Lactococcus lactis and Lactobacillus salivarius differently modulate early immunological response of Wistar rats co-administered with Listeria monocytogenes.

In the light of the increasing resistance of bacterial pathogens to antibiotics, one of the main global strategies in applied science is development of alternative treatments, which would be safe both for the host and from the environmental perspective. Accordingly, the aim of this study was to test whether two lactic acid bacteria (LAB) strains, Lactococcus lactis BGBU1-4 and Lactobacillus salivarius BGHO1, could be applied as safe supplements for Listeria infection. Two major research objectives were set: to compare the effects of BGBU1-4 and BGHO1 on early immune response in gut tissue of W

Protective immunity against influenza H5N1 virus challenge in chickens by oral administration of recombinant Lactococcus lactis expressing neuraminidase.

BACKGROUND: Highly pathogenic H5N1 avian influenza viruses pose a debilitating pandemic threat in poultry. Current influenza vaccines predominantly focus on hemagglutinin (HA) which anti-HA antibodies are often neutralizing, and are used routinely to assess vaccine immunogenicity. However, Neuraminidase (NA), the other major glycoprotein on the surface of the influenza virus, has historically served as the target for antiviral drug therapy and is much less studied in the context of humoral immunity. The aim of this study was to evaluate the protective immunity of NA based on Lactococcus lactis

Statistical optimization of cell disruption techniques for releasing intracellular X-prolyl dipeptidyl aminopeptidase from Lactococcus lactis spp. lactis.

X-prolyl dipeptidyl aminopeptidase (PepX) is an intracellular enzyme from the Gram-positive bacterium Lactococcus lactis spp. lactis NRRL B-1821, and it has commercial importance. The objective of this study was to compare the effects of several cell disruption methods on the activity of PepX. Statistical optimization methods were performed for two cavitation methods, hydrodynamic (high-pressure homogenization) and acoustic (sonication), to determine the more appropriate disruption method. Two level factorial design (2FI), with the parameters of number of cycles and pressure, and Box-Behnken d

Oral treatment with Lactococcus lactis expressing Staphylococcus hyicus lipase enhances lipid digestion in pigs with induced pancreatic insufficiency.

The Staphylococcus hyicus lip gene was cloned in Lactococcus lactis. Pancreatic insufficiency was induced by ligation of the pancreatic duct in pigs. In pigs who had undergone pancreatic ligation, the coefficient of fat absorption was higher after consumption of lipase-expressing L. lactis (91.9% +/- 3.7%) than that after consumption of the inactive control strain (78.4% +/- 2.4%).

Application of group A streptococcal collagen-like protein 1-expressing Lactococcus as a novel immunotherapeutic against pancreatic ductal adenocarcinoma.

Pancreatic ductal adenocarcinoma (PDAC) is a fatal malignancy characterized by an immunosuppressive tumor environment. Neutrophil extracellular traps (NETs) have gained recognition in playing a critical role in cancer progression and formulation of the PDAC TME, making them a target for novel PDAC therapies. Herein, we utilize a Lactococcus strain expressing group A streptococcal collagen-like protein 1 (Scl1) to stimulate anti-tumor immunity and inhibit cancer promoting NETs. Orthotopically implanted male and female C57BL6/J or NET-deficient PAD4−/− mice were intra-tumorally or intra-peritone

Genomic and functional insights into a novel Lactococcus sp. AK05 from Cirrhinus mrigala with potent antagonism against Aeromonas hydrophila.

In aquaculture, addressing antibiotic-resistant fish pathogens underscores the immediate necessity for the identification of candidate probiotic strains with favorable safety and functional attributes as alternatives to antibiotics. We isolated and characterized a new strain of Lactococcus sp., called AK05, from the gut of Cirrhinus mrigala. The phenotypic tests showed that it can survive and grow in harsh conditions, including a pH of 4, and 0.8% bile salts. At 8% NaCl, it survived and exhibited merely a one-log rise in CFU·mL⁻¹, while NaCl levels ≤ 5% facilitated over a 4-log increase in 24

Construction of Lactococcus lactis Expression System via Chromosomal Integration of T7 RNA Polymerase for Levan Production from Fruit Pomace.

Fruit pomace recycling is critical for green biomanufacturing. Lactococcus lactis, a common host for waste valorization, shows poor protein expression in conventional systems. We constructed T7RNAP-driven inducible and constitutive expression systems in L. lactis NZ9000. The two systems increased target gene transcription by 103-fold and 207-fold, and enzyme activity by 5.24-fold and 10.36-fold, respectively. A signal peptide enabled efficient extracellular secretion of recombinant proteins. The engineered strains synthesized levansucrase and levan, achieving yields of 36.27 g/L and 27.53 g/L

d-Methionine and d-Phenylalanine Improve Lactococcus lactis F44 Acid Resistance and Nisin Yield by Governing Cell Wall Remodeling.

Lactococcus lactis encounters various environmental challenges, especially acid stress, during its growth. The cell wall can maintain the integrity and shape of the cell under environmental stress, and d-amino acids play an important role in cell wall synthesis. Here, by analyzing the effects of 19 different d-amino acids on the physiology of L. lactis F44, we found that exogenously supplied d-methionine and d-phenylalanine increased the nisin yield by 93.22% and 101.29%, respectively, as well as significantly increasing the acid resistance of L. lactis F44. The composition of the cell wall in

Oral immunization with recombinant Lactococcus lactis delivering a multi-epitope antigen CTB-UE attenuates Helicobacter pylori infection in mice.

Urease is an essential virulence factor and colonization factor for Helicobacter pylori (H. pylori) and is considered as an excellent vaccine candidate antigen. However, conventional technologies for preparing an injectable vaccine require purification of the antigenic protein and preparation of an adjuvant. Lactococcus lactis NZ9000 (L. lactis) could serve as an antigen-delivering vehicle for the development of edible vaccine. In previous study, we constructed a multi-epitope vaccine, designated CTB-UE, which is composed of the mucosal adjuvant cholera toxin B subunit (CTB), three Th cell epi

Oral immunization of mice with recombinant Lactococcus lactis expressing Cu,Zn superoxide dismutase of Brucella abortus triggers protective immunity.

Brucella infections mainly occur through mucosal surfaces. Thus, the development of mucosal administered vaccines could be instrumental for the control of brucellosis. Here, we evaluated the usefulness of recombinant Lactococcus lactis secreting Brucella abortus Cu-Zn superoxide dismutase (SOD) as oral antigen delivery system, when administered alone or in combination with L. lactis expressing IL-12. To this end, mice were vaccinated by oral route with L. lactis NZ9000 transformed with pSEC derivatives encoding for SOD (pSEC:SOD) and IL-12 (pSEC:scIL-12). In animals receiving L. lactis pSEC:SO

Identification, cloning and sequencing of the replication region of Lactococcus lactis ssp. lactis biovar. diacetylactis Bu2 citrate plasmid pSL2.

The replication region of the 7.8 kilobase (kb) citrate plasmid pSL2 from Lactococcus lactis ssp. lactis biovar. diacetylactis Bu2 was identified. Deletion derivatives of pSL2 were introduced into plasmid-free strain Bu2-60 and tested for their ability to replicate autonomously. The region necessary for replication was identified by comparison of the pSL2 derivatives, cloned and sequenced. No homologies were detected by comparing the putative Rep protein of pSL2 with replicons of other plasmids of Gram-positive bacteria. A part of an IS-element flanking the replication region was found.

Development of donkey milk fermented with Lactococcus lactis subsp. lactis CECT 31096 and characterization of the antioxidant and antihypertensive activity.

Donkey milk (DM) is a nutritionally rich source of bioactive compounds, essential vitamins and minerals. However, its potential for developing functional fermented products remains unexplored. To address this gap, we investigated microbial dynamics during spontaneous fermentation of raw Zamorano-Leonese DM to isolate lactic acid bacteria (LAB) suitable for dairy applications. Raw milk from Zamorano-Leonese breed donkeys, raised on an organic farm in Salamanca (Spain), was spontaneously fermented at 37 °C until reaching pH 4.6. Metataxonomic analysis revealed a clear shift from environmental ba

Nisin, a food preservative produced by Lactococcus lactis, affects the localization pattern of intermediate filament protein in HaCaT cells.

Nisin is a food preservative produced by Lactococcus lactis subsp. lactis. Previous blood biochemical research revealed that nisin has physiological effects in mammals; although the site of action has yet to be identified, keratinocytes have been proposed as a possible target. In this study, we investigated whether nisin affects keratinocytes by examining the effects on eukaryotic intermediate filaments in HaCaT human keratinocytes. Treatment with 93 μg/ml nisin for 24 h decreased the localization of the intermediate filament proteins cytokeratin (CK)5 and CK17 at the cell periphery, which wer
