[DNA diagnosis of fragile X syndrome in a family. A new type of heredity--dynamic mutations].

In typical cases, the diagnosis of fragile X syndrome can be made clinically, but has so far been based on cytogenetic detection of a fragile site on the X chromosome at Xq27.3. Cytogenetic analyses are time-consuming, and false negative results have been a problem. Following the successful molecular cloning of the gene causing the fragile X syndrome, a novel genetic principle was discovered: the "dynamic" DNA mutation. DNA analyses have now the power to distinguish between normal copies of the gene, carrier mutations and fragile X mutations. We have performed both cytogenetic and DNA analyses

[An example of dominant heredity in the transmission of primary open-angle glaucoma in a northwestern region of Greece].

The authors have accomplished an epidemiological clinical and genetic study on primary open-angle glaucoma among 411 persons in the North Western district of Greece (Epirus), belonging to 4 genealogical trees. 112 of the 411 persons were offsprings aged 30 years or more, alive or dead, and they had direct blood-relation to the propositus. 35 of these offsprings were suffering from primary open-angle glaucoma. The classification and the number of the affected subjects suggested that one major gene, expressed by the autosomal dominant character, is the main factor for the heredity of primary ope

Arteriosclerosis, heredity, and some previous infections in the etiology of Parkinson's disease. A case-control study.

A case-control study was performed to investigate the significance of arteriosclerosis, heredity and some infections in the etiology of Parkinson's disease. The study group consisted of all traceable patients with Parkinson's disease living in a defined area, a total of 444 patients, and of control subjects for each patient, matched in sex and age, chosen from among the general population residing in the same area. No significant differences were found between the patients and the controls concerning the occurrence of cardiac insufficiency, coronary heart disease, or stroke. The Parkinsonian p

[A physician from Finnmark who pointed out the significance of heredity in congenital hip dysplasia].

In 1918 A.B. Wessel, district physician in Sør-Varanger, Northern Norway, published an article titled Limping families in the county of Finnmark. Based on his own investigations, he concluded that the congenital dislocation of the hip joint was a hereditary disease, and that the prevalence of the disease was high in the Sami population, especially among Sami women. Dr. Wessel observed an association between tuberculosis and the poverty of the population. The miserable hygienic conditions in small and overcrowded houses were a serious risk factor for the development of tuberculosis. Wessel was

Ascending Aneurysms in Heart Transplant Patients: A Rare Opportunity to Assess Heredity Versus Biological Environment.

Three patients developed severe ascending aortic aneurysm requiring surgical resection after heart transplantation. In all 3 cases, the donor aorta of the transplant remained normal in caliber, despite the development of a large aneurysm in the native upper ascending aorta. The aneurysmal disease did not cross the suture line. (Level of Difficulty: Advanced.).

William Boven's 1915 thesis "Similarity and Mendelism in the heredity of dementia praecox and manic-depressive insanity".

Boven published, in 1915, his MD thesis at the University of Lausanne in which he examined 60 3- to 4-generation pedigrees ascertained from admitted patients with dementia praecox (DP) and manic-depressive insanity (MDI). He asked three questions: (i) were DP and MDI hereditary? (ii) were they the same or distinct conditions? and (iii) were they Mendelian disorders? Based on the rarity of environmental precipitants severe enough to cause disorder onset and the pattern of disorders in relatives, Boven concluded that both disorders were inherited. He found that MDI largely ran in families throug

Heredity, diet and lifestyle as determining risk factors for the esophageal cancer on Nanao Island in Southern China.

In this case-control study we evaluated contribution of environmental and genetic factors for risk of esophageal cancer (EC) by studying populations on Nanao Island (highest risk area for EC in China) and Shanwei (low risk region). Data on lifestyle, diet and family history were collected from the 166 newly diagnosed EC patients on Nanao between 2003 and 2004, from their 1450 first degree relatives and from controls on Nanao and Shanwei. Univariate and logistic regression analysis, family aggregation patterns, standardized incidence ratio (SIR), segregation ratio and heritability index were ev

Development of atopy and wheezing symptoms in relation to heredity and early pet keeping in a Swedish birth cohort.

The role of pet keeping during infancy for the development of allergy and asthma is still controversial. The objective of this population-based birth cohort study was to assess the development of atopy and different wheezing phenotypes during the first 4 yr of life in relation to heredity and early pet keeping. The cohort comprised all 1228 infants living in a Swedish county who were born over a 1-yr period. The parents replied to repeated questionnaires and 817 of the children were skin prick tested both at 1 and 4 yr. Cat keeping during the first year of life was associated with an increased

Congenital muscular torticollis: is heredity a possible factor in a family with five torticollis patients in three generations?

Although the clinical features of congenital muscular torticollis are well known, the cause of this disorder still remains hypothetical. The role of heredity is not well established, because there have been only a few studies on it. We presented a family in which five members suffer from CMT in three generations, two of whom were operated on. According to the pattern in the pedigree, probably an autosomal dominant condition could be considered.

Aspects of heredity, syndromic associations, and course of conditions in which cutaneous lesions occur solitarily or in multiplicity.

Many tumors of the skin, whether originating from ectoderm or mesoderm, exist in either single or multiple form. In the multiple state there are frequently associations with other cutaneous and systemic abnormalities, sometimes serious and even life-threatening. The resultant syndrome is almost always inherited in an autosomal dominant pattern. By contrast, solitary lesions are not heritable, nor are they associated with other stigmata. The solitary forms appear in middle or late life, the multiple, syndromic forms in childhood or early adulthood.

Awareness of heredity in colorectal cancer patients is insufficient among clinicians: a Norwegian population-based study.

OBJECTIVE: The assessment of family history and medical data is crucial in identifying families with Lynch syndrome (LS). Among consecutive colorectal cancer (CRC) patients, we aimed at identifying all patients with a hereditary predisposition, and to study a possible discrepancy with assessments made by the responsible clinicians. METHOD: All consecutively diagnosed patients with CRC from two Norwegian hospitals were included, and information on family history was collected in a detailed interview. We assessed information in medical records, and tumours were examined for LS-associated histop

Effects of heredity, age, weight, puberty, activity, and calcium intake on bone mineral density in children.

Osteoporosis is a disease that plagues older individuals, particularly women. At the usual age of diagnosis, limited therapy is available. By further delineating the factors that influence bone mineral acquisition before peak bone density is achieved, individuals at risk may be identified at an earlier age when therapies may be more effective. This was a study of 16 family units, 16 mothers, eight fathers, and their 28 children between the ages of 5 and 20 years. The evaluation consisted of a focused history, Tanner staging of adolescents, anthropometric data (height, weight), and spinal bone

[Late hereditary degenerative sensorineural hearing loss associated with IgA mesangial glomerulonephritis of probable autosomal dominant heredity].

We studied four generations of a Canary Islands family presenting a tardive heredodegenerative hearing loss, associated with IgA mesangial glomerulonephritis, of probable autosomal dominant heredity. With respect to the family, we revised Alport's syndrome, for possible transmission associated with X chromosome, as well as heredodegenerative hearing loss associated with renal pathology of autosomic transmission currently described; we differentiate these hearing losses from our case study, and we discuss the pathogeny of the auditive affection in the said hereditary syndromes. Lastly, we stres

Long non-coding RNAs could act as vectors for paternal heredity of high fat diet-induced obesity.

Long non-coding RNAs (lncRNAs) play an important role in epigenetic regulation, and abnormalities may lead to male infertility. To investigate whether lncRNAs are involved in intergenerational inheritance of obesity and obesity-induced decline in fertility, we divided mice into obesity (F0 mice fed a high-fat diet, F0-HFD) and non-obese (F0 mice fed normal chow, F0-NC) model groups and their male offspring (F1-HFD and F1-NC, respectively). We examined the differences in the expression levels of lncRNAs and mRNAs in the F0-HFD/F0-NC and F1-HFD/F1-NC groups. The results revealed similar expressi
