Fatty acid oxidation of alternatively activated macrophages prevents foam cell formation, but Mycobacterium tuberculosis counteracts this process via HIF-1α activation.

The ability of Mycobacterium tuberculosis (Mtb) to persist inside host cells relies on metabolic adaptation, like the accumulation of lipid bodies (LBs) in the so-called foamy macrophages (FM), which are favorable to Mtb. The activation state of macrophages is tightly associated to different metabolic pathways, such as lipid metabolism, but whether differentiation towards FM differs between the macrophage activation profiles remains unclear. Here, we aimed to elucidate whether distinct macrophage activation states exposed to a tuberculosis-associated microenvironment or directly infected with

Deep tissue loads in the seated buttocks on an off-loading wheelchair cushion versus air-cell-based and foam cushions: finite element studies.

For wheelchair users, a common injury is a sitting-acquired pressure ulcer (PU) which typically onsets near the interface between the ischial tuberosity (IT) and the overlying soft tissues. The risk of developing PUs can be reduced considerably if an adequate cushion is placed on the wheelchair in order to protect tissues from PUs by minimising interface mechanical loads between the body and cushion and also, exposure to internal soft tissue loads. In this work, we studied the biomechanical performances of an off-loading (OL) cushion with limited adjustability, in comparison to a standard foam

Classical and Alternative Activation and Metalloproteinase Expression Occurs in Foam Cell Macrophages in Male and Female ApoE Null Mice in the Absence of T and B Lymphocytes.

BACKGROUND: Rupture of advanced atherosclerotic plaques accounts for most life-threatening myocardial infarctions. Classical (M1) and alternative (M2) macrophage activation could promote atherosclerotic plaque progression and rupture by increasing production of proteases, including matrix metalloproteinases (MMPs). Lymphocyte-derived cytokines may be essential for generating M1 and M2 phenotypes in plaques, although this has not been rigorously tested until now. METHODS AND RESULTS: We validated the expression of M1 markers (iNOS and COX-2) and M2 markers (arginase-1, Ym-1, and CD206) and the

Tafolecimab mitigates ox-LDL-induced macrophage foam cell formation and inflammation by modulating SR-A/ABCG1 expression and inhibiting the NF-κB/MAPK pathways.

Macrophage foam-cell formation, triggered by excessive uptake of oxidized low-density lipoprotein (ox-LDL) and subsequent intracellular lipid accumulation, represents a critical pathological event in atherosclerotic plaque initiation that drives localized inflammatory responses. The present study investigated the effects of tafolecimab on ox-LDL-induced foam-cell formation and inflammatory responses in murine macrophages, and further explored the underlying molecular mechanisms. Foam-cell models were established by exposing RAW264.7 cells to 100 µg/ml ox-LDL for 24 h. The study groups included

Scutellarin Attenuates Pro-Inflammatory Foam Cell Formation and Facilitates M2 Polarization in Microglia during Copper Homeostasis Imbalance via the MAPK Signaling Pathway.

BACKGROUND: Clinical and experimental evidence indicates that copper has the ability to promote the progressive development of demyelinating diseases such as multiple sclerosis. Microglia-mediated neuroinflammation is believed to play a crucial role in this process. Scutellarin, a flavonoid compound, has anti-inflammatory, antioxidative, and neuroprotective effects. AIM: We investigated the effect of scutellarin on copper-induced inflammatory foam cell formation in microglia. METHODS: We exposed BV2 murine microglial cells to copper, then collected the conditioned medium and co-cultured it w

PPARα/γ signaling pathways are involved in Chlamydia pneumoniae-induced foam cell formation via upregulation of SR-A1 and ACAT1 and downregulation of ABCA1/G1.

Chlamydia pneumoniae (Cpn) has been reported to be involved in the pathogenesis of early atherosclerosis by inducing macrophage-derived foam cell formation in the presence of low-density lipoprotein (LDL). However, the biochemical mechanisms underlying Cpn-induced foam cell formation are still not fully elucidated. The present study showed that in LDL-treated THP-1-derived macrophages, Cpn not only upregulated the expression of scavenger receptor A1 (SR-A1) and acyl-coenzyme A: cholesterol acyltransferase 1 (ACAT1), but it also downregulated the expression of ATP binding cassette transporters

Early-stage Management of Complex Lower Extremity Wounds Using Negative Pressure Wound Therapy With Instillation and a Reticulated Open Cell Foam With Through Holes.

INTRODUCTION: Treatment modalities that overcome stalled wound healing in lower extremity wounds are crucial for reducing lower limb amputations, which have a 5-year mortality rate of an astounding 70%. Recent non-comparative studies have shown negative pressure wound therapy with instillation and dwell time (NPWTi-d) using a dressing comprised of reticulated open cell foam with through holes (ROCF-CC) provides favorable clinical outcomes for various wound types, including complex lower extremity wounds. OBJECTIVE: The objective of this study is to compare NPWTi-d using ROCF-CC dressings (tre

Increased degradation of lipoprotein(a) in atherosclerotic compared with nonlesioned aortic intima-inner media of rabbits: in vivo evidence that lipoprotein(a) may contribute to foam cell formation.

To investigate a potential role of lipoprotein(a) [Lp(a)] in foam cell formation, we have measured the degradation rates of Lp(a) and LDL in the rabbit aorta in vivo. Lp(a) (or LDL) was labeled with both 113I-TC and 125I and injected into 17 rabbits with extensive aortic atherosclerosis and into 16 rabbits without atherosclerosis. As the protein moiety of the doubly labeled lipoproteins is degraded, 131I-TC is trapped in the cell, whereas 125I diffuses out of the cell. Twenty-four hours after injection, 12 samples of the aorta and biopsies from 9 other tissues were removed. The degradation rat

Gamma-ray shielding performance of pressurized xenon-filled Ni-Cr 70/30 and Ni-Mo 70/30 closed-cell metal foams.

Pressurized xenon-filled closed-cell Ni-Cr 70/30 and Ni-Mo 70/30 metal foams are investigated as lightweight gamma-ray shielding materials for nuclear and aerospace applications. The proposed concept combines a nickel-based metallic matrix with xenon gas confined inside closed-cell cavities, aiming to exploit both the structural advantages of metal foams and the high atomic number of xenon for enhanced photon attenuation. Computational material definitions were established for different porosity levels and xenon pressures, and the shielding behavior was evaluated using GEANT4, FLUKA, and WinXC

Gualou-Xiebai-derived Apigenin-7-O-glucoside inhibits VSMCs foam cell formation and alleviates atherosclerosis by targeting Rab7 to drive SNAP29-dependent lipophagy.

BACKGROUND: Foam cell formation by vascular smooth muscle cells (VSMCs) is a key event in atherosclerotic plaque development. Lipophagy, a selective autophagy pathway governed by the small GTPase Rab7, helps maintain cellular lipid homeostasis, yet how Rab7-regulated lipophagy controls VSMC lipid balance in atherosclerosis (AS) remains unclear. The traditional formula Gualou-Xiebai herb pair (GLXB) is used clinically for chest-pain conditions now recognized AS and modulates lipid metabolism, but its molecular targets are undefined. PURPOSE: To define the role of Rab7 in VSMC lipid homeostasis

Double-locked fluorescent probe with in situ labeling of foam cell-derived exosomes and lipid-specific imaging for real-time monitoring the atherosclerotic progression in vitro and in vivo.

In vivo fluorescent labeling strategy has been developed to stain the foam cell-derived exosomes as circulating biomarkers for the in vitro detection of atherosclerotic progression. However, free fluorescent dyes in the blood produce a strong background fluorescence, which may interference their fluorescence signal and thus affect the detection accuracy. Herein, we developed a fluorescent probe that can stain the foam cell-derived exosomes specifically, which are secreted from atherosclerotic plaques into the blood as circulating biomarkers for the in vitro detection of atherosclerosis (AS). Macrophage P2Y6 receptor deletion attenuates atherosclerosis by limiting foam cell formation through phospholipase Cβ/store-operated calcium entry/calreticulin/scavenger receptor A pathways.

BACKGROUND AND AIMS: Macrophage-derived foam cells play a causal role during the pathogenesis of atherosclerosis. P2Y6 receptor (P2Y6R) highly expressed has been considered as a disease-causing factor in atherogenesis, but the detailed mechanism remains unknown. This study aims to explore P2Y6R in regulation of macrophage foaming, atherogenesis, and its downstream pathways. Furthermore, the present study sought to find a potent P2Y6R antagonist and investigate the feasibility of P2Y6R-targeting therapy for atherosclerosis. METHODS: The P2Y6R expression was examined in human atherosclerotic pl

Tumor necrosis factor-α and receptor activator of nuclear factor-κB ligand augment human macrophage foam-cell destruction of extracellular matrix through protease-mediated processes.

By secreting proteases such as cathepsins and matrix metalloproteinases (MMPs), macrophage foam cells may be a major cause of ruptured atherosclerotic plaques. The aims of the present study were to investigate in vitro role of human macrophage foam cells in degrading type I collagen, a major component of extracellular matrix (ECM) in plaques, and to establish whether the pro-inflammatory molecules, tumor necrosis factor (TNF)-alpha, and receptor activator of nuclear factor-κB ligand (RANK-L) increase this degradation. A Case Review of Wound Bed Preparation in an Infected Venous Leg Ulcer Utilizing Novel Reticulated Open Cell Foam Dressing with Through Holes during Negative Pressure Wound Therapy with Instillation.

Chronic venous insufficiency (CVI) and venous leg ulcers (VLUs) have major financial implications for patients and healthcare professionals. VLUs, in particular, require significant care, can be slow to heal, and have a high rate of recurrence. These factors combine to make VLUs a major burden on the healthcare system. Recent estimates show that the cost of treatment of VLUs per patient in the United States is $10,000 to $12,000 per year, with the average lifetime cost of care greater than $40,000. Infected VLUs often require surgical debridement for the removal of bacterial burden and biofilm
