The emergence of azole resistance in Aspergillus fumigatus is a clinically important issue in the management of invasive aspergillosis as it could limit therapeutic options. Accurate measurement of in vitro antifungal activity in terms of minimum inhibitory concentration (MIC) is considered of clinical relevance and often gives useful therapeutic information for physicians. However, the lack of in vitro-in vivo correlation is frequent and the observed in vitro phenotype does not always correlate with the in vivo response. In this regard, a wild-type strain and five A. fumigatus cyp51A mutated

Recent data suggest that opioid receptors are involved in the development of nicotine physical dependence in mammals. Evidence in support of a similar involvement in an invertebrate (Planaria) is presented using the selective opioid receptor antagonist naloxone, and the more receptor subtype-selective antagonists CTAP (D-Phe-Cys-Tyr-D-Trp-Arg-Thr-Pen-Thr-NH2) (μ, MOR), naltrindole (δ, DOR), and nor-BNI (norbinaltorphimine) (κ, KOR). Induction of physical dependence was achieved by 60-min pre-exposure of planarians to nicotine and was quantified by abstinence-induced withdrawal (reduction in sp

Harmful benthic microalgae blooms are an emerging phenomenon causing health and economic concern, especially in tourist areas. This is the case of the Mediterranean Sea, where Ostreopsis ovata blooms occur in summer, with increasing regularity. Ostreopsis species produce palytoxin (PTX) and analogues, and a number of deaths directly associated with the ingestion of PTX contaminated seafood have been reported. PTX is considered one of the most toxic molecules occurring in nature and can provoke severe and sometimes lethal intoxications in humans. So far in temperate areas, O. ovata blooms were

The carrageenan model of pleurisy is described as temporal plasma exudation (1-5 h) with extensive neutrophil infiltration and release of proteinases into the pleural cavity. The aim of this work was to study the effects of serine proteinase inhibitors on the inflammatory process induced by administration of carrageenan to the rat pleural cavity and on release of kinins in pleural exudate. Pleurisy was induced by injecting carrageenan and serine proteinase inhibitors simultaneously into the pleural cavity. The proteinase inhibitors used were: aprotinin, a plasma kallikrein inhibitor; recombina

The widespread use of pesticides in agriculture also results in the exposure of non-target organisms. These non-target organisms can differ widely in their sensitivity to pesticides. The sensitivity of organisms to a compound is typically assessed using standardized ecotoxicological tests that rely on fixed-time metrics such as four-day LC50 values. These metrics ignore the time-dependency of toxicity and provide little mechanistic understanding of the observed differences in species sensitivity to toxic substances. To quantify potential differences in the development of toxicity over time, se

Silver nanomaterials (Ag NMs) are widely used, including in consumer products, and they inevitably enter the environment, with the soil compartment acting as a major sink. However, most available toxicity data focus on the reference Ag NM300K and rely on standard tests, even though long(er)-term exposure tests are recognized as particularly important for assessing the risks to soil invertebrates. Hence, the aim of the present study was to investigate the toxicity of commercial Ag NPs (Ag-Sigma, NPs < 150 nm) to the soil ecotoxicology model Folsomia candida (Collembola). Effects were assessed b

Locomotor behavioral responses are increasingly recognized as sensitive and ecologically relevant indicators for assessing aquatic organism exposure to contaminants. They often occur at concentrations lower than those causing physiological damage, yet their consequences for organisms' fitness and population dynamics remain unclear. Moreover, it is still uncertain how these behavioral endpoints align with traditional toxicological apical endpoints used to assess species sensitivities in risk assessment. This study evaluates the correlation between thresholds triggering locomotor reactions with

Water ecosystems represent main targets of unintentional contamination of nanomaterials, due to industrial waste or other anthropogenic activities. Nanoparticle insult to living organisms may occur in a sequential way, first by chemical interactions of the material with the target membrane, then by progressive internalisation and interaction with cellular structures and organelles. These events trigger a signal transduction, through which cells modulate molecular pathway in order to respond and survive to the external elicitation. Therefore, the analysis of the global changes of the molecular

Proteins containing WAP (whey acidic protein) domains with a characteristic WFDC (WAP four-disulfide core) occur not only in mammals (including marsupials and monotremes) but also in birds, reptiles, amphibians and fish. In addition, they are present in numerous invertebrates, from cnidarians to urochordates. Many of those from non-mammalian groups are poorly understood with respect to function or phylogeny. Those well characterized so far are waprins from snakes, perlwapins from bivalves and crustins from decapod crustaceans. Waprins are venom proteins with a single WAP domain at the C-termin

Cyanobacterial blooms are becoming increasingly frequent and intense worldwide, often dominated by Microcystis aeruginosa, a species capable of producing a wide array of bioactive metabolites beyond microcystins. This study evaluates the ecotoxicological potential of a non-microcystin-producing strain, M. aeruginosa CCIBt3106, using acute immobilization assays with three microcrustacean species: Daphnia similis, Artemia salina, and Parhyale hawaiensis. Biomass was extracted using solvents of varying polarity, and selected extracts (aqueous and 50% methanol) were further fractionated and analyz

Klebsiella is a common dangerous pathogen for humans and animals and is widely present in the digestive system. The genus Klebsiella is ubiquitous, as it is endemic to surface water, soil, and sewage. In this study, 70 samples were obtained from soil-dwelling invertebrates from September 2021 to March 2022 from Taif and Shafa in different altitudinal regions of Saudi Arabia. Fifteen of these samples were identified as Klebsiella spp. The Klebsiella isolates were genetically identified as Klebsiella pneumoniae using rDNA sequencing. The antimicrobial susceptibility of the Klebsiella isolates wa

Chironomids inhabit freshwater benthic ecosystems which are prone to microplastic contamination. This work aimed at understanding the factors and mechanisms influencing microplastic uptake and related adverse effects on Chironomus riparius, by exploring an extensive project database, conducting a literature review, and performing an agent-based model to explore trends in data. Results reveal that high concentrations of small microplastics fill the gut of fourth instar C. riparius (99.7 %). Ingested microplastics had an average size of 38-61 μm, presenting slower elimination rates than undigest

Down syndrome cell adhesion molecule (Dscam) generates tens of thousands of isoforms by alternative splicing, thereby providing crucial functions during immune responses. In this study, a novel Dscam signaling pathway was investigated in crab, which remains poorly characterized in invertebrates. Bacterial infection induced the cytoplasmic cleavage of Dscam intracellular domains (ICDs) by γ-secretase, and then the released ICDs carrying specific alternatively spliced exons could directly interact with IPO5 to facilitate nuclear translocation. Nuclear imported ICDs thus promoted hemocyte prolife

Abrasion of tire wear is one of the largest sources of microplastics to the environment. Although most tire particles settle into soils, studies on their ecotoxicological impacts on the terrestrial environment are scarce. Here, the effects of tire particles (<180 μm) on three ecologically relevant soil invertebrate species, the enchytraeid worm Enchytraeus crypticus, the springtail Folsomia candida and the woodlouse Porcellio scaber, were studied. These species were exposed to tire particles spiked in soil or in food at concentrations of 0.02%, 0.06%, 0.17%, 0.5% and 1.5% (w/w). Tire particles

White spot syndrome virus (WSSV) inflicts severe economic losses on global aquaculture and poses a grave threat to the farming of shrimp, crabs, and other invertebrates. This review synthesizes the molecular pathways through which WSSV infection induces reactive oxygen species (ROS) generation in the host; examines the dual role of oxidative stress, wherein moderate ROS contribute to antiviral defense while excessive ROS drive oxidative injury; and, drawing on the threshold effect concept established in mammalian studies, evaluates its applicability and limitations as a working hypothesis in i

Marine invertebrates harbor diverse bacterial communities that contribute to host defense by producing antibacterial compounds. In this study, 258 cultivable bacterial isolates from corals (Porites panamensis, P. lobata), the sea urchin Echinometra vanbrunti, and the ctenophore Mnemiopsis leidyi were screened against aquaculture and human pathogens. 14% (37 isolates) exhibited antibacterial activity, predominantly from E. vanbrunti and M. leidyi. Multiple assays revealed strong, broad-spectrum inhibition, particularly against aquaculture pathogens such as Listonella anguillarum, Photobacterium

Mercury is a toxic environmental contaminant that is known to undergo long range transport, resulting in deposition in remote and pristine ecosystems. Increases in methylmercury concentration at low trophic levels may result in greater risk to top predators in food webs. As a result, low trophic level invertebrates are important sentinels for ecosystem contamination and overall ecosystem health. Big Meadow Bog, located on Brier Island, Nova Scotia, is a disturbed wetland habitat that was subject to colonization by herring gulls (Larus argentatus), resulting in significant phosphate nutrient in

Host-parasite interactions represent an evolutionary arms race, with parasites evolving strategies to exploit host resources, and hosts deploying mechanisms to resist or tolerate infection. Unlike vertebrates, which possess a sophisticated immune system, invertebrates rely entirely on innate immunity to combat pathogens. Yet, the molecular underpinnings and life-history consequences of immune activation remain little understood in non-insect invertebrates. Here, we combine life-history trait analysis with transcriptomics to investigate immune responses in an aquatic invertebrate exposed to a n

Echinoderms exhibit a wide range of reproductive strategies as adaptations to variable environments. The processes of gonadal development, germ cell differentiation, and spermatogenesis in echinoderms are crucial physiological processes that warrant further in-depth exploration. This review systematically summarizes research from early basic sciences to recent studies on male gonadal development and spermatogenesis, encompassing morphology, histology, physiology, cell biology, developmental biology, and evolutionary biology. We introduce the structural and cellular similarities and differences

Neuroanatomical information on neuropeptidergic neurons provides crucial clues to understanding the functions of neuropeptides and the cerebral structure-function relationships. In this study, we constructed comprehensive two- and three-dimensional atlases of 23 neuropeptides in the central nervous system (CNS) of an invertebrate chordate, Ciona robusta, at the single-cell level using a mass spectrometry imaging workflow. The neuropeptidomic atlases revealed that the cerebral ganglion is divided into three functional areas, dorso-rostral, dorso-caudal, and ventral areas. Particularly, the dist
