Role of type I interferon signaling in human metapneumovirus pathogenesis and control of viral replication.

UNLABELLED: Type I IFN signaling, which is initiated through activation of the alpha interferon receptor (IFNAR), regulates the expression of proteins that are crucial contributors to immune responses. Paramyxoviruses, including human metapneumovirus (HMPV), have evolved mechanisms to inhibit IFNAR signaling, but the specific contribution of IFNAR signaling to the control of HMPV replication, pathogenesis, and adaptive immunity is unknown. We used IFNAR-deficient (IFNAR(-/-)) mice to assess the effect of IFNAR signaling on HMPV replication and the CD8(+) T cell response. HMPV-infected IFNAR(-/

Human metapneumovirus viral load is an important risk factor for disease severity in young children.

BACKGROUND: The role of viral load in human metapneumovirus (HMPV) disease severity has not yet been clearly determined. OBJECTIVE: We evaluated the importance of viral load along with other factors in HMPV disease severity among children aged <3 years old. STUDY DESIGN: HMPV-positive cases were selected from a cohort of outpatients and hospitalized children with lower respiratory tract infections. HMPV groups (A or B) and viral loads were determined in their nasopharyngeal aspirates. Disease severity was defined by assessing risk for hospitalization and by using two validated clinical sever

Treatment of severe human metapneumovirus (hMPV) pneumonia in an immunocompromised child with oral ribavirin and IVIG.

BACKGROUND: The clinical manifestations of human metapneumovirus (hMPV) infection resemble those of respiratory syncytial virus with the most severe disease occurring in infants, the elderly, chronically ill, and immunocompromised hosts. OBSERVATION: We present a case of a 2-year-old girl undergoing intensive chemotherapy for Burkitt lymphoma who developed severe hMPV pneumonia. Rapid and complete recovery was observed after treatment with oral ribavirin and intravenous immunoglobulin. CONCLUSION: As hMPV can cause severe pneumonia in immunocompromised patients and due to the reports of effe

In situ Hybridization to Study the Pathogenicity of North American Avian Metapneumovirus Subtypes A and B in Turkeys.

Avian metapneumovirus (aMPV) is a highly contagious upper respiratory pathogen that causes significant economic losses in both chickens and turkeys. Globally, there are four antigenically distinct subgroups, A, B, C, and D, based on the sequence of glycoprotein G. Subgroups A and B emerged recently in the US, causing several outbreaks in chickens and turkeys, resulting in high morbidity and mortality rates, as well as a decline in egg production. aMPV primarily infects the upper respiratory tract and eyelids/conjunctiva. In-situ Hybridization (ISH) is a molecular tool that localizes specific n

The immune receptors TLR4 and SLAMF1 regulate TNF release by human metapneumovirus in human macrophages.

BACKGROUND: Human metapneumovirus (HMPV) is a major cause of acute respiratory disease in children, the elderly, and immunocompromised individuals. While pro-inflammatory cytokines and type I interferons (IFNs) are important for antiviral defense, excessive tumor necrosis factor (TNF) is associated with severe disease in HMPV and other respiratory infections. Hence, defining regulatory mechanisms by which HMPV induces TNF and IFN-β is important for therapeutic strategies in airway disease. The immunoregulatory receptors Toll-like receptor (TLR)4 and signaling lymphocytic activation molecule fa

Efficacy of a novel avian metapneumovirus live vaccine candidate based on vaccination route and age.

This article describes a series of animal studies for the development of an avian metapneumovirus (aMPV) live vaccine. Although aMPV causes continual economic loss in the poultry industry, there are no live aMPV vaccines available in Korea. Furthermore, information is limited with respect to standard field practices for vaccinations at an early age. Here, the development of an aMPV live vaccine was attempted, and its efficacy was investigated with respect to the vaccination route and age to develop a method for controlling aMPV. Before vaccine development, an animal challenge model was establi

Serosurvey of Avian metapneumovirus, Orithobacterium rhinotracheale, and Chlamydia psittaci and Their Potential Association with Avian Airsacculitis.

Seasonal outbreaks of airsacculitis in China's poultry cause great economic losses annually. This study tried to unveil the potential role of Avian metapneumovirus (AMPV), Ornithobacterium rhinotracheale (ORT) and Chlamydia psittaci (CPS) in avian airsacculitis. A serological investigation of 673 breeder chickens and a case-controlled study of 430 birds were undertaken. Results showed that infection with AMPV, ORT, and CPS was highly associated with the disease. The correlation between AMPV and CPS were positively robust in both layers and broilers. Finally, we determined the co-infection with

Severe acute respiratory distress syndrome associated with human metapneumovirus infection in a morbidly obese adult.

Human metapneumovirus (hMPV) is a recognized cause of respiratory tract infections and can occasionally lead to severe pneumonia and acute respiratory distress syndrome (ARDS) in adults. However, reports describing the clinical course and management of hMPV-associated ARDS in adults remain limited. We report a case of severe ARDS caused by hMPV infection in a morbidly obese adult who achieved a favorable outcome with standard ARDS management. A 58-year-old man with morbid obesity (body mass index 36.6 kg/m²) was admitted to a previous hospital with fever and cough. Due to rapidly progressive h

Protective efficacy of a novel recombinant influenza virus carrying partial human metapneumovirus F protein epitopes.

Human metapneumovirus (HMPV) is a leading cause of acute respiratory tract infections in infants and children. Currently, no approved HMPV vaccine is available. We developed a novel recombinant influenza virus, which carried partial HMPV F protein (HMPV-F) epitopes, utilizing reverse genetics. The novel single-stranded RNA virus, termed rFLU-HMPV/F-NA, was synthesized in the neuraminidase (NA) fragment of influenza virus A/PuertoRico/8/34 (PR8). The morphological characteristics of rFLU-HMPV/F-NA were consistent with the wild-type flu virus. The virus could passage in specific pathogen-free (S

Hospitalizations due to acute respiratory tract infections caused by human metapneumovirus in Costa Rican children.

BACKGROUND: Human metapneumovirus (hMPV) is a leading cause of acute respiratory infection in children, particularly early in life. Despite this, there are few prospective hMPV studies in Latin American children and this is the only of that kind in Costa Rica. METHODS: We conducted an observational descriptive prospective study at the only national tertiary referral pediatric hospital of the country. We included children < 13 years of age with a laboratory-confirmed hMPV episode of acute respiratory tract infection requiring hospitalization from September 1, 2015, to September 30, 2017. Diagn

Clinical significance of human metapneumovirus detection in critically ill adults with lower respiratory tract infections.

BACKGROUND: Unlike other viruses, the pathogenicity of human metapneumovirus (hMPV) in adults remains uncertain. To address this question, a retrospective monocentric cohort including all patients admitted to ICU with hMPV infection between January 1, 2010, and June 30, 2018 was performed. The characteristics of hMPV infected patients were studied and compared to matched influenza infected patients. Consecutively, a systematic review and meta-analyses investigating PUBMED, EMBASE and COCHRANE databases was conducted to explore the hMPV infections in adult patients (PROSPERO number: CRD42018106
