CLEC16A in astrocytes promotes mitophagy and limits pathology in a multiple sclerosis mouse model.

Astrocytes promote neuroinflammation and neurodegeneration in multiple sclerosis (MS) through cell-intrinsic activities and their ability to recruit and activate other cell types. In a genome-wide CRISPR-based forward genetic screen investigating regulators of astrocyte proinflammatory responses, we identified the C-type lectin domain-containing 16A gene (CLEC16A), linked to MS susceptibility, as a suppressor of nuclear factor-κB (NF-κB) signaling. Gene and small-molecule perturbation studies in mouse primary and human embryonic stem cell-derived astrocytes in combination with multiomic analys

Investigating the Crime Scene-Molecular Signatures in Inflammatory Bowel Disease.

Inflammatory bowel diseases (IBD) are without cure and troublesome to manage because of the considerable diversity between patients and the lack of reliable biomarkers. Several studies have demonstrated that diet, gut microbiota, genetics and other patient factors are essential for disease occurrence and progression. Understanding the link between these factors is crucial for identifying molecular signatures that identify biomarkers to advance the management of IBD. Recent technological breakthroughs and data integration have fuelled the intensity of this research. This research demonstrates t

Targeting pyroptosis in intestinal and systemic inflammatory diseases: mechanisms and therapeutic opportunities.

Pyroptosis is a lytic form of programmed cell death driven by gasdermin cleavage and subsequent plasma membrane pore formation, which triggers cell swelling, lysis, and the release of inflammatory cytokines that amplify tissue injury. Once viewed as an innate antimicrobial defense mechanism, pyroptosis is now recognized as a central pathogenic pathway in both acute and chronic inflammatory diseases, including inflammatory bowel disease (IBD). Its execution proceeds through a cascade of druggable events-inflammasome assembly, caspase activation, and gasdermin pore formation-and it operates with

Pyroptosis in triple-negative breast cancer: From inflammasome signaling to tumor immune remodeling and translational therapeutic strategies.

Triple-negative breast cancer (TNBC) remains an aggressive breast cancer subtype marked by molecular heterogeneity, immune evasion, limited actionable targets, and variable responses to immune-based therapy. Pyroptosis, a gasdermin-mediated inflammatory form of regulated cell death, may link tumor-cell injury to remodeling of the tumor immune microenvironment (TIME). However, its role in TNBC should not be interpreted as uniformly antitumor. Instead, pyroptotic signaling represents a context-dependent inflammatory process shaped by inflammasome activation, gasdermin cleavage, cellular source,

Targeting Pyroptotic Cell Death Pathways in Retinal Disease.

Pyroptosis is a gasdermin-mediated, pro-inflammatory form of cell death distinct from apoptosis. In recent years, increasing attention has shifted toward pyroptosis as more studies demonstrate its involvement in diverse inflammatory disease states, including retinal diseases. This review discusses how currently known pyroptotic cell death pathways have been implicated in models of age-related macular degeneration, diabetic retinopathy, and glaucoma. We also identify potential future therapeutic strategies for these retinopathies that target drivers of pyroptotic cell death. Presently, the driv

Baicalein attenuates atherogenesis by suppressing endothelial pyroptosis via cylindromatosis-mediated tumor necrosis factor receptor-associated factor 6 deubiquitination.

BACKGROUND: Endothelial pyroptosis is a critical contributor to the development of atherosclerosis. Baicalein (BAI) exhibits anti-atherosclerotic potential; however, whether it alleviates atherosclerosis by modulating endothelial pyroptosis and the underlying molecular mechanisms remain unclear. PURPOSE: This study aimed to elucidate the mechanism by which baicalein attenuates atherosclerosis, with a particular focus on Cylindromatosis (CYLD)-mediated deubiquitination of tumor necrosis factor receptor-associated factor 6 (TRAF6). METHODS: Atherosclerosis was induced in ApoE⁻/⁻ mice, and ox-L

Structural and molecular principles of DAMP biology.

Damage-associated molecular patterns (DAMPs) are endogenous danger signals. They can be preformed molecules released upon membrane rupture and stress-induced or newly generated factors arising during cell death. These signals link cellular demise to diverse host responses. Rather than passive by-products, DAMPs are actively mobilized through membrane-remodeling proteins, vesicular trafficking and metabolic regulation. Conformational changes, oligomerization and post-translational modifications shape their release and immunogenicity, as illustrated by redox-dependent DAMP states, pore-forming g

MyD88 signaling in myeloid cells induces gastrointestinal tract injury and systemic inflammation after West Nile virus infection.

Humans with autoantibodies to type I interferons (IFNs) are at higher risk for neurological disease after West Nile virus (WNV) infection. IFN signaling deficient WNV-infected mice develop greater inflammation in the brain, which correlates with loss of gut barrier integrity. Here, we investigate the immune pathways that mediate this virus-gut-brain axis of inflammation. Myd88-/- mice administered anti-IFNAR1 antibody and infected with WNV exhibit reduced GI tract damage and inflammation. GI tract injury caused by WNV infection requires gasdermins or IL-1 receptor expression in hematopoietic c

Role for Caspase-8 in the Release of IL-1β and Active Caspase-1 from Viable Human Monocytes during Toxoplasma gondii Infection.

Monocytes are actively recruited to sites of infection and produce the potent proinflammatory cytokine IL-1β. We previously showed that IL-1β release during Toxoplasma gondii infection of primary human monocytes requires the NLRP3 inflammasome and caspase-1 but is independent of gasdermin D and pyroptosis. To investigate mechanisms of IL-1β release, we generated caspase-1, -4, -5, or -8 knockout (KO) THP-1 monocytic cells. Genetic ablation of caspase-1 or -8, but not caspase-4 or -5, decreased IL-1β release during T. gondii infection without affecting cell death. In contrast, TNF-α and IL-6 se

Pyroptosis in urinary malignancies: a literature review.

Urinary neoplasms refer to malignant tumours occurring in any part of the urinary system, including the kidney, renal pelvis, ureter, bladder, prostate, etc. The worldwide incidence of urinary system tumours has been increasing yearly. Available methods include surgical treatment, radiotherapy, chemotherapy, endocrine therapy, molecular targeted therapy, and immune therapy. In recent years, emerging evidence has demonstrated that cell pyroptosis plays an important role in the occurrence and progression of malignant urinary tumours. Pyroptosis is a new type of cell death that involves inflammat

Pyroptosis in Periprosthetic Osteolysis.

Periprosthetic osteolysis (PPO) along with aseptic loosening (AL) caused by wear particles after artificial joint replacement is the key factor in surgical failure and subsequent revision surgery, however, the precise molecular mechanism underlying PPO remains unclear. Aseptic inflammation triggered by metal particles, resulting in the imbalance between bone formation by osteoblasts and bone resorption by osteoclasts may be the decisive factor. Pyroptosis is a new pro-inflammatory pattern of regulated cell death (RCD), mainly mediated by gasdermins (GSDMs) family, among which GSDMD is the best

Pyroptosis, A Double-Edged Sword in Respiratory Diseases: A Narrative Review.

Pyroptosis is an inflammatory form of programmed cell death (PCD) driven by the gasdermin protein family. This process begins when danger- or damage-associated molecular patterns (DAMPs) are recognized, triggering either the canonical Caspase-1 or non-canonical Caspase-4/5/11 pathways. These pathways activate the cleavage of gasdermin D (GSDMD), releasing N-terminal fragments that form pores in cell membranes. This pore formation increases membrane permeability, leading to cell swelling and eventual lytic rupture. Pyroptosis serves as a double-edged sword in innate immunity: it defends against

The Multifaceted Role of Pyroptosis: Molecular Mechanisms, Crosstalk with Other Cell Death Pathways, and Therapeutic Implications.

Pyroptosis is a tightly regulated, proinflammatory form of programmed cell death defined by gasdermin-mediated membrane pore formation and cytokine release. By coupling cell death to innate immune activation, pyroptosis occupies a unique position at the interface of host defense and tissue pathology. Recent advances have demonstrated that pyroptosis is activated through multiple molecular routes, including canonical and noncanonical inflammasome pathways, Caspase-3/8-gasdermin axes, and granzyme-dependent mechanisms. Importantly, pyroptosis does not operate in isolation but engages in extensiv

Neutrophil extracellular traps and pyroptosis: a molecular nexus linking systemic autoimmune diseases to atherosclerosis.

This review aims to elucidate the molecular mechanisms of neutrophil extracellular traps (NETs) and pyroptosis, exploring their synergistic role as a pathological nexus bridging systemic autoimmune diseases with atherosclerosis. By systematically synthesizing recent literature and experimental evidence, we delineate the signaling cascades of these programmed inflammatory processes and analyze their contributions to disease progression in systemic lupus erythematosus, rheumatoid arthritis, and cardiovascular pathology. The analysis reveals a sophisticated bidirectional crosstalk, termed the NET

Enhanced Mitochondrial Targeting and Inhibition of Pyroptosis with Multifunctional Metallopolyphenol Nanoparticles in Intervertebral Disc Degeneration.

Intervertebral disc degeneration (IVDD) is a significant contributor to low back pain, characterized by excessive reactive oxygen species generation and inflammation-induced pyroptosis. Unfortunately, there are currently no specific molecules or materials available to effectively delay IVDD. This study develops a multifunctional full name of PG@Cu nanoparticle network (PG@Cu). A designed pentapeptide, bonded on PG@Cu nanoparticles via a Schiff base bond, imparts multifunctionality to the metal polyphenol particles (PG@Cu-FP). PG@Cu-FP exhibits enhanced escape from lysosomal capture, enabling e

Oropouche virus infection induces pyroptosis in human THP-1 macrophages.

Oropouche fever, an emerging zoonotic viral disease in Central and South America, is caused by Oropouche virus (OROV). While typically self-limiting, severe complications such as aseptic meningoencephalitis, miscarriage, and neonatal malformations can occur. Macrophages are critical in host defense, but the pathological mechanisms underlying OROV infection remain unclear. IL-1β, a key pro-inflammatory cytokine, plays a central role in the febrile response and is regulated by inflammasomes, such as NLRP3. This study investigates NLRP3-mediated IL-1β maturation and pyroptotic cell death in OROV-

Inducers and Inhibitors of Pyroptotic Death of Granulosa Cells in Models of Premature Ovarian Insufficiency and Polycystic Ovary Syndrome.

Granulosa cells (GCs), the largest cell population and primary source of steroid hormones in the ovary, are the important somatic ovarian components. They have critical roles in folliculogenesis by supporting oocyte, facilitating its growth, and providing a microenvironment suitable for follicular development and oocyte maturation, thus having essential functions in maintaining female fertility and in reproductive health in general. Pyroptotic death of GCs and associated inflammation have been implicated in the pathogenesis of several reproductive disorders in females including Premature Ovari

Pyroptosis in Alzheimer's disease: Mechanisms and neuroinflammatory networks.

Alzheimer's disease (AD) is a neurodegenerative disorder pathologically characterized by amyloid-β (Aβ) deposition, tau protein hyperphosphorylation, neuronal loss, and sustained neuroinflammation. In recent years, pyroptosis, a gasdermin-mediated form of inflammatory programmed cell death, has been recognized as a potential mechanism linking innate immune activation to neurodegenerative injury. This review summarizes the major molecular pathways of pyroptosis, including the canonical inflammasome-caspase-1-GSDMD pathway, the noncanonical caspase-4/5/11-GSDMD pathway, and alternative pathways

Antifungal Treatment Aggravates Sepsis through the Elimination of Intestinal Fungi.

Prophylactic antifungal therapy is widely adopted clinically for critical patients and effective in reducing the morbidity of invasive fungal infection and improves outcomes of those diagnosed patients; however, it is not associated with higher overall survival. As intestinal commensal fungi play a fundamental role in the host immune response in health and disease, we propose that antifungal therapy may eliminate intestinal fungi and aggravate another critical syndrome, sepsis. Here, with murine sepsis model, we found that antifungal therapy with fluconazole dismissed intestinal fungal burden

Perturbation of azurophilic granule integrity drives NLRP3-independent IL-1β processing and release in neutrophils.

Interleukin 1-beta (IL-1β) is an inflammatory cytokine produced by myeloid cells in response to infection or sterile tissue damage. Secretion of bioactive IL-1β from macrophages (Mφ) or dendritic cells (DC) downstream of activated NLRP3/caspase-1 inflammasomes is the best characterized model; this is mediated by caspase-1 cleavage of proIL-1β and Gasdermin D. Gasdermin D pores that form in the plasma membrane mediate IL-1β release and pyroptotic cell death. NLRP3 inflammasome assembly is triggered by perturbation of ionic, metabolic or organelle homeostasis via diverse stimuli. A recent report
