Oral administration of Lactococcus lactis-expressing heat shock protein 65 and tandemly repeated IA2P2 prevents type 1 diabetes in NOD mice.

Type 1 diabetes mellitus (T1DM) is a chronic autoimmune disease characterized by the destruction of insulin-secreting β cells upon autoreactive T cell attack. Oral administration of autoantigens is an attractive approach to treating T1DM, but an effective carrier should be used in order to protect antigens. Lactococcus lactis, a safe engineering strain, was used for this task in the present study. Two recombinant L. lactis expressing protein HSP65-6IA2P2 were used and be investigated the effects and mechanisms against T1DM in NOD mice. Our findings demonstrate that recombinant L. lactis strain

Lactococcus lactis-expressing listeriolysin O (LLO) provides protection and specific CD8(+) T cells against Listeria monocytogenes in the murine infection model.

Lactococcus lactis has previously been proposed as a vaccine platform for the safe delivery of heterologous antigens. Here we utilized L. lactis as a live vector for expression of listeriolysin O (LLO), a major Listeria monocytogenes antigen and virulence factor. A variety of plasmid constructs were designed to permit either constitutive or nisin-inducible expression of secreted or non-secreted LLO in L. lactis. Recombinant strains were subsequently tested in a murine model for vaccination efficacy against L. monocytogenes infection. CD8(+) T lymphocytes specific for the LLO(91-99) epitope wer

Function of Yogurt Fermented with the Lactococcus lactis 11/19-B1 Strain in Improving the Lipid Profile and Intestinal Microbiome in Hemodialysis Patients.

BACKGROUND/OBJECTIVES: The number of chronic kidney disease (CKD) patients is increasing in Japan, and this population is at high risk of death from cardiovascular and cerebrovascular diseases. Therefore, prevention of arteriosclerosis as a common underlying cause of these diseases is required. In this study, we examined whether 11/19-B1 yogurt, which has been proven to reduce serum low-density lipoprotein (LDL) levels, can decrease the serum levels of indoxylsulfate and trimethylamine-N-oxide (TMAO), which are produced by intestinal microbiota and known to cause arteriosclerosis, through impr

Construction of recombinant Lactococcus lactis expressing VP1 from duck hepatitis a virus types 1 and 3 and evaluation of its immune effect.

In this study, VP1 genes from duck hepatitis A virus (DHAV) genotypes 1 (DHAV-1) and 3 (DHAV-3) were amplified through reverse transcription polymerase chain reaction (RT-PCR) by using viral RNA as the template. The amplified genes were cloned into the plasmid pNZ8149 through homologous recombination, yielding the recombinant plasmids pNZ8149-DHAV-1 VP1 and pNZ8149-DHAV-3 VP1. These plasmids were electroporated into competent L. lactis NZ3900 cells to generate the recombinant strains L. lactis pNZ8149-DHAV-1 VP1/NZ3900 and L. lactis pNZ8149-DHAV-3 VP1/NZ3900, respectively. 2,3-Butanediol dehydrogenase is more efficient than acetoin reductase at metabolizing reserve carbon to improve carbon cycling pathways in Lactococcus lactis N8.

Acetoin (AC) and 2,3-butanediol (2,3-BDO) are metabolites produced by lactic acid bacteria using glucose as a carbon source. These two metabolites act as carbon reserves and can be reutilised by the cells. In this study, we investigated the enzymatic characteristics of acetoin reductase (ButA) and 2,3-butanediol dehydrogenase (ButB). The performance of butA or/and butB knockout mutants of Lactococcus lactis N8 was evaluated. ButA and ButB were heterologously expressed in E. coli, and their enzymatic characteristics were measured in vitro under different pH, temperature, and metal ion condition

Surface display of spring viremia of carp virus glycoprotein on Lactococcus lactis and its protection efficacy in common carp (Cyprinus carpio L.).

Spring viremia of carp virus (SVCV) causes devastating disease in aquaculture, resulting in significant economic impact. To develop an effective means against SVCV infection, a Lactococcus lactis (L.lactis) based subunit vaccine (pNZ-UGA) was developed based on surface displaying of SVCV glycoprotein using anchoring motif of the cA (C terminus of the peptidoglyvsn-binding) domains of AcmA, a major autolysin from L.lactis. The surface expression of SVCV glycoprotein was verified by indirect immunofluorescence assay. The efficacy of the constructed vaccine was further evaluated in common carp. T

Oral vaccine of Lactococcus lactis harbouring pandemic H1N1 2009 haemagglutinin1 and nisP anchor fusion protein elevates anti-HA1 sIgA levels in mice.

OBJECTIVE: An oral lactococcal-based vaccine which haboured the haemagglutinin1 (HA1) antigen fused to nisP anchor protein for the purpose of surface displaying the HA1 antigen was developed against H1N1 virus. RESULTS: Recombinant L. lactis strains expressed HA1-nisP fusion proteins when induced with nisin, as confirmed through western blotting. However, immunofluorescense did not detect any surface-displayed proteins, suggesting that the protein was either unsuccessfully translocated or improperly displayed. Despite this, oral administration of recombinant L. lactis strains to BALB/c mice r

Engineered invasive Lactococcus lactis as an oral vaccine delivery platform is associated with compartmentalized immune modulation and enhances control of Leishmania infantum infection.

Visceral leishmaniasis caused by Leishmania infantum remains a major challenge for vaccine development due to the intracellular localization of the parasite and the need for effective antigen delivery and cellular immune responses. Genetically engineered lactic acid bacteria have emerged as promising platforms for mucosal vaccination because of their genetic tractability and capacity to interact with the host immune system. Here, we evaluated an engineered invasive Lactococcus lactis strain expressing fibronectin-binding protein A (FnBPA) and carrying the chimeric TryPan antigen-encoding plasm

Comparison of Postbiotics and Probiotics of Lactococcus lactis NZ9000 in Alleviating Liver Injury Induced by Bisphenol A via Binding and Removing Bisphenol A in Mouse Model.

Studies have demonstrated the toxic effects of inevitable exposure of bisphenol A (BPA), especially liver injury via gut-liver axis. Probiotics are increasingly used as adjunctive therapy to ameliorate adversely effects induced by BPA. In view of safety concerns of probiotics, postbiotics are proposed as alternatives to probiotics in clinical applications. At present, no study has directly compared the ameliorative effect of probiotics and postbiotics. This study compares the effect of postbiotics and probiotics derived from Lactococcus lactis NZ9000 employed for BPA-induced liver injury in a
