Reeler Domain-Containing Proteins Involved in the Antibacterial Immunity of Shrimp Litopenaeus vannamei.

Like other invertebrates, Litopenaeus vannamei lacks adaptive immunity and relies mainly on innate immunity for defense against foreign pathogens. In this study, three distinct Reeler domain-containing molecules were discovered in L. vannamei, designated as LvReeler1, LvReeler2, and LvReeler3. Analysis of tissue-specific expression patterns indicated that LvReeler1 showed predominant expression in the stomach, whereas LvReeler2 and LvReeler3 demonstrated peak transcriptional activity within gill tissues. The expression of these molecules was induced by Vibrio parahaemolyticus. In vivo interfer

Evaluation of Antibiotic-Sensitive Bacillus Strain as a Potential Probiotic for Enhanced Growth in Penaeus vannamei.

The safety of probiotics in aquaculture, particularly concerning the transfer of antibiotic resistance genes (ARGs), has become a significant concern. This study aims to isolate antibiotic-sensitive Bacillus strains from the gut of Penaeus vannamei and evaluate the potential for practical application in shrimp farming. The strain was tested for antibiotic sensitivity to 10 commonly used antibiotics, and the minimum inhibitory concentration (MIC) values were determined. Morphological analysis and 16S rRNA sequencing confirmed its identity. The probiotic properties, antibacterial activity, and f

Molecular Characterization, Activation Mechanism, and Antimelanosis Implications of Polyphenol Oxidase in Litopenaeus vannamei.

Polyphenol oxidase (PPO) plays a central role in the quality deterioration of shrimp, particularly in melanosis development. However, the mechanisms triggering melanosis in Litopenaeus vannamei (L. vannamei) remain poorly understood. This study purified natural PPO from L. vannamei and investigated its molecular characteristics and activation mechanism. PPO exhibited phenoloxidase activity in the form of a 210 kDa heterotetramer. Under heat-denaturing conditions, it dissociated into two 62 kDa PPOα chains and two 45 kDa PPOβ chains. During cold storage, the onset of melanosis was accompanied b

Characterization of a novel phage SPX1 and biological control for biofilm of Shewanella in shrimp and food contact surfaces.

Shewanella putrefaciens, commonly found in seafood, forms tenacious biofilms on various surfaces, contributing to spoilage and cross-contamination. Bacteriophages, owing to their potent lytic capabilities, have emerged as novel and safe options for preventing and eliminating contaminants across various foods and food processing environments. In this study, a novel phage SPX1 was isolated, characterized by a high burst size (43.81 ± 3.01 PFU/CFU) and a short latent period (10 min). SPX1 belongs to the Caudoviricetes class, exhibits resistance to chloroform, and sensitivity to ultraviolet. It sh

Shrimp pmo-miR-750 regulates the expression of sarcoplasmic calcium-binding protein facilitating virus infection in Penaeus monodon.

MicroRNAs (miRNAs) regulate gene expression post-transcriptionally and play crucial roles in antiviral responses. Penaeus monodon miR-750 (pmo-miR-750) was found to be strongly up-regulated in the late phase of white spot syndrome virus (WSSV) infection, but its function remains uncharacterized. Herein, the targets that were translationally down-regulated in the shrimp stomach following a pmo-miR-750 mimic injection were identified using two-dimensional gel electrophoresis. Sarcoplasmic calcium-binding protein (Scp) and actin1 (Act1) were revealed to be down-regulated protein spots. The genuin

Impact of pretreatment and atmosphere on quality of lipids extracted from cephalothorax of Pacific white shrimp by ultrasonic assisted process.

Impacts of different pretreatment conditions and atmosphere on yield and oxidative stability of lipids from Pacific white shrimp (Litopenaeus vannamei) cephalothorax using ultrasonic assisted extraction (UAE) process were studied. Cephalothorax was subjected to vacuum-microwave (VM) heating prior to UAE using a mixture of isopropanol/n-hexane (1:1) as solvent. Nitrogen gas was flushed at two flow rates; low (2.15 l/min) and high (4.35 l/min) into the system during ultrasonication. Vacuum-microwave heating resulted in the increase of lipid yield and highest yield was observed in the samples ext

Injected phage-displayed-VP28 vaccine reduces shrimp Litopenaeus vannamei mortality by white spot syndrome virus infection.

White spot syndrome virus (WSSV) is the most important viral pathogen for the global shrimp industry causing mass mortalities with huge economic losses. Recombinant phages are capable of expressing foreign peptides on viral coat surface and act as antigenic peptide carriers bearing a phage-displayed vaccine. In this study, the full-length VP28 protein of WSSV, widely known as potential vaccine against infection in shrimp, was successfully cloned and expressed on M13 filamentous phage. The functionality and efficacy of this vaccine immunogen was demonstrated through immunoassay and in vivo chal

A Nested PCR Assay to Avoid False Positive Detection of the Microsporidian Enterocytozoon hepatopenaei (EHP) in Environmental Samples in Shrimp Farms.

Hepatopancreatic microsporidiosis (HPM) caused by Enterocytozoon hepatopenaei (EHP) is an important disease of cultivated shrimp. Heavy infections may lead to retarded growth and unprofitable harvests. Existing PCR detection methods target the EHP small subunit ribosomal RNA (SSU rRNA) gene (SSU-PCR). However, we discovered that they can give false positive test results due to cross reactivity of the SSU-PCR primers with DNA from closely related microsporidia that infect other aquatic organisms. This is problematic for investigating and monitoring EHP infection pathways. To overcome this probl

Recent insights into host-pathogen interaction in white spot syndrome virus infected penaeid shrimp.

Viral disease outbreaks are a major concern impeding the development of the shrimp aquaculture industry. The viral disease due to white spot syndrome virus (WSSV) observed in early 1990s still continues unabated affecting the shrimp farms and cause huge economic loss to the shrimp aquaculture industry. In the absence of effective therapeutics to control WSSV, it is important to understand viral pathogenesis and shrimp response to WSSV at the molecular level. Identification and molecular characterization of WSSV proteins and receptors may facilitate in designing and development of novel therape

Macroalgal biomonitors of trace metal contamination in acid sulfate soil aquaculture ponds.

Earthen shrimp aquaculture ponds are often impacted by acid sulfate soils (ASS), typically resulting in increased disease and mortality of cultured organisms. Production losses have been attributed to either low pH or to elevated concentrations of toxic metals, both direct products of pyrite oxidation in ASS. The standard farm management practice to minimise effects of pyrite oxidation is to maintain pH of pond waters above 5, based on the assumption that dissolved metal bioavailability is negligible at this pH. This study aimed to test the validity of this assumption, and therefore elucidate

Aflatoxin contamination in shrimp feed and effects of aflatoxin addition to feed on shrimp production.

One hundred fifty samples of shrimp feed were collected from the eastern and southern regions of Thailand, and aflatoxins B1, B2, G1, and G2 (AFB1, AFB2, AFG1, and AFG2) in them were analyzed. AFB1 contamination ranged from a nondetectable level (< 0.003 ppb) to 0.651 ppb. Metabolites of AFB1 were less abundant than AFB1. To study the effects of aflatoxin in feed on shrimp production, black tiger shrimp were divided into four groups of 30 shrimp per group, tested in triplicate, and fed diets containing 0 (control), 5, 10, or 20 ppb of AFB1 for 10 consecutive days. Gonad development characteristics and sex ratio in triploid Chinese shrimp ( Fenneropenaeus chinensis).

This paper details for the first time the gonad development characteristics and sex ratio of triploid shrimp ( Fenneropenaeus chinensis). In triploid shrimp the development of gonad is apparently impaired, especially in females. In the ovary of triploids, germ cells mainly remain at oogonia stage during September through December. From January to February of the next year, partial primary oocytes developed in the ovary lobes. Spermatocytes and spermatids could be observed in the testes of triploids, and a few sperm were observed in the vas deferens and spermatophores. The morphology of sperms

IgE Epitope Mapping of Cross-Reactive Tropomyosin between Shrimp and Edible Cricket (Gryllus bimaculatus).

Tropomyosin is a pan-allergen in crustaceans known to cross-react with homologous proteins from edible insects. However, epitope-level cross-reactivity in edible insects remains poorly defined. This study aimed to identify IgE-binding epitopes of tropomyosin and to evaluate their cross-reactivity between shrimp and the edible two-spotted cricket. Here, sera from shrimp-allergic patients were screened for IgE-binding to proteins from Pacific white shrimp (Penaeus vannamei) and two-spotted cricket (Gryllus bimaculatus). Candidate tropomyosin epitopes from the two-spotted cricket were predicted.

Perfluorooctanoic acid disrupts lysine metabolism and autophagy to promote white spot syndrome virus infection in shrimp.

Perfluorooctanoic acid (PFOA), a globally distributed per- and polyfluoroalkyl substance (PFAS), is increasingly recognized for its immunotoxic and metabolic effects in aquatic organisms; however, its role in viral disease susceptibility remains poorly understood. Using Pacific white shrimp (Penaeus vannamei) and white spot syndrome virus (WSSV) as a model system, we investigated the effects of chronic PFOA exposure on antiviral immunity and viral transmission. Chronic exposure to environmentally relevant concentrations of PFOA resulted in significant accumulation of PFOA in the hepatopancreas

Combined glycosylation processing reduces immunoreactivity of litopenaeus vannamei matrix by modifying allergen epitopes and improves edible quality.

The current study was designed to evaluate the influence of combined glycosylation processing modification on the immune binding activity and edible quality of the litopenaeus vannamei matrix. Three chemical processing methods had been compared, and the glycosylation processing could reduce the IgE/IgG-binding activity of TM to a large extent (25.8% ± 0.1% and 29.9% ± 2.4%, respectively) by destroying the structure and modifying lysine. Subsequently, the optimized combined glycosylation processing of shrimp matrix presented lower IgE-binding activity (75.97% ± 8.74%). As hypothesized, for the
