Zinc-dependent modulation of ceftriaxone killing in tolerant Neisseria gonorrhoeae.

Ceftriaxone tolerance in Neisseria gonorrhoeae is increasingly recognized as a phenotype that allows prolonged bacterial survival at inhibitory drug concentrations without altering minimum inhibitory concentrations (MICs). Host environmental factors are likely to influence such tolerance phenotypes, but experimental evidence remains limited. Zinc is a host-relevant transition metal that shapes bacterial stress responses and ribosomal function. Transcriptomic analyses have previously linked ceftriaxone tolerance in N. gonorrhoeae to a zinc-sparing ribosomal programme. In this study, we examined

Immortal genome assumption significantly underestimates replication and death rates of Mycobacterium tuberculosis in mice and monkeys.

UNLABELLED: Immune correlates of protection against infection with Mycobacterium tuberculosis (Mtb) or against tuberculosis (TB) remain poorly defined. The ratio of colony-forming units (CFUs) to chromosomal equivalents (CEQs), [Formula: see text], recovered either from the whole lung (mice) or individual lesions (monkeys or rabbits) of Mtb-infected animals has been used as a metric for how effectively Mtb is killed in vivo. However, the contribution of bacterial killing to changes in the CFU/CEQ ratio during an infection has not been rigorously investigated. We developed alternative mathemati

Examining the viability of five Salmonella enterica subsp. enterica in thymol at 4°C and 25°C using flow cytometry.

Salmonella spp., a major cause of foodborne illness, requires effective control strategies to improve food safety. Thymol, an antibacterial agent derived from natural essential oils, has been assessed for use as an antimicrobial agent and preservative in the food industry, due to its safety and low cost. This study used flow cytometry and Tryptic Soy Agar (TSA) plate counts, to assess the viability of five serotypes of Salmonella enterica subsp. enterica over 56 days in thymol at 4°C and 25°C, during long-term storage in distilled water. The minimum inhibitory concentration (MIC) of thymol aga

Antibacterial activity of diazepam against planktonic and biofilm strains of methicillin-resistant Staphylococcus aureus.

The aim was to evaluate the antibacterial activity of diazepam against methicillin-susceptible (MSSA) and methicillin-resistant (MRSA) strains of Staphylococcus aureus and its possible mechanism of action. The broth microdilution assay was used to determine the minimum inhibitory concentration (MIC) of diazepam. A checkerboard assay was used to evaluate the interaction of diazepam with different antibiotics. Colorimetric assays with MTT were used to evaluate the effect of diazepam against the biofilms by MSSA and MRSA. Flow cytometry and fluorescence microscopy were used to evaluate the possib

Tetracyclines at subinhibitory concentrations are lethal for NADH peroxidase-deficient mutants of Enterococcus faecium.

OBJECTIVES: Tigecycline is a bacteriostatic antibiotic member of the glycylcycline family that inhibits protein synthesis. Tigecycline is a last-line treatment for infections caused by MDR pathogens like vancomycin-resistant Enterococcus faecium (VR-Efm). We recently explored oxidative stress defences in E. faecium and we here aimed to assess their role in antibiotic resistance. METHODS: Antibiotic susceptibility was evaluated in mutants deficient in primary oxidative stress defences by monitoring bacterial survival after a 24 h treatment. Hydrogen peroxide (H2O2) levels were quantified to li

Cocktail Approach with Polyserotonin Nanoparticles and Peptides for Treatment of Streptococcus mutans.

Dental plaque, formed by a Streptococcus mutans biofilm, is a major contributor to cavity formation. While antimicrobial strategies exist, the growing risk of antibiotic resistance necessitates alternative therapeutic solutions. Polyserotonin nanoparticles (PSeNPs), recently recognized for their photothermal property and promising biomedical applications, open up a new avenue for antimicrobial use. Here, we introduced a UV-initiated synthetic route for PSeNPs with improved yield. Using these PSeNPs, a cocktail treatment to reduce the viability of this cavity-causing bacteria was developed. Thi

Microbial Inactivation Property of Pulsed Corona Discharge Plasma and Its Effect on Chilled Pork Preservation.

Although plasma, especially atmospheric plasma generated by corona discharge, has been proven to be effective in sterilization and food preservation, its disinfection mechanism on chilled pork is poorly understood. In this research, the bactericidal and preservation effect of corona discharge plasma (CDP) was investigated. The maximum bactericidal effect was found after 20 kV 4 kHz CDP treatment, with 2.77 log (colony-forming unit [CFU]/g), 2.41 log (CFU/g), and 1.36 log (CFU/g) reduction for Pantoea agglomerans, Serratia liquefaciens, and Kurthia zopfii, respectively, after 10 min of exposure

Chitosan disrupts biofilm formation and promotes biofilm eradication in Staphylococcus species isolated from bovine mastitis.

Staphylococci are the main pathogens associated with hard-to-control intramammary infections in dairy cattle, and bacterial biofilms are suspected to be responsible for the antimicrobial resistance and persistence of this disease. Biofilms have the ability to resist to higher levels of antibiotics and reduce their efficacy. It is thus necessary to develop strategies targeted to bacterial biofilm infections. Chitosan is a polysaccharide with a proven broad spectrum of antimicrobial activity against fungi and bacteria. The aim of this study was assess the effect of low molecular weight (LMW) chi

5-Fluorouracil sensitivity varies among oral micro-organisms.

5-Fluorouracil (5-FU), a commonly used chemotherapeutic agent, often causes oral mucositis, an inflammation and ulceration of the oral mucosa. Micro-organisms in the oral cavity are thought to play an important role in the aggravation and severity of mucositis, but the mechanisms behind this remain unclear. Although 5-FU has been shown to elicit antibacterial effects at high concentrations (>100 µM), its antibacterial effect at physiologically relevant concentrations in the oral cavity is unknown. This study reports the effect of different concentrations of 5-FU (range 0.1-50 µM) on the growth

Enhancement of the antibiofilm activity of amphotericin B by polyamine biosynthesis inhibitors.

The aim of this study was to investigate the effect of polyamine biosynthesis inhibitors on the activity of amphotericin B (AmB) against Candida albicans biofilms and to clarify the underlying mechanisms. The antibiofilm activity of AmB was significantly enhanced when used in combination with the polyamine biosynthesis inhibitors 1,4-diamino-2-butanone (DAB) and α-difluoromethylornithine (DFMO). Further study showed that DAB and DFMO also enhanced the antibiofilm activity of several other antifungal agents. Moreover, the combination of AmB and polyamine biosynthesis inhibitors resulted in an i

In vitro inhibitory activity of terpenic derivatives against clinical and environmental strains of the Sporothrix schenkii complex.

Sporotrichosis is a subacute or chronic subcutaneous infection, caused by the fungus Sporothrix schenkii complex, occurring in human and animal tissues. Potassium iodide and itraconazole have been used as effective therapy for first-choice treatment, while amphotericin B may be indicated for disseminated infection. However, the adverse effects of potassium iodide and amphotericin B or the long duration of therapy with itraconazole often weigh against their use, leading to the search for alternatives for the treatment of severe infections. Terpinen-4-ol and farnesol are components of essential

Synergistic activity of phenazines isolated from Pseudomonas aeruginosa in combination with azoles against Candida species.

Candidiasis infections are caused by yeasts from the genus Candida. The types of infection range from superficial to systemic. Treatment often requires antifungals such as the azoles; however, increased use of these drugs has led to the generation of yeasts with increased resistance to these drugs. Here, we describe the synergistic anticandidal activity of three phenazines-phenazine-1-ol, phenazine-1-carboxylic acid, and phenazine-1-carboxamide. These phenazines were purified from Pseudomonas aeruginosa in combination with three clinically used azoles-fluconazole, itraconazole, and clotrimazol

Increased persistence of Salmonella enterica serovar Typhi in the presence of Acanthamoeba castellanii.

Salmonella enterica serovar Typhi (S. Typhi) is the etiological agent of the systemic disease typhoid fever. Transmission occurs via ingestion of contaminated food or water. S. Typhi is specific to humans, and no animal or environmental reservoirs are known. As the free-living amoeba Acanthamoeba castellanii is an environmental host for many pathogenic bacteria, this study investigates interactions between S. Typhi and A. castellanii by using cocultures. Growth of both organisms was estimated by cell count, viable count, flow cytometry, and fluorescence microscopy. Results indicate that S. Typ

In vitro synergy of pseudolaric acid B and fluconazole against clinical isolates of Candida albicans.

Candida albicans is the most common fungal pathogen in humans. The emergence of resistance to azole antifungals has raised the issue of using such antifungals in combination to optimise therapeutic outcome. The objective of this study was to evaluate in vitro synergy of pseudolaric acid B (PAB) and fluconazole (FLC) against clinical isolates of C. albicans. The in vitro antifungal activity of PAB, a diterpene acid from Pseudolarix kaempferi Gordon, was evaluated alone and in combination with FLC against 22 FLC-resistant (FLC-R) and 12 FLC-susceptible (FLC-S) C. albicans using the chequerboard

A comparison of the fungicidal activity of amphotericin B and posaconazole against Zygomycetes in vitro.

Zygomycoses is a rapidly progressive infection associated with high mortality. Although amphotericin B (AMB) has been the only treatment option for years, recent studies have demonstrated that posaconazole (PCZ) has good activity against Zygomycetes. Because rapid onset of antifungal activity is crucial in the management and never studied before, we compared the time for maximum fungicidal activity of AMB versus PCZ using time-kill curves. The MIC of AMB and PCZ against clinical isolates of Mucor spp. and Rhizopus spp. was determined by the Clinical and Laboratory Standards Institute M38-A2 me

Competition of Listeria monocytogenes serotype 1/2a and 4b strains in mixed-culture biofilms.

The majority of Listeria monocytogenes isolates recovered from foods and the environment are strains of serogroup 1/2, especially serotypes 1/2a and 1/2b. However, serotype 4b strains cause the majority of human listeriosis outbreaks. Our investigation of L. monocytogenes biofilms used a simulated food-processing system that consisted of repeated cycles of growth, sanitation treatment, and starvation to determine the competitive fitness of strains of serotypes 1/2a and 4b in pure and mixed-culture biofilms. Selective enumeration of strains of a certain serotype in mixed-culture biofilms on sta
