Obesity is a medical condition where excess fat is accumulated in the body. Strategies for treatment include nutritional intervention, drugs, nutraceuticals, satiety peptides. High fat-low carbohydrate food as a therapeutic is one of the strategies studied in the recent past for weight management. This study aimed to evaluate the effects of a plant-based high-fat food formulation (SU001) on lipid accumulation in diet-induced obese (DIO) C57BL/6 mice. SU001, which is mildly ketogenic and characterized by low carbohydrates, high fat and protein content, was administered to DIO mice for 30 days.

Robust dengue virus (DENV) replication requires lipophagy, a selective autophagy that targets lipid droplets. The autophagic mobilization of lipids leads to increased β-oxidation in DENV-infected cells. The mechanism by which DENV induces lipophagy is unknown. Here, we show that infection with DENV activates the metabolic regulator 5' adenosine-monophosphate activated kinase (AMPK), and that the silencing or pharmacological inhibition of AMPK activity decreases DENV replication and the induction of lipophagy. The activity of the mechanistic target of rapamycin complex 1 (mTORC1) decreases in D

The crystallinity of drug-loaded lipid nanoparticles is believed to affect drug release rates; however, effective control over lipid crystallinity has not been achieved by current lipid nanoparticle preparation methods. The present study investigates control over the crystallinity of drug-loaded nanoparticle aerosol lipid matrices (NALM) through differences in evaporation rates of precursor solution drops and the subsequent control over drug release rates from these matrices. Gefitinib-loaded NALM were synthesized in an aerosol reactor using precursor solutions of gefitinib and stearic acid at

The outer membrane of Gram-negative bacteria is an asymmetric bilayer in which lipopolysaccharide (LPS), the principal component of the outer leaflet, promotes tight packing and ordering of the membrane components that are essential for the barrier and load-bearing functions of this membrane. Lipopolysaccharide mobility is known to be restricted in the outer membrane, but this confinement and the underlying biophysical interactions responsible remain to be fully characterized. Here, we apply a bio-orthogonal strategy for in situ site-specific fluorescent labeling of LPS. Using fluorescence mic

A monolayer of 5-nm-sized Ag nanoparticles embedded in a liquid-crystalline lipid membrane undergoes a reversible morphological change during hydration and dehydration of the lipid membrane. High mobility of the encapsulating lipid molecules, chemically bound to the Ag atoms, induces redistribution of metal particles to produce significant and optically detectable changes in nanoparticle morphology. The morphological change occurs on a time scale that enables the Ag-nanoparticle-embedded membrane to be used as a convenient visual sensor for moisture and other organic solvents, as well as for b

A closed-loop extracorporeal system for the removal of atherosclerotic plaque from accessible vascular segments by targeted chemical dissolution in a blood-free environment is proposed. The target segment is isolated by balloon occlusion, drained of blood, and connected to an external bypass that maintains distal perfusion. With the treatment zone converted to a sealed saline chamber, concentrated chemical agents, each targeting a specific plaque component, dissolve the atheroma in situ: ethylenediaminetetraacetic acid (EDTA) for calcified deposits, surfactants for the lipid-rich necrotic core

A hallmark of cancer cells is their remarkable ability to efficiently adapt to favorable and hostile environments. Due to a unique metabolic flexibility, tumor cells can grow even in the absence of extracellular nutrients or in stressful scenarios. To achieve this, cancer cells need large amounts of lipids to build membranes, synthesize lipid-derived molecules, and generate metabolic energy in the absence of other nutrients. Tumor cells potentiate strategies to obtain lipids from other cells, metabolic pathways to synthesize new lipids, and mechanisms for efficient storage, mobilization, and u

Obesity is characterized by an excessive triacylglycerol accumulation in white adipocytes. Various mechanisms allowing the tight regulation of triacylglycerol storage and mobilization by lipid droplet-associated proteins as well as lipolytic enzymes have been identified. Increasing energy expenditure by inducing a mild uncoupling of mitochondria in adipocytes might represent a putative interesting anti-obesity strategy as it reduces the adipose tissue triacylglycerol content (limiting alterations caused by cell hypertrophy) by stimulating lipolysis through yet unknown mechanisms, limiting the

In humans, heparin-binding protein (HBP) and the potent chemotactic lipid leukotriene B(4) (LTB(4)) are important mediators of innate immune responses. Here we show that human neutrophils (PMNs) challenged with LTB(4) (30 s to 5 min) release HBP as determined by Western blot analysis. This response peaks at 100 nM of agonist and is mediated by the BLT1 receptor. Protein phosphatase-1 (30 microM) and wortmannin (0.5 microM) block the LTB(4)-mediated HBP release from PMNs, which suggests involvement of the 1-phosphatidylinositol 3-kinase intracellular pathway during degranulation. Furthermore, p

BACKGROUND: Hormone-sensitive lipase (HSL) is expressed predominantly in adipose tissue, where it plays an important role in catecholamine-stimulated hydrolysis of stored lipids, thus mobilizing fatty acids. HSL exhibits broad substrate specificity and besides acylglycerides it hydrolyzes cholesteryl esters, retinyl esters and lipoidal esters. Despite its role in fatty acid mobilization, HSL null mice have been shown to be resistant to diet-induced obesity. The aim of this study was to define lipid profiles in plasma, white adipose tissue (WAT) and liver of HSL null mice, in order to better un

Connective tissue cells present in periovular granulomas, induced in mice livers by schistosomal infection, were studied in situ and in an in vitro culture after cell migration from explanted granulomas. They were compared to cells of the adjacent hepatic tissue. Connective tissue cells in granulomas and granuloma-derived primary cell lines were characterized as myofibroblasts. The presence of lipid droplets in cells of early granulomas, and comparison with adjacent perisinusoidal cells, indicated their origin through activation and mobilization of lipocytes from the adjacent hepatic tissue. INTRODUCTION: Xanthomatosis is an atypical pseudotumor of connective tissue characterized by the accumulation of lipid-laden histiocytes, often manifesting as encapsulated yellowish masses. It is commonly associated with familial hypercholesterolemia (FH). This case report aims to present a rare instance of bilateral xanthomatosis affecting the Achilles tendons in a 42-year-old male, which underscores the importance of identifying clinical and radiological findings of this condition to facilitate early diagnosis and intervention for related musculoskeletal and metabolic disorders.

Apolipoprotein E (apoE) is an essential protein in lipid and cholesterol metabolism. Although the three common isoforms in humans differ only at two sites, their consequences in Alzheimer's disease (AD) are dramatically different: only the ε4 allele is a major genetic risk factor for late-onset Alzheimer's disease. The isoforms exist as a mixture of oligomers, primarily tetramer, at low μM concentrations in a lipid-free environment. This self-association is involved in equilibrium with the lipid-free state, and the oligomerization interface overlaps with the lipid-binding region. Elucidation o

Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disorder characterized by the progressive loss of motor neurons, muscle atrophy and systemic energy imbalance. Increasing evidence suggests a metabolic shift in ALS from glucose metabolism toward fatty acid utilization; however, the downstream consequences of this reprogramming on disease progression and neuropathology remain poorly defined. We investigated neurometabolic changes in ALS using in vitro and in vivo models of familial ALS expressing the human fused in sarcoma variant R521G (hFUSR521G), along with post-mortem spinal

UNLABELLED: Obese adipose tissue is enriched in unsaturated triacylglycerol (TAG) species, yet the mechanisms driving this lipid remodeling remain incompletely understood. In this study, we conducted comprehensive lipidomic profiling of white adipose tissue from predominantly midlife obese individuals and consistently observed an enrichment of unsaturated TAG species. Acyl chain analysis of both TAGs and glycerophospholipids revealed a predominance of C18 unsaturated fatty acids (FAs), particularly FA 18:1 and FA 18:2. To investigate the mechanistic basis of this selective enrichment, we perf

UNLABELLED: Very low-density lipoprotein (VLDL) secretion provides a mechanism to export triglycerides (TG) from the liver to peripheral tissues, maintaining lipid homeostasis. In nonalcoholic fatty liver disease (NAFLD), VLDL secretion disturbances are unclear. Methionine adenosyltransferase (MAT) is responsible for S-adenosylmethionine (SAMe) synthesis and MAT I and III are the products of the MAT1A gene. Deficient MAT I and III activities and SAMe content in the liver have been associated with NAFLD, but whether MAT1A is required for normal VLDL assembly remains unknown. We investigated the

A systematic study of the retention behavior of isomeric triacylglycerols (TGs) in silver-ion HPLC on a ChromSpher Lipids column has been performed between 10 to 40°C using the most widespread hexane- and dichloromethane-based mobile phases. The randomization of mono-acyl TG standards and the random esterification of glycerol with fatty acids are employed to produce mixtures of TG isomers. The mobile phase composition has no influence on the general retention pattern, but significant differences in the retention order of double bond (DB) positional isomers in hexane and dichloromethane mobile

Lipids play an important role to support the rapid growth of cancer cells, which can be derived from both the endogenous synthesis and exogenous supplies. Enhanced de novo fatty acid synthesis and mobilization of stored lipids in cancer cells promote tumorigenesis. Besides, lipids and fatty acids derived from diet or transferred from neighboring adipocytes also influence the proliferation and metastasis of cancer cells. Indeed, the pathogenic roles of adipocytes in the tumor microenvironment have been recognized recently. The adipocyte-derived mediators or the cross talk between adipocytes and

Melanoma's ability to progress and develop resistance to therapies are the result of both genotypic changes and substantial levels of adaptability at the phenotypic and metabolic level. One of the most clinically relevant adaptive states is the microphthalmia associated transcription factor MITF low, which has been correlated with de-differentiation, invasion potential and drug tolerance, as well as altered redox status. As important regulators of this adaptation, lipid droplets have become involved in coordinating fat stores and mobilization, organelle signaling, membrane lipid composition an
