Hepatic fructose-1,6-diphosphatase deficiency. A cause of lactic acidosis and hypoglycemia in infancy.

An 8-month-old female, maintained on breast feeding for 6 months, experienced numerous attacks of hyperventilation when weaned to baby food and was admitted with severe lactic acidosis (20 mM) and hypoglycemia. Physical examination was negative except for hepatomegaly. Fasting (18 hr) after stabilization on a high carbohydrate diet resulted in hypoglycemia (plasma glucose 40 mg/100 ml), lactic acidosis (6-10 mM), and a rise in plasma alanine. Glucagon produced a glycemic response after 6 hr, but not after 18 hr fasting. Intravenous galactose increased plasma glucose (Delta 45 mg/100 ml) but in

Garlic-Derived Exosomes Alleviate Intestinal Fibrosis in Crohn's Disease by Modulating PFKFB3-Mediated Metabolic Reprogramming.

Crohn's disease (CD), a chronic inflammatory bowel disease, is frequently complicated by intestinal fibrosis, a process driven by extracellular matrix remodeling. Current therapies lack efficacy in controlling or reversing fibrotic progression, underscoring the need for novel treatments. Here, we found that garlic-derived exosomes (GDE) significantly reduced hydroxyproline production and inhibited the expression of fibrosis-related proteins such as COL1A2, COL3A1, and α-SMA in TGF-β1-treated intestinal fibroblasts. Administration of GDE significantly improved the lesions of the muscular mucosa

Renal glucose metabolic enzyme expression during AKI-to-CKD transition.

The kidney is a high-energy-consuming organ, and glucose is one of its principal fuel sources. It has been documented that disturbed renal glucose metabolism occurs in acute kidney injury (AKI), but whether these disturbances are consistent in different types of AKI, and how they change during the transition from AKI to chronic kidney disease (CKD), remain to be addressed. In this study, we used AKI models induced by cisplatin (20 mg/kg, 48 h), lipopolysaccharide (10 mg/kg, 48 h), and ischemia-reperfusion (48 h) to mimic distinct etiologies, and the catalytic enzymes involved in glucose transp

Regulatory functions of AcuK and AcuM transcription factors in fungal metabolic adaptation, stress response, and virulence.

Fungal species thrive in diverse ecological niches and must dynamically adjust their metabolism, growth, and development in response to environmental fluctuations. Transcriptional regulation plays a pivotal role in these adaptive processes, with Zn2Cys6 transcription factors constituting the largest family of fungal-specific regulators that orchestrate metabolism, development, and pathogenicity. Among these, AcuK and AcuM represent a distinct subfamily with diverse key regulatory functions. In this review, we provide a comprehensive analysis of AcuK and AcuM functions across various fungal spe

Pharmacological evaluation of hyperin for antihyperglycemic activity and effect on lipid profile in diabetic rats.

Antihyperglycemic potential of hyperin at 25 and 50 mg/kg doses for 30 days to streptozotocin induced diabetic rats has been reported. In oral glucose tolerance test, hyperin treated rats showed a significant reduction in blood glucose level after 120 min. It was found that hyperin exhibited dose dependent and significant antihyperglycemic activity in streptozotocin induced diabetic rats which were nearly similar with standard drug glybenclamide. Activities of glucose-6-phosphatase, fructose-1,6-bisphosphatase, glycogen phosphorylase, glycosylated haemoglobin and level of serum urea and creati

The essential role of a single PEPCK isoform is presented as a target for disrupting tick embryogenesis.

The cattle tick Rhipicephalus microplus faces periods of nutritional stress that are inherent to its life cycle and during which starvation is a critical physiological challenge demanding specific metabolic adaptations. We used the embryonic cell line BME26 as a model to study metabolic responses during nutritional stress, to investigate the mechanisms underlying tick resilience. Our findings demonstrate that BME26 cells withstand prolonged starvation by activating gluconeogenesis, as evidenced by a marked increase in phosphoenolpyruvate carboxykinase (PEPCK) activity. Follow-up qPCR and enzym

PFKFB2 Gates a Relationship Between Cardiac Glycolytic Regulation and Electrophysiological Function.

BACKGROUND: The cardiac isoform of phosphofructokinase-2/fructose 2,6-bisphosphatase (PFKFB2) is the heart's strongest glycolytic regulator but is degraded in the absence of insulin signaling. This makes PFKFB2 loss critical to understand in metabolic heart disease, of which impaired insulin signaling is a hallmark. Prolongation of the QT interval, risk of arrhythmia, and sudden cardiac death are also augmented in metabolic heart disease, raising a question as to whether potential crosstalk between glycolytic dysregulation and electrophysiological dysfunction exists. ZBTB16 promotes the transcription of glycolytic enzyme PFKFB2 to participate in fibrogenesis in septic lung injury.

BACKGROUND: Sepsis is a severe systemic inflammatory response syndrome that leads to acute lung injury (ALI), which subsequently progresses to lung fibrosis. This study aims to investigate the role of zinc finger and BTB domain containing 16 (ZBTB16), a transcription factor, in septic lung injury. METHODS: The expression of Zbtb16 in the injured lung was evaluated using single-cell sequencing. The signaling pathways that can be regulated by Zbtb16 were predicted using gene set enrichment analysis (GSEA). Then, the functions of ZBTB16 and its potential molecular mechanism in fibrogenesis were

Differential expression of gluconeogenesis-related transcripts in a freshwater zooplankton model organism suggests a role of the Cori cycle in hypoxia tolerance.

Gluconeogenesis (GNG) is the process of regenerating glucose and NAD+ that allows for continued ATP synthesis by glycolysis during fasting or in hypoxia. Recent data from C. elegans and crustaceans challenged with hypoxia show differential and tissue-specific expression of GNG-specific genes. Here we report differential expression of several GNG-specific genes in the head and body of a model organism, Daphnia magna, a planktonic crustacean, in normoxic and acute hypoxic conditions. We predict that GNG-specific transcripts will be enriched in the body, where most of the fat tissue is located, r

PFKFB3-dependent glucose metabolism regulates 3T3-L1 adipocyte development.

Proliferation and differentiation of preadipocytes, and other cell types, is accompanied by an increase in glucose uptake. Previous work showed that a pulse of high glucose was required during the first 3 days of differentiation in vitro, but was not required after that. The specific glucose metabolism pathways required for adipocyte differentiation are unknown. Herein, we used 3T3-L1 adipocytes as a model system to study glucose metabolism and expansion of the adipocyte metabolome during the first 3 days of differentiation. Our primary outcome measures were GLUT4 and adiponectin, key proteins

Metabolomics and correlation network analyses of core biomarkers in type 2 diabetes.

The identification of metabolic pathways and the core metabolites provide novel molecular targets for the prevention and treatment of diseases. Diabetes is often accompanied with multiple metabolic disorders including hyperglycemia and dyslipidemia. Analysis of the variances of plasma metabolites is critical for identifying potential therapeutic targets for diabetes. In the current study, non-diabetic subjects with normal glucose tolerance and diabetics (age 40-60 years; n = 42 per group) were selected and plasma samples were analyzed by GC-MS for various metabolites profiling followed by netw

Low metformin causes a more oxidized mitochondrial NADH/NAD redox state in hepatocytes and inhibits gluconeogenesis by a redox-independent mechanism.

The mechanisms by which metformin (dimethylbiguanide) inhibits hepatic gluconeogenesis at concentrations relevant for type 2 diabetes therapy remain debated. Two proposed mechanisms are 1) inhibition of mitochondrial Complex 1 with consequent compromised ATP and AMP homeostasis or 2) inhibition of mitochondrial glycerophosphate dehydrogenase (mGPDH) and thereby attenuated transfer of reducing equivalents from the cytoplasm to mitochondria, resulting in a raised lactate/pyruvate ratio and redox-dependent inhibition of gluconeogenesis from reduced but not oxidized substrates. Here, we show that

Efficacy of asiatic acid, a pentacyclic triterpene on attenuating the key enzymes activities of carbohydrate metabolism in streptozotocin-induced diabetic rats.

Asiatic acid (AA), a triterpenoid derivative of Centella asiatica, has shown significant biological effects of antioxidant and anti-inflammatory activities. Aim of this investigation was to evaluate the antihyperglycemic effect of AA on the activities of hepatic enzymes of carbohydrate metabolism in streptozotocin (STZ)-induced diabetic rats. To induce diabetes mellitus, rats were injected with streptozotocin intraperitoneally at a single dose of 40 mg/kg b.w. Diabetic rats showed significant (p<0.05) increased in plasma glucose, glycosylated hemoglobin and significant (p<0.05) decreased in ci

Disulfiram and Its Derivatives: An Immortal Phoenix of Drug Repurposing.

Disulfiram (DSF) is a well-established inhibitor of aldehyde dehydrogenases (ALDHs) and an FDA-approved drug for chronic alcoholism. DSF has gained attention as a versatile scaffold for drug repurposing. Its metabolite, diethyldithiocarbamate (DDTC), mediates multiple biological effects via metal chelation and covalent modification of key cysteine residues. Beyond its established anticancer properties, DSF modulates cancer stem cells, reactive oxygen species, proteasome function, and drug-resistance pathways. It also shows promise in metabolic disorders, including type 2 diabetes and obesity,

Pdcd4-Rictor interaction suppresses mTORC2-PFKFB3 axis and tumorigenesis in NSCLC.

Programmed cell death 4 (Pdcd4) is a well-established tumor suppressor as well as an inhibitor of protein translation. Although Pdcd4-mediated translational repression contributes to tumor suppression, emerging evidence suggests that Pdcd4 also exerts translation-independent functions. In this study, we found that Pdcd4 suppresses tumorigenesis through disrupting mTORC2 complex formation by binding with the rapamycin-insensitive companion of mTOR (Rictor), a core component of the mTORC2 complex. Using deletion mapping and site-directed mutagenesis, we defined the Rictor-binding domain of Pdcd4

GSM1 Requires Hap4 for Expression and Plays a Role in Gluconeogenesis and Utilization of Nonfermentable Carbon Sources.

Multiple transcription factors in the budding yeast Saccharomyces cerevisiae are required for the switch from fermentative growth to respiratory growth. The Hap2/3/4/5 complex is a transcriptional activator that binds to CCAAT sequence elements in the promoters of many genes involved in the tricarboxylic acid cycle and oxidative phosphorylation and activates gene expression. Adr1 and Cat8 are required to activate the expression of genes involved in the glyoxylate cycle, gluconeogenesis, and utilization of nonfermentable carbon sources. Here, we characterize the regulation and function of the z

Functional characterization of Yersinia pestis aerobic glycerol metabolism.

Yersinia pestis biovar Orientalis isolates have lost the capacity to ferment glycerol. Herein we provide experimental validation that a 93 bp in-frame deletion within the glpD gene encoding the glycerol-3-phosphate dehydrogenase present in all biovar Orientalis strains is sufficient to disrupt aerobic glycerol fermentation. Furthermore, the inability to ferment glycerol is often insured by a variety of additional mutations within the glpFKX operon which prevents glycerol internalization and conversion to glycerol-3-phosphate. The physiological impact of functional glpFKX in the presence of dys

Neuroprotective Effects of Ethiopian Coffee Beans against Hyperglycemia-induced Brain Injury in Rats.

At least 50% of people with diabetes suffer from one or more complications if their conditions are not adequately managed over time. Diabetic neuropathy is one of the prevalent complications of diabetes, which also includes diabetic nephropathy, retinopathy, cardiomyopathy, and diabetic foot diseases. The present study evaluated the protective effects of Ethiopian coffee beans (Coffea arabica) against glucose-induced brain tissue injury using in vitro, ex vivo, and in silico experimental models. Oxidative injury was induced by incubating brain tissue collected from normal male Sprague-Dawley r
