Effects of NGR1 on the Protective Efficacy and Functional Vision Profile of Retinal Photodamage.

Prolonged exposure to high-intensity light can harm macula vision, particularly affecting the function of Müller cells and cone photoreceptors. Panax notoginseng saponins (PNS) have valuable pharmacological effects on cerebrovascular, neurological, and microcirculatory health. Notoginsenoside R1 (NGR1), derived from PNS, shows potential for treating vascular or ischemia-reperfusion-related retinal issues; however, its impact on cone cells and the functional vision profile is not well understood. This study aimed to explore the effect and efficacy of NGR1 on retinal photodamage in vivo in mice.

Retinoic acid signaling regulates spatiotemporal specification of human green and red cones.

Trichromacy is unique to primates among placental mammals, enabled by blue (short/S), green (medium/M), and red (long/L) cones. In humans, great apes, and Old World monkeys, cones make a poorly understood choice between M and L cone subtype fates. To determine mechanisms specifying M and L cones, we developed an approach to visualize expression of the highly similar M- and L-opsin mRNAs. M-opsin was observed before L-opsin expression during early human eye development, suggesting that M cones are generated before L cones. In adult human tissue, the early-developing central retina contained a m

Physiological Postnatal Neurogenesis, Development, and Maturation in the Regenerable Retina.

The retina enlarges postnatally by ∼440% in axolotls and ∼150% in humans, predicting a reduction in neuron densities by 81.5% and 60%, respectively, if neurogenesis is absent. The concurrent retinal remodeling is poorly characterized. We studied retinas of 2-15 cm axolotls. Results showed that hatchling (∼2 cm) retinas, particularly the peripheral (Peri) had 1-2 more soma rows in the ganglion cell (GCL) layer and outer (ONL) and inner nuclear (INL) layers. Cell density reduced in GCL (GCs, -49%) in juveniles (4-12 cm) but increased in INL (74%) and ONL (55.5%) in young juveniles (4-6 cm), demo

Light-dependent activation of G proteins by two isoforms of chicken melanopsins.

In the chicken pineal gland, light stimuli trigger signaling pathways mediated by two different subtypes, Gt and G11. These G proteins may be activated by any of the three major pineal opsins, pinopsin, OPN4-1 and OPN4-2, but biochemical evidence for the coupling has been missing except for functional coupling between pinopsin and Gt. Here we investigated the relative expression levels and the functional difference among the three pineal opsins. In the chicken pineal gland, the pinopsin mRNA level was significantly more abundant than the others, of which the OPN4-2 mRNA level was higher than t

Thyroid hormone receptor beta-2 (TRβ2) overexpression modulates photoreceptor phenotype diversity in a ligand-dependent manner.

BACKGROUND: Vertebrate color vision results from the specification of photoreceptor subtypes expressing distinct opsins. Thyroid hormone (TH) and its receptor TRβ2 are essential regulators of long-wavelength-sensitive (LWS) cone development, but their ligand-dependent roles in regulating cone subtype fate remain unclear. METHODS: We investigated how varying TH availability and TRβ2 overexpression impact cone photoreceptor diversity and opsin expression using a gain-of-function transgenic zebrafish line (crx:trβ2), which expresses TRβ2 in all photoreceptors, and manipulated TH levels through T

Night blindness and the mechanism of constitutive signaling of mutant G90D rhodopsin.

The G90D rhodopsin mutation is known to produce congenital night blindness in humans. This mutation produces a similar condition in mice, because rods of animals heterozygous (D+) or homozygous (D+/+) for this mutation have decreased dark current and sensitivity, reduced Ca(2+), and accelerated values of tau(REC) and tau(D), similar to light-adapted wild-type (WT) rods. Our experiments indicate that G90D pigment activates the cascade, producing an equivalent background light of approximately 130 Rh* rod(-1) for D+ and 890 Rh* rod(-1) for D+/+. The active species of the G90D pigment could be un

Immunocytochemical demonstration of visual pigments in the degenerate retinal and pineal photoreceptors of the blind cave salamander (Proteus anguinus).

Visual pigments in the regressed eye and pineal of the depigmented neotenic urodele, the blind cave salamander (Proteus anguinus anguinus), were studied by immunocytochemistry with anti-opsin antibodies. The study included light- and electron-microscopic investigations of both the eye and the pineal organ. A comparison was made with the black pigmented subspecies Proteus anguinus parkelj (black proteus), which has a normal eye structure. In the retina of the black proteus, we found principal rods, red-sensitive cones and a third photoreceptor type, which might represent a blue- or UV-sensitive

Diversification of non-visual photopigment parapinopsin in spectral sensitivity for diverse pineal functions.

BACKGROUND: Recent genome projects of various animals have uncovered an unexpectedly large number of opsin genes, which encode protein moieties of photoreceptor molecules, in most animals. In visual systems, the biological meanings of this diversification are clear; multiple types of visual opsins with different spectral sensitivities are responsible for color vision. However, the significance of the diversification of non-visual opsins remains uncertain, in spite of the importance of understanding the molecular mechanism and evolution of varied non-visual photoreceptions. RESULTS: Here, we i

Neuropsin-expressing cells in the retina affect melanopsin expression and response to light in mice.

Neuropsin (OPN5) is an opsin involved in several non-image forming functions in the retina, such as the photoentrainment of the retinal circadian clock. These roles were previously attributed to melanopsin (OPN4) which prompted the hypothesis that OPN4 and OPN5 functionally interact. However, the specific cells expressing OPN5 and their interactions with the broader photoreception system remain unclear. This study aims to characterize OPN5-expressing cells and assess how their absence influences the photoresponses of OPN4-expressing cells. Through the use of genetically encoded reporters, conf

Primary primitive neuroectodermal tumors of the retina and ciliary body in dogs.

We describe the clinical, histological, and immunohistochemical features of primary intraocular primitive neuroectodermal tumors in eight dogs. Four of eight tumors exhibited histological features similar to human retinoblastomas characterized by Flexner-Wintersteiner rosettes, and fleurettes, and demonstrated variable immunoreactivity for retinal markers opsin, S-antigen (S-Ag) and interphotoreceptor retinoid-binding protein (IRBP). All dogs with tumors displaying histological and immunohistochemical features of retinal differentiation were ≤2 years of age. All tumors diagnosed as medulloepit

Rhodopsin-induced experimental autoimmune uveoretinitis in monkeys.

We present the first evidence that purified rhodopsin can induce experimental autoimmune uveoretinitis (EAU) in monkeys. Injection of a highly purified lipid-free rhodopsin preparation provokes severe chorioretinitis with concomitant anterior uveitis. The onset of disease is earlier, its frequency is higher, and the inflammation is considerably more severe than in EAU induced under similar conditions by opsin. The first inflammatory cells are observed in the ciliary body and pars plana. Within a few days the inflammation extends into the anterior chamber, choroid, and retina. Retinitis predomi

Sea urchin larvae utilize light for regulating the pyloric opening.

BACKGROUND: Light is essential for various biological activities. In particular, visual information through eyes or eyespots is very important for most of animals, and thus, the functions and developmental mechanisms of visual systems have been well studied to date. In addition, light-dependent non-visual systems expressing photoreceptor Opsins have been used to study the effects of light on diverse animal behaviors. However, it remains unclear how light-dependent systems were acquired and diversified during deuterostome evolution due to an almost complete lack of knowledge on the light-respon

A novel map of the mouse eye for orienting retinal topography in anatomical space.

Functionally distinct retinal ganglion cells have density and size gradients across the mouse retina, and some degenerative eye diseases follow topographic-specific gradients of cell death. Hence, the anatomical orientation of the retina with respect to the orbit and head is important for understanding the functional anatomy of the retina in both health and disease. However, different research groups use different anatomical landmarks to determine retinal orientation (dorsal, ventral, temporal, nasal poles). Variations in the accuracy and reliability in marking these landmarks during dissectio

Identification and functional characterization of a novel rhodopsin mutation associated with autosomal dominant CSNB.

PURPOSE: Mutations in RHO, PDE6B, and GNAT1 can lead to autosomal dominant congenital stationary night blindness (adCSNB). The study was conducted to identify the genetic defect in a large Swiss family affected with adCSNB and to investigate the pathogenic mechanism of the mutation. METHODS: Two affected cousins of a large Swiss family were examined clinically by standard methods: funduscopy, EOG, ERG, and dark adaptometry. Twelve family members were screened for mutations in RHO. The ability of mutant rhodopsin to activate transducin constitutively was monitored by measuring the catalytic ex

Activating mutations of rhodopsin and other G protein-coupled receptors.

Rhodopsin, the visual pigment of rod photoreceptors cells, is a member of the large family of G protein-coupled receptors. Rhodopsin is composed of two parts: a polypeptide chain called opsin and an 11-cis-retinal chromophore covalently bound to the protein by means of a protonated Schiff base linkage to Lys296 located in the seventh transmembrane segment of the protein. Several mutations have been described that constitutively activate the apoprotein opsin. These mutations appear to activate the protein by a common mechanism of action. They disrupt a salt-bridge between Lys296 and the couteri

Closed-Loop Optogenetic Temperature Control System for Inducing Mild Hypothermia in Rats.

Mild hypothermia is widely recognized as a potent neuroprotective intervention. However, conventional physical cooling methods frequently trigger physiological thermoregulatory defenses, eliciting stress responses that limit clinical utility. Neuromodulation based approaches designed to lower body temperature have attracted interest as a means of circumventing these limitations. The preoptic area (POA) of the hypothalamus is a critical site for mammalian thermoregulation, and the medial preoptic area (MPA) sub region contains a high density of temperature sensitive neurons. Prior work has show

Recovery of Behavioral Impairments in F1 Offspring of Zebrafish (Danio rerio) Induced by Parental Imidacloprid Exposure Linked to Attenuation of Disruptions in Visual and Circadian Systems.

The intergenerational effects on behavior following parental imidacloprid (IMI) exposure were detected in zebrafish F1 offspring at different stages. At 4 dpf, F1 larvae exhibited detectable IMI residues, accompanied by significant behavioral impairments, altered expression levels of opsin and circadian rhythm genes, and reduced levels of retinol-related metabolites. At 2 and 4 months, no statistical variations were detected in the behavioral end points and the levels of retinol-related metabolites. The changes in the mRNA/protein levels of opsins and circadian rhythm were reduced in the numbe

Rpe65 is the retinoid isomerase in bovine retinal pigment epithelium.

The first event in light perception is absorption of a photon by an opsin pigment, which induces isomerization of its 11-cis-retinaldehyde chromophore. Restoration of light sensitivity to the bleached opsin requires chemical regeneration of 11-cis-retinaldehyde through an enzymatic pathway called the visual cycle. The isomerase, which converts an all-trans-retinyl ester to 11-cis-retinol, has never been identified. Here, we performed an unbiased cDNA expression screen to identify this isomerase. We discovered that the isomerase is a previously characterized protein called Rpe65. We confirmed o

Optogenetic Peripheral Nerve Immunogenicity.

Optogenetic technologies have been the subject of great excitement within the scientific community for their ability to demystify complex neurophysiological pathways in the central (CNS) and peripheral nervous systems (PNS). The excitement surrounding optogenetics has also extended to the clinic with a trial for ChR2 in the treatment of retinitis pigmentosa currently underway and additional trials anticipated for the near future. In this work, we identify the cause of loss-of-expression in response to transdermal illumination of an optogenetically active peroneal nerve following an anterior co
