Inverted genomic segments and complex triplication rearrangements are mediated by inverted repeats in the human genome.

We identified complex genomic rearrangements consisting of intermixed duplications and triplications of genomic segments at the MECP2 and PLP1 loci. These complex rearrangements were characterized by a triplicated segment embedded within a duplication in 11 unrelated subjects. Notably, only two breakpoint junctions were generated during each rearrangement formation. All the complex rearrangement products share a common genomic organization, duplication-inverted triplication-duplication (DUP-TRP/INV-DUP), in which the triplicated segment is inverted and located between directly oriented duplica

A kinesin signaling complex mediates the ability of GSK-3beta to affect mood-associated behaviors.

Lithium has been the gold standard in the treatment of bipolar disorder (BPD) for 60 y. Like lithium, glycogen synthase kinase 3 (GSK-3) inhibitors display both antimanic-like and antidepressant-like effects in some animal models. However, the molecular mechanisms of both lithium and GSK-3 inhibitors remain unclear. Here we show that the GSK-3 inhibitor AR-A014418 regulated alpha-amino-3-hydroxyl-5-methyl-4-isoxazole-propionate (AMPA)-induced GluR1 and GluR2 internalization via phosphorylation of kinesin light chain 2 (KLC2), the key molecule of the kinesin cargo delivery system. Specifically,

Oxygen is required for the L-cysteine-mediated decomposition of protein-bound dinitrosyl-iron complexes.

Increasing evidence suggests that iron-sulfur proteins are the primary targets of nitric oxide (NO). Exposure of Escherichia coli cells to NO readily converts iron-sulfur proteins to protein-bound dinitrosyl-iron complexes (DNICs). Although the protein-bound DNICs are stable in vitro under aerobic or anaerobic conditions, they are efficiently repaired in aerobically growing E. coli cells even without new protein synthesis. The cellular repair mechanism for the NO-modified iron-sulfur proteins remains largely elusive. Here we report that, unlike aerobically growing E. coli cells, starved E. col

[Arthralgia and urticaria as immune-complex mediated complications after meningitis with meningococci group B].

A 4-year-old boy was treated for an infection with a meningococcus type B. After a few days of recovery, he felt ill again on day 6. The patient had urticaria and arthralgia without arthritis. The infection parameters did not normalize or increase in severity. No abnormalities other than a lowered CH50 value were found in the extensive diagnostic tests performed. The diagnosis of immune-complex mediated complications following a meningococcal infection was made. The patient was sent home where he continued pain relief and physiotherapy treatment. After 2 months at outpatient follow-up, he show

BKCa-Cav channel complexes mediate rapid and localized Ca2+-activated K+ signaling.

Large-conductance calcium- and voltage-activated potassium channels (BKCa) are dually activated by membrane depolarization and elevation of cytosolic calcium ions (Ca2+). Under normal cellular conditions, BKCa channel activation requires Ca2+ concentrations that typically occur in close proximity to Ca2+ sources. We show that BKCa channels affinity-purified from rat brain are assembled into macromolecular complexes with the voltage-gated calcium channels Cav1.2 (L-type), Cav2.1 (P/Q-type), and Cav2.2 (N-type). Heterologously expressed BKCa-Cav complexes reconstitute a functional "Ca2+ nanodoma

Tiam1-IRSp53 complex formation directs specificity of rac-mediated actin cytoskeleton regulation.

The exchange factor Tiam1 regulates multiple cellular functions by activating the Rac GTPase. Active Rac has various effects in cells, including alteration of actin cytoskeleton and gene expression, via binding to and modulating the activity of diverse effector proteins. How individual Rac effectors are selected for activation and regulated in response to upstream signals is not well understood. We find that Tiam1 contributes to both of these processes by binding to IRSp53, an adaptor protein that is an effector for both Rac and Cdc42. Tiam1 directs IRSp53 to Rac signaling by enhancing IRSp53

Novel checkpoint response to genotoxic stress mediated by nucleolin-replication protein a complex formation.

Human replication protein A (RPA), the primary single-stranded DNA-binding protein, was previously found to be inhibited after heat shock by complex formation with nucleolin. Here we show that nucleolin-RPA complex formation is stimulated after genotoxic stresses such as treatment with camptothecin or exposure to ionizing radiation. Complex formation in vitro and in vivo requires a 63-residue glycine-arginine-rich (GAR) domain located at the extreme C terminus of nucleolin, with this domain sufficient to inhibit DNA replication in vitro. Fluorescence resonance energy transfer studies demonstra

UV-induced ubiquitylation of XPC protein mediated by UV-DDB-ubiquitin ligase complex.

The xeroderma pigmentosum group C (XPC) protein complex plays a key role in recognizing DNA damage throughout the genome for mammalian nucleotide excision repair (NER). Ultraviolet light (UV)-damaged DNA binding protein (UV-DDB) is another complex that appears to be involved in the recognition of NER-inducing damage, although the precise role it plays and its relationship to XPC remain to be elucidated. Here we show that XPC undergoes reversible ubiquitylation upon UV irradiation of cells and that this depends on the presence of functional UV-DDB activity. XPC and UV-DDB were demonstrated to i

Are oxidized LDL/beta2-glycoprotein I complexes pathogenic antigens in autoimmune-mediated atherosclerosis?

The oxidative modification of low-density lipoprotein (LDL) in the intima of arteries contributes to the initiation and progression of atherosclerotic lesions. We have previously reported that oxidized LDL (oxLDL) interacts with an endogenous plasma protein, beta2-glycoprotein I (beta2GPI), to form complexes and that the interaction is mediated by oxLDL specific ligands. We have also demonstrated the presence of oxLDL/beta2GPI complexes in the blood stream of patients with systemic inflammatory and autoimmune diseases. These findings implicate that oxLDL/beta2GPI complexes are possible atherog

CARD11 mediates factor-specific activation of NF-kappaB by the T cell receptor complex.

NF-kappaB is a critical target of signaling downstream of the T cell receptor (TCR) complex, but how TCR signaling activates NF-kappaB is poorly understood. We have developed an expression cloning strategy that can identify catalytic and noncatalytic molecules that participate in different pathways of NF-kappaB activation. Screening of a mouse thymus cDNA library yielded CARD11, a membrane-associated guanylate kinase (MAGUK) family member containing CARD, PDZ, SH3 and GUK domains. Using a CARD-deleted variant of CARD11 and RNA interference (RNAi), we demonstrate that CARD11 mediates NF-kappaB

C5b-9 membrane attack complex mediates endothelial cell apoptosis in experimental glomerulonephritis.

We studied the role of the C5b-9 membrane attack complex in two models of inflammatory glomerulonephritis (GN) initiated by acute glomerular endothelial injury in Piebold-viral-Glaxo (PVG) complement-sufficient rats (C+), C6-deficient rats (C6-), and rats systematically depleted of complement with cobra venom factor (CVF). GN was induced by performing a left nephrectomy and selectively perfusing the right kidney with either 1) the lectin concanavalin A (Con A) followed by complement-fixing anti-Con A (Con A GN) or 2) purified complement-fixing goat anti-rat glomerular endothelial cell (GEN) an

Measurements of cell-mediated cytotoxicity on target cells labelled with the indium-111-oxine complex.

The preparation is given of an 111indium-oxine complex that is non toxic and can label reproductively various types of cells. They retain the label for long periods (24 hours and more). This method gives parallel results for cell-mediated cytotoxicity measurements to Chromium-51 labelling and is found to have wider uses. It can label cells that release 51Cr too fast, makes possible long term measurements of cytotoxicity and accurate counts on small numbers of target cells.

ATG14-Mediated SNARE Complex Activation Promotes ΔFosB Degradation to Ameliorate Levodopa-Induced Dyskinesia.

The chronic accumulation of ΔFosB in striatal medium spiny neurons has been implicated as a pivotal contributor to the pathogenesis of levodopa-induced dyskinesia (LID). While recent studies have implicated autophagy in the degradation of ΔFosB and the amelioration of LID, the precise mechanisms remain elusive. We induced LID in a unilateral 6-hydroxydopamine-lesioned parkinsonism rat model via chronic levodopa treatment. To modulate the autophagy pathway, we overexpressed ATG14 in the striatum of LID rats and administered chloroquine, an autophagy inhibitor, peripherally. We assessed LID seve

Lattice-Directed Spin-Vibronic Coherence-Mediated Ultrafast Intersystem Crossing in Crystalline Diplatinum Complex.

External control on chemical reactivity requires understanding the fundamental forces governing reactions at molecular length scales, where interactions with the environment can affect reaction outcomes. In single crystals, reactivity is governed by lattice-constrained molecular packing and unique electrostatic potentials leveraged by specific system-bath interactions that are absent in the solution phase. However, sample depletion and product accumulation have made studies of ultrafast photochemical reactions in crystals challenging. Here, we present the first report of ultrafast spin-vibroni

SFTSV NSs Sequesters the Complex of SFTSV NP-SAFA to Suppress SAFA Mediated Immune Response.

Severe fever with thrombocytopenia syndrome virus (SFTSV) is an emerging pathogenic Bunyavirus with a fatality rate of up to 30%. SFTSV nonstructural protein (NSs) forms viroplasm-like structures in the cytoplasm of infected cells, sequestering antiviral proteins and inhibiting interferon signaling. Nuclear scaffold attachment factor A (SAFA), a novel RNA sensor, can be retained in the cytoplasm by SFTSV nucleoprotein (NP) and facilitates antiviral immune response. Intriguingly, we discovered that SFTSV NSs triggered the nucleocytoplasmic translocation of SAFA. Nevertheless, it remains unclear

Ligand-complex-based quantification of β2-integrin-mediated affinity and avidity of murine T cells.

BACKGROUND: Integrins facilitate binding to the extracellular matrix and other cells. Their subunit β2 is exclusively expressed by leukocytes, binds to the intercellular cell adhesion molecule 1 (ICAM-1), and is pivotal for their recruitment to sites of inflammation such as the atherosclerotic plaque. METHODS: To investigate β2-integrin-mediated adhesiveness, a well-established assay for human whole blood was adapted for the analysis of murine T cell subsets. Changes in avidity and affinity were assessed by incubation of murine complexes ICAM-1 in murine whole blood and consecutive stimulatio

Photoredox-Mediated Radical Smiles-Truce Rearrangement: From Aryl Migration to Molecular Complexity Generation.

The Smiles-Truce rearrangement represents a uniquely powerful but historically underutilized strategy for arene functionalization, enabling intramolecular C-C bond formation through controlled molecular reorganization. Despite its conceptual appeal as a strategic disconnection, its traditional implementation as a two-electron SNAr process has relied on strong bases and polarized reaction manifolds, which restrict functional group tolerance, limit substrate scope, and thus confine its use to relatively simple molecular settings. The emergence of radical Smiles-Truce rearrangements, particularly

RAD51C-XRCC3 complex regulates FANCM-mediated R-loop resolution to safeguard genome integrity.

Fanconi anemia (FA) is characterized by bone marrow failure, congenital abnormalities, and cancer predisposition. Mutations in RAD51 paralogs have been identified in FA-like disorders and cancers. Although the role of RAD51 paralogs is well established in homologous recombination (HR)-mediated DNA repair, little is known about their role during replication stress responses. Here, we report that the RAD51C-XRCC3 (CX3) complex of RAD51 paralogs participates in the FA pathway of R-loop tolerance mechanism. CX3 complex suppresses R-loops, transcription-replication collisions (TRCs), and associated

Pyridylalkylamine copper complex-mediated oxygen reduction: effect of alkyl chain and substituents on catalytic performance.

The oxygen reduction reaction (ORR) plays an essential role in many biological and artificial systems related to energy conversion. Modifying the organic scaffolds of metal complexes can adjust their catalytic performances toward the ORR. In this study, we have synthesized and characterized three pyridylalkylamines bearing different chains and substituents and their copper complexes, namely [Cu(tmpmen)](ClO4)2 (Cu-tmpmen), [Cu(tpmpn)](ClO4)2 (Cu-tpmpn), and [Cu(etOHtpen)](ClO4)2·CH3CN (Cu-etOHtpen) (tmpmen = N-methyl-N,N',N'-tris[(3-methyl-2-pyridinyl)methyl]-1,2-ethanediamine, tpmpn = N-methy

Ligand field and polarity tuning of Fe(II) complexes for selective ROS-mediated anticancer activity.

A redesigned N5-type ligand that strengthens the donating ability while reducing polarity was utilized to generate an Fe(II) complex. This modification reorganized the metal-centered electronic structure, producing red-shifted π-π* and LMCT transitions and enabling oxidation at a substantially lower potential. The resulting complex exhibited enhanced hydroxyl radical generation and improved cellular uptake, leading to pronounced and selective ROS elevation in renal cancer cells with minimal effects on normal fibroblast cells. These results demonstrate that concurrent tuning of ligand-field str
