Genotyping of Burkholderia mallei from an outbreak of glanders in Bahrain suggests multiple introduction events.

BACKGROUND: Glanders, caused by the gram-negative bacterium Burkholderia mallei, is a highly infectious zoonotic disease of solipeds causing severe disease in animals and men. Although eradicated from many Western countries, it recently emerged in Asia, the Middle-East, Africa, and South America. Due to its rareness, little is known about outbreak dynamics of the disease and its epidemiology. METHODOLOGY/PRINCIPAL FINDINGS: We investigated a recent outbreak of glanders in Bahrain by applying high resolution genotyping (multiple locus variable number of tandem repeats, MLVA) and comparative wh

Comparative evaluation of three commercially available complement fixation test antigens for the diagnosis of glanders.

The sensitivity and specificity of three commercially available complement fixation test (CFT) antigens from c.c.pro (c.c.pro), Central Veterinary Institute of Wageningen UR (CIDC) and the United States Department of Agriculture (USDA) were comparatively evaluated by testing 410 sera collected from glanders-endemic and non-endemic areas (200 true-negative randomly collected sera and 210 sera collected from experimentally immunised animals (12 rabbits, 19 horses), clinically positive (135) and culture-positive (44) horses, donkeys and mules). Immunoblotting (IB) was used as the gold standard te

[Possible use of the indirect hemagglutination reaction for determining the sensitivity of the bacteria of glanders and melioidosis to antibiotics].

The use of IHAT for rapid assay of antibiotic sensitivity of bacteria causing malleus and melioidosis was shown to be possible. Pure bacterial cultures and suspensions of the spleen of infected animals were used as the investigation objects. The materials were additionally grown in meat-peptone broth with 5 per cent of glycerol at 37 degrees C with addition of an antibiotic or without it (control). Antibiotic efficacy was estimated by the difference in the titers in the main and control experiments. The test not requiring isolation of pure bacterial cultures provided a 2-3-fold decrease in the

Surveillance of equine glanders in India (2019-2023): Prevalence estimate, risk analysis and future strategy.

Equine glanders, a notifiable zoonotic disease caused by Burkholderia mallei, continues to pose a significant health and regulatory challenge in India. This study summarizes the outcomes of a nationwide sero surveillance programme conducted between January 2019 and December 2023 to assess the distribution and temporal trends of glanders in equid populations. A total of 158,798 serum samples collected from 391 districts across 19 states and three union territories were screened using Hcp1-based indirect ELISA, followed by confirmatory testing through the complement fixation test. Sequencing-Based Genotyping of Pakistani Burkholderia mallei Strains: A Useful Way for Investigating Glanders Outbreaks.

Burkholderia (B.) mallei is a host-adapted equine pathogen that causes glanders, a re-emerging zoonotic disease, which is endemic in Pakistan and other developing countries and seriously impacts the global equine movement. Due to globalization, the geographical restriction of diseases vanishes and the lack of awareness of and experience with eradicated diseases in industrialized countries also promotes the re-introduction of infections in these regions. Owing to the high equine population, the Pakistani province Punjab is a potential hotspot where several glanders outbreaks have been seen over

Improvement of recombinant-truncated Burkholderia motility protein A (BimA)-based indirect ELISA for equine glanders.

Glanders is a contagious and highly fatal disease of equines with zoonotic potential. It is caused by a Gram-negative, nonmotile bacterium Burkholderia mallei. Complement fixation test (CFT) is one of the most commonly used tests for diagnosis of glanders; however, it has some limitations. A recombinant-truncated Burkholderia intracellular motility A (BimA) protein-based indirect enzyme-linked immunosorbent assay (iELISA) was previously reported by us for glanders diagnosis, which has been re-optimized in this study using a panel of glanders positive (n = 75) and glanders negative (n = 227) se

Evaluation of serological responses in horses challenged with Burkholderia pseudomallei using current diagnostic tests for glanders.

Six horses were challenged experimentally with a strain of Burkholderia pseudomallei isolated from a fatal case of the infection in a dromedary camel years earlier in the Emirate of Dubai. Three horses were inoculated subcutaneously and in 3 the bacterium was administered by the oral route. Four of the horses became serologically positive based on reactions to one or more of the OIE described tests for glanders. B. pseudomallei was re-isolated from the 4 serological positive horses. Only one of the subcutaneously infected horses, developed fever for 3 days. The white blood cell values and the

Seroprevalence and associated epidemiological drivers of glanders in horses: a systematic review and meta-analysis (2002-2026).

Glanders (malleus), a highly contagious zoonotic disease caused by Burkholderia mallei, remains an important transboundary disease of equids despite extensive control efforts in many countries. However, its epidemiological patterns and associated drivers have not been comprehensively synthesized. This study aimed to estimate the seroprevalence of glanders in horses and identify key epidemiological drivers based on studies published up to April 27, 2026. Following PRISMA 2020 guidelines, systematic searches were conducted in PubMed, Scopus, Web of Science, ScienceDirect, Cochrane Library, ProQu

["Acute human glanders". Contribution for the scientific history of the Museum of pathological anatomy established in Trieste Hospital].

The Museum of Pathological Anatomy of the Regina Elena City Hospital of Trieste houses various pathological preparations of infective and contagious diseases, dating back to the early 1900's (ileo-typhus, dysentery, tuberculosis, syphillis, pulmonary plague, etc.) together with their relative diagnostic certificates. These bear witness to the key role of the Hospital's Anatomical Institute (in operation operating since 1872) during the height of the Pasteurian age. In fact, the Institute houses several anatomical-pathological preparations from a fatal clinical case of "acute human glanders". T

[The early laboratory diagnosis of the pulmonary form of glanders and melioidosis by using rapid methods of immunochemical analysis].

The problems of the sanitary guarding of frontiers comprise such aspects as the early recognition of infectious diseases, even those which are considered exotic in the Russian Federation. These diseases include glanders and melioidosis, infections which are endemic for some frontier regions. The study performed on experimental models of the pulmonary form of glanders and melioidosis with the acute course of the diseases demonstrates that the use of such laboratory diagnostic methods as immunosorbent variants of the enzyme-linked, chemiluminescent and immunofluorescent immunoassays permits maki

Protective efficacy of heat-inactivated B. thailandensis, B. mallei or B. pseudomallei against experimental melioidosis and glanders.

Burkholderia pseudomallei and Burkholderia mallei are gram-negative bacilli that are the causative agents of melioidosis and glanders, respectively. Both humans and animals are susceptible to both diseases. There is currently no vaccine available for the prevention of disease. We report the protective efficacy of heat-inactivated Burkholderia thailandensis, B. mallei or B. pseudomallei cells as vaccines against murine melioidosis and glanders. Immunisation with heat-inactivated B. pseudomallei cells provided the highest levels of protection against either melioidosis or glanders. These studies

Burkholderia Hep_Hag autotransporter (BuHA) proteins elicit a strong antibody response during experimental glanders but not human melioidosis.

BACKGROUND: The bacterial biothreat agents Burkholderia mallei and Burkholderia pseudomallei are the cause of glanders and melioidosis, respectively. Genomic and epidemiological studies have shown that B. mallei is a recently emerged, host restricted clone of B. pseudomallei. RESULTS: Using bacteriophage-mediated immunoscreening we identified genes expressed in vivo during experimental equine glanders infection. A family of immunodominant antigens were identified that share protein domain architectures with hemagglutinins and invasins. These have been designated Burkholderia Hep_Hag autotrans
