Measles and mumps vaccination as a model to investigate the developing immune system: passive and active immunity during the first year of life.

Evaluations of neutralizing antibody responses in 6-, 9- and 12-month-old infants given measles or mumps vaccine indicated that 6-month-old infants had diminished humoral immune responses associated with passive antibody effects, but also had an intrinsic deficiency in antiviral antibody production, which was independent of passive antibody effects. In contrast, lower neutralizing antibody titers in 9-month-olds were related only to passive antibody effects. Measles and mumps-specific T-cell proliferation and interferon-gamma (IFNgamma) production were induced by vaccination at 6, 9 or 12 mont

The immune response of the mouse to lymphocytic choriomeningitis virus. II. Active suppression of cell-mediated immunity by infection with high virus doses.

Infection of CBA/J mice with low doses of strain WE lymphocytic choriomeningitis (LCM) virus resulted in high cell-mediated and relatively low humoral virus-specific immune phenomena; anamnestic responses were marked. In contrast, infection with high doses of this virus induced no or low degrees of cell-mediated immune phenomena but higher antibody concentrations. Subsequent challenge of these mice did not reveal cell-mediated immunity. However, transfer of spleen cells from mice suppressed as to cell-mediated immunity together with virus into X-irradiated recipients led to marked anamnestic r

Immune Activation Signatures Associated with Fatigue in Cancer Patients Undergoing Immune Checkpoint Inhibitor Therapy.

UNLABELLED: Fatigue is a common and often debilitating adverse event among patients receiving immune checkpoint inhibitor (ICI) therapy, yet its underlying immunologic mechanisms remain poorly defined. We prospectively collected clinical data and blood samples from patients with solid tumors receiving ICI therapy. Patients were prospectively surveyed at month 2, 4, or 6 on treatment to assess for the presence and severity of fatigue as compared with treatment baseline. We analyzed peripheral lymphocyte populations by cytometry by time of flight and peripheral levels of 39 cytokines with a Lumi

Maternal immune activation during gestation modulates offspring immune profiles in a nonhuman primate model.

BACKGROUND: Maternal Immune Activation (MIA) during pregnancy is an environmental risk factor implicated in neurodevelopmental disorders such as autism spectrum disorder and schizophrenia. While numerous studies have shown that MIA can lead to neuropathological and behavioral abnormalities in offspring, the consequences for immune system development and function are less well characterized. METHODS: To assess the impact of MIA on offspring immune function, we utilized samples from 24 nonhuman primate (NHP) dam-infant pairs. Pregnant dams received either saline (control) or polyinosinic: polyc

Culex pipiens pipiens: characterization of immune peptides and the influence of immune activation on development of Wuchereria bancrofti.

Stimulating or augmenting the innate immune response of insect vectors has been shown to impede or disrupt the development and transmission of eukaryotic pathogens; however, the majority of such studies have utilized model systems and not natural parasite-vector systems. The Culex pipiens complex of mosquitoes functions as a primary urban vector of Wuchereria bancrofti, a causative agent of lymphatic filariasis. To test the effects of immune activation on this vector-parasite interaction, Culex pipiens pipiens from the filariasis-endemic Nile Delta were subjected to bacteria inoculation and su

Active immunization against tetanus in guinea-pigs. An attempt to evaluate "rapid immunization" with tetanus vaccine.

The effect of 3 schedules for tetanus vaccination on the immunity developed by guinea-pigs was investigated, using 1.5 Lf adsorbed tetanus toxoid injected subcutaneously. The following injection schedules were used: a) 5 injections: initially and after 3, 7, 10, and 13 days; b) 3 injections:: initially and after 2 and 4 weeks; c) 2 injections: initially and after 4 weeks. Blood samples were taken after 1, 2, 3, 4, 6, and 8 weeks and the titres of tetanus antitoxin were recorded. No significant difference in the titres was observed within 2 weeks. Immunity was also tested by challenge with diff

Arabidopsis NLR Immune Receptors: Structural Diversification, Signaling Networks, and Emerging Principles of Immune Activation.

Plant intracellular nucleotide-binding leucine-rich repeat receptors (NLRs) are central to effector-triggered immunity (ETI), but the rapid expansion of structural data has outpaced an integrated explanation of how receptor architecture, oligomerization, and network context determine immune output. Recent structural and biochemical studies, particularly in Arabidopsis, have revealed that NLRs operate as dynamic signaling assemblies rather than simple pathogen-recognition switches. Here, we synthesize recent evidence across coiled-coil NLRs (CNLs), Toll/interleukin-1 receptor NLRs (TNLs), and R

Ultrasound-Activated Nanoinhibitors Blocking the Chemo-Immune Checkpoint to Overcome Chemotherapy-Induced Immune Resistance.

Breast cancer features an immunosuppressive tumor microenvironment following chemotherapy and develops resistance to conventional immune checkpoint inhibitors. Through single-cell database mining and in vitro analysis, we demonstrate that chemotherapy upregulates Xkr8 expression and promotes phosphatidylserine (PS) externalization, which mediates macrophage- and dendritic cell (DC)-intrinsic immunosuppression. To address this, we fabricate an ultrasound-activated nanoinhibitor (Sono-TT8) that co-delivers tirapazamine (TPZ) and siXkr8 using a TiO2-derived sonosensitizer functionalized with the

Spatial Immune Coding in Tumor-Draining Lymph Nodes: Functional Compartmentalization of Immune Activation and Immunosuppression.

Tumor-draining lymph nodes (TDLNs) are important immune organs linking primary tumors with systemic immune responses. They support tumor antigen presentation, T-cell priming, and effector immune responses, but under sustained tumor influence they can also be remodeled into microenvironments that promote immune escape and metastatic colonization. Conventional methods, including flow cytometry, bulk RNA sequencing, and routine immunohistochemistry, have advanced our understanding of TDLN immunity but cannot simultaneously preserve tissue architecture, cell identity, and spatial cell-cell relatio

Identification of TRAPPC4 as a Key Autoantigen in Immune-Related Pancytopenia: Epitope Characterization and Immune Activation Mechanisms.

OBJECTIVE: Immune-related pancytopenia (IRP) is characterized by autoantibody-mediated destruction or suppression of bone marrow cells, leading to pancytopenia. This study aimed to explore the role of trafficking protein particle complex subunit 4 (TRAPPC4) as a key autoantigen in IRP, including epitope identification and immune activation mechanisms. MATERIALS AND METHODS: A total of 90 participants were included in the study, divided into four groups: 30 newly diagnosed IRP patients, 25 patients with IRP in remission, 20 patients with other hematological conditions (severe aplastic anemia [

Combination of tumor antigen drainage and immune activation to promote a cancer-immunity cycle against glioblastoma.

While conventional cancer modalities, such as chemotherapy and radiotherapy, act through direct killing of tumor cells, cancer immunotherapy elicits potent anti-tumor immune responses thereby eliminating tumors. Nevertheless, promising outcomes have not been reported in patients with glioblastoma (GBM) likely due to the immune privileged status of the central nervous system and immunosuppressive micro-environment within GBM. In the past years, several exciting findings, such as the re-discovery of meningeal lymphatic vessels (MLVs), three-dimensional anatomical reconstruction of MLV networks,

Immunization with Pneumocystis carinii A121-85 antigen activates immune function against P. carinii.

BACKGROUND: Pneumocystis pneumonia (PcP), which is caused by Pneumocystis carinii, is a life-threatening infection that affects immunocompromised individuals. Unfortunately, chemoprophylaxis and dapsone are only effective for half of the patients with PcP, indicating that additional preventive methods are needed. We predicated the pneumocystis surface protein A12 sequence 1-85 by DNAStar software and BepiPred, and identified it as a potential vaccine candidate by bioresearch. METHODS: We used recombinant A121-85 as antigen to immunized mice and detected serum titer of IgG, expression of infla

The role of rare innate immune cells in Type 2 immune activation against parasitic helminths.

The complexity of helminth macroparasites is reflected in the intricate network of host cell types that participate in the Type 2 immune response needed to battle these organisms. In this context, adaptive T helper 2 cells and the Type 2 cytokines interleukin (IL)-4, IL-5, IL-9 and IL-13 have been the focus of research for years, but recent work has demonstrated that the innate immune system plays an essential role. Some innate immune cells that promote Type 2 immunity are relatively abundant, such as macrophages and eosinophils. However, we now appreciate that more rare cell types including g

The characterization of the exposure to immune mediated apoptosis and the regulation of immune cytotoxic activity in the environment of a neoplasm and in decidua.

Acquiring the immune-mediated apoptosis and the ability to regulate the cytotoxic immune response are the main phenomena playing fundamental roles in such situations as neoplasm survival and creation of immune tolerance during pregnancy. The aim of this study was to investigate these phenomena through the evaluation of metallothionein and RCAS1 proteins in neoplasm and its healthy environment (clear surgical margin), physiological conditions in placenta and its environment (decidua) and the comparison to non-neoplasmatic lesions originating from the environment (nasal polyps, endometriosis). Immune dysfunction and immune restoration disease in HIV patients given highly active antiretroviral therapy.

BACKGROUND: Some immune defects caused by HIV infection resolve following treatment with highly active antiretroviral therapy (HAART), but residual immune dysfunction may cause disease. Problems with the regulation of the restored immune system in the first six months of treatment can lead to atypical presentations of mycobacterial, cytomegalovirus (CMV), hepatitis B virus or hepatitis C virus (HCV) disease. We defined these conditions as immune restoration diseases (IRD) and showed that they occur in 30-40% of individuals who begin HAART from low CD4 T cell counts. OBJECTIVES: Analysis of im

Distribution and phenotype of immune cells in normal human gingiva: active immune response versus unresponsiveness.

The oral cavity, and particularly the gingival mucosa, is continuously exposed to numerous food and bacterial plaque antigens, though evident immunologic reactions are uncommon. It is therefore possible that the mucosal associated lymphoid tissue (MALT) of this region is preferentially biased towards unresponsiveness, rather than immune cell activation. The distribution and phenotype of immune cells in normal human gingiva were examined. Their distribution varied, and high and low cellularity areas could be distinguished in the same specimen. The number of CD3 positive (CD3+) T lymphocytes was
