Interplay between stress granule-like structures and nuclear RNA export in a heat shock-specific manner.

When stress strikes, cells initiate adaptive responses, including perturbation of RNA splicing, modulation of nuclear RNA export, and formation of stress granules (SGs). Despite their central role, the composition and regulation of fungal SGs remain poorly understood. Here, we report the formation of heat shock-induced SG-like structures in Fusarium graminearum. Through purification of SG-like structures followed by RNA-sequencing, we performed a transcriptome-wide characterization of their associated transcripts and found that heat shock induces a distinct enrichment of intron-containing tran

Cryo-Electron Tomography of Candida glabrata Plasma Membrane Proteins.

Fungal plasma membrane proteins have long been recognized as targets for the development of antifungal agents. Despite recent progress in experimental approaches and computational structural predictions, our knowledge of the structural dynamics and spatial distribution of these membrane proteins in the context of their native lipid environment remains limited. By applying cryo-electron tomography (cryoET) and subtomogram analysis, we aim to characterize the structural characteristics and spatial distribution of membrane proteins present in Candida glabrata plasma membranes. This study has resu

Fine-needle aspiration biopsy of disseminated sporotrichosis: a case report.

In this report, we describe a case of disseminated sporotrichosis that was diagnosed by fine-needle aspiration biopsy (FNAB). The cytologic smears exhibited a large number of macrophages, few polymorphonuclear neutrophils and numerous round or oval, sometimes elongated, isolated and scattered yeast-like structures localized extracellularly or inside macrophages. These structures were clearly visualized by Giemsa and Papanicolaou methods. Cultures from skin biopsy material revealed fungal colonies which were subsequently identified as Sporothrix schenckii. The cytologic aspects, the correlation

Histology-Correlated FTIR Chemical Imaging of Fungal Infection-Associated Tissue Compartments in Human Skin: A Proof-of-Concept Study.

Background/Objectives: Fungal skin infections are commonly assessed using clinical examination and conventional histopathology, including hematoxylin-eosin (HE), periodic acid-Schiff (PAS), and Grocott methenamine silver (GMS) staining. However, these methods provide limited spatially resolved biochemical information. This proof-of-concept study investigated whether Fourier transform infrared (FTIR) chemical imaging can identify biochemical patterns associated with histologically defined fungal infection-associated tissue compartments in human skin. Methods: Archived formalin-fixed, paraffin-e

Proteinous Components of Neutrophil Extracellular Traps Are Arrested by the Cell Wall Proteins of Candida albicans during Fungal Infection, and Can Be Used in the Host Invasion.

One of defense mechanisms of the human immune system to counteract infection by the opportunistic fungal pathogen Candida albicans is the recruitment of neutrophils to the site of invasion, and the subsequent production of neutrophil extracellular traps (NETs) that efficiently capture and kill the invader cells. In the current study, we demonstrate that within these structures composed of chromatin and proteins, the latter play a pivotal role in the entrapment of the fungal pathogen. The proteinous components of NETs, such as the granular enzymes elastase, myeloperoxidase and lactotransferrin,

Etiological agents of otomycosis in Nigeria.

A total of 159 suspected cases of otomycosis comprising 101 adults and 58 children, 61 males and 98 females were investigated. Of these, 36 cases were confirmed specifically of mycotic etiology on the basis of microscopic demonstration of fungal structures in epithelial debris/plugs and positive culture. Another 31 cases positive for fungi by culture but negative for direct microscopy were considered of doubtful fungal etiology. The predominant etiological agents in the confirmed cases were Aspergillus niger (13 cases), followed by A. flavus (6), Candida albicans (6) and C. parapsilosis (4). O

Mixed pulmonary infection with Strongyloides stercoralis and Blastomyces dermatitidis.

We report the first case of mixed pulmonary infection with Strongyloides stercoralis and Blastomyces dermatitidis. Histopathology from the lung biopsy showed structures consistent with B. dermatitidis and S. stercoralis. A parasitology exam from a bronchi alveolar lavage yielded an immature rhabditiform larva and female worm. Fungal cultures grew B. dermatitidis.

Insights into the Metabolomic Capacity of Podaxis and Isolation of Podaxisterols A-D, Ergosterol Derivatives Carrying Nitrosyl Cyanide-Derived Modifications.

Cultures of a termite-associated and a free-living member of the fungal genus Podaxis, revived from spores maintained in century-old herbarium collections, were analyzed for their insecticidal and antimicrobial effects. Their secondary metabolomes were explored to uncover possible adaptive mechanisms of termite association, and dereplication of LC-HRMS/MS data sets led to the isolation of podaxisterols A-D (1-4), modified ergosterol derivatives that result from a Diels-Alder reaction with endogenous nitrosyl cyanide. Chemical structures were determined based on HRMS/MS and NMR analyses as well

Bilateral proptosis-an unusual presentation of extensive allergic fungal sinusitis.

Allergic fungal rhinosinusitis (AFRS) is counted as the most common form of fungal sinusitis. It is mainly due to the hypersensitivity reaction to fungal infection. Usually, the patients are atopic or immunocompetent. These patients are usually suffering from signs and symptoms of rhinosinusitis. The expanding mass in the disease leads to bony remodeling and the involvement of adjacent structures. When allergic mucin involves the orbit, many complications may occur. This includes diplopia, telecanthus, unilateral proptosis, malar flattening, epiphora, Asthenopia and even visual loss. The diagn

Distinct Antifungal Ingredients Deploying β-Alkylated (Hetero)arylethylamines Inspired by Fluopyram.

Inspired by the unique profiles of the succinate dehydrogenase inhibitor (SDHI) fungicides empowered by the introduction of β-arylethylamines, we elaborated new antifungal ingredients incorporating β-(hetero)arylethylamines inspired by fluopyram. Thirteen kinds of (hetero)arylethylamines, featuring various heterocycles and concomitant β-modifications, were deployed for achieving β-quinolinyl cyclohexylmethanamine as a novel pharmacophore through an antifungal screen. The subsequent fine-tuning was conducted conveniently by employing commercially inexpensive and easily available acid segments.

The lichen secondary metabolite lichesterinic acid exhibits antibiofilm activity against fungal pathogens.

Lichens are well known for producing unique secondary metabolites, some of which have been shown to exhibit medically relevant bioactivities, including antimicrobial effects. With the increasing prevalence of fungal infections and the growing resistance to commonly used antimycotics, there is an urgent need for new antifungal agents, especially for aged and/or immunocompromised individuals. In this study, we screened a collection of lichen-derived metabolites for antifungal properties in two medically relevant fungal pathogens, Candida albicans and Nakaseomyces glabratus. Several compounds exh

Recurrent Prosthetic Aortic Valve Aspergillus Endocarditis Requiring Fourth Redo Root Surgery.

BACKGROUND: Fungal prosthetic valve endocarditis is rare, frequently culture-negative, and often complicated by embolization, making diagnosis challenging. CASE SUMMARY: A 58-year-old man with prior bioprosthetic aortic valve surgery presented with splenic infarction and prosthetic valve dysfunction. Transesophageal echocardiography demonstrated an immobile echodensity encasing the valve, with elevated gradients. Blood cultures and serologic testing were repeatedly negative, and he was initially treated for presumed valve thrombosis. Despite therapeutic anticoagulation, he developed recurrent

Nanoparticle decoration impacts airborne fungal pathobiology.

Airborne fungal pathogens, predominantly Aspergillus fumigatus, can cause severe respiratory tract diseases. Here we show that in environments, fungal spores can already be decorated with nanoparticles. Using representative controlled nanoparticle models, we demonstrate that various nanoparticles, but not microparticles, rapidly and stably associate with spores, without specific functionalization. Nanoparticle-spore complex formation was enhanced by small nanoparticle size rather than by material, charge, or "stealth" modifications and was concentration-dependently reduced by the formation of

Cell wall nanoparticles from hyphae of Alternaria infectoria grown with caspofungin, nikkomycin, or pyroquilon trigger different activation profiles in macrophages.

Alternaria infectoria causes opportunistic human infections and is a source of allergens leading to respiratory allergies. In this work, we prepared cell wall nanoparticles (CWNPs) as a novel approach to study macrophage immunomodulation by fungal hyphal cell walls. A. infectoria was grown in the presence of caspofungin, an inhibitor of β(1,3)-glucan synthesis; nikkomycin Z, an inhibitor of chitin synthases; and pyroquilon, an inhibitor of dihydroxynaphthalene (DHN)-melanin synthesis. Distinct CWNPs were obtained from these cultures, referred to as casCWNPs, nkCWNPs, and pyrCWNPs, respectively
