Mercury methylation in methanogenic archaea: A protocol for stabilized cultivation and accurate assessment.

Although methanogenic archaea are among the oldest microorganisms capable of mercury methylation, their contribution to methylmercury (MeHg) production has only recently gained attention. Studies with laboratory-cultivated methanogens elucidate the transformation of inorganic mercury (Hg) into MeHg, thereby uncovering underlying microbial methylation mechanisms. However, this field faces challenges such as significant Hg loss and unstable culture systems, which impede accurate assessment of these processes. This study aims to develop a reliable low-Hg-loss cultivation protocol for Hg methylati

Demethylation Pathway of Methylmercury in the Spleen and Peripheral Organs of Bluefin Tuna─Implications for Fish Consumers.

Atlantic bluefin tuna (ABFT, Thunnus thynnus) accumulates and breaks down methylmercury (MeHg) mainly in the spleen, with smaller amounts processed in kidneys and other peripheral organs, and only minimally in the liver. While the spleen is well-known as the body's largest immune organ, its role in detoxification is less recognized. Here, we used high-energy-resolution X-ray absorption spectroscopy to examine the chemical forms of mercury (Hg) in ABFT tissues, building on prior analysis of stable Hg isotopes. We found that Hg binds to selenium (Se) in 28 out of 29 tissues, primarily as Hg-tetr

Eight Unexpected Selenoprotein Families in Organometallic Biochemistry in Clostridium difficile, in ABC Transport, and in Methylmercury Biosynthesis.

The bioinformatics of a nine-gene locus, designated selenocysteine-assisted organometallic (SAO), was investigated after identifying six new selenoprotein families and constructing hidden Markov models (HMMs) that find and annotate members of those families. Four are selenoproteins in most SAO loci, including Clostridium difficile. They include two ABC transporter subunits, namely, permease SaoP, with selenocysteine (U) at the channel-gating position, and substrate-binding subunit SaoB. Cytosolic selenoproteins include SaoL, homologous to MerB organomercurial lyases from mercury resistance loc

Amendments of nitrogen and sulfur mitigate carbon-promoting effect on microbial mercury methylation in paddy soils.

The imbalance of nutrient elements in paddy soil could affect biogeochemical processes; however, how the key elements input influence microbially-driven conversion of mercury (Hg) to neurotoxic methylmercury (MeHg) remains virtually unknown. Herein, we conducted a series of microcosm experiments to explore the effects of certain species of carbon (C), nitrogen (N) and sulfur (S) on microbial MeHg production in two typical paddy soils (yellow and black soil). Results showed that the addition of C alone into the soils increased MeHg production approximately 2-13 times in the yellow and black soi

MeHg production in eutrophic lakes: Focusing on the roles of algal organic matter and iron-sulfur-phosphorus dynamics.

The mechanisms by which eutrophication affects methylmercury (MeHg) production have not been comprehensively summarized, which hinders accurately predicting the MeHg risk in eutrophic lakes. In this review, we first discussed the effects of eutrophication on biogeochemical cycle of mercury (Hg). Special attentions were paid to the roles of algal organic matter (AOM) and iron (Fe)-sulfur (S)-phosphorus (P) dynamics in MeHg production. Finally, the suggestions for risk control of MeHg in eutrophic lakes were proposed. AOM can affect in situ Hg methylation by stimulating the abundance and activit

Sublethal effects of methylmercury on lateral line sensory and ion-regulatory functions in zebrafish embryos.

Methylmercury can interfere with the normal functioning of the nervous system, causing a variety of behavioral and physiological changes in fish. However, the influence of MeHg on the lateral line sensory and ion-regulatory functions of fish is not clear. Zebrafish embryos were utilized as a model to address this question. After exposure to water-borne MeHg (5, 10, 50, or 100 ppb) for 96 h (4-100 h post-fertilization), the survival rate declined by ca. 50 % at 100 ppb. However, MeHg at sublethal concentrations delayed hatching and decreased heart rates and body length. As to effects on the lat

A SYBR Green I-based aptasensor for the label-free, fluorometric, and anti-interference detection of MeHg.

Methylmercury (MeHg+) is a common form of organic mercury that is substantially more toxic than inorganic mercury and is more likely to accumulate in organisms through biological enrichment. Therefore, developing a method to enable the specific and rapid detection of MeHg+ in seafood is important and remains challenging to accomplish. Herein, a rapid, label-free fluorescence detection method for MeHg+ determination was developed based on SYBR Green I. The detection system implemented "add and measure" detection mode can be completed in 10 min. Under optimal assay conditions, the detection plat

Advances on the Influence of Methylmercury Exposure during Neurodevelopment.

Mercury (Hg) is a toxic heavy-metal element, which can be enriched in fauna and flora and transformed into methylmercury (MeHg). MeHg is a widely distributed environmental pollutant that may be harmful to fish-eating populations through enrichment of aquatic food chains. The central nervous system is a primary target of MeHg. Embryos and infants are more sensitive to MeHg, and exposure to MeHg during gestational feeding can significantly impair the homeostasis of offspring, leading to long-term neurodevelopmental defects. At present, MeHg-induced neurodevelopmental toxicity has become a hotspo

Assessing the factors impacting the bioaccessibility of mercury (Hg) in rice consumption by an in-vitro method.

Mercury (Hg) in rice is drawing mounting concern since methylmercury (MeHg) was found capable of accumulating in rice. In-vitro bioaccessibility is a feasible and reliable method to assess the health effects of Hg in rice and has been utilized in a number of studies. This study was done to investigate the impact of cultivar, planting location, and cooking on the total mercury (THg) and MeHg bioaccessibility of rice, for which multiple statistical analysis methods were used to analyze the significance of their effects. The THg concentrations of rice samples taken from non-Hg contaminated areas

Distribution and phylogeny of mercury methylation, demethylation, and reduction genes in the Seto Inland Sea of Japan.

Mercury (Hg) adversely affects human and environmental health. To evaluate the mercury (Hg) speciation (methylation, demethylation, and reduction) of microorganisms in coastal seawater, we analyzed the microbial functional gene sets involved in Hg methylation (hgcA and hgcB), demethylation (merB), and reduction (merA) using a metagenomic approach in the eastern and western parts (the Kii and Bungo channels, respectively) of the Seto Inland Sea (SIS) of Japan. We determined the concentration of dissolved total mercury (dTHg) and methylated mercury (dMeHg) in seawater. The metagenomic analysis d

Demethylation of Methylmercury in Bird, Fish, and Earthworm.

Toxicity of methylmercury (MeHg) to wildlife and humans results from its binding to cysteine residues of proteins, forming MeHg-cysteinate (MeHgCys) complexes that hinder biological functions. MeHgCys complexes can be detoxified in vivo, yet how this occurs is unknown. We report that MeHgCys complexes are transformed into selenocysteinate [Hg(Sec)4] complexes in multiple animals from two phyla (a waterbird, freshwater fish, and earthworms) sampled in different geographical areas and contaminated by different Hg sources. In addition, high energy-resolution X-ray absorption spectroscopy (HR-XANE

Three-Dimensional Printed Dual-Mode Chemical Vapor Generation Point Discharge Optical Emission Spectrometer for Field Speciation Analyses of Mercury and Inorganic Selenium.

Due to the large size and high energy consumption of instruments, field elemental speciation analysis is still challenging so far. In this work, a portable and compact system device (230 mm length × 38 mm width × 84 mm height) was fabricated by using three-dimensional (3D) printing technology for the field speciation analyses of mercury and inorganic selenium. The device comprises a cold vapor generator, photochemical vapor generator, and miniaturized point discharge optical emission spectrometer (μPD-OES). For mercury, inorganic mercury (IHg) was selectively reduced to Hg0 by cold vapor gener

Identification of mercury-containing nanoparticles in the liver and muscle of cetaceans.

Natural mercury-containing nanoparticles (Hg-NPs) have been found in the environment, but the information for Hg-NPs in organisms was still limited. Clarifying the unique roles of Hg-NPs in organisms is crucial to fully understand the health risks of Hg. Herein, liver and muscle tissues of cetaceans were collected to identify the presence and characteristics of Hg-NPs. We found that methylmercury (MeHg) was the dominant species of Hg in muscles, while inorganic Hg (IHg) accounted for 84.4-99.0% (average 94.0%) of Hg in livers. By using transmission electron microscopy (TEM), size exclusion chr

Mitigating the impact of mercury contaminants in fish and other seafood-A review.

Mercury (Hg) is a well-known toxicant which enters the marine environment by both natural and anthropogenic sources. Consumption of fish and other seafood that contain methylmercury (MeHg) is a leading source of Hg exposure in humans. Considerable efforts have been made to mitigate the Hg presence and reduce its risks to humans. In this review the acknowledged methods of mitigation are summarized such as regulation and maximum allowable limits, and culinary treatments. In addition, selected industrial level trials are reviewed, and studies on Hg intoxication and the protective effects of the e

Application of biochar prepared from ethanol refinery by-products for Hg stabilization in floodplain soil: Impacts of drying and rewetting.

This study evaluated three biochars derived from bioenergy by-products - manure-based anaerobic digestate (DIG), distillers' grains (DIS), and a mixture thereof (75G25S) - as amendments to stabilize Hg in contaminated floodplain soil under long-term saturated (up to 200 d) and cyclic drying and rewetting conditions. Greater total Hg (THg) removal (72 to nearly 100%) and limited MeHg production (<65 ng L-1) were observed in digestate-based biochar-amended systems under initial saturated conditions. Drying and rewetting resulted in limited THg release, increased aqueous MeHg, and decreased solid

Mercury Methylation Genes Identified across Diverse Anaerobic Microbial Guilds in a Eutrophic Sulfate-Enriched Lake.

Mercury (Hg) methylation is a microbially mediated process that converts inorganic Hg into bioaccumulative, neurotoxic methylmercury (MeHg). The metabolic activity of methylating organisms is highly dependent on biogeochemical conditions, which subsequently influences MeHg production. However, our understanding of the ecophysiology of methylators in natural ecosystems is still limited. Here, we identified potential locations of MeHg production in the anoxic, sulfidic hypolimnion of a freshwater lake. At these sites, we used shotgun metagenomics to characterize microorganisms with the Hg-methyl

Geochemical Factors Controlling Dissolved Elemental Mercury and Methylmercury Formation in Alaskan Wetlands of Varying Trophic Status.

The transformations of aqueous inorganic divalent mercury (Hg(II)i) to volatile dissolved gaseous mercury (Hg(0)(aq)) and toxic methylmercury (MeHg) govern mercury bioavailability and fate in northern ecosystems. This study quantified concentrations of aqueous mercury species (Hg(II)i, Hg(0)(aq), MeHg) and relevant geochemical constituents in pore waters of eight Alaskan wetlands that differ in trophic status (i.e., bog-to-fen gradient) to gain insight on processes controlling dark Hg(II)i reduction and Hg(II)i methylation. Regardless of wetland trophic status, positive correlations were obser

Review of stable mercury isotopes in ecology and biogeochemistry.

Due to the advent of cold vapor-multicollector-inductively coupled plasma mass spectrometry (CV-MC-ICP-MS) in the past two decades, many research groups studying mercury (Hg) biogeochemistry have integrated stable Hg isotopes into their research. Currently, >200 studies using this technique have been published and this has greatly enhanced our understanding of the Hg biogeochemical cycle beyond what Hg concentration and speciation analyses alone can provide. These studies are largely divided into two groups: (i) controlled experiments investigating fractionation of Hg isotopes and refining too

Methylmercury and inorganic mercury in Chinese commercial rice: Implications for overestimated human exposure and health risk.

China is the largest rice producer and consumer in the world, and mercury (Hg) levels, particularly methylmercury (MeHg), in rice and health exposure risks are public concerns. Total Hg (THg) and MeHg levels in 767 (domestic = 709 and abroad = 58) Chinese commercial rice were investigated to evaluate Hg pollution level, dietary exposures and risks of IHg and MeHg. The mean rice THg and MeHg levels were 3.97 ± 2.33 μg/kg and 1.37 ± 1.18 μg/kg, respectively. The highest daily intake of MeHg and IHg were obtained in younger groups, accounted for 6% of the reference dose-0.1 μg/kg bw/day for MeHg,
