The objective of this study was to develop a short-term experimental infection model for Mycobacterium avium subsp. paratuberculosis (MAP) in cattle, using small oral doses of organisms. Specifically, the effect of dose size was evaluated, as well as specific tissue predilection sites for recovery of MAP. Oral doses as low as 1.5 x 10(6) CFU reliably produced infection that could be detected 3 weeks following infection. Detection of infection required culture of multiple intestinal samples (jejunum and ileum) for MAP. Histological examination did not permit detection at this early stage. Resul

Inflammatory bowel diseases (IBDs) are a major public and veterinary health concern, but the causes are poorly understood. In this review, we discuss the potential of mycobacteria as causative factors for such diseases. We focus on similarities between the most common human IBD, Crohn's disease, and a common IBD in cattle, Johne's disease. Both are multifactorial diseases, leading to a chronic hyperinflammatory immune response of the intestines. However, the underlying genetic and environmental factors, such as variations in the intestinal microbiome, are still poorly understood. While Johne's

MicroRNAs (miRNAs) are small non-coding RNAs that play a crucial role in the post-transcriptional regulation of gene expression. Since miRNAs modulate host immune responses, they represent promising molecular biomarkers of paratuberculosis (PTB), particularly during early or subclinical stages, when conventional diagnostic tests may lack sensitivity. In this context, faecal miRNAs could provide valuable insights into intestinal immune responses and mucosal damage associated with Mycobacterium avium subsp. paratuberculosis (MAP) infection. Although miRNAs have been extensively investigated in s

Paratuberculosis, an infectious and chronic ailment that affects ruminants, causes significant annual economic losses to the livestock industry. Early diagnosis and prompt culling are the primary measures for controlling this disease, highlighting the need for accurate and rapid diagnostic methods. This systematic review and meta-analysis aimed to evaluate the efficacy of quantitative PCR (qPCR) in the diagnosis of paratuberculosis. We searched for and selected articles from databases including PubMed, Web of Science, Elsevier ScienceDirect, Wiley-Blackwell, Springerlink, AGRICOLA, and CAB Abs

Bovine Johne's disease (BJD) is a chronic, debilitating enteritis caused by Mycobacterium avium ssp. paratuberculosis (MAP), with both animal welfare and economic consequences. The detection of MAP in feces is an important measure of transmission from infectious animals through environmental contamination. Therefore, control programs focused on early removal of infectious animals from a herd, combined with management approaches to reduce exposure of susceptible young animals, have a high likelihood of reducing the disease burden. Quantitative PCR (qPCR) tests to detect MAP in feces enable rapi

Paratuberculosis is a major endemic disease caused by Mycobacterium avium subspecies paratuberculosis (MAP) infection and leads to huge economic loss in the dairy sector worldwide. Alternative splicing (AS) events, playing indispensable regulatory roles in many protein functions and biological pathways, are shown to be associated with complex traits and diseases. In this study, by integrating the RNA sequencing (RNA-seq) data of 24 samples from three tissues (peripheral blood, jejunum and salivary gland) of Holstein cows, we obtained 2,706,541,696 uniquely mapped reads in total that represente

Paratuberculosis (ParaTB) in ruminants, commonly known as Johne's disease (JD)is caused by Mycobacterium avium subspecies paratuberculosis (MAP). Characteristic hallmark of JD is chronic inflammation of intestines. Similar infection of MAP in human population is called Crohn's disease (CD). In sub-tropical countries like India, domestic buffaloes are important source of milk and meat and vital for thenutritional security of human beings. Diarrhoea leading to weight loss and weakness invariably leads to reduced productivity, therefore huge financial losses to the livestock farmers. Evaluation o

Fasciola hepatica (liver fluke) is a parasite prevalent in much of the world that causes the economically-important disease of fasciolosis in livestock. The threat that this disease poses extends beyond its direct effects due to the parasite's immunomodulatory effects. Research at this laboratory is focusing on whether this immunoregulation can, in animals infected with liver fluke, exert a bystander effect on concurrent infections in the host. It has already been established that F. hepatica infection reduces cell mediated immune responses to Mycobacterium bovis in cattle, and that the intera

Crohn's disease is a chronic inflammatory bowel disease of unknown cause, affecting approximately 1.4 million North American people. Due to the similarities between Crohn's disease and Johne's disease, a chronic enteritis in ruminant animals caused by Mycobacterium avium paratuberculosis (MAP) infection, MAP has long been considered to be a potential cause of Crohn's disease. MAP is an obligate intracellular pathogen that cannot replicate outside of animal hosts. MAP is widespread in dairy cattle and because of environmental contamination and resistance to pasteurization and chlorination, huma

The diagnosis of infection with Mycobacterium avium subsp. paratuberculosis was studied in 12 cattle from infected dairy herds and four from non-infected herds. A comparison was made of (1) histopathological examination and bacterial culture of tissues, (2) culture of serial samples of rectal contents, and (3) examination of repeated blood samples for interferon-gamma (IFN-gamma) and antibody responses. Tissue samples were taken from the small and large intestine and corresponding mesenteric lymph nodes, and from the pharyngeal tonsil and other lymphoid nodes (retropharyngeal, mediastinal, hep

Paratuberculosis was diagnosed in one 18-month-old and two 30-month-old hinds in a herd of 70 red deer (Cervus elaphus) in Ireland. Loss of condition and intermittent diarrhoea were the main clinical findings. Clumps of acid-fast organisms were found in the faeces of the three deer. Post mortem examination of one deer showed a slight swelling and pallor of the intestinal tract and associated lymph nodes. Histopathology showed a severe, granulomatous enteritis and lymphadenitis, with extensive cellular infiltration, notably with epithelioid macrophages containing numerous acid-fast organisms. M

UNLABELLED: The Mycobacterium avium complex (MAC) is a common causative agent causing nontuberculous mycobacterial (NTM) pulmonary disease worldwide. Whole-genome sequencing was performed on a total of 203 retrospective MAC isolates from respiratory specimens. Phylogenomic analysis identified eight subspecies and species. M. avium subspecies hominissuis (MAH) was the overwhelmingly dominant species (148/203, 72.9%). The other seven identified species were M. intracellulare subsp. yongonense (18/203, 8.9%), M. intracellulare subsp. chimaera (10/203, 4.9%), M. colombiense (11/203, 5.4%), M. para

Integrated control of bovine virus diarrhoea virus, bovine herpesvirus-1, Leptospira interrogans serovar hardjo subtype hardjobovis, Mycobacterium avium subsp. paratuberculosis, and Salmonella dublin in dairy herds may provide economic benefits superior to those obtained by sequential disease control, because, among other things, it allows optimization of voluntary culling. However, in practice there are no adequate instruments to establish priorities in voluntary culling. Therefore, in this study the priorities in decision-making for voluntary culling of infected cattle, as indicated by more

Johne's disease (JD) in cattle is caused by Mycobacterium avium subspecies paratuberculosis (MAP) and is characterized by chronic, progressive enteritis that can lead to substantial weight loss, severe diarrhea, and eventual death. Economic losses due to JD are primarily driven by reduced milk production in subclinical and clinically infected cows, but also include reduced value when sold to slaughter, and costs associated with premature culling. Controlling the transmission of JD within a dairy herd can be achieved through proactive calf management practices and reactive test-based culling. W

Vaccination against paratuberculosis, before or after infection with Mycobacterium avium subsp. paratuberculosis (Map), could affect the progression of paratuberculosis, the development of lesions, the peripheral and local immune response, or the colonization of Map in tissues and its elimination through feces. An experimental study was conducted with thirty-five 1.5-month-old kids, which were separated into 6 experimental groups that include different intervention combinations (vaccinated, non-vaccinated, challenged and non-challenged) at different points and slaughtered at 120 and 330 days p

Systemic immunisation delivered subcutaneously is currently used to control paratuberculosis, a chronic enteritis of ruminants caused by Mycobacterium avium subspecies paratuberculosis (MAP). These vaccines do not provide complete protection and a small cohort of animals still succumb to clinical disease. The aim of this study was to assess mycobacterial infection site-specific variations in immune cells in vaccinated sheep that did or did not develop the disease following controlled exposure to MAP. Immunohistochemical staining of terminal ileum demonstrated that vaccination increased infiltr

Mycobacterium avium ssp. paratuberculosis (MAP) is the causative agent of the paratuberculosis (Para Tb) in ruminants. In addition, this pathogen has been suspected to be implicated in the pathogenesis of Morbus Crohn disease (MC), causing chronic inflammatory intestine changes of humans. The participation of MAP in this illness is discussed intensively and has very contradictory opinions. On the one hand several times succeeded in proving MAP DNA in changed human tissues as well as, in recent time, the bacteria has been isolated from patient's blood. On the other hand there are many publicati

Paratuberculosis (Johne's disease) was suspected in a flock of approximately 1000 sheep after weight losses and scouring had increased in adult animals despite repeated anthelmintic treatment. A total of 10 ewes showing clinical symptoms were submitted to laboratory examination. Gross pathological and histological examination of tissue samples rendered results compatible with Johne's disease in all 10 ewes. The diagnosis could only be confirmed by cultural isolation and identification of Mycobacterium paratuberculosis in the faeces of two ewes. This is considered the first documented isolation

Disseminated Mycobacterium avium complex (DMAC) infection is a major AIDS-defining condition with diagnostic challenges due to nonspecific symptoms. This study, for the first time in Iran, aimed to discriminate MAC organisms in 100 HIV-positive patients directly from clinical specimens and assess their clinical significance, epidemiological characteristics, and associated risk factors through a detailed review of medical and demographic records. Clinical specimens (blood, sputum, and stool) were collected, and routine clinical evaluations were performed. Acid-fast bacilli were detected using Z

Epstein-Barr virus (EBV) is the main environmental agent associated to neuromyelitis optica spectrum disorder (NMOSD). Following to studies reporting an increased prevalence of antibodies against peptides derived from Mycobacterium avium subsp. paratuberculosis (MAP) homologous to EBV and human epitopes (MBP85-98, IRF5424-434) in multiple sclerosis (MS), we investigated whether seroreactivity to these antigens display a NMOSD-specific pattern. The sera of 34 NMOSD patients showed elevated levels of antibodies against MAP and MBP compared to healthy controls (44% vs. 5%, p < 0.0002 and 50% vs.
