The Application of Guanidinium to Improve Biomolecule Quality in Fixed, Paraffin-embedded Tissue.

Neutral buffered formalin (NBF) is the most common fixative in clinical applications. However, NBF damages proteins and nucleic acids, limiting the quality of proteomic and nucleic acid-based assays. Prior studies have demonstrated that BE70, a fixative of buffered 70% ethanol, has many benefits over NBF but the degradation of proteins and nucleic acids in archival paraffin blocks remain a challenge. Thus, we evaluated the addition of guanidinium salts to BE70 with the hypothesis that this may protect RNA and protein. Guanidinium salt supplemented BE70 (BE70G)-fixed tissue is comparable with t

The effect of the formalin-fixed paraffin-embedded process on salivary microbiota profiling.

In recent years, bacterial DNA in formalin-fixed paraffin-embedded (FFPE) samples has been recognized as a valuable bioresource for microbiota studies. This study aimed to examine the effect of the FFPE process on microbiota profiling to evaluate whether FFPE samples could serve as an alternative bioresource to fresh samples in oral microbiota studies. Fresh saliva was collected from nine subjects. The pellets obtained by centrifuging the collected saliva were fixed in formalin, then dehydrated and embedded in paraffin to prepare FFPE samples. The abundance of the hypervariable regions V1-9, V

Enhancement of the Thermal Performance of the Paraffin-Based Microcapsules Intended for Textile Applications.

Phase changing materials (PCMs) microcapsules MPCM32D, consisting of a polymeric melamine-formaldehyde (MF) resin shell surrounding a paraffin core (melting point: 30-32 °C), have been modified by introducing thermally conductive additives on their outer shell surface. As additives, multiwall carbon nanotubes (MWCNTs) and poly (3,4-ethylenedioxyoxythiophene) poly (styrene sulphonate) (PEDOT: PSS) were used in different parts by weight (1 wt.%, 5 wt.%, and 10 wt.%). The main aim of this modification-to enhance the thermal performance of the microencapsulated PCMs intended for textile applicatio

Trypsin digestion on paraffin sections is a useful tool for diagnosis of alport syndrome.

Trypsin digestion for 90 min revealed the alpha(5) chain of type IV collagen along the glomerular basement membrane and Bowman's capsule in paraffin-embedded renal sections of controls. In the 9 patients with the ultrastructures suggestive of Alport syndrome (AS), 8 patients were classified as X-linked dominant type due to the lack or mosaic pattern of alpha(5) chain in paraffin sections of renal biopsies by trypsin digestion, and 1 patient was classified as autosomal recessive type due to the lack of alpha(5) chain in the glomerular basement membrane only. Trypsin digestion is useful for the

Lipid fixation for fat staining in paraffin sections applied to lesions of atherosclerosis.

A new method to fix lipids for staining in paraffin sections was applied here to early lesions of atherosclerosis to test comparability with similar results using frozen-section fat stains. Small blocks of formalin-fixed human coronary artery were exposed to an emulsion of linoleic acid and lecithin in 70% ethylene glycol at 56 degrees C for 3 days. The unsaturated fatty acids partitioned into the tissue lipids for later fixation by chromic acid and could then be processed through paraffin-section for fat staining. Blocks of tissue from the same specimens were also processed for standard froze

Use of patients' sera for immunoperoxidase demonstration of infectious agents in paraffin sections.

Using patients' sera diluted from 1:10 to 1:1,000 as the primary antibodies in indirect immunoperoxidase staining, the authors visualized a variety of infectious agents in formalin-fixed, paraffin-embedded tissue sections. The target lesions included 1) pyoderma caused by Staphylococcus aureus, 2) cryptococcal infection, 3) dermal sporotrichosis, 4) colon ulcer caused by amebic dysentery, 5) cutaneous leishmaniasis, and 6) chronic liver abscess containing ova of Ascaris lumbricoides. The infectious agents were clearly identified in the respective lesions. Paraffin sections of other kinds of in

Identification of Toxoplasma gondii in paraffin-embedded sections by the polymerase chain reaction.

We used the polymerase chain reaction to amplify DNA fragments specific to Toxoplasma gondii. The sensitivity of the technique allowed for the detection of as few as ten cultured T. gondii tachyzoites. We applied the same amplification technique to deparaffinized ocular sections from two cases of ocular toxoplasmosis. Although toxoplasmic cysts could only be seen in one eye by optical microscopy, polymerase chain reaction allowed the identification of the parasite in both cases. Our study indicates the feasibility of a sensitive DNA-based assay to complement pathologic studies of an ocular par

Identification of cryptosporidium in paraffin-embedded tissue sections with the use of a monoclonal antibody.

Recently a monoclonal antibody has been developed against the oocyst form of Cryptosporidium species for use in detecting the organism in fecal material. The authors undertook this study to determine if this antibody could be used in identifying Cryptosporidium species in paraffin-embedded tissue sections. Three biopsies showing Cryptosporidiosis as diagnosed by characteristic appearance by light and electron microscopy were selected from the surgical pathology files at the authors' institution. Paraffin sections were examined with the use of both an indirect immunofluorescence and an avidin-b

The demonstration of B-cell, T-cell and myeloid antigens in paraffin sections.

The monoclonal antibodies MB1 and MT1, which detect B cells and T cells respectively, have been applied to human lymphoid tissues. The distribution of staining within paraffin sections was compared with that observed using frozen sections and was found to be identical. The antibodies were then applied to paraffin sections of 19 B-cell lymphomas and 10 T-cell lesions in which full immunophenotyping had been performed. The B lymphomas all consisted of a large majority of MB1 positive cells with a variable infiltrate of small MT1 positive lymphocytes. The T cell lesions consisted of MT1 positive

Chinese massage (Tuina) combline with paraffin therapy versus tuina or paraffin therapy alone for the treatment of congenital muscular torticollis: A protocol for systematic review and Bayesian network meta-analysis.

BACKGROUND: Current studies in patients with congenital muscular torticollis (CMT) have predominantly focused on the role of tuina or paraffin therapy alone. This systematic review with Bayesian network meta-analysis will be performed to sum up the existing evidence on the effects and safety of tuina plus paraffin therapy for CMT in infants and children. METHODS: The Preferred Reporting Items for Systematic Reviews and Meta-Analyzes reporting guidelines will be followed to conduct this study. MicroRNA differential expression analysis in canine visceral hemangiosarcoma formalin-fixed, paraffin-embedded tissues.

INTRODUCTION: Canine visceral hemangiosarcoma (HSA) is a highly aggressive malignancy of endothelial cells with a poor prognosis and limited diagnostic tools. MicroRNAs (miRNAs) are small non-coding RNAs with emerging utility as diagnostic and prognostic biomarkers due to their stability and disease-specific expression profiles. METHODS: This study aimed to identify miRNA biomarkers for canine splenic and cardiac HSA using small RNA sequencing and quantitative PCR. Formalin-fixed, paraffin-embedded (FFPE) tissues were analyzed from 24 dogs with histologically confirmed HSA (18 splenic, six ca

Detection of Human Prion Seeding Activity in Formalin-Fixed Paraffin-Embedded Archival Tissues.

AIMS: Formalin-fixed paraffin-embedded (FFPE) samples, routinely used in neuropathology, represent an invaluable resource for studying rare diseases like transmissible spongiform encephalopathies (TSE). Despite fixation-induced protein cross-linking, prion seeding activity can be effectively detected using the seeding amplification assays. In this study, we employed the second-generation real-time quaking-induced conversion (RT-QuIC) assay to analyse and quantify human prion seeding activity in FFPE brain tissues. METHODS: FFPE frontal brain tissues were deparaffinised in xylene, followed by

Multispectral Imaging Method for Rapid Identification and Analysis of Paraffin-Embedded Pathological Tissues.

The study of the interaction between light and biological tissue is of great help in the identification of diseases as well as structural alterations in tissues. In the present study, we have developed a tissue diagnostic technique by using multispectral imaging in the visible spectrum combined with principal component analysis (PCA). We used information from the propagation of light through paraffin-embedded tissues to assess differences in the eye tissues of control mouse embryos compared to mouse embryos whose mothers were deprived of folic acid (FA), a crucial vitamin necessary for the gro

Microbiome analysis from formalin-fixed paraffin-embedded tissues: Current challenges and future perspectives.

Formalin-fixed paraffin-embedded (FFPE) tissues stored in thousands of human and animal pathology laboratories around the globe represent mines of stored genetic information. In recent years, the use of FFPE tissues as a viable source of DNA for diverse genetic studies has attracted attention for interrogating microbiomes from this sample type. These studies have proven that 16S rRNA amplicon sequencing-based microbiome studies are possible from FFPE samples but present some particular challenges. In this review, we summarize all aspects of microbiome studies from FFPE tissues including the ch

Optimized High Quality DNA Extraction from Formalin-Fixed Paraffin-Embedded Human Atherosclerotic Lesions.

Formalin-fixed paraffin-embedded (FFPE) tissues represent a valuable source for molecular analyses and clinical genomic studies. These tissues are often poor in cells or difficult to process. Therefore, nucleic acids need to be carefully isolated. In recent years, various methods for DNA isolation have been established for tissues from many diseases, mostly cancer. Unfortunately, genomic DNA extracted from FFPE tissues is highly degraded due to the cross-linking between nucleic acid strands and proteins, as well as random breakings in sequence. Therefore, DNA quality from these samples is mark

Formalin-Fixed and Paraffin-Embedded Samples for Next Generation Sequencing: Problems and Solutions.

Over the years, increasing information has been asked of the pathologist: we have moved from a purely morphological diagnosis to biomolecular and genetic studies, which have made it possible to implement the use of molecular targeted therapies, such as anti-epidermal growth factor receptor (EGFR) molecules in EGFR-mutated lung cancer, for example. Today, next generation sequencing (NGS) has changed the approach to neoplasms, to the extent that, in a short time, it has gained a place of absolute importance and diagnostic, prognostic and therapeutic utility. In this scenario, formaldehyde-fixed
