Valine administration in the hypothalamus alters the brain and plasma metabolome in rainbow trout.

Central administration of valine has been shown to cause hyperphagia in fish. Although mechanistic target of rapamycin (mTOR) is involved in this response, the contributions to feed intake of central and peripheral metabolite changes due to excess valine are unknown. Here, we investigated whether intracerebroventricular injection of valine modulates central and peripheral metabolite profiles and may provide insights into feeding response in fish. Juvenile rainbow trout (Oncorhynchus mykiss) were administered an intracerebroventricular injection of valine (10 µg·µL-1 at 1 μL·100·g-1 body wt), a

Partition of antimicrobial additives in an intravenous emulsion and their effect on emulsion physical stability.

A number of antimicrobial agents are potentially applicable to the preservation of small-volume parenteral emulsions. However, the physical stability of these emulsions is of paramount importance in ensuring their safety, and it is possible that antimicrobial additives could reduce the emulsion stability by a number of mechanisms. We have studied the effects of several antimicrobial agents on the physical stability of Diprivan, an intravenous anaesthetic emulsion. A particular problem is that many antimicrobial additives require an acidic pH in order to be effective (e.g. sodium benzoate, sodi

Subacute sclerosing panencephalitis aggravated during the intraventricular interferon administration therapy.

Human lymphoblastoid interferon-alpha was given by intraventricular administration via an Ommaya reservoir to a woman who entered Jabbour stage III at the age of 25 years. The first dose was of 150,000 IU, and daily doses were gradually increased; the total dose over a 42-day period was 33,550,000 IU. The patient's neurological disability index score before this treatment was 56.3%, and 75% after interferon treatment ended. After other treatment for 6 months, the patient died. Results of the autopsy showed perivascular lymphocyte infiltration in the cerebrum and brain stem, with diffuse gliosi

[Formation of alcohol motivation under blockade of protein synthesis by cycloheximide].

The influence of protein blocker cycloheximide on ethanol intake in rats under condition of development of alcohol motivation was studied. It was found that intracerebroventricular injection of cycloheximide caused inhibition of alcohol intake in rats which had free choice between water and 20% ethanol solution. The blockade action of cycloheximide on the development of alcohol motivation was dependent on initial preference of ethanol in rats and was more strong in rats with originally low preference for ethanol.

Effect of intracerebroventricular injection of IGF-I on circulating growth hormone concentrations in the sheep.

The effect of intracerebroventricular administration of IGF-1 on circulating growth hormone (GH) concentrations has been studied in sheep. Twenty sheep were fitted with jugular vein catheters and with indwelling cerebroventricular cannulae. IGF-I was injected into a lateral cerebral ventricle and changes in the circulating concentrations of GH were measured in jugular vein blood samples. Administration of saline had no effect on circulating GH concentrations over a 3-hr period, and administration of IGF-I (at 1, 3 and 10 micrograms/sheep) also had no significant effect on circulating GH concen

Sodium salicylate centrally augments ventilation through cholinergic mechanisms.

Several different stimuli, including hydrogen ions, may exert their effect on central ventilatory control through cholinergic mechanisms. Salicylates are known to be central respiratory stimulants. Therefore this study explored whether the ventilatory effect of sodium salicylate (SAL) is through cholinergic mechanisms. Ventriculocisternal perfusion was used in spontaneously breathing anesthetized (pentobarbital sodium, 30 mg/kg) mongrel dogs to study the effects of SAL (50 mM), atropine (ATR, 4.8 mM), and SAL-ATR on ventilation. After 15 min of perfusion with mock cerebrospinal fluid, each tes

The roles of glucagon and adrenal epinephrine in mediating hyperglycemia induced by third cerebroventricular injection of bombesin.

The roles of glucagon and adrenal epinephrine in mediating bombesin-induced central hyperglycemia were further studied in anesthetized rats. Bombesin (10(-9) mol) injected into the third cerebral ventricle produced an increase in plasma concentrations of glucose, glucagon, and epinephrine. Prior bilateral adrenalectomy completely prevented the hyperglucagonemic and hyperglycemic responses to third cerebral ventricle injection of bombesin. These results support the view that bombesin-induced increases in plasma glucose and glucagon are fully dependent on adrenal epinephrine secretion. Furthermo

Alteration of food intake following intracerebroventricular administration of plasma from free-feeding domestic fowl.

Four experiments were conducted to investigate the presence of a factor in the plasma of the domestic fowl that alters food intake when injected into the lateral cerebral ventricle. Plasma collected from free-feeding and 24-hour fasted birds was lyophilized and reconstituted to 2, 4, or 5 times normal concentration. Stainless steel guide cannulae were stereotaxically implanted into the lateral cerebral ventricle of 8-week-old Single Comb White Leghorn cockerels. Free-feeding birds received 10 microliter intracerebroventricular (ICV) injections of normal (Experiment 1) and concentrated (Experim

The treatment of subacute sclerosing panencephalitis with interferon: a case report.

A child with subacute sclerosing panencephalitis (SSPE) received intraventricular alpha interferon (IFN) as experimental treatment. The course was monitored by colleagues in pediatric neurology, neuro-opthalmology and clinical psychology, also by monthly EEGS and brain CT scans. Two courses of IFN were administered. During the first course, improvement occurred nearly to the premorbid level of function. About 1 month after this trial was stopped, a severe recrudescence rapidly produced a thalamic state. A second trial of IFN resulted in less improvement. When the brain CT showed severe degener

Changes in body temperature and oxygen consumption rate of conscious mice produced by intrahypothalamic and intracerebroventricular injections of delta 9-tetrahydrocannabinol.

delta 9-Tetrahydrocannabinol (delta 9-THC) was injected into the preoptic area of the anterior hypothalamus or into the third or fourth cerebral ventricle of the conscious mouse through a chronically implanted cannula and the effects on body temperature and oxygen consumption rate were measured. At an ambient temperature of 22 degrees C, injections of delta 9-THC into the fourth ventricle (5 and 10 microgram) produced dose-dependent falls in rectal temperature. Hypothermia was also observed after injections of the drug into the hypothalamus (5 and 10 microgram) or into the third ventricle (10

Hyperthermic effect of prostacyclin injected into the third cerebral ventricle of the cat.

Prostaglandin E2 may not be solely responsible for hyperthermia produced by central administration of sodium arachidonate. To determine its effect, if any, on body temperature prostacyclin sodium salt, another product of prostaglandin endoperoxides, was injected into the third cerebral ventricle of unrestrained, unanesthetized cats while deep body temperature was recorded automatically. Doses of 2--25 microgram, in a volume of 0.05 ml saline solution, did not appreciably alter body temperature. A 100 microgram dose produced hyperthermia in five of six animals. Administration of 1 mg prostacycl

The role of circulating renin in drinking in response to isoprenaline.

1. In nephrectomized rats, S.C. (0.12 mg. kg body wt.(-1)) or intracerebroventricular (I.C.V.: 0.03 mg. kg(-1)), isoprenaline failed to elicit drinking. However, when preceded (5-20 min) by a non-dipsogenic dose of I.V. pig renin, S.C. isoprenaline induced a marked, and I.C.V. isoprenaline a smaller drinking response. 2 hr after I.V. renin, S.C. isoprenaline no longer caused drinking.2. The effect of intraventricular 6-hydroxydopamine on the content of oxytocin and vasopressin in the hypothalamus and pituitary gland of water-deprived rats.

The effect of intraventricular 6-hydroxydopamine on the content of oxytocin and vasopressin in the hypothalamus and pituitary gland of water deprived rats. Acta Physiol. Pol., 1977, 28 (6): 497-504. Rats received one infusion of 200 microgram 6-hydroxydopamine with 25 microgram of ascorbic acid into the lateral cerebral ventricle. After 57 days some rats were deprived of water for 4, 8 or 12 days. Then, the animals were sacrificed by decapitation. Oxytocin was determined in extracts from the posterior pituitary lobe and hypothalamus by the method of Van Dongen and Hays, while the vasopressin c

Nerve growth factor prevents the amblyopic effects of monocular deprivation.

Monocular deprivation early in life causes dramatic changes in the functional organization of mammalian visual cortex and severe reduction in visual acuity and contrast sensitivity of the deprived eye. We tested whether or not these changes could be from competition between the afferents from the two eyes for a target-derived neurotrophic factor. Rats monocularly deprived during early postnatal development were treated with repetitive intraventricular injections or topical administration of nerve growth factor. The effects of monocular deprivation were then assessed electrophysiologically. In

Same-Day Approach for Combined Intravitreal and Intracerebroventricular Enzyme Replacement Therapy to Prevent Retinal Disease Progression in Children With Neuronal Ceroid Lipofuscinosis Type 2.

BACKGROUND: Classic late infantile neuronal ceroid lipofuscinosis (CLN2) is caused by a biallelic mutations of the TPP1 gene. Vision loss begins around age four years, resulting in blindness by age seven to ten years. Intracerebroventricular cerliponase alfa (Brineura; BioMarin) is indicated to slow the loss of ambulation in pediatric patients with CLN2. However, treated children continue to experience visual loss. Intravitreal cerliponase alfa allows the enzyme to target tissues in the eye, offering a treatment option. METHODS: We developed a pathway for same-day administration of both intra
