Reduced efficacy of an anti-toxin vaccine from senescence-driven attenuation of toxin virulence.

It remains unclear why vaccines targeting prominent microbial virulence factors often fail in clinical trials. Because microbial virulence depends on interaction with the host immune system, we investigated how changes in host immune function alter vaccine efficacy. Using a vaccine against Staphylococcus aureus α-toxin (Hla), which targets host metalloprotease ADAM10 on myeloid cells, we show that Hla virulence is reduced in aged mice due to diminished ADAM10 activity and impaired myeloid cell function. Depletion of myeloid cells with cyclophosphamide in young mice similarly reduced toxin viru

The Wolbachia Endosymbionts.

The Wolbachia endosymbionts encompass a large group of intracellular bacteria of biomedical and veterinary relevance, closely related to Anaplasma, Ehrlichia, and Rickettsia. This genus of Gram-negative members of the Alphaproteobacteria does not infect vertebrates but is instead restricted to ecdysozoan species, including terrestrial arthropods and a family of parasitic filarial nematodes, the Onchocercidae. The Wolbachia profoundly impact not only the ecology and evolution but also the reproductive biology of their hosts, through a wide range of symbiotic interactions. Because they are essen

Amblypygid-fungal interactions: The whip spider exoskeleton as a substrate for fungal growth.

Fungi and arthropods represent some of the most diverse organisms on our planet, yet the ecological relationships between them remain largely unknown. In animals, fungal growth on body surfaces is often hazardous and is known to cause mortality. In contrast, here we report the presence of an apparently non-harmful mycobiome on the cuticle of whip spiders (Arachnida: Amblypygi). The associations are not species-specific and involve a diversity of fungal species, including cosmopolitan and local decomposers as well as entomopathogens. We discuss the ecology of the detected fungal species and hyp

Development of Anncaliia algerae (Microsporidia) in Drosophila melanogaster.

Representatives of the genus Anncaliia are known as natural parasites of dipteran and coleopteran insects, amphipod crustaceans, but also humans, primarily with immunodeficiency. Anncaliia algerae-caused fatal myositis is considered as an emergent infectious disease in humans. A. (=Nosema, Brachiola) algerae, the best studied species of the genus, demonstrates the broadest among microsporidia range of natural and experimental hosts, but it has never been propagated in Drosophila. We present ultrastructural analysis of development of A. algerae in visceral muscles and adipocytes of Drosophila m

Host-microbe interactions in the pathogenesis and clinical course of sarcoidosis.

Sarcoidosis is a rare inflammatory disease characterized by the development of granulomas in various organs, especially in the lungs and lymph nodes. Clinics of the disease largely depends on the organ involved and may range from mild symptoms to life threatening manifestations. Over the last two decades, significant advances in the diagnosis, clinical assessment and treatment of sarcoidosis have been achieved, however, the precise etiology of this disease remains unknown. Current evidence suggests that, in genetically predisposed individuals, an excessive immune response to unknown antigen/s

Systematic protein interactome analysis of glycosaminoglycans revealed YcbS as a novel bacterial virulence factor.

Microbial pathogens have evolved several strategies for interacting with host cell components, such as glycosaminoglycans (GAGs). Some microbial proteins involved in host-GAG binding have been described; however, a systematic study on microbial proteome-mammalian GAG interactions has not been conducted. Here, we used Escherichia coli proteome chips to probe four typical mammalian GAGs, heparin, heparan sulphate (HS), chondroitin sulphate B (CSB), and chondroitin sulphate C (CSC), and identified 185 heparin-, 62 HS-, 98 CSB-, and 101 CSC-interacting proteins. Bioinformatics analyses revealed th

Interactions of the bacteriome, virome, and immune system in the nose.

Emerging evidence suggests that the nasal microbiome may influence host susceptibility to initial development and severity of respiratory viral infections. While not as extensively studied as the microbiota of the alimentary tract, it is now clearly established that the microbial composition of this niche is influenced by medical, social and pharmacological influences, predisposing some sub-populations to respiratory infections. The resulting specific microbial profiles may explain variance in susceptibility to viral infection. This review summaries the evolution and constituents of the commen

Salmonella Typhimurium vitamin B12-dependent methionine metabolism regulates Caenorhabditis elegans development and vitellogenesis.

Host-microbe interactions extend beyond infection, involving microbial metabolites, such as short-chain fatty acids, vitamins, and other bioactive compounds, that influence host physiology and development. Using Caenorhabditis elegans, an excellent model organism for host-microbe interactions, we demonstrate that Salmonella Typhimurium significantly accelerates C. elegans growth through a vitamin B12-dependent mechanism affecting methionine metabolism. We measured the reproductive maturity of C. elegans via embryo and oocyte production in response to various Salmonella virulence factor mutants

The role of gut microbiota in insulin resistance: recent progress.

Insulin resistance (IR) is the most critical pathophysiological basis of type 2 diabetes mellitus (T2DM). The mechanisms underlying IR are extremely complex, with variations observed among different patients and different tissues. Current research has gradually revealed characteristics of the gut microbiota and its role in metabolizing major nutrients during IR. The interaction between microbial communities and host metabolism has become a hotspot in modern medical research. This review aims to provide a comprehensive understanding of microbial metabolism related to IR by elucidating the effec

Prebiotic fructan chain length influences enteric microbiota-host GABAergic signaling and intestinal motility.

Dietary fiber ingestion serves as a critical regulator of intestinal motility and the structure and function of the enteric microbiome. Yet, the extent to which subtle structural differences among fibers modulate motility via microbiota-host interactions remains undefined. GABA is a microbial metabolite intimately related to microbial fructan fermentation and host intestinal motility. The purpose of this study was to investigate how fructan chain length influenced microbiota-host signaling underlying ileal and colonic contractions. Male and female mice were pair-fed diets containing no fiber (

Predicting inter-microbial host specificity in oral biofilms using a lightweight relation-aware knowledge graph model.

INTRODUCTION: The human oral cavity hosts a complex microbial ecosystem of bacteria, viruses, bacteriophages, and other microorganisms forming biofilms in different niches. Phage-bacteria host specificity is crucial in shaping microbial community, stability, and dysbiosis. mapping this specificity is limited by experimental constraints and traditional methods can't capture ecological complexity. The goal is to create a graph-based model that treats inter-microbial host specificity as a relational learning problem, integrating taxonomic, ecological, and infection data into a knowledge graph. The gut microbial influence on cholestatic liver disease.

Patients with cholestatic liver diseases like primary sclerosing cholangitis (PSC) and primary biliary cholangitis (PBC) have a different gut microbiome composition than healthy controls. In contrast with PBC, PSC has a strong association with inflammatory bowel disease and is the prototypical disease of the gut-liver axis. Still, there are some distinct overlapping microbial features in the microbiome of patients with PSC and PBC suggesting similarities in cholestatic diseases, although the possible pathogenetic involvement of these shared microbial changes is unknown. Herein, we present an o

Message in a vesicle - trans-kingdom intercommunication at the vector-host interface.

Vector-borne diseases cause over 700,000 deaths annually and represent 17% of all infectious illnesses worldwide. This public health menace highlights the importance of understanding how arthropod vectors, microbes and their mammalian hosts interact. Currently, an emphasis of the scientific enterprise is at the vector-host interface where human pathogens are acquired and transmitted. At this spatial junction, arthropod effector molecules are secreted, enabling microbial pathogenesis and disease. Extracellular vesicles manipulate signaling networks by carrying proteins, lipids, carbohydrates an

Host-initiated microbial association leads to stable ectosymbiosis in an ecological model.

Microbial symbiosis is widespread among metabolically coupled cells; it presumably gave rise to mitochondria. However, how such symbioses emerge, evolve, and stabilize are unknown, particularly in the prokaryotic domain where endosymbiosis is virtually nonexistent. Yet there is growing evidence suggesting that mitochondria originated from such a metabolically driven prokaryotic partnership rather than phagocytotic predation. While prokaryotes almost ubiquitously engage in metabolic syntrophy, it is unknown whether syntrophy alone can enable stable physical associations that could pave the road

Bacteria-induced immunity: from homeostasis to immune subversion in periodontitis.

This review synthesizes current evidence on host immune responses to bacteria, their roles in the development and progression of periodontitis, and emerging immunomodulatory therapeutic strategies designed to restore periodontal homeostasis. Periodontitis develops through interactions between a dysbiotic bacterial community that disrupts host-microbe homeostasis and a chronic, maladaptive interaction with the host immune system. Key immune subversion microorganisms such as Porphyromonas gingivalis trigger inflammation while subverting immune surveillance by evading complement-mediated clearanc

Microbiome network: a robust biomarker of skin health.

The skin microbiome is an integral part of the skin barrier. Its activity is shaped by microbe-microbe and microbe-host interactions. Understanding such interactions is an evolving field essential for uncovering the ecology and functions of a human microbiome community. Various studies have adapted network theory to explore the relationships of the microbiome community and its functional importance in health and diseases. The microbiome network is described as a biomarker depicting the interaction or correlation between two species, rather than their concentrations or abundances as in traditio

Exploring host-virus interaction to improve immunotherapy against Ebola virus.

Recent immunological advances have led to the development of FDA-approved immunotherapies against Ebola virus (EBOV). However, patients with high viral loads have not seen as large a benefit as mild cases. Here we discuss areas of investigation that may lead to adjunctive immune therapy for patients with severe EBOV disease.

Gut-liver axis: barriers and functional circuits.

The gut and the liver are characterized by mutual interactions between both organs, the microbiome, diet and other environmental factors. The sum of these interactions is conceptualized as the gut-liver axis. In this Review we discuss the gut-liver axis, concentrating on the barriers formed by the enterohepatic tissues to restrict gut-derived microorganisms, microbial stimuli and dietary constituents. In addition, we discuss the establishment of barriers in the gut and liver during development and their cooperative function in the adult host. We detail the interplay between microbial and dieta

A system biology perspective on environment-host-microbe interactions.

A vast, complex and dynamic consortium of microorganisms known as the gut microbiome colonizes the human gut. Over the past few decades, we have developed an increased awareness of its important role in human health. In this review we discuss the role of the gut microbiome in complex diseases and the possible causal scenarios behind its interactions with the host genome and environmental factors. We then propose a new analysis framework that combines a systems biology approach, cross-kingdom integration of multiple levels of omics data, and innovative in vitro models to yield an integrated pic
