Development of TaqMan real-time PCR and droplet digital PCR protocols for the detection of Candidatus Midichloria mitochondrii and evaluation of exposure among wildlife.

Candidatus Midichloria mitochondrii (Ca. M. mitochondrii), an endosymbiont intracellular bacterium living in the mitochondria of several tick species, especially ixodid species, has been proposed as a potential marker for monitoring tick-bite exposure. Therefore, the present study aimed to develop two different diagnostic methods, TaqMan-based real-time PCR (rt-PCR) and Droplet Digital PCR (dd-PCR), targeting the 16 S rRNA gene and gyrB gene for the detection of Ca. M. mitochondrii in different wildlife species from several areas of southern Italy. Both techniques were validated using 10-fold

A longitudinal study on the detection of anti-Leishmania antibodies in a captive European mink (Mustela lutreola) population and their correlation with serum protein electrophoresis.

Leishmaniosis, caused by Leishmania infantum and transmitted by sand flies, is a significant zoonotic disease. Dogs are primary reservoirs, but other domestic animals, such as cats and ferrets, and wild species, including Eurasian otters and European mink, can be hosts. This study focused on European mink, a critically endangered species, investigating the seroprevalence of L.infantum antibodies and its correlation with serum protein profiles over 2021-2024. A total of 68 captive minks in Spain were monitored, yielding 105 serum samples. ELISA tests identified 44 seropositive samples (41.9 %),

Trichinella spp. in wild mesocarnivores in an endemic setting.

Human trichinellosis and Trichinella infection in pigs are both still endemic in the Balkans, including Serbia. Because of the flow between the sylvatic and the domestic cycle of Trichinella spp., monitoring wildlife has been recommended for the risk assessment of Trichinella spp. infection in swine. We have previously shown the presence of Trichinella infection in wild carnivores including the wolf and the golden jackal, and here we report on Trichinella infection in several other mesocarnivore species. From a total of 469 animals collected between 1994 and 2013, Trichinella larvae were detec

Subtype analysis of zoonotic pathogen Cryptosporidium skunk genotype.

Cryptosporidium skunk genotype is a zoonotic pathogen commonly identified in surface water. Thus far, no subtyping tool exists for characterizing its transmission in humans and animals and transport in environment. In this study, a subtyping tool based on the 60kDa glycoprotein (gp60) gene previously developed for Cryptosporidium chipmunk genotype I was used in the characterization of Cryptosporidium skunk genotype in animal and storm runoff samples from a watershed in New York. Altogether, 17 positive samples from this watershed and 5 human and animal specimens from other areas were analyzed.

Xenoendocrine disrupters: laboratory studies on male reproductive effects.

Wildlife from ecosystems contaminated with endocrine disrupting chemicals (EDCs) display a variety of reproductive alterations including cryptorchidism in the Florida panther, small baculum in young male otters, small penises in alligators, sex reversal in fish, and altered social behavior in birds. It has been proposed that EDCs also may have contributed to increases in testicular cancer and hypospadias and the reported decline in human sperm counts. Combined in vivo and in vitro studies are necessary to fully characterize EDC induced alterations of reproduction. We have identified several pe

Concentration and retention of Toxoplasma gondii oocysts by marine snails demonstrate a novel mechanism for transmission of terrestrial zoonotic pathogens in coastal ecosystems.

The parasite Toxoplasma gondii is an environmentally persistent pathogen that can cause fatal disease in humans, terrestrial warm-blooded animals and aquatic mammals. Although an association between T. gondii exposure and prey specialization on marine snails was identified in threatened California sea otters, the ability of kelp-dwelling snails to transmit terrestrially derived pathogens has not been previously investigated. The objective of this study was to measure concentration and retention of T. gondii by marine snails in laboratory aquaria, and to test for natural T. gondii contamination

Absence of Wolbachia endobacteria in the human parasitic nematode Dracunculus medinensis and two related Dracunculus species infecting wildlife.

BACKGROUND: Wolbachia endosymbionts are a proven target for control of human disease caused by filarial nematodes. However, little is known about the occurrence of Wolbachia in taxa closely related to the superfamily Filarioidea. Our study addressed the status of Wolbachia presence in members of the superfamily Dracunculoidea by screening the human parasite Dracunculus medinensis and related species from wildlife for Wolbachia. FINDINGS: D. medinensis, D. lutrae and D. insignis specimens were all negative for Wolbachia colonization by PCR screening for the Wolbachia ftsZ, 16S rRNA and Wolbach

First findings and prevalence of adult heartworms (Dirofilaria immitis) in wild carnivores from Serbia.

Heartworm (Dirofilaria immitis) is a parasitic roundworm that causes a zoonotic disease known as dirofilariosis. Little is known about the role of wild carnivores serving as reservoirs in nature. Therefore, we examined 738 hearts and lungs of free ranging wild carnivores from Serbia to determine the presence of adult heartworms. During the period 2009-2013, the prevalence in golden jackals (Canis aureus) was 7.32%, in red foxes (Vulpes vulpes) 1.55%, in wolves (Canis lupus) 1.43%, and in wild cats (Felis silvestris) 7.69%. No adult heartworm specimens were found in beech martens (Martes foina)

Using Ongoing Wildlife Surveillance Programs to Monitor Antibiotic Resistant Bacteria in the Environment.

The need to include wildlife in surveillance programs for antibiotic resistance has been previously highlighted. In the present study, we explored the possibility of sampling Swedish wildlife in conjunction with ongoing monitoring programs for wildlife health and disease to evaluate the occurrence of antibiotic resistance. The animals included in the study represented different trophic levels and degrees of interaction with humans, namely bears, eagles, wolves, foxes, otters, and hares. Samples were screened for ESBL-, pAmpC-, and carbapenemase-producing Escherichia coli using selective media,

Are the ghosts of nature's past haunting ecology today?

Humans have decimated populations of large-bodied consumers and their functions in most of the world's ecosystems. It is less clear how human activities have affected the diversity of habitats these consumers occupy. Rebounding populations of some predators after conservation provides an opportunity to begin to investigate this question. Recent research shows that following long-term protection, sea otters along the northeast Pacific coast have expanded into estuarine marshes and seagrasses, and alligators on the southeast US coast have expanded into saltwater ecosystems, habitats presently th

Air leak syndrome in animals: definition and pathogenesis.

Air leak syndrome (ALS) is described in human medicine as a constellation of clinical disorders including pneumomediastinum, pneumopericardium, pulmonary interstitial emphysema, pneumothorax, pneumoperitoneum, pneumoretroperitoneum and subcutaneous emphysema. The pathogenesis of ALS depends on the anatomy of the mediastinum and its associations with thoracic, abdominal and cervical connective tissues, as well as a physical phenomenon referred to as the Macklin effect. Various animal species develop diverse combinations of these lesions, although ALS has not been recognized in animals. However,

Chubby, Hairy and Fearless. Subcultural Identities and Predictors of Self-Esteem in a Sample of Polish Members of Bear Community.

Bear subculture exists within a larger gay community, which has been recognized by public health experts as disproportionately burdened with stigma and related health adversities. Bears are distinguished by a particular body look-body hirsuteness and heavy-set physique. Previous research documented the various health risks, and the exposure to both sexual minority and weight stigma, of this population. In this study we focused on the determinants of self-esteem in Bears. We explored the significance of such predictors as: perceived sexual minority and weight stigma, age, resilience, and physiq

Discovery of os cordis in the cardiac skeleton of chimpanzees (Pan troglodytes).

Cardiovascular diseases, especially idiopathic myocardial fibrosis, is one of the most significant causes of morbidity and mortality in captive great apes. This study compared the structure and morphology of 16 hearts from chimpanzees (Pan troglodytes) which were either healthy or affected by myocardial fibrosis using X-ray microtomography. In four hearts, a single, hyperdense structure was detected within the right fibrous trigone of the cardiac skeleton. High resolution scans and histopathology revealed trabecular bones in two cases, hyaline cartilage in another case and a focus of mineralis

First report of treatment and short-term follow-up of a European mink (Mustela lutreola) with leishmaniosis in Spain.

Leishmania infantum causes disease endemic in the Mediterranean Basin that affects humans and various mammals, including mustelids. Clinical leishmaniosis has been documented in domestic ferrets (Mustela putorius furo) and in a captive Eurasian otter (Lutra lutra). The European mink (Mustela lutreola), one of the world's most endangered mammals, is experiencing severe population declines linked to habitat degradation, climate change, the introduction of non-native species, and diseases. This report presents the first documented case of clinical leishmaniosis in a captive European mink, detaili

Comparative Genomics of 42 Arcanobacterium phocae Strains.

For the last 13 years, the fur industry in Europe has suffered from epidemic spouts of a severe necrotizing pyoderma. It affects all species currently farmed for fur and causes animal welfare problems and significant losses to the farmers. The causative agent of this disease was identified as Arcanobacterium phocae. Previously, this bacterium has been isolated from seals and other marine mammals, apparently causing wound and lung infections. Attempts at antibiotic treatment have been unsuccessful and the current advice on preventing the disease is to cull all animals with clinical signs. This

[New data on the Irkutsk focus of opisthorchiasis and on the need for its investigation].

The paper gives data on an opisthorchiasis focus in the Taishetsky district, Irkutsk Region. The species affiliation of Bithynia from the Biryusa River basin was established. These mollusks are morphologically close to Opisthorchophorus hispanica (= Bithynia inflata). No O. felineus trematodes were found in the snails examined. Roach, dace, and bream were the second intermediate hosts. Analysis of long-term data revealed that fish and human infection rates have remained comparatively high for almost 20 years. The peak for Opisthorchis infestation in carps were in 1982 (9.3%), 1983 (2.9%), 1998

Further assessment of the binding properties of proteins A, G, and chimeric protein AG to immunoglobulins of multiple host species of marine mammals.

Detection of infectious diseases in marine mammals is crucial to reduce the risk of transmission of zoonotic pathogens to humans. Although serodiagnostic tests such as enzyme-linked immunosorbent assay (ELISA) can detect antibodies against such pathogens, commercial secondary antibodies specific to marine mammal species are limited. Proteins A and AG have been previously found to react strongly to the immunoglobulins of pinnipeds and cetaceans. However, the binding properties of immunoglobulins of other marine mammals remain unclear to these proteins. Using ELISA, this study further assessed t

Brain volumetry from CT-scan endocasts of three neotropical carnivores.

Studies on brain anatomy can clarify specific evolutionary and behavioural aspects of wild animals. The rich diversity in a broad range of habitats makes carnivorans especially eligible for studying the relations between the brain form and behaviour, cognitive, sensorial and motor traits. This study compared the brain's contour and total and segmented brain volumetry in three species of neotropical carnivorans. CT images of 17 skulls of three species were acquired: Conepatus chinga (n = 6), Galictis cuja (n = 6) and Lontra longicaudis (n = 5). Three-dimensional endocasts allowed for estimating

Zoonotic Mutation of Highly Pathogenic Avian Influenza H5N1 Virus Identified in the Brain of Multiple Wild Carnivore Species.

Wild carnivore species infected with highly pathogenic avian influenza (HPAI) virus subtype H5N1 during the 2021-2022 outbreak in the Netherlands included red fox (Vulpes vulpes), polecat (Mustela putorius), otter (Lutra lutra), and badger (Meles meles). Most of the animals were submitted for testing because they showed neurological signs. In this study, the HPAI H5N1 virus was detected by PCR and/or immunohistochemistry in 11 animals and was primarily present in brain tissue, often associated with a (meningo) encephalitis in the cerebrum. In contrast, the virus was rarely detected in the resp
