Sex difference in psychological behavior changes induced by long-term social isolation in mice.

Social isolation can induce psychological behavior changes. It is interesting to know whether there is sex difference in responding to social isolation or not. The present study compared the behavior difference between male and female mice isolated for 1-4 months. The results showed that the isolated male mice had higher accounts of locomotor activity than the isolated female and group-housed ones. Both isolated male and female mice spent shorter time in the dark box than the group-housed mice in the light/dark test, and isolated male mice spent less time in the closed arms than isolated femal

The Use of Behavioral Reconsolidation Interference in Depressive Disorders. A Double-Blinded Randomized Controlled Experimental Registered Report.

Depressive disorders often show recurrent courses that cannot be sufficiently prevented by existing therapeutic protocols. In other affective disorders, recurrence has been linked to three mechanisms -spontaneous recovery, accelerated new/relearning, and reinstatement- which are related to the preservation of disorder-related memory traces even through successful extinction-based interventions. Reconsolidation-interference protocols aim to directly alter such traces by reactivating and destabilizing them before intervention. While this approach has shown benefits in fear, craving, and trauma-r

Reflections on written comments by women on their abuse.

In a study to differentiate injuries of physical abuse from non-physical abuse, 103 women wrote uninvited comments that were analyzed in two ways: Q-sort and phenomenology analyses. Q-sort identified two categories: helplessness and assertiveness. Comparisons with subject responses about current or previous violence indicated that women who wrote assertive comments were less likely to be in a violent relationship than women who wrote helpless comments. Phenomenological analysis indicated that women who were categorized as silent, received or subjective were in abusive relationships; some had b

A theory-based nursing intervention to instill hope in homeless veterans.

This quasi-experimental study sought to determine if a specific nursing intervention to instill hope would positively influence levels of hope, self-efficacy, self-esteem, and depression in homeless veterans. Miller's Model of Patient Power Resources served as the conceptual framework from which a middle-range theory of homelessness-hopelessness was derived to guide the study. Homeless veterans completed pretests on admission to a Veterans Affairs Medical Center, were randomly assigned to treatment or waiting control group, and completed posttests at the end of 4 weeks. There was support for t

From Monoamines to Systems Psychiatry: Rewiring Depression Science and Care (1960s-2025).

Major depressive disorder (MDD) was long framed as a single clinical entity arising from a linear stress-monoamine-hypothalamic-pituitary-adrenal (HPA) axis cascade. This view was shaped by forced swim and learned helplessness tests in animals and by short-term symptom-based trials using scales such as the Hamilton Depression Rating Scale (HAM-D) and the Montgomery-Åsberg Depression Rating Scale (MADRS). This "unitary cascade" view has been dismantled by advances in neuroimaging, immune-metabolic profiling, sleep phenotyping, and plasticity markers, which reveal divergent circuit-level, inflam

Systemic immunization with altered myelin basic protein peptide produces sustained antidepressant-like effects.

Immune dysregulation, specifically of inflammatory processes, has been linked to behavioral symptoms of depression in both human and rodent studies. Here, we evaluated the antidepressant effects of immunization with altered peptide ligands of myelin basic protein (MBP)-MBP87-99[A91, A96], MBP87-99[A91], and MBP87-99[R91, A96]-in different models of depression and examined the mechanism by which these peptides protect against stress-induced depression. We found that a single dose of subcutaneously administered MBP87-99[A91, A96] produced antidepressant-like effects by decreasing immobility in t

Estrous Cycle Phase-Dependent Changes in Anxiety- and Depression-Like Profiles in the Late Adolescent Wistar-Kyoto Rat.

BACKGROUND: Depression often manifests during adolescence when the development and networking of social and emotional brain areas is being influenced by hormones. The Wistar Kyoto (WKY) rat has been proposed as an animal model of adolescent depression with various face, construct, and predictive validities for clinical depression having been established. PURPOSE: The influence of the estrous cycle on anxiety- and depression-like behaviors in female adolescents may be tested out further using this model. METHODS: Female adolescent WKY rats were tested for anxiety- and depression-like behavior

The Prodrug 4-Chlorokynurenine Causes Ketamine-Like Antidepressant Effects, but Not Side Effects, by NMDA/GlycineB-Site Inhibition.

Currently approved antidepressant drug treatment typically takes several weeks to be effective. The noncompetitive N-methyl-d-aspartate (NMDA) receptor antagonist ketamine has shown efficacy as a rapid-acting treatment of depression, but its use is associated with significant side effects. We assessed effects following blockade of the glycineB co-agonist site of the NMDA receptor, located on the GluN1 subunit, by the selective full antagonist 7-chloro-kynurenic acid (7-Cl-KYNA), delivered by systemic administration of its brain-penetrant prodrug 4-chlorokynurenine (4-Cl-KYN) in mice. Glycine site N-methyl-D-aspartate receptor antagonist 7-CTKA produces rapid antidepressant-like effects in male rats.

BACKGROUND: Glutamate N-methyl-D-aspartate (NMDA) receptor antagonists exert fast-acting antidepressant effects, providing a promising way to develop a new classification of antidepressant that targets the glutamatergic system. In the present study, we examined the potential antidepressant action of 7-chlorokynurenic acid (7-CTKA), a glycine recognition site NMDA receptor antagonist, in a series of behavioural models of depression and determined the molecular mechanisms that underlie the behavioural actions of 7-CTKA. METHODS: We administered the forced swim test, novelty-suppressed feeding t

Acute D-serine treatment produces antidepressant-like effects in rodents.

Research suggests that dysfunctional glutamatergic signalling may contribute to depression, a debilitating mood disorder affecting millions of individuals worldwide. Ketamine, a N-methyl-D-aspartate (NMDA) receptor antagonist, exerts rapid antidepressant effects in approximately 70% of patients. Glutamate evokes the release of D-serine from astrocytes and neurons, which then acts as a co-agonist and binds at the glycine site on the NR1 subunit of NMDA receptors. Several studies have implicated glial deficits as one of the underlying facets of the neurobiology of depression. The present study t

Noninvasive and objective near-real-time detection of pain changes during tonic fluctuating noxious heat stimulation.

Chronic pain involves natural intensity fluctuations that patients cannot control, contributing to learned helplessness and functional impairment. Detecting spontaneous pain decreases could enable precisely timed interventions that enhance perceived control. However, existing research on objective pain assessment has focused primarily on estimating static intensity from short, predictable stimuli rather than detecting moment-to-moment changes in ongoing pain. This study investigated whether pain decreases can be detected objectively using easily obtainable physiological signals during fluctuat

The Interplay Between Experimental Heat Pain and Noninvasive Stimulation of the Medial Prefrontal Cortex on Reinforcement Learning With Manipulated Outcome Contingencies.

Pain negatively affects several cognitive abilities, but knowledge about its effect on reinforcement learning (RL) is limited. During RL, instrumental choices can be influenced by heuristic tendencies to approach rewards or inhibit actions when facing potentially aversive events, introducing "Pavlovian bias" in behavior. Recent studies suggest that compromised outcome controllability enhances Pavlovian bias, a phenomenon that may be linked to suboptimal decision-making in learned helplessness (LH). Since LH is common in chronic pain syndromes, this study sought to establish a link between expe

Novel, small molecules targeting the 5-HT4 receptor protect against stress-induced maladaptive behavior with efficacy across age.

BACKGROUND: Stress is a risk factor for developing psychiatric disorders, including major depressive disorder (MDD). Compounds targeting the serotonin type 4 receptor (5-HT4R) hold promise as novel rapid-acting treatments of mood disorders. However, a lack of selectivity and numerous side effects have been limiting factors for their clinical use. Here, we developed and characterized novel-composition 5-HT4R compounds in mouse models of stress. METHODS: Three 5-HT4R-targeting compounds were designed and synthesized based on PF-04995274, a high-affinity 5-HT4R ligand reported to be a partial ag

Antidepressant-like action of agmatine in the acute and sub-acute mouse models of depression: a receptor mechanism study.

Previous studies have shown that agmatine, a potential neuromodulator or co-transmitter, exhibited antidepressant-like action in animal models, yet its mechanism, especially the receptor mechanism, remains unclear. In the present study, using efaroxan, a preferential antagonist of I1 imidazoline receptor (I1R) and yohimbine, an antagonist of α2 adrenergic receptor (α2AR), we investigated the roles of I1R and α2AR in agmatine's antidepressant-like effect in acute and sub-acute depression models in mice. We found that in the tail-suspension test (TST) and the forced swimming test (FST), acute ad

Serotonin deficiency alters susceptibility to the long-term consequences of adverse early life experience.

Brain 5-HT deficiency has long been implicated in psychiatric disease, but the effects of 5-HT deficiency on stress susceptibility remain largely unknown. Early life stress (ELS) has been suggested to contribute to adult psychopathology, but efforts to study the long-term consequences of ELS have been limited by a lack of appropriate preclinical models. Here, we evaluated the effects of 5-HT deficiency on several long-term cellular, molecular, and behavioral responses of mice to a new model of ELS that combines early-life maternal separation (MS) of pups and postpartum learned helplessness (LH

Mice genetically depleted of brain serotonin do not display a depression-like behavioral phenotype.

Reductions in function within the serotonin (5HT) neuronal system have long been proposed as etiological factors in depression. Selective serotonin reuptake inhibitors (SSRIs) are the most common treatment for depression, and their therapeutic effect is generally attributed to their ability to increase the synaptic levels of 5HT. Tryptophan hydroxylase 2 (TPH2) is the initial and rate-limiting enzyme in the biosynthetic pathway of 5HT in the CNS, and losses in its catalytic activity lead to reductions in 5HT production and release. The time differential between the onset of 5HT reuptake inhibi

Early gestational exposure to moderate concentrations of ethanol alters adult behaviour in C57BL/6J mice.

Alcohol consumption during pregnancy has deleterious effects on the developing foetus ranging from subtle physical deficits to severe behavioural abnormalities and is encompassed under a broad umbrella term, foetal alcohol spectrum disorders (FASD). High levels of exposure show distinct effects, whereas the consequences of moderate exposures have been less well studied. The aim of this study was to examine the effects of a moderate dose ethanol exposure using an ad libitum drinking procedure during the first eight days of gestation in mice on the behavioural phenotype of adult offspring. Involvement of AMPA receptor in both the rapid and sustained antidepressant-like effects of ketamine in animal models of depression.

A growing body of evidence has suggested that the dysfunction of glutamatergic systems plays a pivotal role in major depressive disorder (MDD). In clinical studies, an N-methyl-d-aspartate receptor antagonist, ketamine, was shown to exert both rapid and sustained antidepressant effects in patients with treatment-resistant MDD. The objective of the present study was to confirm the rapid onset of action of ketamine and to investigate the mechanisms underlying both the rapid and sustained antidepressant-like effects of ketamine in rodent models of depression.
