Genetic and neural mechanisms of sleep disorders in children with autism spectrum disorder: a review.

BACKGROUND: The incidence of sleep disorders in children with autism spectrum disorder (ASD) is very high. Sleep disorders can exacerbate the development of ASD and impose a heavy burden on families and society. The pathological mechanism of sleep disorders in autism is complex, but gene mutations and neural abnormalities may be involved. METHODS: In this review, we examined literature addressing the genetic and neural mechanisms of sleep disorders in children with ASD. The databases PubMed and Scopus were searched for eligible studies published between 2013 and 2023. RESULTS: Prolonged awak

[The effects of the antagonists of muscarinic acetylcholine receptor subtypes in rat brain on urinary bladder contraction].

BACKGROUND: Pontine micuturition center and storage center are controlled by upper brain. Pharmacological and molecular biological data indicate the distribution of muscarinic receptor subtypes in rat brain, however, the effect of central muscarinic cholinergic mechanisms on the bladder activity has not been evaluated. We investigated the diversity of effect of each subtype in rat brain. METHODS: The muscarinic agents such as muscarine (non-specific agonist), atropine (non-specific antagonist), pirenzepine (M1 receptor antagonist), AF-DX116 (M2 receptor antagonist) and 4-DAMP (M3 receptor ant

The centrally projecting Edinger-Westphal nucleus--I: Efferents in the rat brain.

The oculomotor accessory nucleus, often referred to as the Edinger-Westphal nucleus [EW], was first identified in the 17th century. Although its most well known function is the control of pupil diameter, some controversy has arisen regarding the exact location of these preganglionic neurons. Currently, the EW is thought to consist of two different parts. The first part [termed the preganglionic EW-EWpg], which controls lens accommodation, choroidal blood flow and pupillary constriction, primarily consists of cholinergic cells that project to the ciliary ganglion. The second part [termed the ce

Prenatal nicotine exposure increases GABA signaling and mucin expression in airway epithelium.

Maternal smoking during pregnancy increases the risk of respiratory disease in offspring, but surprisingly little is known about the underlying mechanisms. Nicotinic acetylcholine receptors (nAChRs) expressed in bronchial epithelial cells (BECs) mediate the effects of nicotine on lung development and function. Recently, BECs were also shown to express a GABAergic paracrine loop that was implicated in mucus overproduction in asthma. We therefore investigated the interactions between cholinergic and GABAergic signaling in rhesus macaque BECs, and found that nicotine upregulated GABA signaling in

Diurnal rhythm in ornithine decarboxylase activity and noradrenergic and cholinergic markers in rat submaxillary lymph nodes.

Diurnal variations in lymph node ornithine decarboxylase activity were examined in submaxillary lymph nodes of rats injected with Freund's complete adjuvant or its vehicle. After immunization, lymph node ornithine decarboxylase activity increased by about 10-fold. Both in immunized and non-immunized rats, a significant diurnal variation in ornithine decarboxylase activity was found, with a maximal activity at early (i.e. 13.00 h, vehicle) or late afternoon (i.e. 17.00 h, Freund's adjuvant). Injection of Freund's adjuvant during daylight or at night resulted in similar day-night differences in

Amitraz exposure induces neurobehavioral toxicity in zebrafish associated with cerebrovascular and BBB-related alterations.

Amitraz (AMT) is a formamidine acaricide widely used in agriculture. The incidence of AMT poisoning shows an increasing trend, yet its neurotoxic effects on non-target organisms remain poorly understood. In this study, zebrafish embryos were used to investigate the developmental and neurobehavioral toxicity of AMT and the underlying mechanisms. Embryos were exposed to varying concentrations of AMT (1, 2.5, and 5 μM) from 4 to 120 h post-fertilization. The results showed that AMT concentration-dependently inhibited embryonic development. Behavioral analyses revealed significant impairments in l

Postbiotics alleviate cognitive impairment and neuroinflammation via gut-brain axis and TLR4/MyD88/NLRP3 pathway.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder characterized by cerebral amyloid-β deposition, neurofibrillary tangles of hyperphosphorylated tau, and chronic neuroinflammation. Growing evidence underscores the role of the gut-brain axis in mediating bidirectional communication between the gut and the central nervous system. Postbiotics, non-viable microorganisms and/or ingredients that are beneficial to the host's health, have emerged as promising modulators of brain function via this axis. Herein, we evaluated the neuroprotective effects of three postbiotics derived fro

[Value of the monkey Papio papio for the study of epilepsy].

The baboon Papio papio is the only animal model showing a natural photosensitive epilepsy very similar to that observed in some human epileptic patients. In the baboon, intermittent light stimulation (ILS) induces bilateral and synchronous myoclonic twitches which are associated with paroxysmal discharges (PDs) predominating in the frontal cortex, and can be followed by generalized tonic-clonic seizures. We were able to demonstrate the motor cortical origin of all these manifestations since neuronal generators responsible for paroxysmal discharges are localized there and are activated by visua

Cognitive dysfunction in Parkinson's disease: Hippocampal vulnerability and redox-driven mechanisms.

Cognitive dysfunction is one of the most disabling non-motor manifestations of Parkinson's disease (PD), progressing from mild cognitive impairment to Parkinson's disease dementia. Although multiple pathological processes have been individually implicated, the mechanisms linking neurotransmitter deficits, proteinopathies, circuit vulnerability, and neurodegeneration remain insufficiently integrated. Here, we synthesize current evidence on the pathophysiology of cognitive impairment in PD, emphasizing the convergence of dopaminergic, cholinergic, noradrenergic and serotonergic dysfunction with

Flavonoids improve neurotransmitters for Parkinson's treatment: mechanism and therapeutic potential.

Parkinson's disease (PD) is recognized as the second most common neurodegenerative disorder worldwide, and it stands out as one of the neurological conditions exhibiting the fastest rise in prevalence, disability, and economic consequences. While the degeneration of dopaminergic neurons and the depletion of striatal dopamine are fundamental to the classic motor symptoms of PD, growing evidence suggests that PD is a multifaceted multisystem disorder marked by extensive impairment across various neurotransmitter systems. Beyond dopaminergic impairment, serotonergic, glutamatergic, γ-aminobutyric

Structural characterization of polysaccharide purified from Amanita caesarea and its pharmacological basis for application in Alzheimer's disease: endoplasmic reticulum stress.

Alzheimer's disease (AD) leads to progressive declines in memory and learning. This disease may arise from endoplasmic reticulum stress due to protein misfolding, which promotes inflammatory pathway activation and induces neuronal cell apoptosis. Polysaccharide is one of the main active components of the mushroom Amanita caesarea (A. caesarea) and has been proven to act as an antioxidant, immune regulatory and anti-inflammatory agent with neurodevelopmental effects. In this study, polysaccharide isolated from A. caesarea (ACPS2) was subjected to analysis to determine the main components, homog

Aclidinium inhibits human lung fibroblast to myofibroblast transition.

BACKGROUND: Fibroblast to myofibroblast transition is believed to contribute to airway remodelling in lung diseases such as asthma and chronic obstructive pulmonary disease. This study examines the role of aclidinium, a new long-acting muscarinic antagonist, on human fibroblast to myofibroblast transition. METHODS: Human bronchial fibroblasts were stimulated with carbachol (10(-8) to 10(-5) M) or transforming growth factor-β1 (TGF-β1; 2 ng/ml) in the presence or absence of aclidinium (10(-9) to 10(-7) M) or different drug modulators for 48 h. Small Intestine Neuromuscular Dysfunctions and Neurogliopathy in a Mouse Model of High-Fat Diet-Induced Obesity: Involvement of Toll-Like Receptor 4.

Obesity is associated with enteric dysfunctions, including gut dysmotility and neurodegeneration, which may involve Toll-like receptor 4 (TLR4) signaling. To investigate this relationship, we examined the impact of TLR4 deficiency on the enteric nervous system (ENS) of the small intestine in a mouse model of high-fat diet (HFD)-induced obesity. Male TLR4-/- and wild-type (WT) C57BL/6J mice were fed either a standard diet (SD; 18% kcal fat) or an HFD (60% kcal fat) for 8 weeks. ENS alterations were evaluated using real-time qPCR and confocal immunofluorescence microscopy on longitudinal muscle-

Potential Mechanisms of Tunneling Nanotube Formation and Their Role in Pathology Spread in Alzheimer's Disease and Other Proteinopathies.

Alzheimer's disease (AD) is the most common type of dementia worldwide. The etiopathogenesis of this disease remains unknown. Currently, several hypotheses attempt to explain its cause, with the most well-studied being the cholinergic, beta-amyloid (Aβ), and Tau hypotheses. Lately, there has been increasing interest in the role of immunological factors and other proteins such as alpha-synuclein (α-syn) and transactive response DNA-binding protein of 43 kDa (TDP-43). Recent studies emphasize the role of tunneling nanotubes (TNTs) in the spread of pathological proteins within the brains of AD pa

Stress augments the rewarding memory of cocaine via the activation of brainstem-reward circuitry.

Effects of stress on the reward system are well established in the literature. Although previous studies have revealed that stress can reinstate extinguished addictive behaviors related to cocaine, the effects of stress on the rewarding memory of cocaine are not fully understood. Here, we provide evidence that stress potentiates the expression of rewarding memory of cocaine via the activation of brainstem-reward circuitry using a cocaine-induced conditioned place preference (CPP) paradigm combined with restraint stress in rats. The rats exposed to 30-minute restraint stress immediately before

In vivo and in vitro studies of glycine- and glutamate-evoked acetylcholinesterase release from spinal motor neurones: implications for amyotrophic lateral sclerosis/motor neurone disease pathogenesis.

To investigate the spinal cellular structures and molecular mechanisms involved in acetylcholinesterase (AChE) release evoked by both glycine (GLY) and glutamate (GLU)--responses that might play a role in chronic neurotoxicity--we analysed AChE histochemistry and histology upon systemic administration of aspartate (ASP), and conducted in vitro experiments in synaptosomes and slices prepared from mouse spinal ventral horns. Upon superfusion and incubation exposure of these preparations to GLY- and GLU-receptor agonists, we assayed both tissue content and release of AChE, butyrylcholinesterase a

Mechanisms of vagus nerve stimulation for the treatment of neurodevelopmental disorders: a focus on microglia and neuroinflammation.

The vagus nerve (VN) is the primary parasympathetic nerve, providing two-way communication between the body and brain through a network of afferent and efferent fibers. Evidence suggests that altered VN signaling is linked to changes in the neuroimmune system, including microglia. Dysfunction of microglia, the resident innate immune cells of the brain, is associated with various neurodevelopmental disorders, including schizophrenia, attention deficit hyperactive disorder (ADHD), autism spectrum disorder (ASD), and epilepsy. While the mechanistic understanding linking the VN, microglia, and neu

α7 nicotinic acetylcholine receptor-mediated neuroprotection against dopaminergic neuron loss in an MPTP mouse model via inhibition of astrocyte activation.

BACKGROUND: Although evidence suggests that the prevalence of Parkinson's disease (PD) is lower in smokers than in non-smokers, the mechanisms of nicotine-induced neuroprotection remain unclear. Stimulation of the α7 nicotinic acetylcholine receptor (α7-nAChR) seems to be a crucial mechanism underlying the anti-inflammatory potential of cholinergic agonists in immune cells, including astrocytes, and inhibition of astrocyte activation has been proposed as a novel strategy for the treatment of neurodegenerative disorders such as PD. The objective of the present study was to determine whether nic
