BACKGROUND: SARS-CoV-2 infection depends on the host cell factors angiotensin-converting enzyme 2, ACE2, and the transmembrane serinprotease 2, TMPRSS2. Potential inhibitors of these proteins would be ideal targets against severe acute respiratory syndrome coronavirus type 2 (SARS-CoV-2) infection. Our data opens the possibility that changes within TMPRSS2 can modulate the outcome during a SARS-CoV-2 infection.
RESULTS: We reveal that TMPRSS2 acts not only during viral entry but has also an important role during viral replication. In addition to previous functions for TMPRSS2 during viral ent

The e antigen of hepatitis B (HBeAg) is positively associated with an increased risk of developing liver cancer and cirrhosis in chronic hepatitis B (CHB) patients. Clinical monitoring of HBeAg provides guidance to the treatment of CHB and the assessment of disease progression. We describe here an affinity binding assay for HBeAg, which takes advantage of G-quadruplex aptamers for enhanced binding and stability. We demonstrate a strategy to improve the binding affinity of aptamers by modifying their sequences upon their G-quadruplex and secondary structures. On the basis of predicting a stable

A novel fluorescent aptasensor based on the G-quadruplex induced fluorescent quenching of psoralen and the competitive interactions between 4'-aminomethyl-4,5',8-trimethylpsoralen (AMT), adenosine triphosphate (ATP) and G-rich DNA functionalized split ATP aptamer was proposed. The binding of ATP to the G-rich DNA functionalized split aptamer induced a significant enhancement in fluorescence emission intensity while undergoing excitation at 340 nm. Under the optimal conditions, the developed aptasensor showed high selectivity and good accuracy for detecting ATP. The practicality of the proposed

G-quadruplexes are DNA or RNA secondary structures that can be formed from guanine-rich nucleic acids. These four-stranded structures, composed of stacked quartets of guanine bases, can be highly stable and have been demonstrated to occur in vivo in the DNA of human cells and other systems, where they play important biological roles, influencing processes such as telomere maintenance, DNA replication and transcription, or, in the case of RNA G-quadruplexes, RNA translation and processing. We report for the first time that DNA G-quadruplexes can be detected in the nuclei of the malaria parasite

In this work we use a water-soluble cationic perylene derivative (compound 1) as the G-quadruplex (G4) structure fluorescence indicator to construct a fluorescent biosensor for simple, rapid and label-free detection of Pb(2+). In the absence of Pb(2+), strong electrostatic interactions between compound 1 and the G-rich DNA probe (PW17) induced the aggregation of compound 1 and resulted in the fluorescence quenching. In the presence of Pb(2+), the PW17 formed Pb(2+)-stabilized G4 structure, which reduced the aggregation of compound 1 and gave rise to high fluorescence. This allowed us to use co

This work concerns an analysis of the binding mechanism of a copper phthalocyanine (Alcian Blue-tetrakis(methylpyridinium) chloride, ABTP) to natural calf thymus DNA, G-quadruplexes (G4) and synthetic RNA polynucleotides in the form of double polyriboadenylic·polyribouridylic acid (poly(A)·poly(U)) or triple strands polyriboadenylic·2polyribouridylic acid (poly(A)·2poly(U)). ABTP is a well know dye that might undergo novel applications, but its interaction with DNA is scarcely studied and we lack information on possible RNA or G4 binding. This might be related to system complexity due to the p

Human neutrophils are the first line of defense against bacterial and viral infections. They eliminate pathogens through phagocytosis, which activate the 5-lipoxygenase (5-LOX) pathway resulting in synthesis of leukotrienes. Using HPLC analysis, flow cytometry, and other biochemical methods, we studied the effect of synthetic oligodeoxyribonucleotides (ODNs) able to fold into G-quadruplex structures on the main functions of neutrophils. Designed ODNs contained four human telomere TTAGGG repeats (G4) including those with phosphorothioate oligoguanosines attached to the end(s) of G-quadruplex co

G-quadruplex motifs are known to be present in telomeres of human and other organisms. Recent bioinformatic studies also revealed the widespread existence of these motifs in promoter regions of human genes. Treatment of cultured cells with 5-bromo-2'-deoxyuridine ((Br)dU) is known to result in the substitution of DNA thymidine with (Br)dU; such replacement has been shown to sensitize cells to killing induced by UV light. Our previous studies revealed that the exposure of (Br)dU-carrying duplex DNA or (Br)dU-treated MCF-7 cells to UVB light could lead to the facile formation of intrastrand cros

Targeting G-quadruplexes (G4s) presents a promising strategy for cancer therapy, but early-generation ligands often lack tumor selectivity. This study identified X-3, a novel photoactive coumarin-benzothiazolium conjugate, as a dual-targeted photosensitizer against triple-negative breast cancer (TNBC). X-3 localized to both mitochondria and the nucleus, where it specifically bound to mitochondrial and c-Myc G4s with high affinity. Upon light activation, X-3 demonstrated excellent phototoxicity and minimal dark toxicity. Mechanistically, X-3 disrupted mitochondrial bioenergetics by inhibiting m

A dual channel method is described for the determination of the amyloid-β peptide Aβ(1-40) that is associated with Alzheimer's disease. The method exploits (a) conformational changes of a G-quadruplex that are triggered by Na+ and K+ ions and (b) the strong affinity between Aβ(1-40) and Cu2+. A G-quadruplex DNA forms an antiparallel structure in the presence of Na+ and can catalyze the oxidation of tetramethylbenzidine by H2O2 system in the presence of Cu2+ to form a visible blue color. If, however, Cu2+ binds to Aβ(1-40), the blue color is no longer formed. Measuring the absorption decrease a

RAS mutations are major drivers of tumorigenesis and represent important therapeutic targets; however, most remain resistant to effective pharmacological inhibition. KRAS and NRAS mRNAs contain guanine (G)-rich regions, forming stable G-quadruplexes (G4s) that regulate translation. Targeting and stabilizing these G4 structures with specific ligands may suppress their expression, offering a potential therapeutic strategy for RAS-driven cancers. BYBC-1, a novel G4-RNA-targeting ligand, shows strong affinity (Kd = 0.05-0.28 μM) for G4-RNAs, particularly KRAS and NRAS, highlighting its promise as

INTRODUCTION: RNA G-quadruplexes (rG4s) are guanine-rich secondary RNA structures that regulate multiple aspects of post-transcriptional gene expression, including RNA stability, splicing, localization, and translation. Emerging evidence suggests that rG4s contribute to the pathogenesis of neurodegenerative diseases; however, the transcriptomic landscape profile of experimentally validated rG4-interacting genes in Parkinson's disease (PD) remains poorly characterized. This study aimed to investigate the gene expression profiles of rG4-associated genes in PD and examine the relationships betwee

Mitochondrial localization concentrates photosensitizing molecules near redox-sensitive organelles. In this study, we investigated SPN, a mitochondria-targeted G-quadruplex (G4)-responsive ligand. Photochemical characterization confirmed that SPN generates robust light-triggered ROS and exhibits photooxidative activity in vitro. In cancer cell models under dark conditions, SPN treatment was associated with cell-cycle perturbation, loss of mitochondrial membrane potential, altered mitochondrial morphology, elevated mitochondrial superoxide-sensitive fluorescence, and accumulation of MDC-positiv

RNA G-quadruplexes (rG4s), formed through guanine self-recognition into stacked tetrads, serve as critical regulators of gene expression, yet their comprehensive mapping and dynamic regulation in physiological contexts remain technically challenging. Here, we develop Ultra-low-input rG4-seq (ULI-rG4-seq), enabling precise rG4 detection enabling precise rG4 detection with ∼140 bp resolution in samples as small as 100 oocytes, and reveal notable enrichment of rG4s near crucial regulatory regions, particularly transcription start sites and end sites. This technological advance, combined with Trim

Real-time monitoring of viral replication is essential for infectious disease diagnosis and antiviral drug development. The G-quadruplex (G4), a conserved regulatory element within viral genomes, represents a significant endogenous biomarker for tracking viral activity. However, imaging viral G4s in deep tissues remains a challenge for current optical technologies due to severe photon attenuation and autofluorescence. Herein, we report Lumin680, the first near-infrared (NIR) chemiluminescent probe directly activated by conserved viral G4 conformations. Its chemiluminescence was triggered by pa

Ischemic stroke creates a hypoxic and nutrient-deprived microenvironment that rapidly activates microglia, a central driver of post-ischemic inflammation and a key determinant of secondary tissue damage and neurological recovery. G-quadruplex (G4) structures are non-canonical nucleic acid conformations that can reshape stress responses in diverse settings, yet whether microglial DNA G4 relates to ischemic pathology remains unknown. Here, using a transient middle cerebral artery occlusion (tMCAO) mouse model and an oxygen-glucose deprivation/reoxygenation (OGD/R) primary microglia model, we exa

The G-quadruplex structure in the human telomerase reverse transcriptase (hTERT) promoter has recently emerged as a promising therapeutic target in cancer. However, the development and application are strongly restricted by the lack of selective ligands. Herein, levo-tetrahydropalmatine (l-THP) and its two metabolic derivatives (M1 and M2) were identified as novel ligands of the hTERT G-quadruplex using a combination of fluorescence spectroscopy, UV-visible absorption, circular dichroism (CD), CD-melting assays, electrophoretic mobility shift assays (EMSA), molecular docking, as well as cellul

BACKGROUND: Epigenetic evolution is a common mechanism used by cancer cells to evade the therapeutic effects of drug treatment. In ovarian cancers, epigenetically driven resistance is thought to be responsible for many late-stage patient deaths. DNA secondary structures called G-quadruplexes (G4s) are emerging as potential epigenetic marks of relevance to cancer evolution, but their prevalence and distribution in ovarian cancer models have never been investigated before.
RESULTS: Here, we describe the first investigation of the role of G4s in the epigenetic regulation of drug-resistant ovaria

Expansion of intronic GGGGCC repeats in the C9orf72 gene causes amyotrophic lateral sclerosis (ALS) and frontotemporal dementia. Transcription of the expanded repeats results in the formation of RNA-containing nuclear foci and altered RNA metabolism. In addition, repeat-associated non-AUG (RAN) translation of the expanded GGGGCC-repeat sequence results in the production of highly toxic dipeptide-repeat (DPR) proteins. GGGGCC repeat-containing transcripts form G-quadruplexes, which are associated with formation of RNA foci and RAN translation. Zfp106, an RNA-binding protein essential for motor

Two new ligands (TPB3P and TPB3Py) showing a strong stabilisation effect and good selectivity for G4 over duplex DNAs have been synthesised. The ligands hold three analogous polyamine pendant arms (TPA3P and TPA3Py) but differ in the central aromatic core, which is a triphenylbenzene moiety instead of a triphenylamine moiety. Both TPB3P and TPB3Py exhibit high cytotoxicity in MCF-7, LN229 and HeLa cancer cells in contrast to TPA-based ligands, which exhibit no significant cytotoxicity. Moreover, the most potent G4 binders have been encapsulated in liposomes and AS1411 aptamer-targeted liposome
