Hydrogen sulfide triggered molecular agent for imaging and cancer therapy.

We developed an activatable molecular reagent, PNF, triggered by intracellular H2S in the lysosome to release the therapeutic drug amonafide, which can escape from the lysosome into the nucleus to induce autophagy of cancer cells. PNF exhibits potent inhibitory activity against cisplatin-resistant tumor cell lines.

Collagen-Targeted Peptides for Molecular Imaging of Diffuse Cardiac Fibrosis.

Background Cardiac fibrosis is the excessive deposition of extracellular matrix in the heart, triggered by a cardiac insult, aging, genetics, or environmental factors. Molecular imaging of the cardiac extracellular matrix with targeted probes could improve diagnosis and treatment of heart disease. However, although this technology has been used to demonstrate focal scarring arising from myocardial infarction, its capacity to demonstrate extracellular matrix expansion and diffuse cardiac fibrosis has not been assessed. Methods and Results Here, we report the use of collagen-targeted peptides la

Molecular Imaging for Evaluation of Viable Testicular Cancer Nodal Metastases.

PURPOSE OF REVIEW: Determining the metastatic viability of suspicious retroperitoneal nodes in testicular cancer with conventional imaging is challenging. The aim of this report is to review recent evidence in the utilization of novel imaging modalities to assess viable testicular cancer nodal metastases. RECENT EVIDENCE: Testicular germ cell tumors (TCGTs) follow a predictable lymphatic metastatic spread to the retroperitoneum. Accordingly, retroperitoneal imaging is critical in staging, assessing treatment response, and evaluating for recurrence. Conventional computed tomography (CT) imagin

PBCA-based polymeric microbubbles for molecular imaging and drug delivery.

Microbubbles (MB) are routinely used as contrast agents for ultrasound (US) imaging. We describe different types of targeted and drug-loaded poly(n-butyl cyanoacrylate) (PBCA) MB, and demonstrate their suitability for multiple biomedical applications, including molecular US imaging and US-mediated drug delivery. Molecular imaging of angiogenic tumor blood vessels and inflamed atherosclerotic endothelium is performed by modifying the surface of PBCA MB with peptides and antibodies recognizing E-selectin and VCAM-1. Stable and inertial cavitation of PBCA MB enables sonoporation and permeabilizat

Novel Molecular Imaging Approaches to Abdominal Aortic Aneurysm Risk Stratification.

Selection of patients for abdominal aortic aneurysm repair is currently based on aneurysm size, growth rate, and symptoms. Molecular imaging of biological processes associated with aneurysm growth and rupture, for example, inflammation and matrix remodeling, could improve patient risk stratification and lead to a reduction in abdominal aortic aneurysm morbidity and mortality. (18)F-fluorodeoxyglucose-positron emission tomography and ultrasmall superparamagnetic particles of iron oxide magnetic resonance imaging are 2 novel approaches to abdominal aortic aneurysm imaging evaluated in clinical t

Molecular Imaging and Precision Medicine in Dementia and Movement Disorders.

Precision medicine (PM) has been defined as "prevention and treatment strategies that take individual variability into account." Molecular imaging (MI) is an ideally suited tool for PM approaches to neurodegenerative dementia and movement disorders (MD). Here we review PM approaches and discuss how they may be applied to other associated neurodegenerative dementia and MD. With ongoing major therapeutic research initiatives that include the use of molecular imaging, we look forward to established interventions targeted to specific molecular pathophysiology and expect the potential benefit of MI

Alternative Non-Antibody Protein Scaffolds for Molecular Imaging of Cancer.

The development of improved methods for early detection and characterization of cancer presents a major clinical challenge. One approach that has shown excellent potential in preclinical and clinical evaluation is molecular imaging with small-scaffold, non-antibody based, engineered proteins. These novel diagnostic agents produce high contrast images due to their fast clearance from the bloodstream and healthy tissues, can be evolved to bind a multitude of cancer biomarkers, and are easily functionalized by site-specific bioconjugation methods. Several small protein scaffolds have been verifie

Imaging signatures of molecular pathology in behavioral variant frontotemporal dementia.

Pathology underlying behavioral variant frontotemporal dementia (bvFTD) is heterogeneous, with the most common pathologies being Pick's disease (PiD), corticobasal degeneration (CBD), and FTLD-TDP type 1. Clinical features are unhelpful in differentiating these pathologies. We aimed to determine whether imaging atrophy patterns differ across these pathologies in bvFTD subjects. We identified 15 bvFTD subjects that had volumetric MRI during life and autopsy: five with PiD, five CBD, and five FTLD-TDP type 1. Voxel-based morphometry was used to assess atrophy patterns in each bvFTD group compare

Molecular imaging of Bcr-Abl phosphokinase in a xenograft model.

The purpose of this study was to determine whether the Bcr-Abl tyrosine kinase can be assessed by gamma-imaging using an 111In-labeled anti-phosphotyrosine (APT) antibody, and if the response to treatment with imatinib could be detected using this imaging technique. APT antibody was labeled with 111In using ethylenedicysteine (EC) as a chelator. To determine if 111In-EC-APT could assess a nonreceptor tyrosine kinase, xenografts of the human chronic myelogenous leukemia cell line K562 were used. gamma-Scintigraphy of the tumor-bearing mice, before and after imatinib treatment, was obtained 1, 2

Clinical feasibility of molecular imaging of plaque inflammation in atherosclerosis.

Despite substantial advances in the diagnosis and management of coronary artery disease, acute coronary events continue to occur in many patients. It has been increasingly realized that the lesions responsible for acute events may not necessarily be critically obstructive and hence not be associated with inducible ischemia. Various morphologic features of plaque vulnerability have been described by CT angiography, intravascular ultrasound, and optical coherence tomography. The culprit plaques often demonstrate large plaque and necrotic core volumes, positive vascular remodeling, and attenuatio

Molecular MR imaging and MR-guided ultrasound therapies in cancer.

Imaging in cancer has moved in the last twenty years from morphological detection of diseases to characterization and categorization of different subtypes of tumors. Functional information, based on dynamic contrast-enhanced imaging of tissue perfusion and evaluation of water diffusion, tissue oxygenation, capillary permeability or lymphatic drainage, plays a major role in that field. The next coming steps will concern the differentiation of biological behaviour of tumors according to their phenotypes by identifying specific surface receptors or products of synthesis. These developments allowing

Molecular imaging of neurodegeneration by a novel cross-disease biomarker.

Current pre-mortem diagnosis of neurodegenerative disorders such as Alzheimer's disease (AD) or amyotrophic lateral sclerosis (ALS) is based on clinical assessment of neurological deficits. However, symptoms and signs emerge only late in the disease course, thus indicating an urgent need for novel tools for detection of the underlying neuropathology. NST-729 (MW=310) is a novel molecular imaging probe, which is a member of the ApoSense family of small molecule detectors of apoptosis. We now report on the ability of NST-729, upon its systemic administration in vivo, to detect characteristic neu

MR molecular imaging of early endothelial activation in focal ischemia.

Focal ischemia followed by reperfusion initiates a harmful P- and E-selectin-mediated recruitment of leukocytes in brain microvasculature. In this study, we tested whether a novel magnetic resonance (MR) contrast agent (Gd-DTPA-sLe(x) A), which is designed to bind to activated endothelium could be detected by MR imaging (MRI) in a focal stroke mouse model. MRIs (9.4T) of the brain were acquired 24 hours after transient middle cerebral artery occlusion. T1 maps were acquired repeatedly before and up to 1.5 hours after the intravenous injection of either Gd-DTPA or Gd-DTPA-sLe(x) A. Analysis of

Toward a Structured Imaging Reporting System for Kidney Masses: The Molecular Imaging Perspective on KI-RADS.

Recently, a structured approach to renal mass characterization known as the Kidney Imaging Reporting and Data System (KI-RADS) was proposed. In that proposal, the authors noted the need for imaging characteristics to be rooted in the biological underpinnings of the tumors in order to optimally risk stratify patients with indeterminate renal masses. Although multi-parametric magnetic resonance imaging may be an important initial modality for locoregional staging and lesion characterization, molecular imaging may provide more fundamental biological insight. To date, the mitochondrial imaging age

Magnetic resonance imaging in Parkinson's disease: from routine imaging to molecular biomarkers - an update for radiologists.

Magnetic resonance imaging (MRI) is becoming an increasingly important tool in the diagnostic approach to Parkinson's disease (PD), extending its role beyond the exclusion of secondary causes of parkinsonism. Recent developments in high-resolution imaging techniques - including neuromelanin-sensitive MRI, susceptibility-weighted imaging, quantitative susceptibility mapping, diffusion tensor imaging, and functional MRI - have enabled in vivo assessment of substantia nigra degeneration, iron accumulation, and functional connectivity alterations. These imaging findings correspond to core pathophy

Strategies for the Molecular Imaging of Amyloid and the Value of a Multimodal Approach.

Protein aggregation has been widely implicated in neurodegenerative diseases such as Alzheimer's disease, frontotemporal dementia, Parkinson's disease, and Huntington disease, as well as in systemic amyloidoses and conditions associated with localized amyloid deposits, such as type-II diabetes. The pressing need for a better understanding of the factors governing protein assembly has driven research for the development of molecular sensors for amyloidogenic proteins. To date, a number of sensors have been developed that report on the presence of protein aggregates utilizing various modalities,
