Targeting depression: Pharmacological mechanisms and therapeutic targets.

Depression remains a leading cause of disability worldwide with a rapidly escalating disease burden, yet current pharmacotherapies are limited by delayed onset of action, insufficient efficacy in treatment-resistant depression, and troublesome adverse effects. This review provides a systematic and critical evaluation of pharmacological mechanisms in depression and related therapeutic targets. We examine established targets spanning the neurotransmitters corresponding to serotonergic, glutamatergic, GABAergic, and dopaminergic systems, alongside emerging glial gap junctions and inflammatory per

Parasitism of prehistoric humans and companion animals from Antelope Cave, Mojave County, northwest Arizona.

Previously, we reported a tick recovered from Antelope Cave in extreme northwest Arizona. Further analyses of coprolites from Antelope Cave revealed additional parasitological data from coprolites of both human and canid origin. A second tick was found. This site is the only archaeological locality where ticks have been recovered. We also discovered an acanthocephalan in association with Enterobius vermicularis eggs in the same coprolite. This association shows that the coprolite was deposited by a human. This discovery expands our knowledge of the range of prehistoric acanthocephalan infectio

OpenScientist: evaluating an open agentic AI co-scientist to accelerate biomedical discovery.

BACKGROUND: Advances in medicine depend on analyzing large and complex data sources, but discovery is partly constrained by the limited time and domain expertise of human researchers. Agentic artificial intelligence (agentic AI) can accelerate discovery by automating components of the scientific workflow, including information retrieval, data analysis, and knowledge synthesis. AIM: OpenScientist, an open-source agentic AI co-scientist, aims to accelerate biomedical discovery by semi-autonomously investigating scientist-defined queries and generating clinically relevant, verifiable scientific

[History of scurvy therapy and the injustice done to Dr. Maksim Nikolić-Miskovićev of Sremski Karlovci in the 19th century].

This paper reviews historical aspects regarding medical knowledge on scurvy, vitamin C deficiency, and its etiological factors. This disease used to affect naval-crews on long lasting sea voyages, soldiers during times of war and the poor. Although efficient therapy of scurvy (fresh vegetables, lemon juice) was known in the mid-18th century, its etiology was not known. It was believed to be caused by the cold, moist winds, unhealthy evaporations, malnutrition and it was called "alkaline disease". It was established that acid substances like lemon juice, had beneficial effects on the disease. H

Translational benefits from research on rare genodermatoses.

Significant new discoveries about many of the genodermatoses have been made recently through an improved knowledge of the human genome, advances in molecular screening strategies and also through more comprehensive Internet DNA databases. By 2003, over 350 single gene skin disorders had been characterized at a molecular level. These new data provide more detailed information for patients, allow for more accurate diagnoses, and help improve genetic counselling. Other benefits include the feasibility of carrier screening and DNA-based prenatal testing, as well as a platform for devising new trea

[The transcription factor NF-kappaB in human diseases].

The transcription nuclear factor kappa B (NF-kappaB) was first described in 1986. Currently, the NF-kappaB has been linked to several metabolic and inflammatory diseases as well as to several types of cancer. NF-kappaB will continue to be of interest in many disease processes such as inflammatory, infectious and neoplastic. The discovery of this factor has increased our knowledge about the regulation of inflammation and chronic diseases, as well as the medications used to treat those diseases.

Mental nerve neuropathy as initial symptom of cancer.

Numb chin syndrome is a rare cranial neuropathy. Knowledge of potential cancers that involve the mental nerve is mandatory, since it may lead in many cases to discovery of a neoplasia or to its recurrence. We describe a 55-year-old patient who complained for several days of numbness in the right side of his chin. No dental cause was found. What was found was an undiagnosed pulmonary carcinoma in metastasis. This possibility should be considered in the differential diagnosis.

A standardized educational implementation strategy improves operational Code Sepsis knowledge in a high-complexity hospital in Colombia.

BACKGROUND: Timely recognition and early bundle-based care are essential to improve sepsis outcomes. However, implementation gaps persist in routine hospital practice, particularly in middle-income settings where multidisciplinary coordination and operational knowledge of institutional protocols are critical. This study evaluated the effect of a standardized educational strategy on immediate Code Sepsis knowledge among hospital personnel in a high-complexity hospital in Colombia. METHODS: We conducted a single-group quasi-experimental before-and-after study in 2025 among clinical and non-clin

Accelerating scientific discovery with Co-Scientist.

Scientific discovery is driven by scientists generating novel hypotheses for complex problems that undergo rigorous experimental validation. To augment this process, we introduce Co-Scientist, a multi-agent AI system built on Gemini for structured scientific thinking and hypothesis generation. Co-Scientist aims to help scientists discover new original knowledge. Conditioned on their research objectives and prior scientific evidence, it formulates demonstrably novel research hypotheses for experimental verification. The system's design involves agents continuously generating, critiquing and ref

Shaping the future of early-onset colorectal cancer prevention.

Early-onset colorectal cancer (EOCRC) among individuals under age 50 is increasing globally. Lifestyle factors such as obesity, metabolic comorbid conditions, poor diet and sedentary behaviours are linked to EOCRC, but critical gaps remain in identifying additional risk factors, with growing evidence for early-life and gut microbial-related exposures. In addition, how or when the risk factors act, individually or collectively, to initiate or promote colorectal cancer at much younger ages remains unknown. Compounding the complexity are the unique challenges of developing and implementing effect

Characterization of a dengue NS4B inhibitor originating from an HCV small molecule library.

Dengue is the most important mosquito-transmitted viral disease and a major global health concern. Over the last decade, dengue virus (DENV) drug discovery and development has intensified, however, this has not resulted in approved DENV-specific antiviral treatments yet. DENV and hepatitis C virus (HCV) belong to the same Flaviviridae family and, in contrast to DENV, antiviral treatments for HCV have been licensed. Therefore, applying the knowledge gained on anti-HCV drugs may foster the discovery and development of dengue antiviral drugs. Here, we screened a library of compounds with establis

The discovery of HTLV-1, the first pathogenic human retrovirus.

After the discovery of retroviral reverse transcriptase in 1970, there was a flurry of activity, sparked by the "War on Cancer," to identify human cancer retroviruses. After many false claims resulting from various artifacts, most scientists abandoned the search, but the Gallo laboratory carried on, developing both specific assays and new cell culture methods that enabled them to report, in the accompanying 1980 PNAS paper, identification and partial characterization of human T-cell leukemia virus (HTLV; now known as HTLV-1) produced by a T-cell line from a lymphoma patient. Follow-up studies,

Closing the Empirical Loop: Autonomous AI Agents Conduct End-to-end Research With Human Participants.

Artificial intelligence systems are transforming scientific discovery by accelerating specific research tasks, from protein structure prediction to materials design, yet remain confined to narrow domains requiring substantial human oversight. Exponential growth of scientific literature and increasing domain specialization constrain researchers' capacity to synthesize knowledge across disciplines and develop unifying theories, motivating exploration of general-purpose AI systems for science. Here we show that a domain-agnostic, agentic AI Scientist system can independently navigate the scientif

Lambert-Eaton myasthenic syndrome: from clinical characteristics to therapeutic strategies.

Lambert-Eaton myasthenic syndrome (LEMS) is a neuromuscular autoimmune disease that has served as a model for autoimmunity and tumour immunology. In LEMS, the characteristic muscle weakness is thought to be caused by pathogenic autoantibodies directed against voltage-gated calcium channels (VGCC) present on the presynaptic nerve terminal. Half of patients with LEMS have an associated tumour, small-cell lung carcinoma (SCLC), which also expresses functional VGCC. Knowledge of this association led to the discovery of a wide range of paraneoplastic and non-tumour-related neurological disorders of

Germline Variation and Somatic Alterations in Ewing Sarcoma.

Ewing sarcoma (EwS) is a rare bone or soft tissue tumor that occurs early in life, suggesting genetic variation is likely a major contributor to EwS risk. To date, genetic investigations have identified key somatic mutations and germline variants of importance for EwS risk. While substantial progress has been made in uncovering the genetic etiology of EwS, considerable gaps in knowledge remain. Herein, we provide a summary of methodological approaches for future genomic investigations of EwS. We anticipate that this recommended analytical framework for germline and somatic investigations, alon

Standing on Shoulders: Interferon Research From Viral Interference to Lupus Pathogenesis and Treatment.

The discovery of interferon in the 1950s represents much more than the identification of the first cytokine and the key mediator of antiviral host defense. Defining the molecular nature and complexity of the type I interferon family, as well as its inducers and molecular mechanisms of action, was the work of investigators working at the highest level and producing insights of great consequence. Current knowledge of receptor-ligand interactions, cell signaling, and transcriptional regulation derives from studies of type I interferon. It is on the shoulders of the giants who produced that knowle

Historical perspectives and current global challenges of Trichinella and trichinellosis.

Trichinella spiralis and related species of Trichinella have had a long history of causing human disease, and as a foodborne pathogen have had a major impact on international commerce of pork and other meat animal species which are known to transmit the parasite. Our knowledge of Trichinella has increased substantially over the past few years particularly in the areas of phylogeny, host diversity, epidemiology and control. In this paper, we provide a brief overview of our understanding of Trichinella from its discovery to present time. Past and current challenges of the control of Trichinella

History of Ataxias and Paraplegias with an Annotation on the First Description of Striatonigral Degeneration.

The aim of this paper is to carry out a historical overview of the evolution of the knowledge on degenerative cerebellar disorders and hereditary spastic paraplegias, over the last century and a half. Original descriptions of the main pathological subtypes, including Friedreich's ataxia, hereditary spastic paraplegia, olivopontocerebellar atrophy and cortical cerebellar atrophy, are revised. Special attention is given to the first accurate description of striatonigral degeneration by Hans Joachim Scherer, his personal and scientific trajectory being clarified. Understanding gene-disease associations is important for uncovering pathological mechanisms and identifying potential therapeutic targets. Knowledge graphs can represent and integrate data from multiple biomedical sources, but lack individual-level information on target organ structure and function. Here we develop CardioKG, a knowledge graph that integrates over 200,000 computer vision-derived cardiovascular phenotypes from biomedical images with data extracted from 18 biological databases to model over a million relationships.
