Antineoplastic drug 5-fluorouracil (5-FU) frequently shows cardiotoxic effects, the mechanism of which has not yet been elucidated. The objective of the present study was to explore effects of 5-FU on metabolism of ATP, 2,3-BPG, and oxygen in erythrocytes and to relate these to the phenomenon of 5-FU cardiotoxicity. We determined that 5-FU induced rapid increase in O(2) consumption, which led to drastic changes in the metabolism of phosphate compounds in erythrocytes. Decrease in pO(2) provoked increase in production of 2,3-BPG and subsequent deoxygenation of oxyHb to deoxyHb. However, the mos

INTRODUCTION: Knowledge of physiopathological mechanisms permits understanding of neuro-imaging changes during the various phases of cerebral ischaemia and the mechanisms of action on which many aspects of treatment are based.
DEVELOPMENT: The physiopathology of cerebral ischemia is different in the white and grey matter of the brain. In the grey matter, obstruction of a blood vessel causes an ischemic gradient. In the more peripheral zones, known as the ischemic penumbra, functional alterations occur in the neurons and the glia, although the structural integrity is maintained for some time.

Diacetoxyscirpenol (DAS, anguidine) was given intravenously to swine at 0.0, 0.5, and 1.0 mg/kg body wt. In mitotically and metabolically active tissues such as gastrointestinal epithelium and lymphoid aggregates the effects of DAS mimicked radiation poisoning. A quadratic dose-response relationship between the cytotoxicity of DAS and damage to enterocytes was found. Enterocytes in different anatomical regions of the bowel had differing susceptibilities to the toxic effects of DAS. In lymphoid tissues, DAS was preferentially cytotoxic to B-lymphocyte-rich tissues as compared to T-lymphocyte-ri

The aim of the paper was to define which transepithelial ion transport pathways were activated during stimulation by movement across the colonic epithelium. The experiments were performed with Ussing method on 241 specimens of isolated distal colon wall from 51 rabbits. The tissue was stimulated by a flux from peristaltic pump which caused transient hyperpolarization of the isolated tissue. In the control, the transepithelial potential difference (PD) and its changes during stimulation (dPD) were -3.16 +/- 1.81 mV and -2.16 +/- 0.99 mV, respectively. Preincubation of the tissue in the presence

Physiologically relevant gastrointestinal digestion models for infants, adults, and the elderly are commonly used to explore the fate of food in vitro. However, no consensus protocol exists to simulate the specific conditions observed in the stomach of people using oral proton pump inhibitors (PPIs), a class of widely prescribed medications that reduce gastric acid secretion and may alter food digestion and nutrient absorption. The first objective of this study was to propose an in vitro gastric digestion protocol adapted to model PPI use. This protocol is an extension of the semi-dynamic INFO

Rat submandibular glands have been digested with collagenase P and a crude cellular suspension has been prepared. The [Ca2+]i and the pHi of these cells have been measured using fluorescent probes extracellular ATP increases the [Ca2+]i. At low concentrations ATP binds to a metabotropic receptor coupled to the mobilization of intracellular calcium stores. At high concentrations ATP activates a ionotropic receptor coupled to a non-specific cation channel permeant to calcium, sodium or zinc. The increase of the [Ca2+]i in response to ATP opens an anion permeability leading to a drop of the pHi.

OqxB belongs to the RND (Resistance-Nodulation-Division) efflux pump family, recognized widely as a major contributor towards enhancing antimicrobial resistance. It is known to be predominantly present in all Klebsiella spp. and is attributed for its role in increasing resistance against an array of antibiotics like nitrofurantoin, quinolones, β-lactams and colistin. However, the presence of oqxB encoding this efflux pump is not limited only to Klebsiella spp., but is also found to occur via horizontal gene transfer in other bacterial genera like Escherichia coli, Enterobacter cloacae and Salm

There has been controversy for some time as to whether a posttraumatic influx of calcium ions occurs in stretch/nondisruptively injured axons within the central nervous system in both human diffuse axonal injury and a variety of models of such injury. We have used the oxalate/pyroantimonate technique to provide cytochemical evidence in support of such an ionic influx after focal axonal injury to normoxic guinea pig optic nerve axons, a model for human diffuse axonal injury. We present evidence for morphological changes within 15 min of injury where aggregates of pyroantimonate precipitate occu

1. Mechanisms of ion transport across the choroidal epithelium were investigated using an in vitro preparation of the frog choroid plexus.2. Sodium was actively transported across the plexus from the vascular to the ventricular surface by an ouabain sensitive electrically silent pump. As in other epithelial membranes the rate of sodium transport was stimulated by the presence of bicarbonate ions in the Ringer solutions. Chloride and bicarbonate ions accompany the net flux of sodium across this tissue.3. Some experiments suggest that potassium is actively transported from the ventricular to the

GPR35 is an orphan G-protein coupled receptor that has been implicated in the development of cancer. GPR35 regulates the Na+/K+-ATPase's pump and signalling function. Here we show GPR35's critical role in ion flux that in turn controls cellular osmotic pressure and Na+-dependent transport in HepG2 and SW480 cells. GPR35 deficiency results in increased levels of intracellular Na+, osmotic stress and changes in osmolytes leading to increased cells size and decreased glutamine import in vitro and in vivo. The GPR35-T108M risk variant, which increases risk for primary sclerosing cholangitis and in

In light of the important role of magnesium (Mg) in vascular smooth muscle function, this study was undertaken to examine effects of dietary Mg deficiency on membrane potential (Em), sodium (Na) efflux and distribution, and total ion content in rat aorta. Rats were fed either Mg-sufficient or Mg-deficient diets for 30 days. The aortic arch was removed and a segment incubated in 22Na to monitor kinetics of Na efflux. Another segment was used for measurement of Em under control conditions and during ouabain suffusion. Total tissue water and ion content were also determined. Results of this work

The acidic pH of lysosomes required for function is established by the electrogenic V-ATPase proton pump. How lysosomes prevent hyper-acidification by the pump is not well established. Recently, the Parkinson's disease (PD)-associated protein TMEM175 was proposed as a H+-selective channel to leak protons to counter over-acidification. We rigorously address key findings and predictions of this model and show that, in the lysosome, TMEM175 predominantly conducts K+ and is not a H+-selective channel. The native lysosomal H+ leak is remarkably small, ∼0.02 fA, strongly arguing against major contri

A study was made of rubidium (potassium analog) influxes via ouabain-sensitive Na/K pump, bumetanide-sensitive cotransport and resistant to ouabain bumetanide membrane ion pathways and of the intracellular potassium and sodium contents in red blood cells from patients with high gentamicin sensitivity (GSP) and healthy patients (HP). It is found that red blood cells from two groups of donors do not differ in both intracellular potassium and sodium contents and pump-mediated rubidium influxes, however, bumetanide-sensitive rubidium influxes were twice as low in GSP as compared to HP (0.28 agains

Leg cramping is a common side effect of hemodialysis, and this is frequently treated by the administration of carnitine, but this is not effective in every patient. Alkalosis is a key component of the etiology of leg cramping during hemodialysis sessions. This is mediated through the binding of calcium ions to serum albumin, which causes hypocalcemia, and an increase in the release of calcium ions from the sarcoplasmic reticulum. Normally the calcium pump on the sarcoplasmic reticulum consumes ATP and quickly reuptakes the released calcium ions, which rapidly stops excessive muscle contraction

Cells exhibit a membrane potential due to the differential distribution of ions and the density of channels, pumps, and exchangers. It is known in larval Drosophila that the membrane potential gravitates towards the equilibrium potential of K+ due to the high density of K2P channels. Higher extracellular Ca2+ ([Ca2+]o) tends to drive the resting membrane potential to a more negative state, while lowering it has an opposite effect. The expression of the K2P channel and NALCN was altered genetically to determine the sensitivity to changes in [Ca2+]o, and computational simulations using the theor

BACKGROUND & AIMS: To determine how interferon (IFN)-gamma inhibits epithelial barrier and ion transport functions, intestinal T84 cells were studied.
METHODS: Acute and chronic effects of IFN-gamma on T84 barrier function, Na+,K+-adenosine triphosphatase (ATPase) activity, and certain ion transport and tight junctional proteins were determined. To assess the role of Na+,K+-ATPase and intracellular Na+, similar studies with the Na+,K+-ATPase inhibitor ouabain and Na+ ionophore monensin were performed. To determine the role of nitric oxide (NO), the NO donor SPER-NO was used.
RESULTS: IFN-gam

Proton pumps are membrane-bound enzymes important in generating gradients that help in maintaining cellular ion homeostasis, cell membrane potential, water, and solute transport across the cell surface. This study investigated the modulatory role of vitamin E on proton pump activity and reproductive parameters in cadmium-induced testicular damage. Twenty (20) male Wistar rats weighing between 180 and 200 g were sorted into 4 groups of five rats each. Group I served as the control and was given normal saline orally, Group II rats were treated with a single dose of 2 mg/kg BW cadmium chloride (C

Respiratory Complex I powers oxidative phosphorylation by a long-range proton-coupled electron transfer (PCET) reaction, with mutations linked to more than half of all human mitochondrial disorders. Yet, the molecular principles underlying the functional impairment remain difficult to test, as most mutations impede both the proton pumping and oxidoreductase activities due to the tightly coupled PCET process. Here, we probe how key disease mutations in the terminal ND5 subunit (NuoL/Nqo12), linked to the development of Leigh's syndrome (LS) and LHON/MELAS (F124L, M252T, D393N), affect the proto

Currently, only a few chemical drug classes are available to control the global burden of nematode infections in humans and animals. Most of these drugs exert their anthelmintic activity by interacting with proteins such as ion channels, and the nematode neuromuscular system remains a promising target for novel intervention strategies. Many commonly-used phenotypic readouts such as motility provide only indirect insight into neuromuscular function and the site(s) of action of chemical compounds. Electrophysiological recordings provide more specific information but are typically technically cha
