Chronic Constriction Injury of the Rat's Infraorbital Nerve (IoN-CCI) to Study Trigeminal Neuropathic Pain.

Animal models are important tools to study the pathophysiology and pharmacology of neuropathic pain. This manuscript describes the surgical and behavioral procedures to study trigeminal neuropathic pain in rats. To meet the specificity of trigeminal neuropathic pain syndromes, the infraorbital nerve (IoN) is subjected to a chronic constriction injury (CCI) by loosely ligating the nerve. An intra-orbital approach is presented here to expose and ligate the IoN in the orbital cavity. After IoN ligation, rats exhibit changes in spontaneous behavior and in response to von Frey hair stimulation that

The relationship of obsessive-compulsive disorder to possible spectrum disorders: results from a family study.

BACKGROUND: The familial relationship between obsessive-compulsive disorder (OCD) and "obsessive-compulsive spectrum" disorders is unclear. This study investigates the relationship of OCD to somatoform disorders (body dysmorphic disorder [BDD] and hypochondriasis), eating disorders (e.g., anorexia nervosa and bulimia nervosa), pathologic "grooming" conditions (e.g., nail biting, skin picking, trichotillomania), and other impulse control disorders (e.g., kleptomania, pathologic gambling, pyromania) using blinded family study methodology. METHODS: Eighty case and 73 control probands, as well as

Auto-Reactive Th17-Cells Trigger Obsessive-Compulsive-Disorder Like Behavior in Mice With Experimental Autoimmune Encephalomyelitis.

Th17-lymphocytes are well known for their deleterious role in autoimmunity. But does the notoriety of this repertoire extend beyond autoimmunity? In the present study we employed experimental autoimmune encephalomyelitis as model system to study the role auto-reactive Th17 cells in neuropsychiatric disorders. The mice with experimental autoimmune encephalomyelitis exhibited exaggerated grooming activity. The observed behavioral anomaly resembled obsessive compulsive disorder (OCD) upon analysis of grooming microstructure, induced grooming, marble burying and nestlet shredding. The observed OCD

Oligodeoxynucleotide antisense to the D1 dopamine receptor mRNA inhibits D1 dopamine receptor-mediated behaviors in normal mice and in mice lesioned with 6-hydroxydopamine.

The effects of intracerebroventricular (i.c.v.) administration of a 20-mer phosphorothioate oligodeoxynucleotide antisense to the D1 dopamine receptor mRNA (D1 antisense) on D1 dopamine receptor-mediated behaviors were studied in normal mice and in mice with unilateral 6-hydroxydopamine-induced lesions of corpus striatum. Treating mice with D1 antisense inhibited grooming behavior induced by the D1 dopamine receptor agonist SKF 38393 [1-phenyl-2,3,4,5-tetrahydro-1H-3 benzazepine-7,8-diol hydrochloride], the reduction in grooming being related to the amount and length of time D1 antisense was g

Cortical synchronization of spontaneous trigeminal neuropathic pain monitored with in vivo optical imaging of calcium activity in freely moving mice.

OBJECTIVE: To determine the impact of neuropathic lesion on cortical synchronization in processing spontaneous pain-like behavior. BACKGROUND: In vivo optical monitoring of neuronal activity may provide insightful mechanisms underlying the complexity of spontaneous pain-like behavior in freely moving animals. METHODS: We examined the synchronized pattern of the pyramidical neurons in anterior cingulate cortex during spontaneous grooming behavior using optical monitoring of Ca2+ activity. A chronic constriction injury of infraorbital nerve model was performed to induce trigeminal neuropathic

The Partial M1 Muscarinic Cholinergic Receptor Agonist, CDD-0102A, Differentially Modulates Glutamate Efflux in Striatal Subregions during Stereotyped Motor Behavior in the BTBR Mouse Model of Autism.

The BTBR T+ Itpr3tf/J (BTBR) mouse displays elevated repetitive motor behaviors. Treatment with the partial M1 muscarinic receptor agonist, CDD-0102A, attenuates stereotyped motor behaviors in BTBR mice. The present experiment investigated whether CDD-0102A modifies changes in striatal glutamate concentrations during stereotyped motor behavior in BTBR and B6 mice. Using glutamate biosensors, change in striatal glutamate efflux was measured during bouts of digging and grooming behavior with a 1 s time resolution. Mice displayed both decreases and increases in glutamate efflux during such behavi

Opioids and attachment in rhesus macaque (Macaca mulatta) abusive mothers.

This study investigated the role of the endogenous opioid system in maternal and affiliative behavior of group-living rhesus macaque (Macaca mulatta) mothers with a history of abusive parenting. Eighteen mothers received an injection of the opioid antagonist naltrexone or saline for 5 days per week for the first 4 weeks of the infant's life. After treatment, mother-infant pairs were focally observed. Naltrexone did not significantly affect infant abuse or other measures of maternal behavior. Naltrexone increased the amount of grooming received by mothers from other group members and reduced th

Wider range of allodynia in a rat model of repeated dural nociception compared with infraorbital nerve chronic constriction injury.

BACKGROUND: To identify differences in allodynia and grooming behaviours between rat models of either repeated dural nociception with inflammatory soup (IS) or infraorbital nerve chronic constriction injury (IoN-CCI). METHODS: Repeated dural nociception was induced via the application of IS to the dural meninges and IoN-CCI was applied to model neuropathic pain. All surgeries were performed on the right side and a sham operation was performed on the control group. Mechanical and thermal withdrawal thresholds were tested on different facial areas and hindpaw during the interictal period and gr

Beta adrenoceptor blockade mimics effects of stress on motor activity in mice.

Reduced central noradrenergic function has been implicated as a factor in reduced behavioral activity after stress. The present studies examined the role of reduced beta adrenergic neurotransmission in mediating this effect. This was done by testing the ability of beta receptor antagonists to mimic the behavioral actions of stress. Mice were subjected to stress or given various beta antagonists and tested for swimming behavior, locomotor activity, or grooming behavior. As previously reported, stress reduced swimming and locomotor activity and increased grooming. Both the nonselective antagonis

Acute administration of the NMDA receptor antagonists ketamine and MK-801 reveals dysregulation of glutamatergic signalling and sensorimotor gating in the Sapap3 knockout mouse model of compulsive-like behaviour.

Obsessive-compulsive disorder (OCD) is characterised by excessive intrusive thoughts that may cause an individual to engage in compulsive behaviours. Frontline pharmacological treatments (i.e., selective serotonin reuptake inhibitors (SSRIs)) leave approximately 40% of patients refractory to treatment. To investigate the possibility of novel pharmacological therapies for OCD, as well as the potential mechanisms underlying its pathology, we used the Sapap3 knockout (KO) mouse model of OCD, which exhibits increased anxiety and compulsive grooming behaviours. Firstly, we investigated whether admi

Linking energy availability, movement and sociality in a wild primate (Papio ursinus).

Proximate mechanisms of 'social ageing', i.e. shifts in social activity and narrowing of social networks, are understudied. It is proposed that energetic deficiencies (which are often seen in older individuals) may restrict movement and, in turn, sociality, but empirical tests of these intermediary mechanisms are lacking. Here, we study wild chacma baboons (Papio ursinus), combining measures of faecal triiodothyronine (fT3), a non-invasive proxy for energy availability, high-resolution GPS data (movement and social proximity) and accelerometry (social grooming durations). Higher (individual me

Clozapine generates obsessive compulsive disorder-like behavior in mice.

Clozapine is thought to induce obsessive compulsive symptoms (OCS) in schizophrenic patients. However, because OCS are often comorbid with schizophrenia regardless of clozapine treatment, it remains unclear whether clozapine can generate OCS de novo. Thus, it has been difficult to establish a causal link between clozapine and OCS in human studies. To address this question, we asked whether chronic treatment with clozapine can induce obsessive compulsive disorder (OCD)-like behavior in mice. We injected mice with long-term continuous release pellets embedded with clozapine four times at 60-day

Increasing Water Access for Individuals Living with Spinal Cord Injury Using an Innovative Smart Faucet.

This project aimed to assess the functionality of an innovative fountain faucet, called the Access-H2O™ faucet, to enable and empower individuals living with spinal cord injury (SCI) to gain water access independently for activities of daily living (ADLs). The faucet integrates electromechanical controls, software programs, and remote sensors to control the water arch stream. Eighteen SCI subjects were recruited to test the faucet's functionality by using eye gaze, voice, and motion sensors to activate fountain mode, washing mode, and grooming mode for three ADLs: drinking, rinsing, and groomi

Disruption of prepulse inhibition is associated with compulsive behavior severity and nucleus accumbens dopamine receptor changes in Sapap3 knockout mice.

Obsessive compulsive disorder (OCD) is associated with disruption of sensorimotor gating, which may contribute to difficulties inhibiting intrusive thoughts and compulsive rituals. Neural mechanisms underlying these disturbances are unclear; however, striatal dopamine is implicated in regulation of sensorimotor gating and OCD pathophysiology. The goal of this study was to examine the relationships between sensorimotor gating, compulsive behavior, and striatal dopamine receptor levels in Sapap3 knockout mice (KOs), a widely used preclinical model system for OCD research. We found a trend for di

Differential drug effects on spontaneous and evoked pain behavior in a model of trigeminal neuropathic pain.

PURPOSE: Baclofen and morphine have shown efficacy against mechanical allodynia after infraorbital nerve chronic constriction injury (IoN-CCI). No drug effects have yet been reported on spontaneous trigeminal neuropathic pain. It has been proposed that the directed face grooming behavior that also develops following IoN-CCI offers a measure of spontaneous trigeminal neuropathic pain. SUBJECTS AND METHODS: We examined the effects of a continuous 1-week infusion of 30 mg/day carbamazepine (the first-line drug treatment for trigeminal neuralgia), 1.06 mg/day baclofen, 4.18 mg/day clomipramine, a

Behavioral responses to visual overstimulation in the cockroach Periplaneta americana L.

In the visual systems of insects, different types of photoreceptors contribute to specialized visual channels that mediate distinct functions and behaviors. Large compound eyes of Periplaneta americana contain photoreceptors of two spectral classes, broadband green-sensitive photoreceptors and narrow-band UV-sensitive photoreceptors. Here, we investigated how visual stimulation by UV and green light affects locomotor, resting, and grooming behaviors in P. americana under conditions when light avoidance is not possible. We show that green but not UV light stimulates locomotor activity, inducing

Inhibiting social support from massage-like stroking increases morphine dependence.

Our previous studies showed that altering solely the drug experience of the cage mates with which rodents are housed affects the development of morphine dependence. In this study, we used designer receptors exclusively activated by designer drugs to artificially increase or decrease the activity of peripheral dorsal root ganglia sensory neurons expressing the G-protein-coupled receptor MRGPRB4. This is because sensory MRGPRB4-expressing neurons were shown to specifically detect the sensation of massage-like stroking resulting from social grooming, which is an important affiliative social behav

Motor and cognitive stereotypies in the BTBR T+tf/J mouse model of autism.

The BTBR T+tf/J inbred mouse strain displays a variety of persistent phenotypic alterations similar to those exhibited in autism spectrum disorders (ASDs). The unique genetic background of the BTBR strain is thought to underlie its lack of reciprocal social interactions, elevated repetitive self-directed grooming, and restricted exploratory behaviors. In order to clarify the existence, range, and mechanisms of abnormal repetitive behaviors within BTBR mice, we performed detailed analyses of the microstructure of self-grooming patterns and noted increased overall grooming, higher percentages of

[The effect of the intraventricular administration of oxytocin on the functional activity of the epiphysis, adrenals and gonads and on the behavior of rats].

Intraventricular administration of oxytocin (OT) facilitated horizontal locomotion and inhibited their decrease in rats. OT also increased the number of face grooming responses but did not affect the genital grooming. The OT seems to suppress the pineal gland activity, whereas the plasma concentration of testosterone did not change and the corticosterone concentration increased. The OT seems to exert a complex effect on the nervous and endocrine systems.

Ground substrate affects activity budgets and hair loss in outdoor captive groups of rhesus macaques (Macaca mulatta).

How the captive environment influences the behavior of animals is relevant to the well-being of captive animals. Captivity diverges from the natural environment in many ways, and one goal of enrichment practices is to encourage species-typical behavior in these unnatural environments. This study investigated the influence of grass vs. gravel substrate on activity budgets and degree of hair loss in seven groups of captive rhesus macaques housed in outdoor enclosures at the California National Primate Research Center. Groups having grass substrate spent a greater proportion of their time foragin
