Glutamate and GABA-metabolizing enzymes in post-mortem cerebellum in Alzheimer's disease: phosphate-activated glutaminase and glutamic acid decarboxylase.

Enzymes of glutamate and GABA metabolism in postmortem cerebellum from patients with Alzheimer's disease (AD) have not been comprehensively studied. The present work reports results of original comparative study on levels of phosphate-activated glutaminase (PAG) and glutamic acid decarboxylase isoenzymes (GAD65/67) in autopsied cerebellum samples from AD patients and matched controls (13 cases in each group) as well as summarizes published evidence for altered levels of PAG and GAD65/67 in AD brain. Altered (decreased) levels of these enzymes and changes in links between amounts of these enzym

Spinal cord GLT-1 glutamate transporter and blood glutamic acid alterations in motor neuron degeneration (Mnd) mice.

This study characterizes for the first time neurochemical mechanisms in Mnd mice, initially described as a model of motor neuron disease and more recently proposed as a model for neuronal ceroid lipofuscinosis. A selective decrease (-30%) of [3H]glutamate uptake was found in spinal cord but not cortical synaptosomes of Mnd mice aged 28 weeks, when they show histopathological alterations, complete blindness and moderate neurological deficits. In spite of the widespread presence of stored material in neurons in many brain regions and spinal cord, the active transport of [3H]serotonin, [3H]dopami

Immunoinflammatory Profile of FGF-18, IL-35 and Glutamic Acid Decarboxylase in Patients With Diabetic Foot Ulcers.

Diabetic foot ulcer (DFU) is a serious problem that may cause amputation of the lower extremities in patients with diabetes. The present research aimed to assess the localised tissue expression and potential immunoinflammatory crosstalk among fibroblast growth factor-18 (FGF-18), glutamic acid decarboxylase (GAD) and interleukin-35 (IL-35) in DFU compared to non-diabetic controls (NDCs), while also examining the systemic serum levels of FGF-18 and IL-35. Venous blood was collected from 80 patients with DFU and 100 NDC, and the concentrations of serum IL-35, FGF-18 and blood haemoglobin A1c (Hb

Functional recovery of demineralized dentin using a glutamic acid-modified electrospun scaffold: A multimodal in vitro characterization.

BACKGROUND: Dentin remineralization poses a significant challenge in restorative dentistry due to the complex structure and mineral-organic composition of dentin. Electrospun nanofibrous scaffolds present a biomimetic platform to facilitate mineral deposition. Functionalization of the scaffold, achieved through the incorporation of biomolecules like glutamic acid, which mimics non-collagenous proteins, may improve remineralization. OBJECTIVE: To assess the remineralization potential of a glutamic acid-loaded polycaprolactone/nanohydroxyapatite (PCL/nHA/Glu) scaffold on acid-demineralized dent

Glutamic acid decarboxylase 65-positive autoimmune encephalitis presenting with gelastic seizure, responsive to steroid: A case report.

BACKGROUND: Anti-glutamic acid decarboxylase (GAD) antibody is known to cause several autoimmune-related situations. The most known relationship is that it may cause type I diabetes. In addition, it was also reported to result in several neurologic syndromes including stiff person syndrome, cerebellar ataxia, and autoimmune encephalitis. Decades ago, isolated epilepsy associated with anti-GAD antibody was first reported. Recently, the association between temporal lobe epilepsy and anti-GAD antibody has been discussed. Currently, with improvements in examination technique, many more autoimmune-

Insight into the rheological behaviors of a polyanionic collagen fabricated with poly(γ-glutamic acid)-NHS ester.

The rheological behaviors of a polyanionic collagen, fabricated using poly(γ-glutamic acid)-N-hydroxysuccinimide (γ-PGA-NHS) as a novel modifier, were investigated in this study. It was found that both of the native and modified collagen solutions were pseudoplastic fluids, as shown from the steady-shear tests. While the storage modulus and loss modulus of collagen increased with increasing the amount of γ-PGA-NHS, or with increasing the degree of esterification of γ-PGA-NHS; meanwhile, the dynamic denaturation temperature determined by dynamic temperature sweep was also increased, indicating

The preparation of poly(γ-glutamic acid)-NHS ester as a natural cross-linking agent of collagen.

γ-PGA-NHS ester, which was prepared using poly(γ-glutamic acid) (γ-PGA) and N-hydroxysuccinimide (NHS) as the raw materials, was synthesized to be a novel cross-linker of collagen. Fourier transform infrared spectra analysis suggested that the products displayed the characteristic absorption peak of ester. Results from nuclear magnetic resonance analysis indicated that the esterification degree of γ-PGA-NHS ester was increased with the increase of NHS. Modified collagen was prepared and characterized. The results of circular dichroism analysis indicated modified collagen retained the triple he

Glutamine and glutamic acid enhance thyroid-stimulating hormone β subunit mRNA expression in the rat pars tuberalis.

Thyroid-stimulating hormone (TSH)-producing cells of the pars tuberalis (PT) display distinct characteristics that differ from those of the pars distalis (PD). The mRNA expression of TSHβ and αGSU in PT has a circadian rhythm and is inhibited by melatonin via melatonin receptor type 1; however, the detailed regulatory mechanism for TSHβ expression in the PT remains unclear. To identify the factors that affect PT, a microarray analysis was performed on laser-captured PT tissue to screen for genes coding for receptors that are abundantly expressed in the PT. In the PT, we found high expression o

Diabetes Antibody Standardization Program: evaluation of assays for autoantibodies to glutamic acid decarboxylase and islet antigen-2.

AIMS/HYPOTHESIS: Islet autoantibodies are important in diabetes classification and risk assessment, and as endpoints in observational studies. The Diabetes Autoantibody Standardization Program (DASP) aims to improve and standardise measurement of autoantibodies associated with type 1 diabetes. We report results for glutamic acid decarboxylase autoantibodies (GADA) and islet antigen-2 autoantibodies (IA-2A) from three DASP workshops (2002--2005). METHODS: Up to 60 laboratories in 18 countries participated in each workshop. Participants received coded serum aliquots from 50 patients with newly

The lysine- and glutamic acid-rich protein KERP1 plays a role in Entamoeba histolytica liver abscess pathogenesis.

The parasite Entamoeba histolytica colonizes the large bowel where it may persist as an asymptomatic luminal gut infection, which changes to virulence. Parasite invasion of the intestine leads to dysentery and spreads to the liver, where amoebae form abscesses. We took advantage of changes in virulence that occurs after long-term in vitro culture of E. histolytica strains. Using microarrays, we concluded that virulence correlates with upregulation of key genes involved in stress response, including molecular chaperones, ssp1 and peroxiredoxin; as well as the induction of unknown genes encoding

Selective loss of Purkinje cells in a patient with anti-glutamic acid decarboxylase antibody-associated cerebellar ataxia.

Anti-glutamic acid decarboxylase antibody is associated with the development of progressive cerebellar ataxia and slowly progressive insulin-dependent diabetes mellitus. Previously, the neurophysiological characteristics of IgG in the cerebrospinal fluid of a patient with anti-glutamic acid decarboxylase antibody-associated progressive cerebellar ataxia and slowly progressive insulin-dependent diabetes mellitus were reported. Using a voltage-gated whole-cell recording technique, it was observed that the IgG in the cerebrospinal fluid of the patient selectively suppressed the inhibitory postsyn

GABA and histogenesis in fetal and neonatal mouse brain lacking both the isoforms of glutamic acid decarboxylase.

Recent in vitro investigations have suggested that GABA is involved in the development of the mammalian central nervous system. To evaluate the roles of GABA in neurogenesis in vivo, we generated mice lacking both the isoforms of glutamic acid decarboxylase (GAD), GAD65 and GAD67, by mating GAD65- and GAD67-mutant mice generated by homologous recombination in this laboratory. Similar to GAD67-deficient mice, the GAD65/67-deficient mice did not survive after birth because of cleft palate. We thus analyzed these mice at the fetal and newborn stages. GABA was scarcely detectable in the GAD65/67-d

Light and electron microscopic immunolocalization of rat submandibular gland mucin glycoprotein and glutamine/glutamic acid-rich proteins.

We studied the subcellular localization of two major secretory products of adult rat submandibular gland (RSMG), blood group A-reactive mucin glycoprotein and glutamine/glutamic acid-rich protein (GRP), by light and electron microscopic immunocytochemistry. The structure of the major neutral oligosaccharide of the mucin was shown to be: GalNAc alpha 1,3(Fuc alpha 1,2)Gal beta 1,3GalNAc. A mouse monoclonal antibody (1F9) with specificity for blood group A determinants was prepared against the mucin. The antibody recognized a single band of approximately 114 KD on Western blots of RSMG extract.

Neurotransmitter enzyme abnormalities in senile dementia. Choline acetyltransferase and glutamic acid decarboxylase activities in necropsy brain tissue.

Reductions in 2 neurotransmitter synthesizing enzymes in brain, glutamic acid decarboxylase (GAD) and choline acetyltransferase (CAT), have been found in dementias of different origins, including senile dementia (Alzheimer type). Significant reductions in cerebral GAD have also been found in depression (unipolar). The GAD reductions did not generally appear to be localised in any specific region of the brain examined. However, the reduction of CAT in the hippocampus, relative to reductions in other areas examined, was substantially greater in the brains with Alzheimer-type changes. GAD and CAT
