Androgenic overactivation and epigenetic remodeling drive intergenerational toxicity of bisphenol S in zebrafish.

Bisphenol S (BPS), a widespread plasticizer and endocrine-disrupting compound, can adversely affect steroidogenesis and the hypothalamic-pituitary-gonadal (HPG) axis. This study exposed adult male zebrafish (Danio rerio) to an environmentally relevant BPS concentration (0.5 µg/L) for 14 days (d), assessing its effects on 11-ketotestosterone (11-KT) levels, spermatogenesis, and sperm quality. Additionally, we examined paternal transmission of BPS effects by breeding exposed males with untreated females and evaluating hatching rates, development, survival, and gene expression in offspring. Early second-hand smoke exposure and child and adolescent mental health: evidence from Hong Kong's 'Children of 1997' birth cohort.

BACKGROUND AND AIMS: Second-hand smoke (SHS) exposure is associated negatively with childhood behavioural problems in western settings. In a developed non-western setting, we estimated the associations of early SHS exposure during the prenatal and postnatal periods with several aspects of adolescent mental health. DESIGN: Multivariable linear regression was used to estimate the adjusted associations of pre- and postnatal SHS exposure with adolescent mental health. SETTING: Hong Kong. PARTICIPANTS: Population-representative "Children of 1997" birth cohort. Behavioural problems at ~11 years w

Pregnancy and foetal outcomes following anti-tumor necrosis factor alpha therapy: A prospective multicentre study.

OBJECTIVE: As many inflammatory rheumatic diseases affect patients in childbearing age, some concern has been expressed about the safety of biologic drugs during pregnancy. This study evaluated the effects of anti-tumor necrosis factor alpha (TNFα) agents on pregnancy/foetal outcomes. METHODS: Thirty-eight pregnancies were followed prospectively from November 2008 to February 2015. Information about the patients' exposure to anti-TNFα, disease activity, DMARD therapy, pregnancy/foetal outcomes were registered. RESULTS: Twenty-four/38 (71.1%) pregnancies were exposed to anti-TNFα at conceptio

Investigating the Association Between Infantile Colic and Parental Migraine.

BACKGROUND: To estimate the prevalence of infantile colic and determine its associated factors. Previous studies demonstrate that maternal migraine is significantly associated with increased odds of infantile colic. However, paternal migraine is not significantly associated with infantile colic. METHODS: We conducted a cross-sectional study of infants and parents. We surveyed all parents who brought healthy babies aged ≥3 months to our clinic. We determined the association of infantile colic with the following: child being the first- or nonfirst-born, breastfed or mixed- or formula-fed, sleep

Cohort-based analysis of paternal opioid use in relation to offspring's BMI and plasma lipid profile.

A growing body of evidence suggests that opioid use may affect consumer's offspring by second-hand passive smoke exposure, as well as by transgenerational impacts mediated by genetic and epigenetic alterations of paternal gametes. In human studies, these effects are limited to investigating the neural, behavioral and cognitive characteristics of offspring. Only animal studies have investigated the metabolic parameters influenced by passive opium smoke exposure. Here, we conducted population-based analyses aimed to estimate the association of paternal opioid consumption, started before or after

Parental preconceptual α-cypermethrin exposure alters embryonic brain transcriptomics in mice: Implications for autism spectrum disorder and stress vulnerability.

Pyrethroid insecticides are widely used in agriculture and households, and their exposure can affect neurodevelopment. Few studies have evaluated how preconception parental exposure could also affect this process. To address this knowledge gap, adult C57Bl6/J mice were gavaged daily with α-cypermethrin at a human relevant low (0.3 mg/kg) or high (10 mg/kg) dose in corn oil for four weeks prior to conception. Offspring embryonic day 16 dorsal forebrain was extracted for transcriptomic analysis. In offspring forebrains of exposed compared to unexposed parents, there was increasing number of diff

Parental occupational exposure to metals and risk of cancer in the offspring: A register-based case-control study from Sweden.

BACKGROUND: Cancer risks in the offspring of mothers and fathers exposed to metals are unknown. We estimated the relative risks of childhood cancer, overall and by type, associated with parental occupational exposure to arsenic, cadmium, chromium, nickel, and lead. METHODS: We conducted a nested case-control study (1960-2015) of children born in Sweden aged 0-19 years diagnosed with cancer (National Cancer Register) matched 25:1 to controls on birth year and sex. We obtained parental occupational data around their birth from censuses and a nationwide register and identified exposure to each m

Paternal smoking cessation before pregnancy reduces the risk of spontaneous abortion: a population-based retrospective cohort study.

BACKGROUND: During preconception counselling, pregnant women who smoke are advised to quit smoking. While the adverse effects of paternal smoking on pregnancy and perinatal outcomes have been increasingly recognised, the health benefits of paternal smoking cessation prior to conception remain understudied. METHODS: The current study involved non-smoking reproductive-aged Chinese women who participated two times in the National Free Prepregnancy Checkups Project during 2010-2018. A total of 158 986 pregnancies were included, with husbands reporting smoking at the preconception examination stag

[Transgenerational effects of parental exposure to ionizing radiation].

The deleterious effects of exposure to ionizing radiation on human health have been known for many years. The main long term effect of this exposure is an increase in the risk for developing benign and malignant tumors in the exposed population. This review summarizes the existing data on the possible effects of ionizing radiation on the second generation, focusing on cancer risk. The two mechanisms through which parents' exposure to radiation could affect their children's health are intrauterine exposure of the fetus and preconception exposure of the gonads. Intrauterine exposure to ionizing

Paternal Sperm Gnas-ICR Epigenetic Programming Contributes to PPP-Like Phenotypes in Female Offspring.

The incidence of peripheral precocious puberty (PPP) in females has been rising steadily, emerging as a significant public health concern. However, the paternal developmental origins of PPP remain poorly understood. Using a rat model, we found that paternal preconception caffeine exposure (PPCE) induced PPP-like phenotypes in female offspring and showed paternal-line persistence to the F2 generation. Mechanistic analyses showed that PPCE induced a paternal glucocorticoid-elevated state and was associated with hypermethylation of the Gnas imprinting control region (ICR) in sperm. In offspring o

Positive reappraisal coping mediates the relationship between parental abuse and lack of affection on adulthood generalized anxiety severity.

Exposure to parental abuse and lack of parental affection during childhood are risk factors for adulthood psychopathology. Tendency to engage in positive reappraisal may be a plausible mechanism underlying this relationship. The current study examined if positive reappraisal coping mediated the relationship between maternal/paternal abuse/affection and adulthood generalized anxiety disorder (GAD) symptoms. Participant data (N = 3294) from the Midlife Development in the United States study was collected in three waves, spaced nine years apart. Longitudinal structural equation mediation modeling

Association of Periconception Paternal Body Mass Index With Persistent Changes in DNA Methylation of Offspring in Childhood.

IMPORTANCE: While prenatal nutrition and maternal obesity are recognized as important contributors to epigenetic changes and childhood obesity, the role of paternal obesity in the epigenome of offspring has not been well studied. OBJECTIVES: To test whether periconception paternal body mass index (BMI) is associated with DNA methylation patterns in newborns, to examine associations between maternal and paternal BMI and the epigenome of offspring, and to examine persistence of epigenetic marks at ages 3 and 7 years. DESIGN, SETTING, AND PARTICIPANTS: Project Viva is a prebirth cohort study of

Association of Parental Preconception Exposure to Phthalates and Phthalate Substitutes With Preterm Birth.

IMPORTANCE: Although phthalate exposure during pregnancy has been associated with preterm birth, the association of preconception exposure in either parent with preterm birth constitutes a knowledge gap. OBJECTIVE: To examine the association of paternal and maternal preconception urinary concentrations of biomarkers of phthalates and phthalate substitutes with singleton preterm birth. DESIGN, SETTING, AND PARTICIPANTS: This study, conducted at an academic fertility center in Boston, Massachusetts, included a prospective preconception cohort of subfertile couples comprising 419 mothers and 22

tsRNAs Mediate Intergenerational Inheritance by Concertedly Suppressing Wnt and PPAR Pathways.

OBJECTIVE: The Developmental Origins of Health and Disease hypothesis posits that paternal environmental exposures program offspring metabolic health via sperm epigenetic factors, particularly tRNA-derived small RNAs (tsRNAs). Cadmium (Cd) is a ubiquitous environmental pollutant known to impair reproductive health, but epigenetic links to offspring metabolic disorders are unclear. Using paternal Cd exposure, we elucidate sperm tsRNA pathogenic mechanisms in intergenerational inheritance. METHODS: We validated candidate tsRNA functions via zygotic microinjection of agomirs in mice and transfec

Paternal occupation and anencephaly.

It has been suggested that paternal occupational exposures to pesticides and solvents increase the risk of neural tube defects in offspring. With the use of Texas livebirth, fetal death, and linked livebirth-death records, the authors conducted a population-based case-control study among 1981-1986 Texas births to examine the association between paternal occupation and anencephalic births. Fathers employed in occupations associated with solvent exposure were more likely to have offspring with anencephaly (odds ratio (OR) = 2.53), with painters having the highest risk (OR = 3.43). A lesser assoc
