Astrocyte Infection during Rabies Encephalitis Depends on the Virus Strain and Infection Route as Demonstrated by Novel Quantitative 3D Analysis of Cell Tropism.

Although conventional immunohistochemistry for neurotropic rabies virus (RABV) usually shows high preference for neurons, non-neuronal cells are also potential targets, and abortive astrocyte infection is considered a main trigger of innate immunity in the CNS. While in vitro studies indicated differences between field and less virulent lab-adapted RABVs, a systematic, quantitative comparison of astrocyte tropism in vivo is lacking. Here, solvent-based tissue clearing was used to measure RABV cell tropism in infected brains. Immunofluorescence analysis of 1 mm-thick tissue slices enabled 3D-se

Resilience to the contralateral visual field bias as a window into object representations.

Viewing images of manipulable objects elicits differential blood oxygen level-dependent (BOLD) contrast across parietal and dorsal occipital areas of the human brain that support object-directed reaching, grasping, and complex object manipulation. However, it is unknown which object-selective regions of parietal cortex receive their principal inputs from the ventral object-processing pathway and which receive their inputs from the dorsal object-processing pathway. Parietal areas that receive their inputs from the ventral visual pathway, rather than from the dorsal stream, will have inputs that

Circular orbital manipulation of positive-acoustic-contrast particles employing double-ring perfect acoustic-vortex beams.

Non-contact acoustic-vortex (AV) tweezers have gained prominence for high-precision manipulation. However, conventional AV beams are limited by their topological charge (TC)-dependent radii. While perfect AV beams address this radius issue, existing implementations struggle to achieve both flexible field control and stable trapping of particles with positive acoustic contrast. To overcome these limitations, this study introduces a robust method for constructing double-ring perfect acoustic vortex (DR-PAV) fields, enabling stable circular manipulation of positive-contrast particles along low-pr

Contrast-Enhanced MR Fingerprinting With Delta-Relaxometry: Investigating a New Avenue for Tumor Characterization.

BACKGROUND: MRI contrast agents enhance lesion characterization by altering tissue relaxation properties. However, quantitative assessment of contrast enhancement is limited by variability in contrast administration parameters, and lack of efficient and precise contrast concentration independent relaxivity (r 1, r 2) measurement techniques. MR Fingerprinting (MRF) rapidly, simultaneously and accurately measures T 1 and T 2, enabling for the first time efficient clinical estimation of relaxivity ratios (r 1/r 2). PURPOSE: To introduce an MRF-derived delta-relaxometry method for mapping contras

Reassessing B cell contributions in multiple sclerosis.

There is growing recognition that B cell contributions to normal immune responses extend well beyond their potential to become antibody-producing cells, including roles at the innate-adaptive interface and their potential to modulate the responses of other immune cells such as T cells and myeloid cells. These B cell functions can have both pathogenic and protective effects in the context of central nervous system (CNS) inflammation. Here, we review recent advances in the field of multiple sclerosis (MS), which has traditionally been viewed as primarily a T cell-mediated disease, and we conside

Antiphasic Purkinje cell responses in mouse uvula-nodulus are sensitive to static roll-tilt and topographically organized.

Two vestibular pathways converge at the uvula-nodulus to modulate the discharge of Purkinje cell complex and simple spikes (CSs and SSs). In the mouse, vestibular primary afferent mossy fibers originate from each of the end organs of the ipsilateral labyrinth and terminate in the granule cell layers of folia 9c-10. Vestibular climbing fiber projections originate from the contralateral beta-nucleus and dorsomedial cell column (dmcc) and terminate directly on Purkinje cells. CSs and SSs could be regulated independently or they could be co-dependent. Here we examine how the discharges of CSs and

Fundamentals of pulsed field ablation at the individual cardiomyocyte level.

BACKGROUND: Pulsed field ablation (PFA) is a rapidly adopted catheter ablation method that employs high-voltage pulses to treat cardiac arrhythmias by cardiomyocyte electroporation. OBJECTIVE: We systematically characterized cardiomyocyte responses in vitro to different PFA waveforms to better understand the transient electrophysiological effects observed in clinical PFA. METHODS: Isolated adult rat ventricular cardiomyocytes were exposed to clinically relevant waveforms: monophasic 100 μs, biphasic 2 μs, and monophasic 200 ns pulses. Transmembrane voltage, intracellular Ca2+, and sarcomere

B cell depletion therapies in autoimmune diseases: Monoclonal antibodies or chimeric antigen receptor-based therapy?

Immune system detects foreign pathogens, distinguishes them from self-antigens and responds to defend human body. When this self-tolerance is disrupted, the overactive immune system attacks healthy tissues or organs and the autoimmune diseases develop. B cells and plasma cells contribute a lot to pathogenesis and persistence of autoimmune diseases in both autoantibody-dependent and autoantibody-independent ways. Accumulating data indicates that treatments aiming to eliminate antibody-secreting cells (B cells or plasma cells) are effective in a wide spectrum of autoimmune diseases. Monoclonal a

Premature vision drives aberrant development of response properties in primary visual cortex.

Development of the mammalian visual system is thought to proceed in two stages. In the first stage, before birth in primates and before eye opening in altricial mammals, spontaneous activity generated by the retina and cortex shapes visual brain circuits in an activity-dependent but experience-independent manner. In the second stage, visual activity generated by sensory experience refines receptive fields. Here we investigated the consequences of altering this sequence of events by prematurely opening one or both eyes of ferrets and examining visual receptive fields in monocular cortex after t

Stimulus-specific recruitment of human amygdala neurons predicts episodic memory encoding success.

Controlling whether a given experience is encoded into long-term memory and thus later remembered is a crucial component of our memory system whose failure is often at the root of memory disorders. One brain area that takes part in controlling which experiences are remembered is the amygdala, but the mechanisms by which it does so remain poorly understood. Here we examined single-neuron activity and local field potentials as human participants performed recognition memory tasks with visual stimuli. Category-selective amygdala neurons exhibited elevated firing rates during encoding of later rem

NMDA receptor blockade in the superior colliculus increases receptive field size without altering velocity and size tuning.

Neonatal brain injury triggers compensatory processes that can be adaptive or detrimental, but little is known about the mechanisms of compensation or how they might affect the response properties of neurons within the injured region. We have studied this issue in a rodent model. Partial ablation of the hamster superior colliculus (SC) at birth results in a compressed but complete visual field map in the remaining SC and a compensatory conservation of receptive field (RF) size and stimulus velocity and size tuning. The circuit underlying stimulus tuning in this system or its preservation after

B cell receptor signaling in chronic lymphocytic leukemia.

B cell receptor (BCR) signaling plays an important pathogenic role in chronic lymphocytic leukemia (CLL) and B cell lymphomas, based on structural restrictions of the BCR, and BCR-dependent survival and growth of the malignant B cells. In CLL and lymphoma subtypes, ligand-independent ('tonic') and ligand-dependent BCR signaling have been characterized, which can involve mutations of BCR pathway components or be triggered by (auto)antigens present in the tissue microenvironment. In CLL, based on high response rates and durable remissions in early-stage clinical trials, there is rapid clinical d

Photodegradation of ethylenediaminetetraacetic acid (EDTA) and ethylenediamine disuccinic acid (EDDS) within natural UV radiation range.

The rate of photodegradation of two chelating agents, ethylenediaminetetraacetic acid (EDTA) and an isomeric mixture of ethylenediamine disuccinic acid (EDDS), was analysed in humic lake water and in distilled water using exposure to sunlight, and in the laboratory using lamps emitting UV radiation in the range 315-400 nm. Degradation was studied using Fe(III) complexes and sodium salts of chelates. Fe(III) complexes were illuminated at pH 3.1 and 6.5. The results demonstrated that the rate of photodegradation of Fe(III)-EDTA and Fe(III)-EDDS complexes seems to be pH dependent. In the laborato

Tensor structure and transport of centrifugal distortion constants: A representation-independent framework connecting spectroscopy and quantum chemistry beyond the linear regime.

Centrifugal distortion constants provide a key link between high-resolution rotational spectroscopy and quantum-chemical rovibrational calculations. Within Watson's Hamiltonian, the underlying structure is described in terms of tensorial objects, whereas fitted spectroscopic parameters become representation-dependent upon projection, complicating direct comparison between experiment and theory. Building on the tensorial formulation of centrifugal distortion, we show that standard quartic and sextic constants can be consistently described within a representation-independent linear tensor sector

Signals from posterior parietal area 5 to motor cortex during locomotion.

Area 5 of the parietal cortex is part of the "dorsal stream" cortical pathway which processes visual information for action. The signals that area 5 ultimately conveys to motor cortex, the main area providing output to the spinal cord, are unknown. We analyzed area 5 neuronal activity during vision-independent locomotion on a flat surface and vision-dependent locomotion on a horizontal ladder in cats focusing on corticocortical neurons (CCs) projecting to motor cortex from the upper and deeper cortical layers and compared it to that of neighboring unidentified neurons (noIDs). We found that up

Device-Independent Discrimination of Falsified Amoxicillin Capsules Using Heterogeneous Near-Infrared Spectroscopic Devices for Training and Testing of a Support Vector Machine.

The objective of this work is to demonstrate the potential of near-infrared spectroscopy for common screening of falsified medicines in the field by means of a device-independent universal discrimination approach. In order to provide a useful discrimination tool to protect people from low-quality medical products, not only is a low-cost and portable screening device necessary, but a reference library is also essential. The authors believe that a device-dependent reference library inhibits near-infrared spectroscopy from becoming a popular screening tool. In this study, to develop a device-inde

Surfactant enhanced recovery of tetrachloroethylene from a porous medium containing low permeability lenses. 2. Numerical simulation.

A numerical model of surfactant enhanced solubilization was developed and applied to the simulation of nonaqueous phase liquid recovery in two-dimensional heterogeneous laboratory sand tank systems. Model parameters were derived from independent, small-scale, batch and column experiments. These parameters included viscosity, density, solubilization capacity, surfactant sorption, interfacial tension, permeability, capillary retention functions, and interphase mass transfer correlations. Model predictive capability was assessed for the evaluation of the micellar solubilization of tetrachloroethy

Thermally Tunable Fading Memory Enabled by Opto-Ionic-Electronic Coupling in AgInP2S6-Gated Two-Dimensional Field-Effect Transistors.

In conventional field-effect transistors (FETs), the gate dielectric primarily provides electrostatic control of the channel and is engineered to remain chemically and electronically inert to ensure stable operation. Emerging studies, however, suggest that functional dielectrics integrated with two-dimensional (2D) semiconductors can introduce additional physical mechanisms that influence device behavior. Here, we investigate AgInP2S6 (AIPS), a high-κ van der Waals thiophosphate, as a functional top-gate dielectric in MoS2 field-effect transistors. The AIPS/MoS2 heterointerface exhibits couple
