Nanoplatform-mediated metabolic reprogramming to overcome resistance mechanisms in antitumor therapies.

Therapeutic resistance remains a major bottleneck in achieving effective antitumor treatment. Many tumors exploit aberrant metabolic behaviors (e.g., excessive glycolysis, oxidative phosphorylation, autophagy, and anabolic pathways) to evade therapy-induced stress and sustain proliferation. Consequently, reprogramming tumor metabolism has emerged as a promising strategy for sensitizing tumors to various treatments. In this review, we outline a nanoplatform-mediated metabolic reprogramming framework as a unifying strategy to reverse diverse resistance mechanisms in tumor therapy. We systematica

Nanotechnology-mediated metabolic reprogramming for tumour ferroptosis: mechanisms and therapeutic prospects.

Ferroptosis, an iron-dependent form of regulated cell death driven by lipid peroxidation, has emerged as a promising therapeutic strategy for tumours resistant to conventional treatments. However, tumour cells frequently develop ferroptosis resistance through adaptive metabolic reprogramming mechanisms, such as enhancing antioxidant defences and altering nutrient utilization, both of which help maintain redox homeostasis and reduce oxidative damage. By leveraging the advantageous properties of nanomaterials, including tumour-targeting ability, controlled drug release, and multimodal synergisti

Suppression of Tumor Aggression Through Metabolic Reprogramming via Oxamate Targeting LDHA.

Lactate dehydrogenase (LDH) is a key glycolytic enzyme that catalyzes the interconversion of pyruvate and lactate, with LDHA gaining particular attention for its overexpression in various malignancies and pivotal role in the Warburg effect-driven metabolic reprogramming. Elevated LDHA activity supports rapid ATP production under hypoxic conditions, maintains NAD+ regeneration, and promotes lactate accumulation, creating an acidic tumor microenvironment (TME) that favors invasion and immune evasion. Accumulating evidence demonstrates that LDHA is essential for primary tumor growth and criticall

Tumor-Tropic Bacterial-Nanoparticle Hybrids for Metabolic Reprogramming and Cancer Immunotherapy.

Metabolic dysregulation and immune suppression are intrinsically linked within the solid tumor microenvironment, yet effective strategies to simultaneously reprogram tumor metabolism and restore antitumor immunity remain challenging. Here, we present a tumor-tropic bacterial-nanoparticle hybrid (SW@CDFM) that integrates metabolic reprogramming with immune activation for synergistic cancer treatment. The designed biohybrid system is constructed by covalently conjugating Shewanella oneidensis bacteria with tumor cell membrane-camouflaged DOX-Fe/CpG nanoparticles. Leveraging the intrinsic hypoxia

BCAA metabolic reprogramming identifies BCKDK as a therapeutic target in cancer.

INTRODUCTION: Branched-chain keto acid dehydrogenase kinase (BCKDK) is a mitochondrial kinase that suppresses branched-chain amino acid (BCAA) oxidative catabolism by phosphorylating and inhibiting the branched-chain α-keto acid dehydrogenase complex. Beyond this canonical metabolic function, accumulating evidence indicates that aberrant BCKDK activation contributes to tumor metabolic rewiring, signaling adaptation, malignant progression and therapy resistance. AREAS COVERED: This review summarizes the regulatory position of BCKDK in BCAA catabolism, its context-dependent functions across tum

Mechanosignaling Promotes Macrophage Apoptosis Resistance in Pulmonary Fibrosis via Metabolic Reprogramming.

The mechanisms underlying the progression of pulmonary fibrosis in idiopathic pulmonary fibrosis (IPF) and other interstitial lung diseases remain unclear. Increased extracellular matrix stiffness is a hallmark of fibrotic lung diseases. Monocyte-derived macrophages can promote fibrosis progression. However, there is limited understanding of how the mechanical properties of the fibrotic microenvironment influence macrophage phenotypes. Profibrotic macrophages are apoptosis-resistant, and this phenotype is modulated by enhanced mitochondrial bioenergetics. The objective of the study was to dete

Lipid metabolic reprogramming in ovarian cancer: molecular mechanisms and therapeutic implications.

Lipids play diverse roles in cellular homeostasis, and dysregulation of lipid metabolism is implicated in a wide spectrum of diseases, including cancer. Ovarian cancer cells exhibit profound metabolic alterations that support autonomous proliferation under conditions of hypoxia and nutrient limitation. Although tumor angiogenesis occurs, insufficient and fragile neovasculature often limits nutrient supply, necessitating metabolic adaptation. Among these adaptations, lipid metabolic reprogramming has emerged as a critical feature that supports tumor growth, survival, invasion, and therapeutic r

Cylindrospermopsin-induced metabolic reprogramming mapped in rat tissues by MALDI imaging.

Cylindrospermopsin (CYN) is a cyanobacterial toxin with hepatotoxic and nephrotoxic properties; however, its spatial distribution and molecular effects in mammalian tissues remain unclear. In this study, matrix-assisted laser desorption/ionisation mass spectrometry imaging (MALDI-MSI) was employed to assess toxin-induced lipid alterations in the stomach, liver, and kidneys of rats orally exposed to 500 μg CYN/kg body weight and euthanised at 0, 2, 4, 6, and 24 h. Cryosections were analysed under optimised conditions for CYN detection, followed by molecular annotation and lipidomic profiling. S

Mapping metabolic reprogramming in lung and breast cancer through integrative bioinformatics.

Metabolic reprogramming is central to cancer biology, enabling tumor cells to sustain rapid proliferation, resist stress, and adapt to therapy. However, these alterations are highly heterogeneous across cancer types, and current treatments rarely exploit subtype-specific metabolic vulnerabilities. To address this gap, we developed a unified bioinformatics framework that integrates transcriptomic profiling (UALCAN), drug-gene interactions (DGIdb), gene-disease associations (Open Targets), pathway enrichment (Enrichr), and protein-protein interaction networks (STRING/Cytoscape). This pipeline wa

GLUL drives metabolic reprogramming and confers docetaxel resistance in prostate cancer.

Docetaxel is a first-line chemotherapeutic agent for advanced and castration-resistant prostate cancer (CRPC), yet acquired resistance limits its long-term efficacy. Metabolic reprogramming has emerged as a central mechanism of therapeutic resistance; however, the metabolic determinants of docetaxel resistance remain incompletely defined. Here, we identify glutamate-ammonia ligase (GLUL), the key enzyme mediating de novo glutamine synthesis, as a critical regulator of docetaxel resistance. Integrated transcriptomic, metabolomic, and single-cell RNA sequencing analyses of clinical specimens rev

Elevated plasma cholesterol improves sepsis outcome by promoting hepatic metabolic reprogramming.

BACKGROUND: Sepsis is a life-threatening condition with high mortality and limited therapeutic options. This study investigated the association between plasma cholesterol levels and sepsis survival and explored the mechanisms by which elevated cholesterol confers protection. METHODS: We analyzed 2,787 sepsis patients from the MIMIC-IV database, comparing cholesterol levels between 28-day survivors and non-survivors and assessing mortality risk using multivariable Cox regression. To test causality, C57BL/6J mice were fed either a high-cholesterol diet (HCD) or a regular diet (RD) before cecal

Metabolic reprogramming in aortic diseases: insights from metabolomics and therapeutic opportunities.

Aortic diseases (aortic aneurysm, aortic dissection, atherosclerosis) represent a substantial clinical and economic burden due to the lack of effective early diagnostic tools and mechanism-based therapies. Metabolomics, the systematic study of low-molecular-weight metabolites, has emerged as a powerful approach for elucidating pathogenesis, screening candidate biomarkers, and discovering novel drug targets. Rather than merely cataloging metabolic perturbations, this review highlights how metabolic reprogramming, particularly involving amino acid pathways, mitochondrial dysfunction, and gut mic

Geometrically adaptive hydrogel attenuates osteoarthritis via metabolic reprogramming and defect sealing.

Effective treatment for osteoarthritis (OA) remains challenging because current therapies do not simultaneously restore biomechanical support and modulate the pathological intra-articular microenvironment. Here, we developed an injectable dual-network hydrogel that combines thermosensitive poly(N-isopropylacrylamide)-graft-gelatin (PNIPAAm-g-Gelatin) with a dynamic carboxymethyl chitosan/oxidized hyaluronic acid (CMCS/oxHA) network, enabling rapid in situ gelation, self-healing, and defect filling. Beyond providing structural support, the hydrogel created a biomimetic matrix environment that w

Microglial metabolic reprogramming in Alzheimer's disease: Pathways, mechanisms, and therapeutic implications.

In recent years, the immune metabolism of central nervous system cells has gained increasing attention from researchers. Microglia (MG) are innate immune cells of the central nervous system. They can metabolize a wide range of energy substrates. The pathways and products generated through these processes play a critical role in the onset and progression of Alzheimer's disease (AD). This paper provides a comprehensive review of metabolic reprogramming in MG during AD. It focuses on the three primary energy substrates: glucose, fatty acids, and amino acids. It delves deeply into the molecular si

Metabolic Reprogramming Supports Neonicotinoid Resistance in the Brown Planthopper, Nilaparvata lugens.

Metabolic resistance is commonly attributed to the overexpression of detoxification enzymes, whereas the metabolic systems that sustain detoxification remain less well resolved. Here, we integrated widely targeted metabolomics and transcriptomics to compare the clothianidin-resistant brown planthopper strain CLR with the susceptible strain CLS and validated candidate genes in an independent nitenpyram-resistant background. CLR and CLS exhibited distinct metabolomic profiles. Differential genes and metabolites converged on carbon metabolism, glycolysis/gluconeogenesis, the tricarboxylic acid cy

Methotrexate potentiates oxidative stress-mediated transdifferentiation and metabolic reprogramming of neutrophils.

Rheumatoid arthritis (RA) is associated with inflammation, oxidative stress and infiltration of immune cells (~ 90% neutrophils) into inflamed joints. Neutrophils have demonstrated the ability to transdifferentiate into 'neutrophil-dendritic cell hybrids' or N-DCs, acquire antigen presenting properties (HLA-DR/CD80/CD86) and demonstrate enhanced oxidative stress. Classical neutrophils rely mainly on glycolysis, but the bioenergetics of N-DCs remains poorly defined. Accordingly, this study aimed to assess the impact of methotrexate (MTX) on neutrophils with regard to their transdifferentiation

CD4+T cell metabolic reprogramming as therapeutic targets in neurodegenerative diseases.

Neurodegenerative diseases are a group of disorders characterized by the progressive loss of structure and function of neurons in the brain and/or peripheral nervous system. The main pathological feature of neurodegenerative disease in the central nervous system (CNS) is the selective neuronal loss in the brain and spinal cord, leading to cognitive and/or motor dysfunction. The immune system plays a variety of roles in the pathophysiology of neurodegenerative diseases. CD4+T cells are being recognized as important immunometabolic modulators in the pathophysiology of neurodegenerative disorders

Mitochondrial Energy Metabolic Reprogramming Facilitates the Malignant Progression of Intrahepatic Cholangiocarcinoma.

Mitochondrial function plays a crucial role in cancer development, with mitochondrial energy metabolism-related genes (MEMRGs) contributing to carcinogenesis. This study investigates the role of MEMRGs in intrahepatic cholangiocarcinoma (ICC) by analyzing RNA-seq data from TCGA and GEO databases to identify differentially expressed MEMRGs. Functional enrichment and KEGG pathway analyses revealed their significant involvement in metabolic pathways. Using weighted gene co-expression network analysis (WGCNA) and consensus clustering, two distinct ICC subtypes were identified. Tumor mutational bur

Metabolic reprogramming in ischemic stroke: when glycolytic overdrive meets lipid storm.

Ischemic stroke, a leading cause of global disability and mortality, remains inadequately treated beyond reperfusion, with persistent translational failures in neuroprotection. We posit metabolic reprogramming in ischemic stroke (MRIS) as the unifying pathophysiological driver, where acute compensatory glycolysis collides with enzymatic lipid peroxidation to ignite neuroinflammation and early deficits. This metabolic crisis transcends neuron-centric models, integrating single-cell heterogeneity with bidirectional brain-peripheral crosstalk: hepatic ketogenesis releases neuroprotective β-hydrox

Macrophage Metabolic Reprogramming in Inflammatory Bowel Diseases: From Pathogenesis to Therapy.

The Janus kinase 2 (JAK2) V617F mutation activates the transcription pathway and has been well characterized as a driver of myeloproliferative neoplasms (MPNs). Recently, there has been a heightened interest in understanding germ line predisposition to hematologic malignancies such as MPN, including several reports of familial MPN. Here, we retrospectively analyzed medical records and data from genetic testing to describe 12 patients with germ line variants at amino acid position 564 of JAK2. This includes 3 supportive cases adding to prior literature regarding the germ line JAK2 R564Q associa
