Background: Marburg virus disease (MVD) is a highly virulent viral hemorrhagic fever with reported case fatality rates of up to 90%. It is part of the same family as the Ebola virus (Filoviridae). MVD, originally identified in 1967 in the context of outbreaks associated with African green monkeys, has been reported sporadically in Africa. Recent outbreaks, including those in Equatorial Guinea and Rwanda, underscore the need for robust preparedness systems and global response. Objectives: This narrative review focuses on the pathogenesis, clinical manifestations, diagnostic challenges and treat

Marburg viruses (MARV) are negative-stranded RNA viruses belonging to filoviridae family. This virus causes severe haemorrhagic fever in humans and non-human primates, with a high fatality rate. Currently, there are no vaccines or drugs have been approved to induce productive immunity or control viral infection. As a result, vaccination against this virus is essential for reducing mortality rates. The current study aimed to identify CTL (cytotoxic T lymphocytes) and B-cell epitopes of MARV using in silico tools. A total of 3697 CTL epitopes and 4577 B-cell epitopes were predicted in the viral

Marburg virus (MARV) is a negative-sense, single-stranded RNA virus that belongs to the Filoviridae family. Despite having caused numerous outbreaks of severe hemorrhagic fever with high case fatality rates, there are still no clinically approved therapeutics or vaccines to treat or prevent MARV disease. Recombinant vesicular stomatitis viruses (rVSVs) expressing heterologous viral glycoproteins have shown remarkable promise as live-attenuated vaccine vectors, with an rVSV-based Ebola virus vaccine having received regulatory approval in the United States and numerous other countries. Zaire Ebola virus (EBOV) is a member of the Filoviridae family of negative sense, single-stranded RNA viruses. EBOV infection causes Ebola virus disease (EVD), characterized by coagulopathy, lymphopenia, and multi-organ failure, which can culminate in death. In 2019, the FDA approved the first vaccine against EBOV, a recombinant live-attenuated viral vector wherein the G protein of vesicular stomatitis virus is replaced with the glycoprotein (GP) of EBOV (rVSV-EBOV-GP, Ervebo® by Merck). This vaccine demonstrates high efficacy in nonhuman primates by providing prophylactic, rapid, and post-exp

Ebola virus disease is a serious global health concern given its periodic occurrence, high lethality, and the lack of approved therapeutics. Certain drugs that alter intracellular calcium, particularly in endolysosomes, have been shown to inhibit Ebola virus infection; however, the underlying mechanism is unknown. Here, we provide evidence that Zaire ebolavirus (EBOV) infection is promoted in the presence of calcium as a result of the direct interaction of calcium with the EBOV fusion peptide (FP). We identify the glycoprotein residues D522 and E540 in the FP as functionally critical to EBOV's

Ebola (EBOV) and Marburg (MARV) viruses are members of the Filoviridae family which cause outbreaks of hemorrhagic fever. The filovirus VP40 matrix protein is essential for virus assembly and budding, and its PPxY L-domain motif interacts with WW-domains of specific host proteins, such as Nedd4 and ITCH, to facilitate the late stage of virus-cell separation. To identify additional WW-domain-bearing host proteins that interact with VP40, we used an EBOV PPxY-containing peptide to screen an array of 115 mammalian WW-domain-bearing proteins. Using this unbiased approach, we identified BCL2 Associ

Ebola virus is a Filoviridae that causes hemorrhagic fever in humans and induces high morbidity and mortality rates. Filoviruses are classified as "Category A bioterrorism agents", and currently there are no licensed therapeutics or vaccines to treat and prevent infection. The Filovirus glycoprotein (GP) is sufficient to protect individuals against infection, and several vaccines based on GP are under development including recombinant adenovirus, parainfluenza virus, Venezuelan equine encephalitis virus, vesicular stomatitis virus (VSV) and virus-like particles. Here we describe the developmen

The coronavirus disease 2019 pandemic illustrated the need to develop medical countermeasures against emerging infectious diseases. As viruses rely on cellular machinery for replication, host-directed antivirals (HDAs) may complement conventional antiviral strategies in a manner that offers broad-spectrum efficacy, including against novel viruses, and a potentially higher barrier to viral escape. Despite their potential, HDAs are under-represented as therapeutics, partly due to a lack of consensus on druggable host targets. To address this, we have performed a meta-analysis of 62 functional ge

INTRODUCTION: Ebolavirus (EBOV) is a highly pathogenic member of the Filoviridae family, causing severe hemorrhagic fever with high mortality rates and is persistent threat to global health with recurrent outbreaks. Although advances, including vaccine and monoclonal antibody therapeutic development, have improved survival chances, challenges such as cost and availability remain as well as potential viral evolution. Consequently, there is an urgent need for alternative, cost-effective and scalable therapeutics, which has driven interest in computational drug discovery as rapid response strateg

To investigate the antiviral innate immune responses of natural reservoir bats, we conducted a histopathological analysis of Egyptian rousettes (Rousettus aegyptiacus) experimentally infected with either of two divergent RNA viruses they naturally host, Marburg virus (MARV; family Filoviridae) and Kasokero virus (KASV; family Nairoviridae). Bats were serially euthanized at similar post-infection time points and tissue-based analyses focused on the liver, the primary target for both viruses. At 3 days post-infection (DPI), in situ hybridization (ISH) signal for replicating MARV was 300x less ev

The Egyptian rousette bat (ERB) is a natural reservoir host for Marburg virus (MARV); however, the mechanisms by which MARV is transmitted bat-to-bat and to other animals are unclear. Here we co-house MARV-inoculated donor ERBs with naive contact ERBs. MARV shedding is detected in oral, rectal and urine specimens from inoculated bats from 5-19 days post infection. Simultaneously, MARV is detected in oral specimens from contact bats, indicating oral exposure to the virus. In the late study phase, we provide evidence that MARV can be horizontally transmitted from inoculated to contact ERBs by fi

BACKGROUND: Recent studies of viral entry proteins from influenza, measles, human immunodeficiency virus, type 1 (HIV-1), and Ebola virus have shown, first with molecular modeling, and then X-ray crystallographic or other biophysical studies, that these disparate viruses share a coiled-coil type of entry protein.
RESULTS: Structural models of the transmembrane glycoproteins (GP-2) of the Arenaviruses, lymphochoriomeningitis virus (LCMV) and Lassa fever virus, are presented, based on consistent structural propensities despite variation in the amino acid sequence. The principal features of the

Bats are natural reservoirs for over 4400 viruses across 110 recognized viral families due to their high species diversity, long lifespans, and unique physiological adaptations. Their tolerance to viral infections without clinical disease stems from constitutive interferon-alpha (IFN-α) activity, high metabolic rates during flight, and tightly regulated inflammatory responses. Anthropogenic pressures including deforestation and urbanization intensify human-bat contact, facilitating viral spillover. Beyond direct transmission, bat-associated ectoparasites - ticks, bat flies, and mosquitoes - ma

Taï Forest virus (TAFV) is a negative-sense RNA virus in the Filoviridae family. TAFV has caused only a single human infection, but several disease outbreaks in chimpanzees have been linked to this virus. Limited research has been done on this human-pathogenic virus. We sought to establish an animal model to assess TAFV disease progression and pathogenicity at our facility. We had access to two different viral stock preparations from different institutions, both originating from the single human case. Type I interferon receptor knockout mice were inoculated with TAFV stock 1 or stock 2 by the

In 1976 Ebola revealed itself to the world, marking the beginning of a series of localized outbreaks. However, it was the Ebola outbreak that began in 2013 that incited fear and anxiety around the globe. Since then, our comprehension of the virus has been steadily expanding. Ebola virus (EBOV), belonging to the Orthoebolavirus genus of the Filoviridae family, possesses a non-segmented, negative single-stranded RNA genome comprising seven genes that encode multiple proteins. These proteins collectively orchestrate the intricate process of infecting host cells. It is not possible to view each pr

The current Marburg virus (MARV) outbreak in Tanzania served as a stark reminder of the ongoing threat posed by emerging infectious diseases and the urgent need for global health security. The Tanzanian Ministry of Health (MoH) officially declared the outbreak on March 21, 2023. Eight cases in all, five of which included fatalities, have been reported in the country at present. The virus is a member of the Filoviridae family closely related to the widely known Ebola virus. Similar to other filoviruses, MARV causes acute and lethal hemorrhagic fever in both human and nonhuman primates with high

This article addresses the recent use of mathematical epidemiological SIR or SEIR models in plague research. This use of S(E)IR models is highly problematic, but the problems are not presented and considered. Serious problems show in that such models are used to "prove" that historical plague was a (1) Filoviridae disease and (2) a bacterial disease caused by Yersinia pestis which was transmitted by human fleas and lice. (3) They also support early-phase transmission (by fleas). They purportedly consistently disprove (4) the conventional view that plague is/was a rat-and-rat-flea-borne disease

BACKGROUND: Sudan virus and Marburg virus are high priority biological threat pathogens with fatality rates of 25-90%. Recent outbreaks in Uganda, Equatorial Guinea, and Rwanda between 2022 and 2025 emphasise the critical need for effective vaccines. The aim of this phase 1b study was to determine clinical safety, tolerability, and immunogenicity of these monovalent vaccines. Plasmapheresis was used to collect hyper-immune plasma for use in future assay standardisation.
METHODS: Participants received a single intramuscular injection of either cAd3-Sudan or cAd3-Marburg at 1 × 1011 particle un

A detailed understanding of the circulating pathogens in a particular geographic location aids in effectively utilizing targeted, rapid diagnostic assays, thus allowing for appropriate therapeutic and containment procedures. This is especially important in regions prevalent for highly pathogenic viruses co-circulating with other endemic pathogens such as the malaria parasite. The importance of biosurveillance is highlighted by the ongoing Ebola virus disease outbreak in West Africa. For example, a more comprehensive assessment of the regional pathogens could have identified the risk of a filov
