From Food Additives and Contaminants to the Human Exposome: A Precision Toxicology Framework for Next-Generation Food Safety.

Human exposure to foodborne xenobiotics, including environmental contaminants that enter the food chain, is no longer adequately represented by isolated chemicals. Individuals encounter complex and dynamic mixtures of food additives, pesticide residues, veterinary drugs, mycotoxins, processing contaminants, packaging migrants, environmental pollutants, and other dietary chemicals that interact with genetics, metabolism, the gut microbiome, nutritional status, and lifestyle throughout life. The dietary exposome therefore represents a critical interface linking food contaminants, internal chemic

Integrating genomic and exposomic data identifies endocrine disruptors potentially associated with chronic obstructive pulmonary disease.

BACKGROUND: Chronic obstructive pulmonary disease (COPD) is a complex disorder driven by both genetic susceptibility and environmental exposures. Endocrine-disrupting chemicals (EDCs) are widespread environmental contaminants that interfere with hormonal and immune pathways, yet their molecular links to COPD remain insufficiently defined. METHODS: We integrated exposomic and genomic data to systematically evaluate EDC-related molecular mechanisms in COPD. Chemical-gene interactions were curated from the TEDX and CTD to identify EDC-associated genes. Two-sample Mendelian randomization (MR) was

ExchangeXplorer: an R/Shiny application for processing and visualization of LC-HDX-MS data in metabolomics, lipidomics, and exposomics.

BACKGROUND: Reliable annotation remains a major challenge in untargeted LC-MS-based metabolomics, lipidomics, and exposomics. Liquid chromatography-hydrogen/deuterium exchange-mass spectrometry (LC-HDX-MS) provides orthogonal structural information by revealing the number of exchangeable hydrogens within a molecule, thereby supporting functional-group assignment, distinguishing isomeric structures, and reducing false-positive annotations. However, broader adoption of LC-HDX-MS for small-molecule analysis has been limited by the lack of dedicated software for systematic data processing and inte

Column switching liquid chromatography dual mass spectrometry system for simultaneous untargeted metabolomics and targeted exposomics.

Exposome-wide association studies (ExWAS) require the detection of metabolites and exposures with diverse chemical properties across wide concentration ranges, a task that typically demands multiple analytical methods. To address this challenge, we develop an integrated column-switching two-dimensional liquid chromatography-dual mass spectrometry (2DLC-dual-MS) system. This system employs a 2DLC setup to sequentially separate polar and non-polar compounds with log P ranging from -8 to 15. The separated fractions are directed via a three-way valve to a high-resolution MS (HRMS) and a triple qua

Oculomics meets exposomics: a roadmap for applying multi-modal ocular biomarkers in precision environmental health research.

Precision environmental health (PEH) is an emerging field that seeks to understand how diverse environmental exposures interact with individual biological and genetic factors to influence health outcomes. While recent advances in exposomics have enabled systematic characterization of the exposome, the integrated compilation of all physical, chemical, biological, and psychosocial influences that affect biology and health, identifying and developing sensitive biomarkers remains a critical challenge. The human eye offers unique potential for non-invasive biomarker discovery. Ocular biomarkers can

Holistic evaluation of the impact of pregnancy urban exposome on infant wheezing and chest infections: an outcome-wide approach.

BACKGROUND: Few studies have considered the complex interplay of the urban exposome in association with multiple respiratory outcomes during infancy. AIM: Utilizing an outcome-wide exposome approach, we aimed to assess associations of pregnancy urban exposome with offspring wheezing and chest infections at different time points within the first 18 months of life. METHODS: The analysis included data from 1032 mother-child pairs from the Barcelona Life Study Cohort (BiSC) (2018-2021). In total, 44 urban exposome factors were assessed during pregnancy, including air pollution, noise, temperatur

Validation of external and internal exposome of the findings associated to cerebral small vessel disease: A Mendelian randomization study.

The exposome characterizes all environmental exposures and their impact on a disease. To determine the causally-associated components of the exposome for cerebral small vessel disease (CSVD), we performed mendelian randomization analysis of 5365 exposures on six clinical and subclinical CSVD measures. We found statistically significant evidence (FDR-corrected P < 0.05) that hypertension, high cholesterol, longer television-watching time, lower educational qualifications, younger age of first sexual intercourse, smoking, reduced pulmonary function, higher subjective overall health rating, and f

Brain imaging-derived phenotypes influence risk of bipolar disorder: comprehensive exposome-wide association and wide-angled genetic analyses.

BACKGROUND: Despite many reported imaging markers for bipolar disorder (BD), systematic assessment across diverse brain imaging-derived phenotypes (IDPs) is lacking. We used epidemiological and genetic analyses to progress from association to causality and identify candidate biomarkers and potential pathways of BD. METHODS: We conducted an Exposome-Wide Association Study (ExWAS) in 35,680 UK Biobank participants to assess associations of 686 brain IDPs with BD. Genetic correlations were estimated via linkage disequilibrium score regression (LDSC). Bidirectional two-sample Mendelian randomizat

Early postoperative hospital exposome stress, multisystem recovery trajectories, and long-term oncologic outcomes after curative colorectal cancer surgery.

BACKGROUND: Postoperative recovery after colorectal cancer surgery is highly heterogeneous and incompletely explained by tumor stage or operative factors. Whether early postoperative inpatient stress exposure and subsequent multisystem recovery dynamics are associated with ctDNA-defined molecular residual disease and long-term oncologic outcomes remains unclear. METHODS: In this prospective longitudinal cohort study, 1200 patients undergoing curative-intent resection for stage II-III colorectal cancer (2018-2020) were followed for up to 60 months. Early postoperative hospital exposome stress

From Exposome-Scale Screening to Foam Cell Formation: Identifying Environmental Chemicals with Atherogenic Potential via Nuclear Receptor Perturbation.

Cardiovascular diseases driven by atherosclerosis are a leading global cause of mortality. However, currently identified risk chemicals account for only a small fraction of the environmental chemical burden contributing to atherosclerosis. To address this gap, we established a multi-source prioritization framework to identify pro-atherogenic chemicals among 6346 exogenous compounds predicted in human blood. The framework integrates bioactivity data for atherosclerosis-relevant nuclear receptors, including peroxisome proliferator-activated receptor γ (PPARγ), liver X receptor α (LXRα), farnesoi

Vichy volcanic mineralizing water has unique properties to strengthen the skin barrier and skin defenses against exposome aggressions.

Exposome aggressions are known to weaken certain skin functions, such as skin barrier and skin defense functions. Vichy volcanic mineralizing water (VVMW) percolates through volcanic and magmatic rocks in the Auvergne region in France to create a pure, highly mineralized water containing 15 minerals for a total mineral concentration of 5.2 g/L. Here, we provide an overview of the main results of in vitro and ex vivo studies (keratinocyte cultures, 3D reconstructed skin model, skin explants) and clinical studies to evaluate the effect of VVMW on key skin functions to help elucidate how it count

The Nutri-Exposome Intelligence Framework: Integrating Multi-Omics, Machine Learning, and Digital Nutrition for Precision Chronic Disease Prevention.

Background/Objectives: Precision nutrition is moving beyond population-based guidance and isolated gene-diet interactions toward integrative models of dietary response. However, current approaches remain fragmented across nutrigenomics, microbiome research, multi-omics profiling, digital health, and machine learning. This review proposes the Nutri-Exposome Intelligence Framework as a conceptual, data science-driven model for integrating cumulative dietary, environmental, microbial, molecular, clinical, and digital exposures for precision chronic disease prevention. Methods: This conceptual rev

Fueling Early Tumorigenesis: Interplay Between the Exposome, Obesity-Driven Metabolic Dysfunction and Early-Onset Colorectal and Breast Cancer.

Early-onset cancers (EOCs), defined as cancers diagnosed before the age of 50, are increasing globally. This rise cannot be fully explained by improved detection, suggesting the involvement of evolving environmental and lifestyle-related risk factors. In parallel, the global prevalence of obesity has increased substantially, particularly among younger populations. Obesity is increasingly recognized not simply as excess adiposity, but as a systemic metabolic state characterized by insulin resistance, hyperinsulinemia, chronic low-grade inflammation, altered lipid metabolism, hormonal dysregulat

Lessons learned from aldehyde dehydrogenases as non-P450 aldehyde-oxidizing enzymes: implications for the 'exposome' and human health.

INTRODUCTION: Aldehyde dehydrogenases (ALDHs) are critical enzymes that protect against cellular damage by metabolizing reactive aldehydes derived from both endogenous processes and environmental exposures. Although cytochrome P450 enzymes dominate metabolic toxicology discussions, the non-P450 ALDH superfamily plays a unique underrecognized role in mitigating the health impacts of the exposome - the totality of lifetime environmental exposures. AREAS COVERED: This Special Report highlights key insights from recent research on ALDHs, with a focus on their enzymatic diversity, disease-relevant

Exposome-Wide Association Study of Body Mass Index Using a Novel Meta-Analytical Approach for Random Forest Models.

BACKGROUND: Overweight and obesity impose a considerable individual and social burden, and the urban environments might encompass factors that contribute to obesity. Nevertheless, there is a scarcity of research that takes into account the simultaneous interaction of multiple environmental factors. OBJECTIVES: Our objective was to perform an exposome-wide association study of body mass index (BMI) in a multicohort setting of 15 studies. METHODS: Studies were affiliated with the Dutch Geoscience and Health Cohort Consortium (GECCO), had different population sizes (688-141,825), and covered th

Clinical decisions in fetal-neonatal neurology I. reproductive and pregnancy health influence the neural exposome over multiple generations.

Interdisciplinary fetal neonatal neurology (FNN) training requires integration of reproductive health factors into evaluations of the maternal-placental-fetal (MPF) triad, neonate, and child over the first 1000 days. Serial events that occur before one or multiple pregnancies impact successive generations. A maternal-child dyad history highlights this continuity of health risk, beginning with a maternal grandmother's pregnancy. Her daughter was born preterm and later experienced polycystic ovarian syndrome further complicated by cognitive and mental health disorders. Medical problems during he

Maternal-Fetal Exposome and Early-Life Microbiota-Pattern Recognition Receptors Signaling in Inflammatory Bowel Disease Susceptibility: A Mechanistic Review.

Inflammatory bowel disease (IBD) develops through interactions among genetic susceptibility, intestinal microbiota, environmental exposures, and dysregulated mucosal immunity. Increasing evidence suggests that these interactions may begin before diagnosis-and possibly before birth-when maternal signals, microbial assembly, epithelial maturation, and immune education intersect. This narrative review synthesizes findings from human observational and cohort studies, experimental animal and cellular models, and mechanistic literature to examine how prenatal and early-life exposures may influence l
