Application of low-intensity ultrasound to enhance simultaneous nitrification/iron-based autotrophic denitrification.

Intermittent ultrasound with an intensity of 0.2 W/ml was applied during simultaneous nitrification/iron-based autotrophic denitrification to evaluate its impacts on total nitrogen (TN) removal efficiency and microbial characteristics during low carbon/nitrogen ratio (C/N) wastewater treatment. At an optimal dissolved oxygen (DO) concentration of 1.2 mg/L, the TN removal rate was 91 ± 4%, and the TN concentration in the effluent decreased by 31% owing to the ultrasound treatment. In addition, the number of iron-compounds that formed in the sludge and wastewater increased by 55% and 37%, respec

Bacterial nitrite production oxidizes Fe(II) bioremediating acidic abandoned coal mine drainage.

UNLABELLED: Passive remediation systems (PRSs) treating either acidic or neutral abandoned coal mine drainage (AMD) are colonized by bacteria that can bioremediate iron (Fe) through chemical cycling. Due to the low pH in acidic AMD, iron oxidation from soluble Fe(II) to precipitated Fe(III) is mainly directed by microbial oxidation. Less well described are biotic reactions that lead to iron remediation through abiotic secondary reactions. We describe here iron oxidation in acidic AMD that is mediated by the bacterial reduction of nitrate to nitrite followed by the geochemical oxidation of Fe(I

Ammonia in the crosshairs: microbial targets for metabolic dysfunction-associated steatohepatitis prevention.

Metabolic dysfunction-associated steatohepatitis (MASH) is increasingly linked to disruptions of the gut/liver axis, yet the microbial mechanisms driving disease progression remain incompletely defined. Here, Qu et al. have identified ileal microbial ammonia production by Clostridium perfringens as a mechanistic driver of epithelial barrier dysfunction and hepatic CD8+ T cell remodeling in MASH. In nonhuman primate and mouse models of MASH, the authors demonstrated that the glycine-based tripeptide DT-109 restored gut barrier integrity and attenuated FosB-mediated CCL5 expression in CD8+ T cel

Opportunities for mesoporous nanocrystalline SnO2 electrodes in kinetic and catalytic analyses of redox proteins.

PFV (protein film voltammetry) allows kinetic analysis of redox and coupled-chemical events. However, the voltammograms report on the electron transfer through a flow of electrical current such that simultaneous spectroscopy is required for chemical insights into the species involved. Mesoporous nanocrystalline SnO(2) electrodes provide opportunities for such 'spectroelectrochemical' analyses through their high surface area and optical transparency at visible wavelengths. Here, we illustrate kinetic and mechanistic insights that may be afforded by working with such electrodes through studies o

[Nitric oxide metabolism in the inclusion of N-acetylcysteine into the complex therapy of patients with community-acquired pneumonia].

Thirty-five patients with community-acquired pneumonia were examined. Studies of red blood cells and expired air condensate revealed significant nitric oxide metabolic disturbances in them. In a group of 17 patients, the use of N-acetylcysteine in the complex therapy resulted in the normalization of most parameters that characterized nitric oxide metabolism (nitrates, nitrites, peroxynitrite, NADP-H-diaphorase, and nitrate reductase activity). The positive changes were less significant in the control group receiving mucaltin instead of N-acetylcysteine. The established regularities in the bala

Enantioselective formation and ring-opening of epoxides catalysed by halohydrin dehalogenases.

Halohydrin dehalogenases catalyse the conversion of vicinal halohydrins into their corresponding epoxides, while releasing halide ions. They can be found in several bacteria that use halogenated alcohols or compounds that are degraded via halohydrins as a carbon source for growth. Biochemical and structural studies have shown that halohydrin dehalogenases are evolutionarily and mechanistically related to enzymes of the SDR (short-chain dehydrogenase/reductase) superfamily. In the reverse reaction, which is epoxide-ring opening, different nucleophiles can be accepted, including azide, nitrite a

Uptake of nitrite by Neurospora crassa.

Like the nitrate transport system, the nitrite uptake system in Neurospora crassa is induced by either nitrate or nitrite. This induction is prevented by cycloheximide, puromycin, or 6-methyl purine. The K(m) for nitrite of the induced nitrite uptake system is 86 muM, and the V(max) is 100 mumol of nitrite per g (wet weight) per h. Nitrite uptake is inhibited by metabolic poisons such as arsenate, dinitrophenol, cyanide, and antimycin A. No repression or inhibition of the nitrite transport system by ammonia, nitrate, or Casamino Acids was observed.

Altered Oral Nitrate Reduction and Bacterial Profiles in Hypertensive Women Predict Blood Pressure Lowering Following Acute Dietary Nitrate Supplementation.

BACKGROUND: The efficacy of dietary nitrate supplementation to lower blood pressure (BP) in pregnant women is highly variable. We aimed to investigate whether differences in oral microbiota profiles and oral nitrate-reducing capacity may explain interindividual differences in BP lowering following nitrate supplementation. METHODS: Participants recruited for this study were both pregnant and nonpregnant women, with or without hypertension (n=55). Following an overnight fast, plasma, saliva, and tongue scraping samples were collected for measurement of nitrate/nitrite concentrations, oral NaR (

Proton NMR of Escherichia coli sulfite reductase: studies of the heme protein subunit with added ligands.

The heme protein subunit of sulfite reductase (SiR-HP; M(r) 64,000) from Escherichia coli as isolated contains the isobacteriochlorin siroheme exchange-coupled to a [4Fe-4S] cluster in the 2+ oxidation state. SiR-HP in the presence of a suitable electron donor can catalyze the six-electron reductions of sulfite to sulfide and nitrite to ammonia. Paramagnetic 1H NMR was used to study the low-spin complexes of SiR-HP formed by binding the exogenous inhibitor cyanide or the substrates sulfite and nitrite. As a model, the cyanide complex of purified siroheme was also prepared. The NMR spectrum of

Hemoglobin as a nitrite anhydrase: modeling methemoglobin-mediated N2O3 formation.

Nitrite has recently been recognized as a storage form of NO in blood and as playing a key role in hypoxic vasodilation. The nitrite ion is readily reduced to NO by hemoglobin in red blood cells, which, as it happens, also presents a conundrum. Given NO's enormous affinity for ferrous heme, a key question concerns how it escapes capture by hemoglobin as it diffuses out of the red cells and to the endothelium, where vasodilation takes place. Dinitrogen trioxide (N(2)O(3)) has been proposed as a vehicle that transports NO to the endothelium, where it dissociates to NO and NO(2). Although N(2)O(3

Synergistic effects of biochar and plants can reduce greenhouse gas emissions from salt affected soil.

Applying biochar and using cover crops are two potential approaches to reduce greenhouse gas (GHG) emissions. However, the effectiveness of these methods, individually or in combination, in salt-affected soils remains unclear. Thus, the objective was to determine the impact of barley (Hordeum vulgare) and biochar on N2O and CO2 emissions from salt-affected soil. During the 28-day replicated study, GHG emissions were measured near-continuously and the number of nirK, nirS, qnorB, and nosZ gene copies were measured 12 and 28 days after planting (DAP). Biochar accelerated barley emergence and red

Regulation of NO-Generating System Activity in Cucumber Root Response to Cold.

Nitric oxide (NO) functions as a signaling molecule in plant adaptation to changing environmental conditions. NO levels were found to increase in plants in response to low temperatures (LTs). However, knowledge of the pathways involved in enhanced NO production under cold stress is still limited. For this reason, we aimed to determine the role of different NO sources in NO generation in cucumber roots exposed to 10 °C for short (1 d) and long (6 d) periods. The short-term treatment of seedlings with LT markedly increased plasma membrane-bound nitrate reductase (PM-NR) activity and induced the

Cerium relieves the inhibition of nitrogen metabolism of spinach caused by magnesium deficiency.

Magnesium is one of the essential elements for plant growth and cerium is a beneficial element for plant growth. However, the effects of the fact that cerium improves the nitrogen metabolism of plants under magnesium deficiency is poorly understood. The main aim of the study was to determine the role of cerium in the amelioration of magnesium-deficiency effects in spinach plants. Spinach plants were cultivated in Hoagland's solution. They were subjected to magnesium deficiency and to cerium chloride administered in the magnesium-present media and magnesium-deficient media. Spinach plants grown

A comparative proteomic analysis of Desulfovibrio vulgaris Hildenborough in response to the antimicrobial agent free nitrous acid.

Sulfate reducing bacteria (SRB) can contribute to facilitating serious concrete corrosion through the production of hydrogen sulfide in sewers. Recently, free nitrous acid (FNA) was discovered as a promising antimicrobial agent to inhibit SRB activities thereby limiting hydrogen sulfide production in sewers. However, knowledge of the bacterial response to increasing levels of the antimicrobial agent is unknown. Here we report the proteomic response of Desulfovibrio vulgaris Hildenborough and reveal that the antimicrobial effect of FNA is multi-targeted and dependent on the FNA levels. This was

Metabolism of reactive nitrogen species in pea plants under abiotic stress conditions.

Nitric oxide (*NO) is a key signaling molecule in different physiological processes of animals and plants. However, little is known about the metabolism of endogenous *NO and other reactive nitrogen species (RNS) in plants under abiotic stress conditions. Using pea plants exposed to six different abiotic stress conditions (high light intensity, low and high temperature, continuous light, continuous dark and mechanical wounding), several key components of the metabolism of RNS including the content of *NO, S-nitrosothiols (RSNOs) and nitrite plus nitrate, the enzyme activities of l-arginine-dep

The role of nitric oxide and nitrogen in mediating copper stress in Brassica juncea L.

Copper (Cu) holds a significant importance in plant metabolism as it serves as an essential micronutrient but becomes toxic at higher concentrations. Nitric oxide (NO), a key signaling molecule, and nitrogen (N) play essential roles in combating toxicity of some metals. This study explores the potential of interactive effects of NO as 100 µM SNP (sodium nitroprusside, NO source) and N (80 mg N kg-1 soil) in mitigating Cu (100 mg Cu kg-1 soil) stress in mustard (Brassica juncea L.) plants. The impaired physio-biochemical changes, photosynthetic efficiency, and the expression level of genes asso

Evidence for post-translational regulation of NrtA, the Aspergillus nidulans high-affinity nitrate transporter.

Here, influx and efflux of (13)NO(3)(-), and net fluxes of (14)NO(3)(-) and (14)NO(2)(-), were measured in Aspergillus nidulans mutants niaD171 and niiA5, devoid of nitrate reductase (NR) and nitrite reductase (NiR) activities, respectively. Transcript and protein abundances of NrtA, the A. nidulans principal high-affinity NO(3)(-) transporter, were determined using semiquantitative reverse transcription-polymerase chain reaction and western blots, respectively. (13)NO(3)(-) influx in niaD171 was negligible relative to wild-type values, whereas efflux to influx ratios increased nine-fold. Neve

Expanding the Regulon of the Bradyrhizobium diazoefficiens NnrR Transcription Factor: New Insights Into the Denitrification Pathway.

Denitrification in the soybean endosymbiont Bradyrhizobium diazoefficiens is controlled by a complex regulatory network composed of two hierarchical cascades, FixLJ-FixK2-NnrR and RegSR-NifA. In the former cascade, the CRP/FNR-type transcription factors FixK2 and NnrR exert disparate control on expression of core denitrifying systems encoded by napEDABC, nirK, norCBQD, and nosRZDFYLX genes in response to microoxia and nitrogen oxides, respectively. To identify additional genes controlled by NnrR and involved in the denitrification process in B. diazoefficiens, we compared the transcriptional p

The octahaem MccA is a haem c-copper sulfite reductase.

The six-electron reduction of sulfite to sulfide is the pivot point of the biogeochemical cycle of the element sulfur. The octahaem cytochrome c MccA (also known as SirA) catalyses this reaction for dissimilatory sulfite utilization by various bacteria. It is distinct from known sulfite reductases because it has a substantially higher catalytic activity and a relatively low reactivity towards nitrite. The mechanistic reasons for the increased efficiency of MccA remain to be elucidated. Here we show that anoxically purified MccA exhibited a 2- to 5.5-fold higher specific sulfite reductase activ
