Convenient alternative synthesis of the Malassezia-derived virulence factor malassezione and related compounds.

Lipophilic yeasts of the genus Malassezia are commensal fungi that constitute the normal skin microbiota but may become pathogenic. These fungi, especially M. furfur, convert tryptophan into various alkaloid indoles such as malassezione, which may serve as virulence factors. To facilitate testing of malassezione as an aryl hydrocarbon receptor agonist and potential glucokinase activator, we developed a convenient synthetic route from commercially available indole-3-acetic acid. Treatment of the N-Boc-protected indole-3-acetic acid with N-ethyl-N'-(3-dimethylaminopropyl)carbodiimide (EDC) in th

Sequential electrophilic trifluoromethanesulfanylation-cyclization of tryptamine derivatives: synthesis of C(3)-trifluoromethanesulfanylated hexahydropyrrolo[2,3-b]indoles.

A practical and efficient synthesis of C(3)-trifluoromethanesulfanylated hexahydropyrrolo[2,3-b]indoles 5 from tryptamine derivatives was described. The features of this synthesis included electrophilic activation of C(3) of tryptamine derivatives with "CF(3)S(+)" and cascade ring cyclization by carbamate nucleophile attacking at C(2). Surprisingly, when Lewis acid (BF(3)·OEt(2)) was used as activator instead of proton acid (TsOH·H(2)O) for the electrophilic trifluoromethanesulfanylation of tryptamine derivatives, the uncyclized product 6 was formed preferentially. This sequential trifluoromet

Iboga Inspired N-Indolylethyl-Substituted Isoquinuclidines as a Bioactive Scaffold: Chemoenzymatic Synthesis and Characterization as GDNF Releasers and Antitrypanosoma Agents.

The first stage of the drug discovery process involves the identification of small compounds with biological activity. Iboga alkaloids are monoterpene indole alkaloids (MIAs) containing a fused isoquinuclidine-tetrahydroazepine ring. Both the natural products and the iboga-inspired synthetic analogs have shown a wide variety of biological activities. Herein, we describe the chemoenzymatic preparation of a small library of novel N-indolylethyl-substituted isoquinuclidines as iboga-inspired compounds, using toluene as a starting material and an imine Diels-Alder reaction as the key step in the s

Silver-catalyzed asymmetric [3+2] cycloaddition via dynamic kinetic resolution: access to chiral oxazolines bearing C-N axial chirality.

A silver-catalyzed asymmetric [3+2] cycloaddition of N-aryl indole aldehydes/ketones with isocyanoacetates is reported. Facilitated by a cinchona alkaloid-derived ligand, this protocol enables the simultaneous construction of C-N axial chirality and two stereocenters in oxazolines under mild conditions. The reaction affords products in excellent yields (up to 99%) with high diastereo- and enantioselectivities (up to >20 : 1 dr, 99% ee).

Sarniensine, a mesembrine-type alkaloid isolated from Nerine sarniensis, an indigenous South African Amaryllidaceae, with larvicidal and adulticidal activities against Aedes aegypti.

A new mesembrine-type alkaloid, named sarniensine, was isolated together with tazettine, lycorine, the main alkaloid, and 3-epimacronine from Nerine sarniensis, with the last two produced for the first time by this plant. This Amaryllidaceae, which is indigenous of South Africa, was investigated for its alkaloid content, because the organic extract of its bulbs showed strong larvicidal activity with an LC50 value of 0.008μgμL-1 against first instar Aedes aegypti larvae and with an LD50 value 4.6μg/mosquito against adult female Ae. aegypti, which is the major vector for dengue, yellow fever and

OxaD: A Versatile Indolic Nitrone Synthase from the Marine-Derived Fungus Penicillium oxalicum F30.

Indole alkaloids are a diverse class of natural products known for their wide range of biological activities and complex chemical structures. Rarely observed in this class are indolic nitrones, such as avrainvillamide and waikialoid, which possess potent bioactivities. Herein the oxa gene cluster from the marine-derived fungus Penicillium oxalicum F30 is described along with the characterization of OxaD, a flavin-dependent oxidase that generates roquefortine L, a nitrone-bearing intermediate in the biosynthesis of oxaline. Nitrone functionality in roquefortine L was confirmed by spectroscopic

Reactivity and synthesis inspired by the Zincke ring-opening of pyridines.

The century-old Zincke process for ring-opening of pyridinium salts produces 5-amino-2,4-pentadienals, a type of donor-acceptor dienes known as Zincke aldehydes. Inspired by this reasonably general and often efficient process for dearomatization, my laboratory has used pyridines as a starting point for heterocycle synthesis, which resulted in unusual syntheses of indoles, pyrrolines, and a formal synthesis of the natural product porothramycin A. Furthermore, our study of the reactivity of Zincke aldehydes has led to accidental discoveries of pericyclic cascade reactions that produce Z-α,β-unsa

Biocatalytic Oxidative [3 + 2] Cycloaddition Enables Divergent Synthesis of (-)-α-Cyclopiazonic Acid and Derived Alkaloids.

The asymmetric construction of highly congested polycyclic architectures remains a formidable challenge in chemical synthesis. Biocatalysis offers exquisite stereocontrol, yet the repertoire of enzyme-catalyzed cycloadditions remains notably limited. Here, we elucidate the intriguing oxidative [3 + 2] cycloaddition capability of a flavoenzyme CpaO and develop it as a robust biocatalyst for asymmetric construction of the pentacyclic scaffold of (-)-α-Cyclopiazonic acid ((-)-α-CPA), a nanomolar inhibitor of sarco/endoplasmic reticulum calcium(II)-dependent ATPase. Crosstalk between melatonin and nitric oxide restrains Cadmium-induced oxidative stress and enhances vinblastine biosynthesis in Catharanthus roseus (L) G Don.

Nitric oxide functions downstream of the melatonin in adjusting Cd-induced osmotic and oxidative stresses, upregulating the transcription of D4H and DAT genes, and increasing total alkaloid and vincristine contents. A few studies have investigated the relationship between melatonin (MT) and nitric oxide (NO) in regulating defensive responses. However, it is still unclear how MT and NO interact to regulate the biosynthesis of alkaloids and vincristine in leaves of Catharanthus roseus (L.) G. Don under Cd stress. Therefore, this context was explored in the present study. Results showed that Cd t
