A collision gradient method to determine the immersion depth of nitroxides in lipid bilayers: application to spin-labeled mutants of bacteriorhodopsin.

Ten mutants of bacteriorhodopsin, each containing a single cysteine residue regularly spaced along helix D and facing the lipid bilayer, were derivatized with a nitroxide spin label. Collision rates of the nitroxide with apolar oxygen increased with distance from the membrane/solution interface. Collision rates with polar metal ion complexes decreased over the same distance. Although the collision rates depend on steric constraints imposed by the local protein structure and on the depth in the membrane, the ratio of the collision rate of oxygen to those of a polar metal ion complex is independ

Formation of sparsely tethered bilayer lipid membrane on a biodegradable self-assembled monolayer of poly(lactic acid).

The utilization of biomimetic membranes supported by advanced self-assembled monolayers is gaining attraction as a promising sensing tool. Biomimetic membranes offer exceptional biocompatibility and adsorption capacity upon degradation, transcending their role as mere research instruments to open new avenues in biosensing. This study focused on anchoring a sparsely tethered bilayer lipid membrane onto a self-assembled monolayer composed of a biodegradable polymer, functionalized with poly(ethylene glycol)-cholesterol moieties, for lipid membrane integration. Real-time monitoring via quartz cry

Lepromatous leprosy patients produce antibodies that recognise non-bilayer lipid arrangements containing mycolic acids.

Non-bilayer phospholipid arrangements are three-dimensional structures that form when anionic phospholipids with an intermediate structure of the tubular hexagonal phase II are present in a bilayer of lipids. Antibodies that recognise these arrangements have been described in patients with antiphospholipid syndrome and/or systemic lupus erythematosus and in those with preeclampsia; these antibodies have also been documented in an experimental murine model of lupus, in which they are associated with immunopathology. Here, we demonstrate the presence of antibodies against non-bilayer phospholipi

Hydrophobic barriers of lipid bilayer membranes formed by reduction of water penetration by alkyl chain unsaturation and cholesterol.

The hydrophobicity profiles across phosphatidylcholine (PC)-cholesterol bilayer membranes were estimated in both frozen liposome suspensions and fluid-phase membranes as a function of alkyl chain length, unsaturation, and cholesterol mole fraction. A series of stearic acid spin labels, with the probe attached to various positions along the alkyl chain, cholesterol-type spin labels (cholestane and androstane spin labels), and Tempo-PC were used to examine depth-dependent changes in local hydrophobicity, which is determined by the extent of water penetration into the membrane. Local hydrophobici

Enzyme Sandwich of Planar Unsupported Porous Polymer-Lipid Hybrid Bilayer for Single-Molecule Nanopore Enzymology.

Electrochemical recording of traditional planar unsupported lipid bilayer is crucial for screening ion channel blocking drugs and studying the biophysical properties of protein channels. However, the fragility and instability of lipid bilayer limit their development and applications. In this work, we developed a robust polydopamine (PDA)-lipid hybrid bilayer platform, which allows membrane proteins to easily insert and form stable channels. Furthermore, individual enzyme molecules can be tightly confined between a porous polymer aerogel film and a lipid monolayer to enable a nanopore-based enz

Investigation of benzodiazepines (BZDs) in a DPPC lipid bilayer: Insights from molecular dynamics simulation and DFT calculations.

Toxicity is an essential parameter for drug development process and drug design. In this context, the effects of concentration of two Benzodiazepine drugs (diazepam, clonazepam) on fully hydrated dipalmitoylphosphatidylcholine (DPPC) has been studied at 323 K using molecular dynamics simulations. Various properties of bilayer such as membrane area per lipid, mass density distributions, order parameters, radial distribution functions, lateral diffusion, and electrostatic potential have been examined at three different concentrations of each drug. The location of drugs in the membrane are estima

Kinetics of lipid bilayer permeation of a series of ionisable drugs and their correlation with human transporter-independent intestinal permeability.

For low molecular weight drugs, lipid bilayer permeation is considered the major route for in vivo cell barrier passage. We recently introduced a fluorescence assay with liposomes to determine permeation kinetics of ionisable compounds across the lipid bilayer by monitoring drug-induced pH changes inside the liposomes. Here, we determined the permeability coefficients (PFLipP, FLipP for "Fluorescence Liposomal Permeability") across 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC) bilayers of 35 ionisable drugs at pH6.0 and compared them to available in vivo human jejunal permeability (P

Membrane Sterols Modulate the Binding Mode of Amphotericin B without Affecting Its Affinity for a Lipid Bilayer.

Membrane-active antibiotics are known to selectively target certain pathogens based on cell membrane properties, such as fluidity, lipid ordering, and phase behavior. These are in turn modulated by the composition of a lipid bilayer and in particular by the presence and type of membrane sterols. Amphotericin B (AmB), the golden standard of antifungal treatment, exhibits higher activity toward ergosterol-rich fungal membranes, which permits its use against systemic mycoses; however, the selectivity for fungal membranes is far from satisfactory leading to severe side effects. Despite decades of

[The reconstruction of ion channels created by protein neurotoxins in bilayer lipid membranes as a research tool for exocytosis processes].

The direct transport of inorganic ions and much highly organized biomolecules across native membranes by the specific membrane proteins--ionic channels provides the functioning of a number of important biochemical processes like depolarization, Ca(2+)-release and exocytosis in nerve or muscle cells at the synaptic transmission. Therefore, the investigation of the structure and ion-conducting properties for neurotoxin channels and different factors affecting channels themselves and neuro-muscular transmission plays an important role in the research of pathologies that appear in intoxicated nerv

Salicylates and proton transport through lipid bilayer membranes: a model for salicylate-induced uncoupling and swelling in mitochondria.

Mechanisms of proton transport were investigated in phospholipid bilayer membranes exposed to salicylates and benzoates. Membranes were formed from diphytanoyl phosphatidylcholine in decane plus chlorodecane (50% vol/vol). Proton and anion conductances (GH and GA) were calculated from the total conductances and the H+ or A diffusion potentials produced by transmembrane H+ or A gradients. At low pH salicylate caused a GH which was proportional to the square of the total weak acid concentration, and GH was maximum when pH = pK. At neutral to alkaline pH salicylate caused a GA which was proportio

[Cation-anion selectivity and conductivity of the channels formed by black widow spider whole venom in lipid bilayer].

It is established that BWSV (black widow spider venom) is capable of forming ion channels in lipid bilayer (BPhM). This effect is minimal upon pH value close to 5.0 and is increased either upon oxidation or alkalization of the medium. The amplitudes of the channels are monotonously decreased upon the reduction of pH with simultaneous narrowing of their dispersion. The degree of the change of BPhM conductivity by concentration of BWSV is close to 2 at different pH. Apparently the absorption of the active component of venom on bilayer surface is accompanied by doubling of the degree of its aggre

Complexes of star-shaped block copolymers of poly(2-alkyl-2-oxazine)s and curcumin can affect lipid bilayers mimicking biomembranes.

Poly(2-alkyl-2-oxazine)s (PAlOz) are promising tools for developing delivery systems due to their high biocompatibility, resistance to enzyme hydrolysis, and ability to degrade in biological environments. Here, we investigated the effects of hexaaza[26]cyclophane (CPh6), poly(2-methyl-2-oxazine) (PMedOz), a block copolymer of poly(2-ethyl-2-oxazine) (PEtOz) and poly(2-isopropyl-2-oxazine) (PiPrOz), star-shaped block copolymers with six PAlOz arms and a CPh6 branching center (CPh6-PAlOz), and the complexes of all these macromolecules with curcumin on lipid bilayers mimicking the membranes of no

Dynamics of Klebsiella pneumoniae OmpA transmembrane domain: the four extracellular loops display restricted motion behavior in micelles and in lipid bilayers.

The transmembrane domain of Klebsiella pneumoniae OmpA (KpOmpA) possesses four long extracellular loops that exhibit substantial sequence variability throughout OmpA homologs in Enterobacteria, in comparison with the highly conserved membrane-embedded β-barrel core. These loops are responsible for the immunological properties of the protein, including cellular and humoral recognition. In addition to key features revealed by structural elucidation of the KpOmpA transmembrane domain in detergent micelles, studies of protein dynamics provide insight into its function and/or mechanism of action. Pressure effects on the structure and phase behavior of DMPC-gramicidin lipid bilayers: a synchrotron SAXS and 2H-NMR spectroscopy study.

The influence of Gramicidin D (GD) incorporation on the structure and phase behavior of aqueous dispersions of DMPC lipid bilayers has been studied using small-angle x-ray scattering (SAXS) and (2)H-NMR spectroscopy. The experiments covered a temperature range from -10 degrees C to 60 degrees C and a pressure range of 0.001-4 kbar. Pressure was used to be able to tune the lipid bilayer conformational order and phase state and because high pressure is an important feature of certain natural biotopes. The data show that, depending on the GD concentration, the structure of the temperature- and pr

Effects of cholesterol on pore formation in lipid bilayers induced by human islet amyloid polypeptide fragments: a coarse-grained molecular dynamics study.

Disruption of the cellular membrane by the amyloidogenic peptide, islet amyloid polypeptide (IAPP), has been considered as one of the mechanisms of β-cell death during type 2 diabetes. The N-terminal region (residues 1-19) of the human version of IAPP is suggested to be primarily responsible for the membrane-disrupting effect of the full-length hIAPP peptide. However, the detailed assembly mode of hIAPP1-19 with membrane remains unclear. To gain insight into the interactions of hIAPP1-19 oligomer with the model membrane, we have employed coarse-grained molecular dynamics self-assembly simulati

Biomimetic 3D DNA Nanomachine via Free DNA Walker Movement on Lipid Bilayers Supported by Hard SiO2@CdTe Nanoparticles for Ultrasensitive MicroRNA Detection.

Herein, a novel three-dimensional (3D) DNA nanomachine with high walking efficiency via free DNA walker movement on biomimetic lipid bilayers supported by hard silica@CdTe quantum dots (SiO2@CdTe) was constructed for ultrasensitive fluorescence detection of microRNA. The synthesized SiO2@CdTe nanoparticles were adopted as the fluorescence indicator and spherical carrier of lipid bilayers, and then the DNA substrates were anchored on lipid bilayers with biomimetic fluidity through the cholesterol-lipid interaction. Once target microRNA-141 interacted with the 3D DNA nanomachine to release chole

Interaction Profiles and Stability of Rigid and Polymer-Tethered Lipid Bilayer Models at Highly Charged and Highly Adhesive Contacts.

Understanding interaction force versus distance profiles of supported lipid bilayers (SLBs) is relevant to a number of areas, which rely on these model systems, including, e.g., characterization of ligand/receptor interactions or bacterial adhesion. Here, the stability of 4 different SLB architectures was compared using the surface forces apparatus (SFA) and atomic force microscopy (AFM). Specifically, the outer envelope of the bilayer systems remained constant as 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC). The inner layer was varied between DPPC and 1,2-dipalmitoyl-3-trimethylammonium

Adaptation Independent Modulation of Auditory Hair Cell Mechanotransduction Channel Open Probability Implicates a Role for the Lipid Bilayer.

The auditory system is able to detect movement down to atomic dimensions. This sensitivity comes in part from mechanisms associated with gating of hair cell mechanoelectric transduction (MET) channels. MET channels, located at the tops of stereocilia, are poised to detect tension induced by hair bundle deflection. Hair bundle deflection generates a force by pulling on tip-link proteins connecting adjacent stereocilia. The resting open probability (P(open)) of MET channels determines the linearity and sensitivity to mechanical stimulation. Classically, P(open) is regulated by a calcium-sensitiv
