Night blindness and the mechanism of constitutive signaling of mutant G90D rhodopsin.

The G90D rhodopsin mutation is known to produce congenital night blindness in humans. This mutation produces a similar condition in mice, because rods of animals heterozygous (D+) or homozygous (D+/+) for this mutation have decreased dark current and sensitivity, reduced Ca(2+), and accelerated values of tau(REC) and tau(D), similar to light-adapted wild-type (WT) rods. Our experiments indicate that G90D pigment activates the cascade, producing an equivalent background light of approximately 130 Rh* rod(-1) for D+ and 890 Rh* rod(-1) for D+/+. The active species of the G90D pigment could be un

Multifocal oscillatory potentials in CSNB1 and CSNB2 type congenital stationary night blindness.

We evaluated the function of the inner retina in patients with congenital stationary night blindness of the complete (CSNB1) and the incomplete type (CSNB2) by recording multifocal oscillatory potentials (mf-OPs). The VERIS system was used to record mf-OPs from 61 areas of the central retina from 5 CSNB1 patients (4 with NYX gene mutation), 6 CSNB2 patients (2 with CACNA1F mutation) and 11 control subjects. For each subject group, the first- and second-order kernel responses for one eye were analysed and the amplitudes and implicit times of their major components compared to 5 concentric rings

Complete congenital stationary night blindness maps on Xp11.4 in a Sardinian family.

X-linked congenital stationary night blindness (CSNBX) is a hereditary non-progressive retinal disorder, which can appear in two different clinical forms, complete and incomplete, associated with CSNB1 and CSNB2 loci on Xp. We describe a Sardinian family with complete CSNBX and define better the limits of the CSNB1 genetic locus on Xp11.4 through linkage analysis. Haplotype analysis showed two key recombinants, which restrict the CSNB1 locus to a region of about 3 cM limited by markers DSX1068 and DSX6810 respectively. The locus that we describe is included in the CSNB1 locus defined by previo

A naturally occurring mouse model of X-linked congenital stationary night blindness.

PURPOSE: To describe a naturally occurring X-linked recessive mutation, no b-wave (nob), that compromises visual transmission between photoreceptors and second-order neurons in mice. METHODS: Affected mice were identified by recording the light-evoked response of the retina, the electroretinogram (ERG). To evaluate visual transmission, cortical potentials were recorded with a scalp electrode. The inheritance pattern for nob was defined by breeding nob animals with normal mice. Retinal histologic analysis was performed by light microscopy. RESULTS: Although the photoreceptor-mediated ERG comp

[Acute reversible night blindness caused by vitamin A deficiency in pancreatic carcinoma].

BACKGROUND: Vitamin A deficiency is a leading cause of blindness in childhood in the developing countries. In developed countries xerophthalmia occurs mainly as secondary hypovitaminosis due to malabsorption in chronic alcoholism or intestinal disorders. PATIENT AND METHODS: A 24-year-old female patient complained of night blindness since one week. There was a history of pancreas tail resection combined with radiation due to a papillary low grade cystic carcinoma at the age of 15. During a review period of 9 years of follow-up there was a slow disseminated metastatic spread including the live

Undetectable S cone electroretinogram b-wave in complete congenital stationary night blindness.

AIMS: The short wavelength sensitive (S) cone electroretinograms (ERGs) were examined in two patients with the complete type of congenital stationary night blindness (CSNB). METHODS: Ganzfeld spectral flashes in the presence of strong white adapting fields were used to elicit the S cone ERGs. RESULTS: The S cone ERG b-wave was not detectable to short wavelength stimuli, while the mixed long (L) and middle (M) wavelength sensitive cone responses appeared normal in waveforms with normal amplitude in both patients. Both patients had normal colour vision on the Farnsworth Panel D-15. CONCLUSION

Protein energy malnutrition, vitamin A deficiency and night blindness in Bangladeshi children.

The occurrence of night blindness and serum vitamin A concentrations among children in rural Bangladesh were studied in relation to protein energy malnutrition, dietary habits and intake of vitamin A capsules. In 1992, 124 night-blind children were registered in a cross-sectional survey in the northern part of Bangladesh, and age-, sex- and neighbourhood-matched controls were selected. Of these, the first reported night-blind child from a household (n = 105) and their controls were included in the analyses. Our results showed that night blindness was associated with protein energy malnutrition

[Suppressive rod-cone interaction in complete and incomplete congenital stationary night blindness].

Dark-adapted rods have a suppressive effect on cone response (suppressive rod-cone interaction: SRCI). This phenomenon has been applied to analyze the pathological mechanism of congenital stationary night blindness (CSNB), but the results have been discrepant among the investigators. Alexander et al reported that 2 individuals with CSNB exhibited a normal SRCI, but Arden and Hogg reported an absence of SRCI in 4 cases of CSNB. We evaluated SRCI in complete and incomplete CSNB. SRCI was tested by measuring luminance threshold for flicker detection (685nm, 1.7 degrees in diameter, 20Hz) across t

The electroretinographic diagnosis of the incomplete form of congenital stationary night blindness.

Fifteen patients with the incomplete form of congenital stationary night blindness (iCSNB) were reviewed to better characterize their electroretinographic (ERG) findings in view of differential diagnosis with other retinal conditions also presenting with negative bright-flash ERG responses. In all 15 patients, in dark-adapted conditions, the bright-flash ERG response had a normal a-wave followed by a subnormal b-wave. Oscillatory potentials (OPs) observed on the ascending limb of the b-wave, although delayed in implicit time, were of large amplitude. The response to a long-wavelength stimulus

The Briard dog: a new animal model of congenital stationary night blindness.

Congenital stationary night blindness (CSNB), apparently inherited in an autosomal recessive manner, was observed in a litter of Briard dogs in Sweden. Of nine litter mates five had nyctalopia. The results of different clinical tests, including electroretinography (ERG), were compared with the results found in four human cases of CSNB, three of which were most probably associated with autosomal recessive inheritance and one with X-linked inheritance. The congenital and stationary nature of the disease, ophthalmoscopically normal appearing fundi, and recordable but reduced photopic flicker resp

Improved mobility and independence of night-blind people using night-vision goggles.

PURPOSE: To investigate whether the use of night-vision goggles (NVGs) by night-blind people improves their mobility and sense of independence under dark circumstances. METHODS: Twenty night-blind subjects with retinitis pigmentosa were requested to walk predetermined routes at night with and without NVGs. The number of unintended contacts with obstacles (hits) and the percentage of preferred walking speed (PPWS) en route were assessed in three different situations: a darkened indoor corridor; a moderately lit outdoor residential area; and a well-lit outdoor shopping area. Assessments were pe

Rod-cone interactions in night-blinding disease.

When rods are dark-adapted they reduce cone flicker sensitivity as though a low-pass filter had been inserted in the visual pathway. This process is exaggerated in certain individuals and accounts for their complaint of night blindness, though ordinary clinical testing shows no abnormality. This rod-cone interaction occurs in the ERG and is absent in disease of the outer retina, which argues that the pathway involved is via the horizontal cell feedback onto the cones. Various anomalies of rod-cone interaction can be seen in retinitis pigmentosa. These suggest that it is the rod dark current wh

Dark continuous noise from mutant G90D-rhodopsin predominantly underlies congenital stationary night blindness.

Congenital stationary night blindness (CSNB) is an inherited retinal disease that causes a profound loss of rod sensitivity without severe retinal degeneration. One well-studied rhodopsin point mutant, G90D-Rho, is thought to cause CSNB because of its constitutive activity in darkness causing rod desensitization. However, the nature of this constitutive activity and its precise molecular source have not been resolved for almost 30 y. In this study, we made a knock-in (KI) mouse line with a very low expression of G90D-Rho (equal in amount to ~0.1% of normal rhodopsin, WT-Rho, in WT rods), with
