington disease. We employed whole-cell patch clamping to examine the effects of rapamycin on neuronal electrophysiological chara

High-frequency stimulation-induced synaptic potentiation in dorsal and ventral CA1 hippocampal synapses: the involvement of NMDA receptors, mGluR5, and (L-type) voltage-gated calcium channels.

The ability of the ventral hippocampus (VH) for long-lasting long-term potentiation (LTP) and the mechanisms underlying its lower ability for short-lasting LTP compared with the dorsal hippocampus (DH) are unknown. Using recordings of field excitatory postsynaptic potentials (EPSPs) from the CA1 field of adult rat hippocampal slices, we found that 200-Hz stimulation induced nondecremental LTP that was maintained for at least 7 h and was greater in the DH than in the VH. The interaction of NMDA receptors with L-type voltage-dependent calcium channels appeared to be more effective in the DH than

Inhibition of apamin-sensitive calcium dependent potassium channels facilitate the induction of long-term potentiation in the CA1 region of rat hippocampus in vitro.

Using field potential recording in the CA1-region of rat hippocampal slices we investigated the effect of apamin; a specific antagonist of small conductive calcium activated potassium channels on long-term potentiation (LTP). The experiments revealed that LTP of excitatory postsynaptic potentials induced by a single 100 Hz tetanization was intensified by extracellular application of apamin in a concentration range of 1-200 nM. No effects of apamin on LTP induced by triple 100 Hz tetanization were seen. We conclude that the positive modulation of LTP by apamin is effective in a nanomolar concen

Enhanced amygdala inhibitory neurotransmission and its vulnerability to hyperthermic stress in Atp1a2-deficient heterozygous mice.

The sodium pump (Na,K-ATPase, NKA) is a membrane-bound enzyme crucial for maintaining Na+/K+ electrochemical gradients across plasma membranes. NKA constitutes catalytic α and auxiliary β subunits, of which four α and three β isoforms have been identified. The physiological roles of the isoforms are not fully understood; nevertheless, mutations in the human α2 subunit gene ATP1A2 have been linked to various neurological disorders, including familial hemiplegic migraine type 2 (FHM2), alternating hemiplegia of childhood (AHC), and epilepsy syndromes, with symptoms typically triggered by physica

Mild chronic perturbation of inhibition severely alters hippocampal function.

Pentameric GABAA receptors mediate a large share of CNS inhibition. The γ2 subunit is a typical constituent. At least 11 mutations in the γ2 subunit cause human epilepsies, making the role of γ2-containing receptors in brain function of keen basic and translational interest. How small changes to inhibition may cause brain abnormalities, including seizure disorders, is unclear. In mice, we perturbed fast inhibition with a point mutation T272Y (T6'Y in the second membrane-spanning domain) to the γ2 subunit. The mutation imparts resistance to the GABAA receptor antagonist picrotoxin, allowing ver

Electrophysiological properties of the somatotopic organization of the vestibulospinal system in the frog.

In experiments on the preparation of a frog perfused brain (Rana ridibunda), field and intracellular potentials were recorded from neurons of the vestibular nuclear complex following stimulation of the ipsilateral vestibular nerve and different levels of the spinal cord. Stimulation of the vestibular nerve evoked mono- and polysynaptic excitatory postsynaptic potentials and orthodromic action potentials. In parallel, an antidromic activation of vestibular neurons sending their axons to the labyrinth was recorded. Vestibulospinal neurons sending their axons to the cervical (C neurons) and lumba

5-hydroxytryptamine 2C receptors tonically augment synaptic currents in the nucleus tractus solitarii.

The nucleus tractus solitarii (nTS) is the primary termination and integration point for visceral afferents in the brain stem. Afferent glutamate release and its efficacy on postsynaptic activity within this nucleus are modulated by additional neuromodulators and transmitters, including serotonin (5-HT) acting through its receptors. The 5-HT(2) receptors in the medulla modulate the cardiorespiratory system and autonomic reflexes, but the distribution of the 5-HT(2C) receptor and the role of these receptors during synaptic transmission in the nTS remain largely unknown. In the present study, we

Refractoriness of sural nerve in humans.

High-frequency stimulation of peripheral nerve bundles is frequently used in clinical tests and physiologic experiments to study presynaptic and postsynaptic effects. To understand the postsynaptic effects, it is important to ensure that each pulse in the train is equally effective in stimulating the presynaptic nerve bundle; however, the optimal interpulse interval (IPI) and the stimulus intensity at which each pulse is equally effective in stimulating the same number of axons are not known. The magnitude of the compound action potential produced by each pulse in a train was tested on the sur

Changes in the calcium dependence of glutamate transmission in the hippocampal CA1 region after brief hypoxia-hypoglycemia.

Using the model of hypoxia-hypoglycemia (HH) in rat brain slices, we asked whether glutamate transmission is altered following a brief HH episode. The HH challenge was conducted by exposing slices to a glucose-free medium aerated with 95% N2-5% CO2, for approximately 4 min, and glutamate transmission in the hippocampal CA1 region was monitored at different post HH times. In slices examined </=8 h post HH, CA1 synaptic field potentials are comparable in amplitude to controls, but are less sensitive to experimental manipulations designed to attenuate intracellular Ca2+ signals, as compared with

Fast and slow synaptic potentials produced in a mammalian sympathetic ganglion by colon distension.

Radial distension of the large intestine produced a slow depolarization in a population of neurons in the inferior mesenteric ganglion of the guinea pig. The slow potentials often occurred simultaneously with cholinergic fast potentials [( excitatory postsynaptic potentials (EPSPs]) yet persisted in the presence of nicotinic and muscarinic cholinergic antagonists when all fast EPSPs were absent. The amplitude of the distension-induced noncholinergic slow depolarization increased with increasing distension pressure. For distensions of 1-min duration at pressures of 10-20 cm of water, the mean d

Differential effects of phorbol ester on AMPA and NMDA components of excitatory postsynaptic currents in dentate neurons of rat hippocampal slices.

Protein kinase C (PKC) is present abundantly in the mammalian central nervous system, and is involved in a variety of neuronal functions. Phorbol esters mimic the role of diacylglycerol, the physiological activator of PKC. We examined effects of phorbol 12,13-diacetate (PDAc) on excitatory synaptic transmission in neurons in the dentate granule cell layer of rat hippocampal slices using the whole-cell patch clamp technique. Excitatory postsynaptic currents (EPSCs) evoked by stimulation of the perforant path (pp) consisted of AMPA and NMDA receptor-mediated components. The application of PDAc p

Argiotoxin-636 blocks effects of N-methyl-D-aspartate on lateral line of Xenopus laevis at concentrations which do not alter spontaneous or evoked neural activity.

Activity of primary afferent neurons in acousticolateralis organs can be modulated by excitatory amino acids (EAA) and their antagonists, and EAA are among the better candidates for the transmitter(s) between hair cells and afferent neurons. Argiotoxin-636 (ATX) is a spider venom toxin that is a selective antagonist of the N-methyl-D-aspartate (NMDA) subtype of excitatory amino acid receptors in vertebrates. In the present study, the effects of NMDA alone and in combination with ATX on resting firing rate (spontaneous activity) and mechanically stimulated activity of lateral line afferent neur

Control of locomotion in marine mollusc Clione limacina. X. Effects of acetylcholine antagonists.

The swimming central pattern generator (CPG) of the pteropod mollusc Clione limacina is located in the pedal ganglia. It consists of three groups of interneurons (7, 8, and 12) which generate the rhythmical activity and determine the temporal pattern of the motor output, that is, phasic relations between different groups of motor neurons supplying dorsal (group 1 and 3 motor neurons) and ventral (group 2 and 4 motor neurons) muscles of the wings. In this work peripheral and central effects of acetylcholine (ACh) antagonists on the swimming control in C. limacina has been studied. The ACh antag

Prolonged incubation with Δ9-tetrahydrocannabinol but not with cannabidiol induces synaptic alterations and mitochondrial impairment in immature and mature rat organotypic hippocampal slices.

Cannabis derivatives are among the most widely used psychoactive substances in the world, which leads to growing medical concerns regarding its chronic use and abuse especially among adolescents. Exposure to THC during formative years produces long-term behavioral alterations that share similarities with symptoms of psychiatric and neurodevelopmental disorders. In this study, we have analyzed the functional and molecular mechanisms that might underlie these alterations. Rat organotypic hippocampal slices were cultured for 2 days (immature) or 10 days (mature) in vitro and then exposed for 7 da

Bridging the gap between presynaptic hair cell function and neural sound encoding.

Neural diversity can expand the encoding capacity of a circuitry. A striking example of diverse structure and function is presented by the afferent synapses between inner hair cells (IHCs) and spiral ganglion neurons (SGNs) in the cochlea. Presynaptic active zones at the pillar IHC side activate at lower IHC potentials than those of the modiolar side that have more presynaptic Ca2+ channels. The postsynaptic SGNs differ in their spontaneous firing rates, sound thresholds, and operating ranges. While a causal relationship between synaptic heterogeneity and neural response diversity seems likely

Acute Cocaine Reduces Excitatory Synaptic Transmission in Pyramidal Neurons of the Mouse Medial Prefrontal Cortex.

The medial prefrontal cortex (mPFC) plays critical roles in the development of cocaine addiction. Numerous studies have reported about the effects of cocaine on neuronal and synaptic activities in the nucleus accumbens and ventral tegmental area, which are brain regions associated with cocaine addiction; however, a limited number of studies have reported the effect of cocaine on mPFC neuronal activity. In this study, using whole-cell patch-clamp recordings in brain slices, we present that under the condition where synaptic transmission is enhanced by increasing extracellular K+ concentration,

Electrophysiological properties and input-output organization of callosal neurons in cat association cortex.

Intracellular recordings from association cortical areas 5 and 7 were performed in cats under barbiturate or ketamine-xylazine anesthesia to investigate the activities of different classes of neurons involved in callosal pathways, which were electrophysiologically characterized by depolarizing current steps. Excitatory postsynaptic potentials (EPSPs), inhibitory postsynaptic potentials (IPSPs), and/or antidromic responses were elicited by stimulating homotopic sites in the contralateral cortical areas. Differential features of EPSPs related to latencies, amplitudes, and slopes were detected in

Disrupted prefrontal neuronal oscillations and morphology induced by sleep deprivation in young APP/PS1 transgenic AD mice.

Emerging evidence suggests that sleep deprivation (SD) is a public health epidemic and increase the risk of Alzheimer's disease (AD) progression. However, the underlying mechanisms remain to be fully investigated. In this study, we investigate the impact of 72 h SD on the prefrontal cortex (PFC) of 3∼4-months-old APP/PS1 transgenic AD mice - at an age before the onset of plaque formation and memory decline. Our results reveal that SD alters delta, theta and high-gamma oscillations in the PFC, accompanied by increased levels of excitatory postsynaptic signaling (NMDAR, GluR1, and CaMKII) in AD

Pre- and post-inspiratory neurons change their firing properties in female rats exposed to chronic intermittent hypoxia.

Obstructive sleep apnea patients face episodes of chronic intermittent hypoxia (CIH), which has been suggested as a causative factor for increased sympathetic activity (SNA) and hypertension. Female rats exposed to CIH develop hypertension and exhibit changes in respiratory-sympathetic coupling, marked by an increase in the inspiratory modulation of SNA. We tested the hypothesis that enhanced inspiratory-modulation of SNA is dependent on carotid bodies (CBs) and are associated with changes in respiratory network activity. For this, in CIH-female rats we evaluated the effect of CBs ablation on

Oligomeric amyloid beta associates with postsynaptic densities and correlates with excitatory synapse loss near senile plaques.

Synapse loss correlates with a cognitive decline in Alzheimer's disease (AD), but whether this is caused by fibrillar deposits known as senile plaques or soluble oligomeric forms of amyloid beta (Abeta) is controversial. By using array tomography, a technique that combines ultrathin sectioning of tissue with immunofluorescence, allowing precise quantification of small structures, such as synapses, we have tested the hypothesis that oligomeric Abeta surrounding plaques contributes to synapse loss in a mouse model of AD. We find that senile plaques are surrounded by a halo of oligomeric Abeta. A
