Enhanced Intramuscular mRNA Activity of Peptide-Lipid Nanoparticles with Hydrophobized Head Groups.

A library of xenopeptide-based ionizable lipids (XP-ILs) was evaluated in mRNA lipid nanoparticles (LNPs) to investigate the impact of modifications within the polar oligoamine headgroup. Modifications included hydrophobic or bulky side chains or a reduced number of protonatable amines. The resulting XP-ILs were characterized by their key physicochemical properties (logD, logP, HLB, CMC), which were correlated with LNP transfection efficiency and cytotoxicity. The inverse relationship between computational logP and experimental logD5.5 is consistent with the HLB, indicating a lower tendency fo

Immunological, anti-inflammatory, and anti-oxidant effects of Fig and Olive leaves extracts nanoparticles against Schistosoma mansoni in C57BL/6 mice.

Schistosomiasis, caused by worms of the genus Schistosoma, especially S. mansoni, remains a major global health challenge, with over 200 million people infected annually. Notably, sterile immunity does not naturally develop, and drug selection pressure has intensified due to the extensive use of praziquantel (PZQ) in endemic areas for nearly four decades, in addition to the emergence of resistance and lack of sterile immunity. This study investigated the effects of nanoparticle-loaded with natural plant extracts-Ficus carica (Fig) and Olea europaea (Olive)-administered in vivo at all S. manson

The role of selected nutraceuticals in management of prediabetes and diabetes: An updated review of the literature. Part II.

We have already published a review about the results of clinical trials evaluating the effects of selected nutraceuticals on glycemia in humans. In this second part, we describe the role of other nutraceuticals involved in dysglycemia. The available evidence showed promising hypoglycemic effects of the nutraceuticals reviewed both for their efficacy and safety profile. However, contradictory results as regard the efficacy of some supplements such as Allium sativum, Juglans regia, and Lycium barbarum on glucose homeostasis have emerged from some clinical studies. Other nutraceuticals including

Delineation of the Olive Pollen Proteome and Its Allergenome Unmasks Cyclophilin as a Relevant Cross-Reactive Allergen.

Olive pollen is a major allergenic source worldwide due to its extensive cultivation. We have combined available genomics data with a comprehensive proteomics approach to get the annotated olive tree (Olea europaea L.) pollen proteome and define its complex allergenome. A total of 1907 proteins were identified by LC-MS/MS using predicted protein sequences from its genome. Most proteins (60%) were predicted to possess catalytic activity and be involved in metabolic processes. In total, 203 proteins belonging to 47 allergen families were found in olive pollen. A peptidyl-prolyl cis-trans isomera

Microwave-assisted synthesis, anti-inflammatory and anti-proliferative activities of new maslinic acid derivatives bearing 1,5- and 1,4-disubstituted triazoles.

In this work, 40 analogs with a natural maslinic acid core (from Olea europaea L.) and various aromatic azides were synthesized. A regiospecific, facile and practical synthesis of 1,5-triazolyl derivatives by Ru(II)-catalyzed azide-alkyne cycloaddition (RuAAC), and mono-, bis- and tri-1,4-triazolyl derivatives by Cu(I)-catalyzed azide-alkyne cycloaddition (CuAAC) was described. All the reactions were assisted by microwave irradiation avoiding toxic reagents and solvents. The new products were obtained from the reaction mixture by simple purification in almost quantitative yields and the reacti

Effects of non-thermal techniques on polyphenols in table olives (cv. Nocellara del Belice).

Processing technologies strongly affect the nutritional and phytochemical quality of table olives. This study investigated the effects of thermal treatment (TT), high-pressure processing (HPP), pulsed electric fields (PEF), and cold plasma (CP) on the polyphenol composition of table olives (Olea europaea L., cv. Nocellara del Belice) using UHPLC-PDA, NMR profiling, and LC-HR-MS/MS. Processing technology influenced polyphenol profiles more strongly than parameter variation. TT, HPP, and PEF promoted the conversion of glycosylated polyphenols into non-glycosylated aglycones, with oleuropein decr

From Olive Tree to Treatment: Nano-Delivery Systems for Enhancing Oleuropein's Health Benefits.

Oleuropein is a natural polyphenolic compound isolated from olive trees (Olea europaea). Besides the strong antioxidant effect of oleuropein, it has many pharmacological activities such as anticancer, antidiabetic, anti-inflammatory, antihypertensive, and many other activities. Thus, oleuropein could be used alone or with other drugs to prevent and treat many diseases. Despite its promising health benefits, oleuropein is highly prone to hydrolysis inside and outside the human body, in addition to a poorly identified pharmacokinetic profile and poor bioavailability. Many nanocarrier delivery sy

Valorization of olive tree leaves: Extraction of oleanolic acid using aqueous solutions of surface-active ionic liquids.

The global olive oil industry annually generates approximately 750,000-1,500,000 tons of Olea europaea leaves as waste that are typically burned for energy production. Yet, this agricultural by-product is a rich source of oleanolic acid, a high value triterpenic acid with outstanding pharmaceutical and nutraceutical activities. The present study focuses on the extraction of oleanolic acid from dried O. europaea leaves using aqueous solutions of surface-active ionic liquids as alternative solvents. A number of imidazolium-based ionic liquids with variable chain length, different anions and opti

From Waste to Value: Solubility and Dissolution Enhancement of Bioactive Extracts from Olive Leaves Using Poloxamers.

The European Union, producing over 2.5 billion tons of waste annually, has prompted the European Parliament to implement legal measures and encourage the shift towards a circular economy. Millions of tons of biowaste from olive plant leaves are generated annually, resulting in environmental and economic challenges. To address this, the biowaste of olive leaves was valorized, resulting in the extraction of valuable components, triterpenes and polyphenols, which hold potential pharmaceutical, food, or cosmetic applications. Our research involved the formulation of a triterpene extract (TTP70, 70

Ultrasound-Assisted Eutectic Solvent-Based Process Intensification for Sustainable Recovery of Oleuropein from Olive Leaves.

Olive leaves, a significant source of agri-food waste, can be valorized as feedstock in biorefineries due to their high content of antioxidant compounds, mainly polyphenols. This study aims to valorize olive leaves through an efficient solid/liquid extraction of oleuropein, its main polyphenol, using green solvents and advanced technologies. Accordingly, three natural eutectic solvents formed with 1,2-butanediol and choline chloride, betaine, or proline, which incorporated water or ethanol as cosolvents, and ultrasound-assisted extraction to enhance the process were used in this work.
