Why Do We Lose Bone During Weight Loss: Can It Be Prevented?

PURPOSE OF REVIEW: Weight loss is recommended for obesity management, but it also increases bone turnover and reduces bone mineral density (BMD) and bone quality (microarchitecture, geometry, and strength). This review examines mechanisms and the multiple factors that influence skeletal responses to caloric restriction. RECENT FINDINGS: Bone loss during weight loss likely reflects multiple interacting mechanisms, including alterations in endocrine signaling, impaired calcium absorption, reduced mechanical loading, and alterations in the gut microbiota. The extent of bone loss is further influ

In vitro activity and in vivo efficacy of sulopenem against Bacillus anthracis in inhalational anthrax mouse and rabbit models.

Bacillus anthracis is a gram-positive, spore-forming bacterium and the etiological agent of anthrax. As a Tier 1 Select Agent, the pathogen can be isolated from the natural environment, and the endospore is a significant biological threat that can be produced in large quantities, stored, and disseminated by aerosolization. The intentional release of B. anthracis spores has the potential for mass casualties and is a serious threat to public and military health. Fluoroquinolones and tetracyclines are common antibiotics used for post-exposure prophylaxis and treatment in the United States (U.S.),

PEGylated thrombopoietin mimetic, JNJ‑26366821 a novel prophylactic radiation countermeasure for acute radiation injury.

Thrombopoietin (TPO) is the primary regulator of platelet generation and a stimulator of multilineage hematopoietic recovery following exposure to total body irradiation (TBI). JNJ‑26366821, a novel PEGylated TPO mimetic peptide, stimulates platelet production without developing neutralizing antibodies or causing any adverse effects. Administration of a single dose of JNJ‑26366821 demonstrated its efficacy as a prophylactic countermeasure in various mouse strains (males CD2F1, C3H/HeN, and male and female C57BL/6J) exposed to Co-60 gamma TBI. A dose dependent survival efficacy of JNJ‑26366821

Safety protocols, precautions, and countermeasures aboard the International Space Station to prevent ocular injury.

The International Space Station (ISS) is a $100 billion epicenter of human activity in the vacuum of space that displays mankind's collective endeavor to explore the cosmic frontier. Even within the marvels of technological sophistication aboard the ISS, the human eye remains a highly vulnerable structure. In the absence of multiple layers of protection and risk assessments, crewmembers would face a substantial increase in vulnerability to ocular injury. Aside from stringent preflight screening criteria for astronauts, the ISS is equipped with ophthalmic medications, environmental control and

K18-hACE2 mice develop respiratory disease resembling severe COVID-19.

UNLABELLED: SARS-CoV-2 emerged in late 2019 and resulted in the ongoing COVID-19 pandemic. Several animal models have been rapidly developed that recapitulate the asymptomatic to moderate disease spectrum. Now, there is a direct need for additional small animal models to study the pathogenesis of severe COVID-19 and for fast-tracked medical countermeasure development. Here, we show that transgenic mice expressing the human SARS-CoV-2 receptor (angiotensin-converting enzyme 2 [hACE2]) under a cytokeratin 18 promoter (K18) are susceptible to SARS-CoV-2 and that infection resulted in a dose-depen

A rationale for space motion sickness.

Space motion sickness (SMS) still remains a problematic nuisance for a majority of fliers during their first 1 to 3 days in space and is an enigma to all concerned after almost half a century of spaceflight and extensive ground and in-flight studies. There is no known etiology consistent with observed signs and symptoms, and attempts to produce pre- and in-flight countermeasures have largely failed. Some medications for motion sickness (MS) have been partially effective, but must be used with caution due to their side effects. A hypothesis regarding SMS that is consistent with current knowledg

The Global Health Security Challenges Posed by Recurring Nipah Virus Outbreaks.

Nipah virus (NiV) has emerged as a high-consequence zoonotic pathogen, posing a significant threat to global health security. Characterised by extreme virulence and case-fatality rates that can exceed 70%, it is recognized as a WHO priority pathogen. This review examines the multifaceted challenges posed by recurring NiV outbreaks, spanning the complex dynamics of unpredictable zoonotic spillover from Pteropus fruit bats, the rising risk of sustained human-to-human transmission, and the profound gaps in medical countermeasures. The epidemiological shift from events mediated by intermediate hos

Regenerating murine CD8+ lung tissue resident memory T cells after targeted radiation exposure.

Radiation exposure occurs during medical procedures, nuclear accidents, or spaceflight, making effective medical countermeasures a public health priority. Naïve T cells are highly sensitive to radiation-induced depletion, although their numbers recover with time. Circulating memory CD8+ T cells are also depleted by radiation; however, their numbers do not recover. Critically, the impact of radiation exposure on tissue-resident memory T cells (TRM) remains unknown. Here, we found that sublethal thorax-targeted radiation resulted in the rapid and prolonged numerical decline of influenza A virus

Nuclear Disaster Medical Response and Preparedness: Perceived Radiation Risk in Ongoing Decommissioning Work and Future Strategies.

This study aimed to assess the situations involving potential risks of over-exposure to radiation faced by workers during the decommissioning of the Fukushima Daiichi Nuclear Power Plant (FDNPP), and to prepare effective medical countermeasures. While deaths have occurred at regular intervals at the FDNPP site, they cannot be attributed to direct effects of radiation. Radiation exposure among decommissioning workers is strictly maintained within regulatory limits, and no deterministic or stochastic radiation effects have been observed. High-risk areas include the reactor buildings of Units 1-4

Population-based study of asymptomatic infection with Chlamydia trachomatis among female and male students.

There are few epidemiological studies of asymptomatic chlamydial infection among students in non-medical settings with minimal bias and improved accuracy; thus, useful data from screening among students are limited. We aimed to obtain accurate epidemiological information about asymptomatic chlamydial infection among students in non-medical settings. A population-based cross-sectional survey of 10,440 >or=18-year-old asymptomatic students who volunteered for a urine screening test for chlamydia was conducted. The prevalences of asymptomatic infection were 9.5% for women and 6.7% for men. Multiv

Challenges in biodefense research and the role of US Army veterinary pathologists.

For years the nation's development of medical countermeasures to biowarfare agents has primarily existed as the domain of the United States military, but it has taken on increased urgency in the last few years. The realization that the civilian population is also at risk from biological agents has resulted in the institution of new biodefense programs at a variety of nonmilitary organizations. USAMRIID, a long-time leader in the nation's biodefense effort, will soon be joined by other US government agencies as part of a planned National Interagency Biodefense Campus at Fort Detrick Maryland. Characterization of rhesus macaque model for cobalt-60 gamma-radiation source without use of blood product.

Despite significant radiobiological advancements following World War II, only a limited number of medical countermeasures (MCMs) have been approved by the United States Food and Drug Administration (US FDA) for acute radiation exposure related illnesses. Accordingly, well-characterized and validated animal models, both large and small, are still very much needed to develop safe and effective countermeasures. Animal models that are used for such purposes need to reflect not only the clinical and pathogenic features of those seen in radiation exposed humans, but also comparable radiation dose- a

2018 Ebola virus disease outbreak in Équateur Province, Democratic Republic of the Congo: a retrospective genomic characterisation.

BACKGROUND: The 2018 Ebola virus disease (EVD) outbreak in Équateur Province, Democratic Republic of the Congo, began on May 8, and was declared over on July 24; it resulted in 54 documented cases and 33 deaths. We did a retrospective genomic characterisation of the outbreak and assessed potential therapeutic agents and vaccine (medical countermeasures). METHODS: We used target-enrichment sequencing to produce Ebola virus genomes from samples obtained in the 2018 Équateur Province outbreak. Combining these genomes with genomes associated with known outbreaks from GenBank, we constructed a max

Polypharmacy to Mitigate Acute and Delayed Radiation Syndromes.

There is a need for countermeasures to mitigate lethal acute radiation syndrome (ARS) and delayed effects of acute radiation exposure (DEARE). In WAG/RijCmcr rats, ARS occurs by 30-days following total body irradiation (TBI), and manifests as potentially lethal gastrointestinal (GI) and hematopoietic (H-ARS) toxicities after >12.5 and >7 Gy, respectively. DEARE, which includes potentially lethal lung and kidney injuries, is observed after partial body irradiation >12.5 Gy, with one hind limb shielded (leg-out PBI). The goal of this study is to enhance survival from ARS and DEARE by polypharmac

Antibodies against In Vivo-Expressed Antigens Are Sufficient To Protect against Lethal Aerosol Infection with Burkholderia mallei and Burkholderia pseudomallei.

Burkholderia mallei, a facultative intracellular bacterium and tier 1 biothreat, causes the fatal zoonotic disease glanders. The organism possesses multiple genes encoding autotransporter proteins, which represent important virulence factors and targets for developing countermeasures in pathogenic Gram-negative bacteria. In the present study, we investigated one of these autotransporters, BatA, and demonstrate that it displays lipolytic activity, aids in intracellular survival, is expressed in vivo, elicits production of antibodies during infection, and contributes to pathogenicity in a mouse

Delayed renal injury in survivors of hematologic acute radiation syndrome.

PURPOSE: A mass casualty disaster involving radiological or nuclear agents continues to be a public health concern which requires consideration of both acute and late tissue toxicities in exposed victims. With the advent of advanced treatment options for the mitigation of hematological injuries, there are likely to be survivors of total body irradiation (TBI) exposures as high as 8-10 Gy. These survivors are at risk for a range of delayed multi-organ morbidities including progressive renal failure. MATERIAL AND METHODS: Here, we established the WAG/RijCmcr rat as an effective model for the ev

Unlocking the surge in demand for personal and protective equipment (PPE) and improvised face coverings arising from coronavirus disease (COVID-19) pandemic - Implications for efficacy, re-use and sustainable waste management.

Currently, there is no effective vaccine for tackling the ongoing COVID-19 pandemic caused by SARS-CoV-2 with the occurrence of repeat waves of infection frequently stretching hospital resources beyond capacity. Disease countermeasures rely upon preventing person-to-person transmission of SARS-CoV2 so as to protect front-line healthcare workers (HCWs). COVID-19 brings enormous challenges in terms of sustaining the supply chain for single-use-plastic personal and protective equipment (PPE). Post-COVID-19, the changes in medical practice will drive high demand for PPE. Important countermeasures

Validation of a pan-orthopox real-time PCR assay for the detection and quantification of viral genomes from nonhuman primate blood.

BACKGROUND: In 1980, smallpox disease was eradicated from nature and Variola virus, the etiological agent of smallpox, was confined to two laboratories, one located in Russia (Moscow) later moved to VECTOR (Novosibirsk, Siberia) and one in the United States (CDC Atlanta). Vaccinations among the general public ceased shortly after the successful eradication campaign, resulting in an increasingly immunologically susceptible population. Because of the possibility of intentional reintroduction of Variola virus and the emergence of other pathogenic poxviruses, there is a great need for the developm
