Distinct roles for intrinsic osteocyte abnormalities and systemic factors in regulation of FGF23 and bone mineralization in Hyp mice.

X-linked hypophosphatemia (XLH) is characterized by hypophosphatemia and impaired mineralization caused by mutations of the PHEX endopeptidase (phosphate-regulating gene with homologies to endopeptidases on the X chromosome), which leads to the overproduction of the phosphaturic fibroblast growth factor 23 (FGF23) in osteocytes. The mechanism whereby PHEX mutations increase FGF23 expression and impair mineralization is uncertain. Either an intrinsic osteocyte abnormality or unidentified PHEX substrates could stimulate FGF23 in XLH. Similarly, impaired mineralization in XLH could result solely

Dentin matrix proteins and soluble factors: intrinsic regulatory signals for healing and resorption of dental and periodontal tissues?

PURPOSE: Targeted radionuclide therapy (TRT), specifically peptide receptor radionuclide therapy (PRRT) with [177Lu]Lu-DOTA-TATE is an established treatment for somatostatin receptor-expressing neuroendocrine tumors across different grades; however, its efficacy in other tumors expressing somatostatin receptors remains less defined. This study aimed to evaluate the safety and efficacy of PRRT with [177Lu]Lu-DOTA-TATE in patients with medullary thyroid cancer (MTC) who presented with progressive recurrence or metastases refractory to standard therapies. METHODS: Ten patients with MTC (3 female

Enhancing the Intrinsic Antiplasmodial Activity and Improving the Stability and Selectivity of a Tunable Peptide Scaffold Derived from Human Platelet Factor 4.

The control of malaria, a disease caused by Plasmodium parasites that kills over half a million people every year, is threatened by the continual emergence and spread of drug resistance. Therefore, new molecules with different mechanisms of action are needed in the antimalarial drug development pipeline. Peptides developed from host defense molecules are gaining traction as anti-infectives due to theood of inducing drug resistance. Human platelet factor 4 (PF4) has intrinsic activity against P. falciparum, and a macrocyclic helix-loop-helix peptide derived from its active domain recapitulates

Serum Concentrations of Gastric Intrinsic Factor-A Pilot Study on Patients Undergoing Bariatric Surgery, Presenting with Portal Hypertension, or Suspected Pernicious Anemia.

Background/Objectives: Small quantities of gastric parietal cell-derived Intrinsic Factor (IF) are present in circulation. Here, we explore circulating concentrations of IF in different medical conditions affecting the stomach, anticipating finding both low and high concentrations depending on the state of the parietal cells. Methods: We measured serum IF in (I) patients before and after bariatric surgery; (II) patients diagnosed with portal hypertension; and (III) patients with suspected pernicious anemia referred for measurement of IF autoantibodies. Results: Patients undergoing bariatric su

Congenic mice provide in vivo evidence for a genetic locus that modulates intrinsic transforming growth factor β1-mediated signaling and bone acquisition.

Osteoporosis, the most common skeletal disorder, is characterized by low bone mineral density (BMD) and an increased risk of fragility fractures. BMD is the best clinical predictor of future osteoporotic fracture risk, but is a complex trait controlled by multiple environmental and genetic determinants with individually modest effects. Quantitative trait locus (QTL) mapping is a powerful method for identifying chromosomal regions encompassing genes involved in shaping complex phenotypes, such as BMD. Here we have applied QTL analysis to male and female genetically-heterogeneous F(2) mice deriv

Analysis of an ethylnitrosourea-generated mouse mutation defines a cell intrinsic role of nuclear factor kappaB2 in regulating circulating B cell numbers.

The number of circulating follicular B lymphocytes is normally kept within a precise range despite their dispersion through the body and daily overproduction of precursors in the bone marrow. By establishing a genome wide recessive mutation screen in C57BL/6 mice to identify critical components of immune system regulation, we identified a mutant strain with selective deficiency in recirculating B cells but not immature or peritoneal B1 cells. Analysis of mixed bone marrow chimeras established that the mutation affects a cell autonomous process within B cells that is required for their accumula

Evaluation of a new assay kit for intrinsic factor blocking antibody (type I) as an aid in the diagnosis of pernicious anemia.

Sera from 39 patients with pernicious anemia and 251 patients with various other diseases and 42 healthy normal subjects were tested for intrinsic factor blocking antibody by a Corning commercial kit. Twenty-five (64%) out of 39 pernicious anemia patients showed a positive reaction to the intrinsic factor blocking antibody. The positive incidence of intrinsic factor blocking antibody by this method agreed well with the results obtained in this laboratory over the past 10 years using the charcoal method. No sera from normal subjects tested were positive for the intrinsic factor blocking antibod

Intrinsic factor, free of R proteins, can be prepared from mouse stomach and used in a ligand assay specific for "true" cobalamin.

A ligand assay specific for cobalamin that uses mouse stomach as the source of intrinsic factor has been developed. When mouse stomach extract incubated with radiocobalamin is fractionated by gel chromatography, the radioactive complex elutes as a single peak with apparent molecular weight of 54,900. Formation of the complex is greater than 98% inhibited by human anti-intrinsic factor antibody. When the equivalent of 10,000 pg/ml of cobinamide is added to serum, the apparent cobalamin concentration detected averages 8.5 pg/ml. Correlation with the Lactobacillus leichmannii microbiologic assay

Clinical evaluation of simultaneously administered 58Co labelled vitamin B12 and 57Co labelled vitamin B12 bound to intrinsic factor in patients with pernicious anaemia.

The clinical and laboratory data from 75 patients with altered vitamin B 12 absorption were reviewed. In 36 cases the diagnosis of pernicious anaemia had been established. Of these, 14 patients showed malabsorption of radiolabelled vitamin B 12, but the absorption of vitamin B 12 bound to the intrinsic factor (IF) was normal (Group A). The other 22 patients with pernicious anaemia showed altered free and IF-bound vitamin B 12 absorption (Group B). Laboratory and clinical data and the absorption tests (xylose and fat excretion) were more abnormal in group B than in group A. Finally, 39 patients

Cross-sectional analysis of factors associated with low intrinsic capacity among Malaysians ageing with HIV in ambulatory care.

OBJECTIVE: Intrinsic capacity is a component of WHO's model for Health Ageing and a key determinant of an individual's functional ability as they age. However, little is known about what factors negatively influence intrinsic capacity among people with HIV (PWH). DESIGN: Cross-sectional study. METHOD: Participants included PWH [≥35 years and on antiretroviral therapy (ART) for a minimum of 12 months] attending HIV care in University Malaya Medical Centre. Intrinsic capacity scores were computed based on five key domains (sensory, mobility, cognition, vitality, and mood). Pareto analyses were

Fracture Patterns in Fatal Free Falls: A Systematic Review of Intrinsic and Extrinsic Risk Factors and the Role of Postmortem CT.

Background: Free fatal falls (FFF) represent a distinct form of blunt force trauma (BFT) that is frequently encountered in forensic practice. Distinguishing FFF injuries from other forms of BFT, such as motor vehicle accidents (MVAs), can pose challenges. Despite its growing usage, the role of postmortem computed tomography (PMCT) in diagnosing FFF and its comparison with autopsy remains underexplored. Purpose: This review synthesizes fracture patterns in FFF, examining both extrinsic and intrinsic variables that influence skeletal injuries. It also compares PMCT and autopsy findings to establ

An Efficient Screen for Cell-Intrinsic Factors Identifies the Chaperonin CCT and Multiple Conserved Mechanisms as Mediating Dendrite Morphogenesis.

Dendritic morphology is inextricably linked to neuronal function. Systematic large-scale screens combined with genetic mapping have uncovered several mechanisms underlying dendrite morphogenesis. However, a comprehensive overview of participating molecular mechanisms is still lacking. Here, we conducted an efficient clonal screen using a collection of mapped P-element insertions that were previously shown to cause lethality and eye defects in Drosophila melanogaster. Of 280 mutants, 52 exhibited dendritic defects. Further database analyses, complementation tests, and RNA interference validatio

Chronic Inflammation in Response to Injury: Retention of Myeloid Cells in Injured Tissue Is Driven by Myeloid Cell Intrinsic Factors.

Chronic inflammation is a hallmark of impaired healing in a plethora of tissues, including skin, and is associated with aging and diseases such as diabetes. Diabetic chronic skin wounds are characterized by excessive myeloid cells that display an aberrant phenotype, partially mediated by stable intrinsic changes induced during hematopoietic development. However, the relative contribution of myeloid cell-intrinsic factors to chronic inflammation versus aberrant signals from the local environmental was unknown. Moreover, identification of myeloid cell intrinsic factors that contribute to chronic

What intrinsic factors influence responsiveness to acupuncture in pain?: a review of pre-clinical studies that used responder analysis.

BACKGROUND: Not many studies have investigated individual sensitivity to acupuncture. To explore the intrinsic factors related to individual responses to acupuncture, we reviewed published pre-clinical studies using responder analysis on pain. METHODS: We searched the PubMed and EMBASE databases to June 2015. We included pre-clinical reports describing responders and non-responders to anti-nociceptive and analgesic effects of acupuncture in animal study. We identified the potential intrinsic factors which might be related with the response to acupuncture. RESULTS: Totally, 216 potentially re

Role of Intrinsic (Graft) Versus Extrinsic (Host) Factors in the Growth of Transplanted Organs Following Allogeneic and Xenogeneic Transplantation.

In our studies of life-supporting α-1,3-galactocyltransferase knockout (GalT-KO) pig-to-baboon kidneys, we found that some recipients developed increased serum creatinine with growth of the grafts, without histological or immunological evidence of rejection. We hypothesized that the rapid growth of orthotopic pig grafts in smaller baboon recipients may have led to deterioration of organ function. To test this hypothesis for both kidneys and lungs, we assessed whether the growth of outbred (Yorkshire) organ transplants in miniature swine was regulated by intrinsic (graft) or extrinsic (host env

Multidrug Intrinsic Resistance Factors in Staphylococcus aureus Identified by Profiling Fitness within High-Diversity Transposon Libraries.

UNLABELLED: Staphylococcus aureus is a leading cause of life-threatening infections worldwide. The MIC of an antibiotic against S. aureus, as well as other microbes, is determined by the affinity of the antibiotic for its target in addition to a complex interplay of many other cellular factors. Identifying nontarget factors impacting resistance to multiple antibiotics could inform the design of new compounds and lead to more-effective antimicrobial strategies. We examined large collections of transposon insertion mutants in S. aureus using transposon sequencing (Tn-Seq) to detect transposon mu

Longitudinal variability in airway responsiveness in a population-based sample of children and young adults. Intrinsic and extrinsic contributing factors.

The degree and long-term variability of airway responsiveness to eucapneic hyperventilation with cold air was assessed in 287 children and young adults (less than 23 yr of age) (735 challenge tests) enrolled in a longitudinal study of pulmonary function, 179 of whom underwent two to five cold-air challenge tests between 1982 and 1986. Survey-to-survey variability in airway responsiveness was assessed with computation of adjusted within-subject correlation coefficients for continuous measures of response (percent decrease in FEV1 after challenge) and with adjusted odds ratios for dichotomous ou

Monolithic tantalum pentoxide athermal microrings with intrinsic Q factors exceeding 5 × 106 without SiO2 cladding.

Tantalum pentoxide (Ta2O5), as a silicon-photonic-compatible material platform, has garnered significant attention for high-performance wafer-scale integrated photonics due to its exceptional properties: a broad transparency window spanning from 0.28 µm to 8 µm, a moderate refractive index of 2.05 at 1550 nm, an impressive nonlinear refractive index of 1.0 × 10-14 cm2/W, and thick-film deposition with low residual stress at room-temperature. Despite these advantages, achieving low-loss fabrication of monolithic microrings on the Ta2O5 platform remains challenging due to its inherent hardness a

Refrigeration-reheating induced fishy odor in cooked Chinese mitten crab (Eriocheir sinensis): Formation and intrinsic factors.

This study investigated odor changes in cooked Chinese mitten crab (Eriocheir sinensis) during refrigeration and reheating, with an emphasis on volatile flavor compounds and lipid oxidation. Sensory evaluation indicated that fishy odor emerged during refrigeration and persisted after reheating. A total of 52 odor compounds were identified, mainly aldehydes, alcohols, and ketones, with nonanal and 1-octen-3-ol increasing during refrigeration. Odor activity value (OAV) analysis and partial least squares discriminant analysis (PLS-DA) identified nonanal, hexanal, and (E)-2-octenal as key differen
