Dual control of hydrogen cyanide biosynthesis by the global activator GacA in Pseudomonas aeruginosa PAO1.

The global response regulator GacA of Pseudomonas aeruginosa PAO1 positively controls the production of the quorum sensing signal molecule N-butanoyl-homoserine-lactone (C4-HSL) and hence the synthesis of several C4-HSL-dependent virulence factors, including hydrogen cyanide (HCN). This study presents evidence that GacA positively influences the transcription of the rhlI gene, specifying C4-HSL synthase, explaining the quorum sensing-dependent transcriptional control of the HCN biosynthetic genes (hcnABC). In addition, GacA was found to modulate hcn gene expression positively at a post-transcr

Kinetic studies of the reaction of ferric soybean leghemoglobins with hydrogen peroxide, cyanide and nicotinic acid.

A kinetic study of the reaction of two soybean leghemoglobins (components a and c) with hydrogen peroxide to form the oxidized compound (leghemoglobin IV) has been carried out over the pH range 2.5--10. Three different ionization processes of leghemoglobins with pKa values of 3,4.7 +/- 0.2 and 8.2 +/- 0.1 are required to explain the rate/pH profiles. Protonation of the former group and ionization of the latter cause a decrease in the rate of reaction of the hemoproteins with H2O2. The results are compared to those obtained for the reactions of plant peroxidases and myoglobin with H2O2. The res

The inhibition of mitochondrial cytochrome oxidase by the gases carbon monoxide, nitric oxide, hydrogen cyanide and hydrogen sulfide: chemical mechanism and physiological significance.

The four gases, nitric oxide (NO), carbon monoxide (CO), hydrogen sulfide (H(2)S) and hydrogen cyanide (HCN) all readily inhibit oxygen consumption by mitochondrial cytochrome oxidase. This inhibition is responsible for much of their toxicity when they are applied externally to the body. However, recently these gases have all been implicated, to greater or lesser extents, in normal cellular signalling events. In this review we analyse the chemistry of this inhibition, comparing and contrasting mechanism and discussing physiological consequences. The inhibition by NO and CO is dependent on oxyg

Mineral-facilitated aqueous synthesis of hydrogen cyanide from prebiotically abundant amino acids for chemical evolution.

Recent advances in prebiotic chemistry suggest that hydrogen cyanide (HCN) serves as a fundamental precursor for nearly all essential biomolecules and protometabolic processes for life's emergence. Although prevailing models of endogenous cyanide production rely on atmospheric methane, the availability of methane remains uncertain. Unlike methane, amino acids were likely prebiotically abundant, forming through multiple synthetic pathways under a methane-free, nonreducing atmosphere. Here, we demonstrate that HCN can form from proteinogenic amino acids in anaerobic aqueous solutions facilitated

Rapid 'on-column' preparation of hydrogen [11C]cyanide from [11C]methyl iodide via [11C]formaldehyde.

Hydrogen [11C]cyanide ([11C]HCN) is a versatile 11C-labelling agent for the production of 11C-labelled compounds used for positron emission tomography (PET). However, the traditional method for [11C]HCN production requires a dedicated infrastructure, limiting accessibility to [11C]HCN. Herein, we report a simple and efficient [11C]HCN production method that can be easily implemented in 11C production facilities. The immediate production of [11C]HCN was achieved by passing gaseous [11C]methyl iodide ([11C]CH3I) through a small two-layered reaction column. The first layer contained an N-oxide an

Intramolecular C-H⋅⋅⋅O Hydrogen-Bonded Solid Emitter as Colorimetric and Fluorometric Cyanide-Selective Chemodosimeter.

An indole-indanedione-based small molecule, compound 1Me, comprising an intramolecular C-H⋅⋅⋅O hydrogen-bond (H-bond) and with an intramolecular charge transfer (ICT) characteristic was prepared for the colorimetric and fluorometric detection of CN- . Compound 1Me is virtually nonfluorescent in solution but emits a strong yellow fluorescence in the solid state because of the intramolecular C-H⋅⋅⋅O H-bond, which inhibits the free rotation of the exocyclic C-C single bond of 1Me. The nucleophilic addition of CN- to the β-conjugated carbon of the indanedione group of 1Me, which breaks the intramo

Synergetic effect between Fe and Ti species on Fe-Ti-Ox for hydrogen cyanide purification.

The Fe-Ti-Ox catalysts with the different Fe contents were used for the catalytic hydrolysis of hydrogen cyanide (HCN) in the presence of H2O, which investigated the roles of Fe chemical valence and oxygen species in HCN removal and the production (NH3 and CO). The results implied that more amounts of Fe3+ species over Fe-Ti-Ox could increase the catalytic hydrolysis activity of HCN while Fe2+ species contributed to the formation of NH3 at high temperatures. Furthermore, the abundance of surface oxygen species was in favour of the catalytic performance of HCN.

CuO nanostructures as quartz crystal microbalance sensing layers for detection of trace hydrogen cyanide gas.

In this work, quartz crystal microbalance (QCM) sensors for detection of trace hydrogen cyanide (HCN) gas were developed based on nanostructural (flower-like, boat-like, ellipsoid-like, plate-like) CuO. Responses of all the sensors to HCN were found to be in an opposite direction as compared with other common volatile substances, offering excellent selectivity for HCN detection. The sensitivity of these sensors is dependent on the morphology of CuO nanostructures, among which the plate-like CuO has the highest sensitivity (2.26 Hz/μg). Comparison of the specific surface areas of CuO nanostruct

Investigation of hydrogen cyanide generation from the cyanoacrylate fuming process used for latent fingermark detection.

Cyanoacrylate fuming is one of the most common techniques employed for the detection of latent fingermarks on non-porous surfaces such as plastic and glass. The technique is generally applied by exposing items of interest to the vapours generated by heating a suitable quantity of commercial cyanoacrylate adhesive. In this study, the potential for highly toxic hydrogen cyanide (HCN) to be generated from the overheating of cyanoacrylate was investigated. Two commercial cyanoacrylate adhesives and two quantitative methods for the determination of HCN were employed: (i) the sodium picrate method;

Purity analysis of hydrogen cyanide, cyanogen chloride and phosgene by quantitative (13)C NMR spectroscopy.

Hydrogen cyanide, cyanogen chloride and phosgene are produced in tremendously large quantities today by the chemical industry. The compounds are also particularly attractive to foreign states and terrorists seeking an inexpensive mass-destruction capability. Along with contemporary warfare agents, therefore, the US Army evaluates protective equipment used by warfighters and domestic emergency responders against the compounds, and requires their certification at > or = 95 carbon atom % before use. We have investigated the (13)C spin-lattice relaxation behavior of the compounds to develop a quan

Unconventional hydrogen bonding to organic ions in the gas phase: stepwise association of hydrogen cyanide with the pyridine and pyrimidine radical cations and protonated pyridine.

Equilibrium thermochemical measurements using the ion mobility drift cell technique have been utilized to investigate the binding energies and entropy changes for the stepwise association of HCN molecules with the pyridine and pyrimidine radical cations forming the C5H5N(+·)(HCN)n and C4H4N2 (+·)(HCN)n clusters, respectively, with n = 1-4. For comparison, the binding of 1-4 HCN molecules to the protonated pyridine C5H5NH(+)(HCN)n has also been investigated. The binding energies of HCN to the pyridine and pyrimidine radical cations are nearly equal (11.4 and 12.0 kcal/mol, respectively) but wea

Improved Selectivity of CeMnOx/Pt@SnO2 Laminated MOS Sensor for Hydrogen Cyanide Under Temperature Dynamic Modulation.

Poor selectivity is one of the main bottlenecks restricting the development of metal oxide semiconductor (MOS) sensors. In this paper, using hydrogen cyanide (HCN) as the target gas, CeMnOx as the catalytic layer material and Pt@SnO2 as the gas-sensitive layer material, we have proposed a scheme to improve the selectivity of a catalytic layer/gas-sensitive layer-laminated MOS sensor under dynamic temperature modulation. We tested HCN and 12 kinds of battlefield environment simulation gases, and the results showed that the CeMnOx/Pt@SnO2 sensor, under the condition of temperature dynamic modula

Exploring adsorption mechanism of hydrogen cyanide and cyanogen chloride molecules on arsenene nanoribbon from first-principles.

The SIESTA package is employed in examining the electronic and adsorption features of the toxic asphyxiants hydrogen cyanide (HCN) and cyanogen chloride (NCCl) on armchair arsenene (As-arm) nanoribbon. Based on the generalized gradient approximation (GGA) with the Perdew-Burke-Ernzerhof (PBE) level of theory, we explored the adsorption of HCN and NCCl on As-arm nanoribbon. The significant parameters necessary to validate the electronic and stable nature of As-arm nanoribbon are computed with the help of formation energy and energy gap change. The density of states (DOS) spectrum and the energy

Formation of highly toxic hydrogen cyanide upon ruby laser irradiation of the tattoo pigment phthalocyanine blue.

Since laser treatment of tattoos is the favored method for the removing of no longer wanted permanent skin paintings, analytical, biokinetics and toxicological data on the fragmentation pattern of commonly used pigments are urgently required for health safety reasons. Applying dynamic headspace-gas chromatography with mass spectrometric detection (DHS-GC/MS) and comprehensive two-dimensional gas chromatography coupled to time-of-flight mass spectrometry (GCxGC-ToF-MS), we identified 1,2-benzene dicarbonitrile, benzonitrile, benzene, and the poisonous gas hydrogen cyanide (HCN) as main fragment

Isocitrate lyase supplies precursors for hydrogen cyanide production in a cystic fibrosis isolate of Pseudomonas aeruginosa.

Pseudomonas aeruginosa colonizes and can persist in the lungs of cystic fibrosis (CF) patients for decades. Adaptation of P. aeruginosa to the CF lung environment causes various genotypic and phenotypic alterations in the bacterium that facilitate persistence. We showed previously that isocitrate lyase (ICL) activity is constitutively upregulated in the P. aeruginosa CF isolate FRD1. We show here that high ICL activity in FRD1 contributes to increased hydrogen cyanide (HCN) production by this isolate. Disruption of aceA, which encodes ICL, results in reduced cyanide production by FRD1 but does

Reduction of Hydrogen Cyanide in Cassava Peel With Sodium Bicarbonate and Heating Using a Stirrer Chamber Unit.

Cassava peel, a by-product of cassava root processing, can be used as food if processed properly. It is important to note that the high-cyanogenic glycoside content in cassava plants has the potential to produce hydrogen cyanide (HCN), which has been proven to cause health problems. Therefore, it would be beneficial to consider a method that could help reduce HCN levels. This study explored the impact of sodium bicarbonate (NaHCO3) and heating on the HCN of cassava peel during the initial stage of food processing. This study is experimental in nature, as a follow-up to previous research. The c
