Carbon Stable Isotope Analysis of Methylmercury Toxin in Biological Materials by Gas Chromatography Isotope Ratio Mass Spectrometry.

A critical component of the biogeochemical cycle of mercury (Hg) is the transformation of inorganic Hg to neurotoxic monomethylmercury (CH3Hg). Humans are exposed to CH3Hg by consuming marine fish, yet the origin of CH3Hg in fish is a topic of debate. The carbon stable isotopic composition (δ(13)C) embedded in the methyl group of CH3Hg remains unexplored. This new isotopic information at the molecular level is thought to represent a new proxy to trace the carbon source at the origin of CH3Hg. Here, we present a compound-specific stable isotope analysis (CSIA) technique for the determination of

Site-specific ontogenetic drivers of mercury concentrations in American alligators.

Contaminant body burdens are determined by complex interactions between contaminant inputs into the environment, local ecological dynamics, and organismal ontogeny. Although a naturally occurring element, mercury (Hg) is a potent neurotoxin, commonly assessed in contaminant body burdens. Anthropogenic activity has affected its spatial distribution, resulting in regional "hotspots" with elevated Hg concentrations. Due to the propensity of methylated Hg to bioaccumulate within individuals over time and biomagnify across trophic levels, long-lived apex predators can carry substantial body burdens

Natural Hg isotopic composition of different Hg compounds in mammal tissues as a proxy for in vivo breakdown of toxic methylmercury.

In the last decade, specific attention has been paid to total mercury (HgT) stable isotopic composition, especially in natural samples such as aquatic organisms, due to its potential to track the cycle of this toxic element in the environment. Here, we investigated Hg Compound Specific stable Isotopic Composition (CSIC) of natural inorganic Hg (iHg) and methylmercury (MMHg) in various tissues of aquatic mammals (Beluga whale from the Arctic marine environment and seals from the freshwater lake Baikal, Russia). In seals' organs the variation in mass dependent fractionation (MDF, δ(202)Hg) for t

Methyl mercury interactions with phospholipid membranes as reported by fluorescence, 31P and 199Hg NMR.

Methylmercury (CH3Hg(II)) interactions with multilamellar vesicles of dimyristoyl(DM)- and dipalmitoyl(DP)-phosphatidylcholine (PC), -phosphatidic acid (PA), -phosphatidylglycerol (PG), -phosphatidylserine (PS) and -phosphatidylethanolamine (PE) have been investigated from the metal viewpoint by solution 199Hg-NMR and from the membrane side by diphenylhexatriene fluorescence polarization and solid state 31P-NMR. Results can be summarized as follows: (1) CH3Hg(II) strong binding to membranes results in a progressive decrease of the free CH3HgOH 199Hg-NMR isotropic signal and because of a slow e

The application of (199)Hg NMR and (199m)Hg perturbed angular correlation (PAC) spectroscopy to define the biological chemistry of Hg(II): a case study with designed two- and three-stranded coiled coils.

The use of de novo designed peptides is a powerful strategy to elucidate Hg(II)-protein interactions and to gain insight into the chemistry of Hg(II) in biological systems. Cysteine derivatives of the designed alpha-helical peptides of the TRI family [Ac-G-(L(a)K(b)A(c)L(d)E(e)E(f)K(g))(4)-G-NH(2)] bind Hg(II) at high pH values and at peptide/Hg(II) ratios of 3:1 with an unusual trigonal thiolate coordination mode. The resulting Hg(II) complexes are good water-soluble models for Hg(II) binding to the protein MerR. We have carried out a parallel study using (199)Hg NMR and (199m)Hg perturbed an

Flood-pulse and trophic position modulate mercury concentrations in fishes from an Amazon floodplain lake.

Mercury (Hg) bioaccumulation can be influenced by biological characteristics (e.g., trophic position) and environmental changes (e.g., seasonal flood-pulse), as it can create site conditions conducive to methylation and alter the availability of food to biota. This study aimed to relate total mercury (THg) and methylmercury (MeHg) concentrations in fishes of an Amazonian lake (Catalão Lake) to their dietary changes during different hydrological seasons (rising-, high-, falling-, and low-water). Levels of THg, MeHg, and stable isotopes of nitrogen and carbon (δ15N and δ13C) were determined in t

Higher mass-independent isotope fractionation of methylmercury in the pelagic food web of Lake Baikal (Russia).

Mercury undergoes several transformations that influence its stable isotope composition during a number of environmental and biological processes. Measurements of Hg isotopic mass-dependent (MDF) and mass-independent fractionation (MIF) in food webs may therefore help to identify major sources and processes leading to significant bioaccumulation of methylmercury (MeHg). In this work, δ(13)C, δ(15)N, concentration of Hg species (MeHg, inorganic Hg), and stable isotopic composition of Hg were determined at different trophic levels of the remote and pristine Lake Baikal ecosystem. Muscle of seals

Enhanced Susceptibility of Methylmercury Bioaccumulation into Seston of the Laurentian Great Lakes.

Mercury concentrations in the Laurentian Great Lakes waters are among the lowest reported in the literature, while game fish concentrations approach consumption advisory limits, particularly in Lakes Superior, Huron, and Michigan, indicating efficient methylmercury transfer from water to game fish. To determine if increased transfer efficiency is evident within the lower food web, we measured (2010-2018) mercury and dissolved organic carbon (DOC) in water, and in size-sieved seston, dietary tracers (carbon and nitrogen isotope ratios), phytoplankton methylmercury bioaccumulation, and methylmer

Determination of mercury methylation potentials in the water column of lakes across Canada.

A stable isotope technique was used to trace the formation of methylmercury in lake water incubation assays at in situ conditions in five lakes across Canada. Methylation activity was only detected in the anoxic hypolimnia of lakes. The stable isotope was methylated at varying rates between lakes and depths within lakes ranging from 0.56%/day to 14.8%/day. A peak in methylation potential was typically observed just below the oxycline, which decreased with increasing depth. The depth and rates of methylation potential changed seasonally with no methylation activity occurring after fall turnover

Difference of mercury bioaccumulation in red mullets from the north-western Mediterranean and Black seas.

The relationships between total mercury (Hg) concentration and stable nitrogen isotope ratio (delta(15)N) were evaluated in Mullus barbatus barbatus and M. surmuletus from the Mediterranean Sea and M. barbatus ponticus from the Black Sea. Mercury concentration in fish muscle was six times higher in the two Mediterranean species than in the Black Sea one for similar sized animals. A positive correlation between Hg concentration and delta(15)N occurred in all species. Increase in Hg concentration with delta(15)N was high and similar in the two Mediterranean fishes and much lower in the Black Sea

Sooty terns as large-scale bioindicator of mercury contamination in marine ecosystems: A pantropical approach.

Mercury (Hg) is a toxic contaminant, which poses serious threats to both wildlife and humans worldwide. Although tropical oceans receive half of the world's atmospheric Hg depositions, its biomonitoring is largely overlooked in these ecosystems. This study provides a comprehensive, pantropical assessment of Hg contamination of marine ecosystems, using the sooty tern (Onychoprion fuscatus) as bioindicator. Total Hg concentrations were measured from feathers collected in 28 colonies across all tropical oceans (2003-2022), from adults (n = 564) and chicks (n = 407), to evaluate longer- and shorte

Terrestrial diet influences mercury bioaccumulation in zooplankton and macroinvertebrates in lakes with differing dissolved organic carbon concentrations.

Dietary uptake is a key step in conveying both toxic mercury (Hg; particularly as highly bioavailable methylmercury, MeHg) and essential dietary biochemicals, such as polyunsaturated fatty acids (PUFA), across trophic levels within aquatic food webs. Using stable isotopes and fatty acids we evaluated the role of food sources in size-fractioned plankton and littoral macroinvertebrates for the bioaccumulation of total Hg and MeHg in six oligotrophic and one mesotrophic Swedish lakes with differing concentrations of dissolved organic carbon (DOC). We found that the consumption of both algal and t

Methylmercury bioaccumulation and biomagnification in streams within forested catchments defoliated by spruce budworm.

Though outbreaks of defoliating insects are a widespread, natural disturbance in Canadian boreal forests and their magnitude and duration are increasing in recent years, we have little understanding of the impacts on stream ecosystems. Herein we examined the fate of mercury (Hg), a toxic element affected by other landscape-scale forest disturbances, in twelve stream food webs in the Gaspé Peninsula (Québec, Canada) that ranged in their severity of watershed defoliation from spruce budworm (Choristoneura fumiferana). Basal food sources (coarse and fine particulate organic matter, and biofilms),

Biomagnification of mercury in an estuarine food web.

Methylmercury is a toxin of local, regional, and global concern, with estuarine habitats possessing ecological characteristics that support conversion of inorganic mercury into this methylated form. We monitored Hg concentrations in species within the food web of the lower Cape Fear River (CFR) estuary in 2018-2020. Samples were analyzed for Hg concentrations and nitrogen isotopes (a measure of trophic level), and we found a positive relationship within this food web each year (p < 0.0001), indicating biomagnification is occurring. The highest Hg concentrations were among the upper trophic lev

Dying in the Sun: Direct evidence for elevated UV-B radiation at the end-Permian mass extinction.

Land plants can adjust the concentration of protective ultraviolet B (UV-B)-absorbing compounds (UACs) in the outer wall of their reproductive propagules in response to ambient UV-B flux. To infer changes in UV-B radiation flux at Earth's surface during the end-Permian mass extinction, we analyze UAC abundances in ca. 800 pollen grains from an independently dated Permian-Triassic boundary section in Tibet. Our data reveal an excursion in UACs that coincide with a spike in mercury concentration and a negative carbon-isotope excursion in the latest Permian deposits, suggesting a close temporal l

History of mercury use and environmental contamination at the Oak Ridge Y-12 Plant.

Between 1950 and 1963 approximately 11 million kilograms of mercury (Hg) were used at the Oak Ridge Y-12 National Security Complex (Y-12 NSC) for lithium isotope separation processes. About 3% of the Hg was lost to the air, soil and rock under facilities, and East Fork Poplar Creek (EFPC) which originates in the plant site. Smaller amounts of Hg were used at other Oak Ridge facilities with similar results. Although the primary Hg discharges from Y-12 NSC stopped in 1963, small amounts of Hg continue to be released into the creek from point sources and diffuse contaminated soil and groundwater

Pulsed volcanic sulfur emissions linked to the end-Triassic terrestrial crisis.

The end-Triassic mass extinction (ETE) was triggered by Central Atlantic Magmatic Province (CAMP) volcanism, which released voluminous carbon dioxide, sulfur dioxide, and halogens into the atmosphere, affecting marine and terrestrial ecosystems, but the mechanism driving terrestrial crisis remains unclear. Here, we investigate two terrestrial Triassic-Jurassic sections in high- and low/middle- paleolatitudes, finding synchronous anomalies of mercury concentration, sulfur (S) isotopes, S-associated molecular fossils, and biomarkers (retene, pimanthrene, and coronene), indicating that peak volca

Organochlorines, brominated flame retardants and mercury levels in six seabird species from the Gulf of St. Lawrence (Canada): relationships with feeding ecology, migration and molt.

Concentrations of organochlorines (OCs), brominated flame retardants (BFRs) and mercury (Hg) were measured in eggs of six seabird species breeding in the Gulf of St. Lawrence, Canada. Stable nitrogen (delta15N) and carbon (delta13C) isotopes were used as ecological tracers to measure trophic level and connectivity with benthos, respectively. Concentrations, patterns as well as ecological tracers varied significantly between species. The sum of polychlorinated biphenyls (SigmaPCBs) was the most important group measured in all seabird species based on concentration followed generally by the sum
