Investigating the behavioral impact of tropical almond (Terminalia catappa L.) consumption: Anxiolytic effects and memory preservation in aged rats.

Terminalia catappa L. (TC) has been traditionally used for its medicinal properties, including antioxidant and neuroprotective effects. Considering the increased vulnerability to cognitive decline and anxiety in aging, we investigated the potential impact of TC on anxiety-like behavior, memory, and oxidative stress in aged Wistar rats. Male Wistar rats were used, including young adults (70 days old) and aged animals (17 months old). The aged rats were allocated into three groups: control (n = 11), TC500 (n = 12), and TC1000 (n = 12). TC was administered by oral gavage at doses of 500 mg/kg or

Effects of aging on behavioral assessment performance: implications for clinically relevant models of neurological disease.

OBJECT: Despite the role of aging in development of neurological and neurodegenerative diseases, the effects of age are often disregarded in experimental design of preclinical studies. Functional assessment increases the clinical relevance of animal models of neurological disease and adds value beyond traditional histological measures. However, the relationship between age and functional impairment has not been systematically assessed through a battery of functional tests. METHODS: In this study, various sensorimotor and behavioral tests were used to evaluate effects of aging on functional pe

Electroacupuncture protected pyramidal cells in hippocampal CA1 region of vascular dementia rats by inhibiting the expression of p53 and Noxa.

AIM: Clinically electroacupuncture (EA) is proved an effective therapy for vascular dementia (VD), but their mechanisms remain uncertain. The aim of this study was to determine whether EA protects pyramidal cells from apoptosis in hippocampus of a VD rat model by inhibiting the expression of p53 and Noxa. METHODS: One month after a VD animal model was established by bilateral occlusion of common carotid arteries, EA treatment was given at "Baihui" (DU20), "Dazhui" (DU14), and "Shenshu" (BL23). The learning and memory ability was assessed by Morris water maze. Neuronal apoptosis in hippocampus

Synergistic Neuroprotective Effects of Traditional Chinese Medicine and Levodopa in Parkinson's Disease: A Behavioral and Pharmacokinetic Study.

Parkinson's disease (PD) is a complex neurological disorder that causes debilitating motor symptoms such as tremors, rigidity, and bradykinesia. TCM formulations, such as Zhengan Xifeng Decoction (ZXFD) in combination with Western medicine (Levodopa), are being examined for Parkinson's disease treatment. However, the absence of reliable and interpretable experimental models has hampered mechanism elucidation. This study evaluated the neuroprotective effects of ZXFD in a rat model of PD, assessing its efficacy and safety when combined with Western medicine (WM). 240 mice with MPTP-induced PD-li

Bexarotene Impairs Cognition and Produces Hypothyroidism in a Mouse Model of Down Syndrome and Alzheimer's Disease.

All individuals with Down syndrome (DS) eventually develop Alzheimer's disease (AD) neuropathology, including neurodegeneration, increases in β-amyloid (Aβ) expression, and aggregation and neurofibrillary tangles, between the third and fourth decade of their lives. There is currently no effective treatment to prevent AD neuropathology and the associated cognitive degeneration in DS patients. Due to evidence that the accumulation of Aβ aggregates in the brain produces the neurodegenerative cascade characteristic of AD, many strategies which promote the clearance of Aβ peptides have been assesse

Neonatal lesion of forebrain cholinergic neurons: further characterization of behavioral effects and permanency.

Intraventricular injections of 192 IgG-saporin in the neonatal rat caused severe loss of basal forebrain cholinergic neurons and ectopic hippocampal ingrowths. These were evident at 24 months of age and thus, were lifelong consequences of the 192 IgG-saporin treatment. When tested as young adults on a novel water-escape radial arm maze, the rats with this lesion were slower to learn the task, committing significantly more working and reference memory errors before they achieved control level of performance. It is unlikely that this was a result of attentional impairment as the lesioned rats pe

Protective effects of curcumin and resveratrol on neurodegeneration and cognitive dysfunction in a streptozotocin-induced alzheimer rat model.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder that is characterized by memory loss, cognitive decline, and behavioral disturbances. Given the limited efficacy of current treatments, there is an urgent need for new neuroprotective strategies. In this study, the therapeutic potential of curcumin and resveratrol was evaluated in a rat AD model induced by intracerebroventricular (ICV) administration of streptozotocin (STZ) (3 mg/kg). Subsequent to model induction, rats were administered curcumin, resveratrol, or a combination of both. The behavioral analyses, which included

Zhuyu Annao decoction promotes angiogenesis in mice with cerebral hemorrhage by inhibiting the activity of PHD3.

Some traditional Chinese decoctions, such as Zhuyu Annao, exert favorable therapeutic effects on acute cerebral hemorrhage, hemorrhagic stroke, and other neurological diseases, but the underlying mechanism remains unclear. This study aimed to determine whether Zhuyu Annao decoction (ZYAND) protects the injured brain by promoting angiogenesis following intracerebral hemorrhage (ICH) and elucidate its specific mechanism. The effect of ZYAND on the nervous system of mice after ICH was explored through behavioral experiments, such as the Morris water maze and Rotarod tests, and its effects on oxid

Isoflurane-delayed preconditioning reduces immediate mortality and improves striatal function in adult mice after neonatal hypoxia-ischemia.

BACKGROUND: Exposure to hypoxia and isoflurane (Iso) before hypoxia-ischemia has been found to be neuroprotective in neonatal rats. We investigated the long-term effects of delayed preconditioning with Iso, hypoxia, or room air on motor and cognitive function in mice that had 65 min of hypoxia-ischemia on postnatal day 10. METHODS: Nine-day-old C57x129T2 F1 mice received either 1.8% Iso, hypoxic (10% O2 in N2), or sham (room air) preconditioning. The following day, the mice were subjected to permanent right common carotid ligation or sham ligation followed by 65 min of hypoxia, or room air. A

Increased expression of WNK3 during the perinatal period in newborn rats with hypoxic-ischemic encephalopathy.

OBJECTIVES: Neonatal hypoxic-ischemic encephalopathy (HIE) is brain damage caused by reduced blood/oxygen supply during the perinatal period. There is no adequate treatment currently. The kinase WNK3 is associated with cerebral edema and stroke prognosis, so we assessed its expression in a neonatal rat model of HIE. METHODS: The Rice method was used to induce HIE in 7-day-old rat pups by ligating the left carotid artery followed by hypoxia exposure. Rats were divided into sham, 6 h, 12 h, 24 h, and 48 h groups (n = 5 each). Neurological function was evaluated by negative geotaxis, righting re

Brain natriuretic peptide improves long-term functional recovery after acute CNS injury in mice.

There is emerging evidence to suggest that brain natriuretic peptide (BNP) is elevated after acute brain injury, and that it may play an adaptive role in recovery through augmentation of cerebral blood flow (CBF). Through a series of experiments, we tested the hypothesis that the administration of BNP after different acute mechanisms of central nervous system (CNS) injury could improve functional recovery by improving CBF. C57 wild-type mice were exposed to either pneumatic-induced closed traumatic brain injury (TBI) or collagenase-induced intracerebral hemorrhage (ICH). After injury, either n

The effects of ethanol on spatial and nonspatial memory in adolescent and adult rats studied using an appetitive paradigm.

Adolescent rats have been reported to be less sensitive than adults to many acute ethanol effects, including ethanol-induced sedation and motor impairment, but conversely more sensitive to ethanol-induced disruptions in spatial memory in a Morris water maze (Markwiese et al., 1998). The present study examined adolescent and adult rats trained for 6 days under spatial or nonspatial versions of a presumably less stressful sand box maze. Moderately food-deprived animals were given 0, 0.5, or 1.5 g/kg ethanol intraperitoneally 30 min before training each day, but were tested without ethanol or rei

The effects of apelin-13 in a mouse model of post-traumatic stress disorder.

The objective is to investigate the effects of apelin-13 in models of post-traumatic stress disorder (PTSD). Mature male CD1 mice were subjected to the single prolonged stress method to induce PTSD-related behaviors. These behaviors were then evaluated using the elevated plus maze test, Morris water maze test, and open field test. Hippocampal neural cell death was assessed using propidium iodide labeling. The expression of hippocampal autophagy pathway-associated proteins was determined through immunoblotting analysis, and LC3 levels were also measured via quantitative real-time reverse transc

Schisantherin A recovers Aβ-induced neurodegeneration with cognitive decline in mice.

Schisantherin A (STA) is a main bioactive lignan isolated from Schisandra chinensis (Turcz.) Baill., which has been widely used as a tonic in traditional Chinese medicine for many years. Lots of studies have reported that STA exhibited anti-inflammatory and antioxidant effects. This paper was designed to investigate the effects of STA on cognitive function and neurodegeneration in the mouse control of Alzheimer's disease (AD) induced by Aβ1-42. It was found that successive intracerebroventricular (ICV) administration of STA (0.01 and 0.1mg/kg) for 5days significantly attenuated Aβ1-42-induced

Floralozone improves cognitive impairment in vascular dementia rats via regulation of TRPM2 and NMDAR signaling pathway.

Vascular dementia (VD) is the second largest type of dementia after Alzheimer's disease. At present, the pathogenesis is complex and there is no effective treatment. Floralozone has been shown to reduce atherosclerosis in rats caused by a high-fat diet. However, whether it plays a role in VD remains elusive. In the present study, the protective activities and relevant mechanisms of Floralozone were evaluated in rats with cognitive impairment, which were induced by bilateral occlusion of the common carotid arteries (BCCAO) in rats. Cognitive function, pathological changes and oxidative stress c
