Role of protein tyrosine phosphorylation in rat corneal neovascularization.

BACKGROUND: Recent studies have suggested that tyrosine kinase pathways that are activated by angiogenic growth factors may play a role in corneal neovascularization. METHODS: Corneal neovascularization was induced in rat corneas by chemical cauterization. At 6, 24, 48, 96, and 168 h after chemical cauterization the rat corneas without the corneal epithelium were prepared for gel electrophoresis. Total protein profiles of the corneal samples were examined by staining gels with Coomassie brilliant blue. Tyrosine-phosphorylated proteins, three angiogenic growth factors (basic fibroblast growth

Fibroblast growth factor receptor-3 expression in meningiomas with stimulation of proliferation by the phosphoinositide 3 kinase-Akt pathway.

OBJECT: Fibroblast growth factor receptors (FGFRs)-1, -2, and -3 are expressed in the developing brain and may participate in CNS neoplasia. Fibroblast growth receptor-3 has not been demonstrated in the human CNS or its tumors. Nonetheless, it has been implicated in the pathogenesis of several other forms of neoplasia. METHODS: Twenty-four human meningiomas were evaluated using Western blot analysis for expression of FGFR3, its ligand acidic FGF, and concomitant phosphorylation/activation of p44/42 mitogen-activated protein kinase (MAPK), Akt, and STAT3. Mutations in exons 7 and 10 of the FGF

Fibroblast growth factor-2 drives and maintains progressive corneal neovascularization following HSV-1 infection.

Herpes simplex virus type 1 (HSV-1) infection of the cornea induces vascular endothelial growth factor A (VEGF-A)-dependent lymphangiogenesis that continues to develop well beyond the resolution of infection. Inflammatory leukocytes infiltrate the cornea and have been implicated to be essential for corneal neovascularization, an important clinically relevant manifestation of stromal keratitis. Here we report that cornea infiltrating leukocytes including neutrophils and T cells do not have a significant role in corneal neovascularization past virus clearance. Antibody-mediated depletion of thes

Renal thrombotic microangiopathy during nintedanib treatment for idiopathic pulmonary fibrosis
.

Agents that block vascular endothelial growth factor (VEGF) and its downstream pathway have been reported to be associated with nephrotoxicity including hypertension, proteinuria, and renal dysfunction. Bevacizumab, a monoclonal antibody against VEGF, is known to cause thrombotic microangiopathy (TMA), while tyrosine kinase inhibitors (TKIs) that block VEGF downstream are mainly associated with minimal change disease or focal segmental glomerulosclerosis. The question regarding the source of the diverse phenotypes of nephrotoxicity associated with these agents remains enigmatic. Nintedanib, a

Gonadal cell apoptosis: hormone-regulated cell demise.

It has become evident that apoptosis, an active form of cell 'suicide', plays an important role in the normal function of all tissues. A balance of cell proliferation and apoptosis is maintained in a healthy individual and any imbalance of the two processes could lead to pathological changes. In both sexes, massive apoptosis accounts for the demise of a majority of gonadal cells (ovarian granulosa cells and male germ cells) during reproductive life. Recent studies have indicated the important role of gonadotrophins as survival factors in both the ovary and the testis. Furthermore, intra-gonada

Asperosaponin VI ameliorates osteoporosis via skeletal muscle-derived FGF-2-mediated osteoblast differentiation.

BACKGROUND: Skeletal muscle functions as a dynamic endocrine organ, secreting myokines that contribute to systemic physiological homeostasis. Despite the established osteoprotective effects of Asperosaponin VI (AVI), it remains unknown whether these effects are mediated via the modulation of muscle-derived factors that subsequently regulate bone metabolism. PURPOSE: The present study aims to investigate whether AVI alleviates ovariectomy (OVX)-induced bone loss by modulating muscle-derived signals that exert regulatory effects on osteogenesis. METHODS: Female C57BL/6 OVX mouse models and the

Pro-inflammatory Cytokines: Cellular and Molecular Drug Targets for Glucocorticoid-induced-osteoporosis via Osteocyte.

Glucocorticoids are widely used to treat varieties of allergic and autoimmune diseases, however, long-term application results in glucocorticoid-induced osteoporosis (GIOP). Inflammatory cytokines: tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) play important regulatory roles in bone metabolism, but their roles in GIOP remain largely unknown. Osteocytes can modulate the formation and function of both osteoblasts and osteoclasts, directly via gap junctions, or indirectly by transferring molecule signaling. Apoptotic osteocytes release RANKL, HMGB1 and pro-inflammatory cytokines to sti

The effects of sugar phosphates in reducing hif-1? Under hypoxic conditions.

Preliminary research has shown evidence of the presence of connective tissue growth factor CTGF in the tenosynovium of patients with carpal tunnel syndrome, a finding which supports the fibrotic pathophysiological progression of the disease. Connective tissue growth factor (CTGF) expression is regulated through the actions of two distinct molecular signals; transforming growth factor-beta (TGF-ß) and hypoxia inducible factor-1 alpha (HIF-1a). Mannose-6 phosphate is a natural inhibitor of TGF-ß and can also is converted to fructose 6 phosphate which we have shown is capable of reducing HIF-1 a.

The effect of p38MAPK on cyclic stretch in human facial hypertrophic scar fibroblast differentiation.

Hypertrophic scars (HTS), the excessive deposition of scar tissue by fibroblasts, is one of the most common skin disorders. Fibroblasts derived from surgical scar tissue produce high levels of α-smooth muscle actin (α-SMA) and transforming growth factor-β1 (TGF-β1). However, the molecular mechanisms for this phenomenon is poorly understood. Thus, the purpose of this study was to evaluate the molecular mechanisms of HTS and their potential therapeutic implications. Fibroblasts derived from skin HTS were cultured and characterized in vitro. The fibroblasts were synchronized and randomly assigned

Chitosan hydrogel loaded with mesenchymal stem cell-derived exosomes significantly accelerated wound closure and enhanced tissue regeneration in a full thickness porcine skin defect model.

There is an urgent need for more effective wound healing therapies, particularly wound dressings that not only promote wound closure and reduce scar formation but also retain antibacterial properties. Chitosan, a natural copolymer with inherent hemostatic and antibacterial properties, and mesenchymal stem cell derived exosomes, which promote rapid re-epithelialization, collagen maturation, and scar reduction, were integrated into a single biofunctional dressing. This study evaluated the therapeutic efficacy and underlying mechanisms of this combined approach using a porcine full thickness skin

Human peritoneal fibroblast proliferation in 3-dimensional culture: modulation by cytokines, growth factors and peritoneal dialysis effluent.

Structural and functional alterations of the peritoneal membrane are a significant problem in long-term peritoneal dialysis patients. The present study has established a 3-dimensional (3D) cell culture system to study the human peritoneal fibroblast (HPFB) and to examine its proliferative responses to cytokines and growth factors as well as dialysis effluent obtained from patients during peritoneal infection. PDGF-AB, basic FGF and IL-1 beta induced a time and dose dependent increase in 3D-HPFB proliferation. At day 9 proliferation, as assessed by MTT uptake, was increased by 2.4-, 2.3- and 1.

Bartonella henselae Persistence within Mesenchymal Stromal Cells Enhances Endothelial Cell Activation and Infectibility That Amplifies the Angiogenic Process.

Some bacterial pathogens can manipulate the angiogenic response, suppressing or inducing it for their own ends. In humans, Bartonella henselae is associated with cat-scratch disease and vasculoproliferative disorders such as bacillary angiomatosis and bacillary peliosis. Although endothelial cells (ECs) support the pathogenesis of B. henselae, the mechanisms by which B. henselae induces EC activation are not completely clear, as well as the possible contributions of other cells recruited at the site of infection. Mesenchymal stromal cells (MSCs) are endowed with angiogenic potential and play a

Patterns and localization of gene expression during intramembranous bone regeneration in the rat femoral marrow ablation model.

Tissue formation and repair are dependent upon cascades of biological events, but the signals involved and the possible gene coexpression patterns during intramembranous bone repair are only poorly understood. We sought to place this mode of regeneration in context by profiling quantitative gene expression for a panel of 39 genes between days 1 and 14 following rat femoral marrow ablation. In situ hybridization was employed to localize a subset of genes. Additionally, principal components analysis was conducted to identify underlying factors suggestive of coexpression patterns. During inflamma
