Researchers are often interested in understanding the disease subtype heterogeneity by testing whether a risk exposure has the same level of effect on different disease subtypes. The polytomous logistic regression (PLR) model provides a flexible tool for such an evaluation. Disease subtype heterogeneity can also be investigated with a case-only study that uses a case-case comparison procedure to directly assess the difference between risk effects on two disease subtypes. Motivated by a large consortium project on the genetic basis of non-Hodgkin lymphoma (NHL) subtypes, we develop PolyGIM, a p

BACKGROUND: Consent2Share (C2S) is an open source software created by the Office of the National Coordinator Data Segmentation for Privacy initiative to support electronic health record (EHR) granular segmentation. To date, there are no published formal evaluations of Consent2Share.
METHOD: Structured data (e.g. medications) codified using standard clinical terminologies (e.g. RxNorm) was extracted from the EHR of 36 patients with behavioral health conditions from study sites. EHRs were available through a health information exchange and two sites. The EHR data was already classified into dat

The debate continues about whether people have a duty to pass on the positive results of their genetic tests to relatives who are at risk from the same disease, and, should they refuse, whether physicians and genetic counselors then have the duty to do so. To date, the role and views of nurses in this debate have not been investigated. In our study, a sample of Israeli nurses, untrained in genetics, were asked for their theoretical opinions and what practical steps they would take in the case of patients' refusal to disclose. The nurses were very sure that patients should inform their families

BACKGROUND: Azoospermia, defined as the absence of sperm in the ejaculate, represents the most severe form of male infertility. Advancements in assisted reproductive technologies (ART) have greatly improved treatment options, allowing men with azoospermia to have biological children. While these men are significantly likely to experience psychosocial challenges due to the dual impact of infertility and ART treatment, few studies have explored their lived experiences in East Asian societies. Our study aimed to explore lived psychosocial experiences of ART treatment among men with azoospermia in

We describe a novel recessive and nonlethal pigmentation mutant in Xenopus tropicalis. The mutant phenotype can be initially observed in tadpoles after stage 39/40, when mutant embryos display markedly reduced pigmentation in the retina and the trunk. By tadpole stage 50 almost all pigmented melanophores have disappeared. Most interestingly, those embryos fail entirely to make pigmented iridophores. The combined reduction/absence of both pigmented iridophores and melanophores renders these embryos virtually transparent, permitting one to easily observe both the developing internal organs and n

BACKGROUND: Leukemia is the most common malignancy in children, while type 1 diabetes mellitus (T1DM) is one of the most prevalent autoimmune diseases among children. The etiology of both conditions is still largely unexplained, yet emerging evidence suggests certain similarities in environmental exposures and genetic predispositions. In this case-series, we sought a new potential factor behind the co-occurrence of leukemia and T1DM by thoroughly reviewing the medical records of patients diagnosed with both diseases.
METHODS: We conducted a retrospective case series at Tampere University Hosp

BACKGROUND: Cancer screening is a cornerstone of effective cancer prevention. The 2022 European Council Recommendation on cancer screening encourages risk-based approaches, including risk stratification, when appropriate, to improve the balance of screening benefits, harms, and resource use. To address conceptual heterogeneity and fragmented guidance across cancer sites and countries, the EUCanScreen Joint Action convened a multidisciplinary working group from 22 European countries to develop a shared conceptual framework for risk-stratified cancer screening, as a specific operational approach

BACKGROUND: The interpretation of pediatric electrocardiograms (ECGs) and management of childhood arrhythmias represent specialized clinical disciplines complicated by age-dependent physiological evolution. While artificial intelligence (AI) has transformed adult cardiology, its application to pediatric electrophysiology remains largely in the research phase.
OBJECTIVE: This review critically appraises the current evidence, methodological rigor, clinical readiness, and translational challenges of AI in pediatric arrhythmia detection, risk stratification, and management.
METHODS: A synthesis

INTRODUCTION: Fetal Alcohol Spectrum Disorder (FASD) is one of the leading non-genetic causes of developmental disability worldwide and is thought to be particularly common in the UK. Despite this, there is a lack of data on FASD in the UK.
OBJECTIVE: To conduct public and professional involvement work to establish stakeholder views on the feasibility, acceptability, key purposes, and design of a national linked longitudinal research database for FASD in the UK.
METHODS: We consulted with stakeholders using online workshops (one for adults with FASD [and their supporters] N = 5; one for care

Bioethics apparently suffers from a misdistribution of research resources analogous to the '10/90' divide in medical research. Though infectious disease should be recognized as a topic of primary importance for bioethics, the general topic of infectious disease has received relatively little attention from the discipline of bioethics in comparison with things like abortion, euthanasia, genetics, cloning, stem cell research, and so on. The fact that the historical and potential future consequences of infectious diseases are almost unrivalled is one reason that the topic of infectious disease wa

BACKGROUND: Previous studies have shown positive tangible health benefits from using online support communities for informational support, daily living support, and emotional support. The specifics of how these communities can be improved have not been studied in detail.
OBJECTIVE: This study will investigate various sources of information that patients with genetic disorders use to learn more about their condition. We will be using patients with Ehlers-Danlos Syndrome (EDS) as a typical representation of the wider group of patients with genetic disorders. This study will also investigate the

INTRODUCTION: Lifelong genomic medicine, in which stored genomic data informs a lifetime of care, is moving from vision toward reality, with dozens of newborn sequencing projects underway worldwide and reanalysis of data already in clinical practice. Professional perspectives on its value and feasibility have not been examined.
METHODS: We conducted and thematically analyzed semi-structured interviews with 52 US-based professionals, including clinical geneticists, genetic counselors, primary care clinicians, laboratory personnel, and genomic screening implementers. Interviews explored deliver

BACKGROUND: World Health Organisation (WHO) in 2024 identified that approximately one in 100 children globally has autism spectrum disorder (ASD). ASD is a collection of neurodevelopmental disorders that impact a person's ability to socially interact and communicate, which can typically be noticed in early childhood. While 'autism' as a term was initially used for schizophrenic patients, later psychiatrists Dr. Kanner and paediatrician Dr. Asperger introduced it as a syndrome in children with behavioural differences in social interaction and communication with restrictive and repetitive intere

In recent years, global health security has been threatened by the geographical expansion of vector-borne infectious diseases such as malaria, dengue, yellow fever, Zika and chikungunya. For a range of these vector-borne diseases, an increase in residual (exophagic) transmission together with ecological heterogeneity in everything from weather to local human migration and housing to mosquito species' behaviours presents many challenges to effective mosquito control. The novel use of drones (or uncrewed aerial vehicles) may play a major role in the success of mosquito surveillance and control p
