Hepatotoxic alterations induced by subchronic exposure of rats to formulated fenvalerate (20% EC) by nose only inhalation.

OBJECTIVE: Fenvalerate (20% EC) is a synthetic pyrethroid, which is commonly used in India by farmers for the protection of many food and vegetable crops against a wide variety of insects. However, its inhalation toxicity data is very limited in the literature due to the fact that the exposure levels associated with these effects were usually not reported. Hence, inhalation exposure was carried out to investigate the hepatotoxic effects. METHOD: Adult male rats were exposed to fen for 4 h/day, 5 days a week for 90 days by using Flow Past Nose Only Inhalation Chamber. Sham treated control rats

Inhalation toxicokinetics of 1,2,4-trimethylbenzene in volunteers: comparison between exposure to white spirit and 1,2,4-trimethylbenzene alone.

The objective of this study was to compare the toxicokinetics of inhaled 1,2,4-trimethylbenzene (1,2,4-TMB) in man after exposure to white spirit with that observed after exposure to 1,2,4-TMB alone. TMBs occur mainly in petroleum products and the TMBs or their metabolites have been suggested as suitable biomarkers of exposure to white spirit and other distillation products. The toxicokinetics were studied in 9 male, healthy volunteers exposed to solvent vapours in an exposure chamber for 2 h during a work load of 50 W. The subjects were exposed to 11 mg/m3 of 1,2,4-TMB on two occasions; durin

Low flow anaesthesia reduces occupational exposure to inhalation anaesthetics. Environmental and biological measurements in operating room personnel.

In the present study we evaluated the occupational exposure to N2O and isoflurane during open circuit (OC) (fresh gas flow > or = minute volume) and low flow (LF) (fresh gas flow = 1.5 l/min) anaesthesia. The effects of active scavenging and of a charcoal filter positioned on the exhausting branch of the ventilator on environmental and urinary concentrations of inhalation anaesthetics were also investigated. The study was carried out in the same operating room provided with non-recirculating air changes (10 per hour). It involved anaesthetists and nurses during routine activity. N2O and isoflu

Effects of prenatal alcohol exposure in mice: influence of an ADH inhibitor and a chronic inhalation study.

In order to examine the effects of an alcohol dehydrogenase inhibitor on the embryotoxic effects of ethanol, pregnant ICR mice were treated with 100 mg/kg pyrazole prior to ethanol injection. I.p. treatment with 2 or 4 g/kg ethanol on day 7 of gestation increased the prenatal mortality rate and produced external and skeletal malformations in the offspring, and the embryotoxic effects were potentiated by pyrazole pretreatment, suggesting that ethanol rather than its metabolites is mainly responsible for the embryotoxicity. In the second experiment, we housed pregnant mice in an ethanol-vapor bo

Occupational exposure to inhalational anesthetics and the risk of spontaneous abortion: a systematic review and meta-analysis.

BACKGROUND: Healthcare professionals, particularly anesthesiologists, nurses, and dental staff, are routinely exposed to inhalational anesthetic agents such as nitrous oxide and halogenated gases. Although scavenging and ventilation systems have reduced ambient levels, concerns remain regarding reproductive risks associated with chronic exposure. Previous studies have yielded inconsistent results. A systematic review and meta-analysis of published observational studies was conducted to clarify the relationship, assessing reproductive outcomes among healthcare workers who were exposed. OBJECTI

Benzalkonium chloride accumulates and causes toxicity in the heart and pancreas following the repeated inhalation exposures.

Benzalkonium chloride (BKC) is used as a disinfectant and preservative in aerosolized formulations, including various household cleaning products. Although primary organ toxicity of BKC has been reported, secondary organ toxicity after inhalation exposure remains unclear. This study evaluated the biodistribution of radiolabeled [14C] BKC using quantitative whole-body autoradiography in rats. Autoradiography after a single intranasal administration revealed significant accumulation and prolonged retention of radioactivity in the heart and pancreas, suggesting potential secondary organ toxicity

Childhood asthma, inhaled corticosteroid exposure, and risk of cataract in adulthood: a register-based study.

BACKGROUND: Cataract is the leading cause of blindness worldwide, with corticosteroid treatment being a known risk factor. The long-term impact of childhood asthma and, particularly, inhaled corticosteroid (ICS) use in adulthood on cataract development remains unclear. METHODS: This register-based study investigated the prevalence and risk of cataract in Danish adults diagnosed with childhood asthma who, between 1950 and 1979, spent four months at an asthma care facility in Kongsberg, Norway. Follow-up was conducted in 2021 using Danish national health registries (2006-2018). These individual

Exposure effects of inhaled nickel nanoparticles on the male reproductive system via mitochondria damage.

Nickel nanoparticles (Ni NPs) have a wide range of application prospects, however there is still a lack of their safety evaluation for the reproductive system. Nowadays, male reproductive health has been widely concerned for the increasing incidence of male infertility. To investigate the male reproductive toxicity induced by Ni NPs and its relation with the mitochondrial fission and mitophagy, male mice were administered with or without 5, 15, and 45 mg/kg of Ni NPs by intratracheal instillation. At the end of intervention, sex hormone level, sperm abnormality rate, pathological morphology of

Demonstrating the Protective Efficacy of the Novel Fluoroquinolone Finafloxacin against an Inhalational Exposure to Burkholderia pseudomallei.

Burkholderia pseudomallei is the causative agent of melioidosis, a serious disease endemic in Southeast Asia and Northern Australia. Antibiotic treatment is lengthy and relapse often occurs. Finafloxacin is a novel fluoroquinolone with increased antibacterial activity in acidic conditions in contrast to other fluoroquinolones which demonstrate reduced activity at a lower pH. Therefore, finafloxacin may have improved efficacy against B. pseudomallei, which can survive within host cells where the local pH is acidic. In vitro analysis was performed using MICs, minimal bactericidal concentrations

Reconstructing population exposures from dose biomarkers: inhalation of trichloroethylene (TCE) as a case study.

Physiologically based pharmacokinetic (PBPK) modeling is a well-established toxicological tool designed to relate exposure to a target tissue dose. The emergence of federal and state programs for environmental health tracking and the availability of exposure monitoring through biomarkers creates the opportunity to apply PBPK models to estimate exposures to environmental contaminants from urine, blood, and tissue samples. However, reconstructing exposures for large populations is complicated by often having too few biomarker samples, large uncertainties about exposures, and large interindividua

Inhalational anthrax after bioterrorism exposure: spectrum of imaging findings in two surviving patients.

The radiographic and computed tomographic (CT) findings in two patients with documented inhalational anthrax resulting from bioterrorism exposure are presented. Chest radiographs demonstrated mediastinal widening, adenopathy, pleural effusions, and air-space disease. Chest CT images revealed enlarged hyperattenuating mediastinal and hilar lymph nodes and edema of mediastinal fat. Chest CT findings are helpful for making the initial diagnosis. To the authors' knowledge, the spectrum and follow-up of CT findings have not been previously described.

Th2 responses induced by epicutaneous or inhalational protein exposure are differentially dependent on IL-4.

Atopic individuals are predisposed to mounting vigorous Th2-type immune responses to environmental allergens. To determine the factors responsible, animal models that closely mimic natural modes of allergen exposure should prove most informative. Therefore, we investigated the role of IL-4, a known Th2-promoting cytokine, in generation of Th2 responses after exposure of either the skin or airway to soluble protein. Compared with wild-type (WT) mice, IL-4-deficient (IL-4(-/-)) mice showed markedly impaired Th2 activation after primary exposure to inhaled ovalbumin (OVA), with decreased OVA-spec

Effect of exposure to low levels of ozone on the response to inhaled allergen in allergic asthmatic patients.

STUDY OBJECTIVES: In a previous study published by our group, six out of nine subjects with mild allergic asthma were shown to have an enhanced response to allergen challenge following a 1-h exposure in an 0.8-m3 exposure chamber (modified from a body plethysmograph) to an average of 120 parts per billion (ppb) ozone at rest. Other studies failed to confirm this effect. In the present study, using a similar design, we reexamined this effect using a larger group of asthmatics and a larger chamber allowing minimal fluctuations in ozone levels during exposures. DESIGN: Prospective, randomized si

[Changes in lipid metabolism in pregnant white rats and their fetuses in inhalational exposure to carbon disulfide].

It is known from the literature that carbon bisulphide affects lipid metabolism, but observations in this field are quite controversial. In the present work the authors studied the influence of carbon bisulphide on some parameters of lipid metabolism in pregnant albino rats, following inhalatory exposure to carbon bisulphide concentrations of 50, 100 and 200 mg/m3, 8 hours daily throughout the whole gestation period. The lipid metabolism changes were followed up both in two subsequent generations. The offsprings were examined at birth and after they had reached sexual maturity, at the end of g

Prevalence and patterns of inhalational exposures in patients with interstitial lung diseases: a cross-sectional study.

Given the etiologic importance of inhalational exposures in specific interstitial lung disease (ILD) subtypes like hypersensitivity pneumonitis (HP) and pneumoconiosis, this study systematically evaluates exposure patterns across all ILD subtypes and assesses the utility of a standardized questionnaire for improved exposure identification.  In this cross-sectional study, ILD patients were recruited between July 2023 and March 2024. Demographic, clinical, pulmonary function, radiological, and immunological parameters were recorded, and ILD subtypes were assigned by multidisciplinary discussion.
