Toscana, West Nile, Usutu and tick-borne encephalitis viruses: external quality assessment for molecular detection of emerging neurotropic viruses in Europe, 2017.

BackgroundNeurotropic arboviruses are increasingly recognised as causative agents of neurological disease in Europe but underdiagnosis is still suspected. Capability for accurate diagnosis is a prerequisite for adequate clinical and public health response.AimTo improve diagnostic capability in EVD-LabNet laboratories, we organised an external quality assessment (EQA) focusing on molecular detection of Toscana (TOSV), Usutu (USUV), West Nile (WNV) and tick-borne encephalitis viruses (TBEV).MethodsSixty-nine laboratories were invited. The EQA panel included two WNV RNA-positive samples (lineages

Chandipura Virus: A Neglected Sandfly-Borne Neurotropic Virus in Children - Insights From Seven Decades of Evidence.

Chandipura virus, a neurotropic RNA virus transmitted by Phlebotomus sandflies, remains an under-recognised cause of acute paediatric encephalitis in India. The 2024 outbreak, with 245 confirmed cases and 82 deaths, underscores its ongoing public health relevance. This review provides an integrated analysis of CHPV, encompassing its epidemiological distribution in India, host-agent-environment transmission dynamics, molecular and cellular pathogenesis, clinical presentation, diagnostic modalities, therapeutic options, and vaccine research. Evidence from 1950 to 2025, drawn from peer-reviewed s

Chandipura virus: A comprehensive review.

Chandipura virus (CHPV) is an emerging zoonotic pathogen of significant public health concern, particularly in the Indian subcontinent. The virus was first identified in 1965 during a dengue-like febrile outbreak in the village of Chandipur, Maharashtra, India. CHPV is a bullet- shaped enveloped rhabdovirus with a single-stranded, negative-sense RNA genome. CHPV infections are characterised by fatal acute encephalitis, specifically in the paediatric population under the age of 15, with a case fatality rate (CFR) of 57%-70% observed during outbreaks. CHPV transmission is believed to occur via s

Unusual presentation of a severely ill patient having severe fever with thrombocytopenia syndrome: a case report.

BACKGROUND: Severe fever with thrombocytopenia syndrome is an emerging infectious disease caused by a novel phlebovirus belonging to the family Bunyaviridate. Emergence of encephalitis/encephalopathy during severe fever with thrombocytopenia syndrome progression has been identified as a major risk factor associated with a poor prognosis. Here we report the case of a severely ill patient with severe fever with thrombocytopenia syndrome virus-associated encephalitis/encephalopathy characterized by a lesion of the splenium, which resolved later. CASE PRESENTATION: A 56-year-old Japanese man pres

Bovine lactoferrin inhibits Toscana virus infection by binding to heparan sulphate.

Toscana virus is an emerging sandfly-borne bunyavirus in Mediterranean Europe responsible for neurological diseases in humans. It accounts for about 80% of paediatric meningitis cases during the summer. Despite the important impact of Toscana virus infection-associated disease on human health, currently approved vaccines or effective antiviral treatments are not available. In this research, we have analyzed the effect of bovine lactoferrin, a bi-globular iron-binding glycoprotein with potent antimicrobial and immunomodulatory activities, on Toscana virus infection in vitro. Our results showed

Severe fever with thrombocytopenia syndrome, Shandong Province, China, 2011.

Severe fever with thrombocytopenia syndrome (SFTS) is an emerging infectious disease in China. The incidence and clinical and laboratory characteristics of SFTS are not clearly defined. During May 22-October 2, 2011, a total of 24 patients with fever, thrombocytopenia, and leukopenia were clinically diagnosed as having SFTS in Yiyuan County, Shandong Province, China. We conducted laboratory tests for these SFTS patients. SFTS virus (SFTSV) infection was confirmed in 22 patients by using reverse transcription PCR and ELISA by acute-phase and convalescent-phase serum samples. Clinical and labora

Seroprevalence of Sandfly-Borne Phleboviruses Belonging to Three Serocomplexes (Sandfly fever Naples, Sandfly fever Sicilian and Salehabad) in Dogs from Greece and Cyprus Using Neutralization Test.

Phleboviruses transmitted by sandflies are endemic in the Mediterranean area. The last decade has witnessed the description of an accumulating number of novel viruses. Although, the risk of exposure of vertebrates is globally assessed, detailed geographic knowledge is poor even in Greece and Cyprus where sandfly fever has been recognized for a long time and repeatedly. A total of 1,250 dogs from mainland Greece and Greek archipelago on one hand and 422 dogs from Cyprus on the other hand have been sampled and tested for neutralising antibodies against Toscana virus (TOSV), Sandfly fever Sicilia

First detection of Toscana virus in Corsica, France.

Toscana virus (TOSV) was detected for the first time from Phlebotomus perniciosus sandflies in Corsica, a French Mediterranean island. Genetic analysis showed that Corsican TOSV belongs to lineage A, together with Italian, Tunisian, Turkish and other French strains. The demonstration of TOSV in Corsica indicates that autochthonous and tourist populations are at risk of infection. Hence, physicians must consider TOSV as a possible cause of aseptic meningitis and unidentified febrile illness during the warm season.

Infections due to sandfly fever virus serotype Toscana in Spain.

Of the sandfly fever viruses known to be human pathogens (serotypes Toscana [TOS], Sicilian [SFS], and Naples [SFN]), only TOS has demonstrated neurotropic activity. Infections by TOS have been reported in Mediterranean countries, but the virus was previously isolated only in Italy and Portugal. We isolated 15 strains of TOS between 1988 and 1996 from the cerebrospinal fluid of patients with acute aseptic meningitis in Granada, Spain. This finding led us to study the presence of antibodies to TOS, SFS, and SFN in 1,181 adults and 87 children from different regions of Spain. We found that the p

Vector-borne diseases and climate change: a European perspective.

Climate change has already impacted the transmission of a wide range of vector-borne diseases in Europe, and it will continue to do so in the coming decades. Climate change has been implicated in the observed shift of ticks to elevated altitudes and latitudes, notably including the Ixodes ricinus tick species that is a vector for Lyme borreliosis and tick-borne encephalitis. Climate change is also thought to have been a factor in the expansion of other important disease vectors in Europe: Aedes albopictus (the Asian tiger mosquito), which transmits diseases such as Zika, dengue and chikungunya

Neuroinvasive phlebovirus infection in Greece: a case report.

OBJECTIVE: Sandfly fever phleboviruses are endemic in Mediterranean countries. We report a febrile phlebovirus case in a Greek patient who presented signs of neuroinvasive infection. METHODS: In summer 2010, a 20-year-old male was admitted to hospital with fever and lethargy; he was a resident of central Macedonia, northern Greece, where a large outbreak of West Nile virus (WNV) infections occurred at that time. Since there was no laboratory evidence of WNV infection, the patient's serum and cerebrospinal fluid were tested for a probable phlebovirus infection. RESULTS: High titers of IgM and

[Seroprevalence survey of Toscana virus infection in Oporto region].

UNLABELLED: Toscana virus (TOSV) endemic in central Italy, has been documented in several European countries of the Mediterranean region. It is a neurotropic virus and in some of these countries studies to investigate seroprevalence have been done. In a recent etiologic study of meningitis we chose 106 of 308 samples to be tested for TOSV using a nested RT-PCR assay, and found six (5.6%) cases of meningitis by Toscana virus. AIM: To investigate the seroprevalence of antibodies against TOSV in a cohort of a population attended in our Hospital. MATERIAL AND METHODS: In serum samples collected

Seroprevalence of Toscana virus in dogs from Corsica, France.

BACKGROUND: Toscana virus (TOSV) is an arbovirus belonging to the Bunyaviridae, a family of negative-stranded, enveloped RNA viruses. The virus can be transmitted to humans through the bite of an infected female sand fly of the genus Phlebotomus. Infections are usually asymptomatic but the virus is known to cause aseptic meningitis and/or meningo-encephalitis in the Mediterranean countries. Dogs are good sentinels for detection of viral circulation and are more easily accessible than wild animals. FINDINGS: In 2013 and 2014, we collected sera from 231 adult dogs living in 26 counties in two d

Phlebotomine sandflies and the spreading of leishmaniases and other diseases of public health concern.

Phlebotomine sandflies transmit pathogens that affect humans and animals worldwide. We review the roles of phlebotomines in the spreading of leishmaniases, sandfly fever, summer meningitis, vesicular stomatitis, Chandipura virus encephalitis and Carrión's disease. Among over 800 species of sandfly recorded, 98 are proven or suspected vectors of human leishmaniases; these include 42 Phlebotomus species in the Old World and 56 Lutzomyia species in the New World (all: Diptera: Psychodidae). Based on incrimination criteria, we provide an updated list of proven or suspected vector species by endemi

Urinary detection of toscana virus nucleic acids in neuroinvasive infections.

BACKGROUND: Toscana virus (TOSV) is a sandfly-borne pathogen causing febrile diseases and neuroinvasive infections in humans. Definitive diagnosis of TOSV infections frequently requires the detection of viral RNA in cerebrospinal fluid (CSF) or in circulation, which can be achieved prior to seroconversion. OBJECTIVES: To evaluate TOSV excretion in urine and impact of urine as a diagnostic specimen. STUDY DESIGN: A total of 82 plasma, CSF and urine samples were collected from 24 individuals with a preliminary diagnosis of atypical viral encephalitis, where frequent bacterial fungal and viral

Flavivirus RNA in phlebotomine sandflies.

Sandfly-transmitted phleboviruses, such as Toscana, sandfly fever Sicilian, and sandfly fever Naples, can cause human disease and circulate at high rates in Mediterranean countries. Previous studies have also established that viruses other than phleboviruses may be detected in and isolated from sand flies. The recent detection and isolation (in a large variety of mosquito species) of insect-only flaviviruses related to cell fusing agent virus has indicated that the latter is not an evolutionary remnant but the first discovered member of a group of viruses, larger than initially assumed, that h

[Virological study of cases of sandfly fever in Afghanistan].

Three strains (Af-1008, Af-1038, and Af-130) of Neapolitan sandfly fever virus (NSFF) and 2 strains (Af-1028 and Af-83) of Sicilian sandfly fever virus (SSFF) were isolated from febrile patients among soldiers of the limited military contingent of Soviet troops in Afghanistan in May-August, 1986-1987. The viruses were isolated in neonate mice and identified by indirect immunofluorescence, complement-fixation tests and plaque reduction neutralization test in Vero E6 and SPEV cells. The new NSFF strains in serological tests differed slightly from the original Sabin strain and completely differed

Elevated glycosyltransferase activities in infected or traumatized hosts: nonspecific response to inflammation.

Streptococcus pneumoniae infection leads to multifold increases in sialyltransferase, galactosyltransferase, alpha 2-fucosyltransferase, and alpha 3-fucosyltransferase activity of rat liver. Such changes may reflect an increased demand for glycosylation of acute-phase proteins synthesized and secreted by the liver during inflammatory processes. Serum sialyltransferase became elevated in bacteria-infected or burned rats and sandfly fever-infected humans, but did not correlate with acute-phase serum protein changes. These data suggest that nonparenchymal liver cells, such as macrophages, may con
